[Human tumor xenografted into SCID mice and human immune function reconstitution].

OBJECTIVE: In order to observe (1) The behavior of growth and metastasis of PG and PGPTS7 in SCID mice and human immune function reconstituted mice; (2) The ability of interleukin-6 autosecreted from PGTS7 in enhancing the anti-tumor activity of the peripheral blood lymphocytes (PBL). METHODS: Xenografting of PG and PGTS7 into the subepithelial space of the SCID mice, and in some of these animals, human PBL were administrated simultaneously into the peritoneal cavities. The latent period, taken rate, growth speed, volume of the grafted tumor, incidences of metastasis in lungs and lymph nodes and serum level of human immunoglobulin of the immunity reconstituted mice were examined. RESULTS: Although tumor growth had been detected in all the experimental animals, the latent period of grafted PGTS7 was postponed and the volume of tumor mass as well as the incidence of lymph node metastasis all became lower in the immunity reconstituted mice accompanied simultaneously with a higher serum level of human immunoglobulin (HIg). CONCLUSIONS: (1) The SCID mice are good as an appropriated hosts in studying the behavior of growth and metastasis of PG and PGTS7. (2) IL-6 autosecreted from PGTS7 stimulates the proliferation and promotes the activating of PBL. It seems also able to enhance the liberation of human immunoglobulin: to suppress the growth of tumor cells and to reduced the rate of lymph node metastasis.

[p53 alterations in human lung cancer and their correlation with clinicopathological features and prognosis].

OBJECTIVE: To investigate p53 protein accumulations and p53 mutations in squamous cell carcinoma and adenocarcinoma of the lung and their correlation with clinicopathological features and prognosis. METHODS: Immunohistochemistry (LSAB) and non-isotopic PCR-SSCP were used to detect p53 abnormalities in lung cancer. RESULTS: The general positive rates for immunohistochemistry using anti-p53 monoclonal antibody Do-7 (DAKO) as first antibody was 50.5% in 95 cases of lung cancer, 56.5% (26/46) in squamous cell carcinoma and 44.9% (22/49) in adenocarcinoma. PCR-SSCP results showed that 39 cases (41.1%) showed abnormal bands. The mutation rate in squamous cell carcinoma was 47.8% (22/46) and 34.7% (17/49) in adenocarcinoma. In 15 (38.5%) cases, the mutations were located on exon 5, 3 (7.7%) were on exon 6, 13 (33.3%) on exon 7 and 8 (20.5%) on exon 8, respectively. The concordance rate of immunohistochemistry and PCR-SSCP was 77.9% (74/95). The mutation rates of p53 gene in N0, N1, N2 staged lung cancer were 22.6% (7/31), 44.2% (19/43) and 61.9% (13/21), respectively. The difference was statistically significant (P < 0.025). The 3 year survival rate in patients with p53 mutations was significantly lower than those without p53 mutations (P < 0.025). CONCLUSION: The location of p53 mutations in squamous cell carcinoma and adenocarcinoma of the lung were more commonly seen on exon 5 and exon 7. p53 mutations were associated with increased lymph nodes metastasis and reduced survival rate in squamous cell carcinoma and adenocarcinoma of the lung.

[Zymographic analysis of MMPs in human carcinoma cell lines with different metastasis potential].

OBJECTIVE: To investigate the correlation between matrix metalloproteinases (MMPs) activities and metastatic potential of several groups of human carcinoma cells with different metastatic potential. METHODS: Several groups of human carcinoma cell lines (human lung carcinoma, human prostatic carcinoma and melanoma) with different metastatic potential were selected. By using cell culture, collection and concentration of conditioned media and zymographic analysis method the difference of MMPs production and activity among those cell lines were detected. RESULTS: The MMPs production capabilities of carcinoma cells rose following the increase of their invasive and metastatic potentials: that of PG is much higher than PAa's, and BE1 is higher also than CL3 and LH7. Advanced stage melanoma cell WM983a and WM451 product MMP-9, but primary stage cells don't. There are MMP-9 in the conditioned medium of prostatic carcinoma cell PC-3M, the metastatic clone of PC-3 which don't express MMP-9. CONCLUSION: The expression of MMPs especially MMP-9 of carcinoma cells is closely correlated to the metastatic and invasive potential.

[The influence of posture on nasal geometry].

To investigate the effect of posture on nasal cavity geometry, nasal cavity geometry were measured by acoustic rhinometry in 35 healthy adults (17 males and 18 females, 26-50 years old, mean age 32 years) after 5 min in changing postures of head and body. The minimum cross-sectional area and volume between the nostril and 8 cm posteriorly were measured on both sides. When changing from sitting to lateral recumbent with the wider side of the nasal cavity downwards both total volume and total minimum cross-sectional area decreased 18%, volume and minimum cross-sectional area of the wider sides showed that decreased 32% and 26%. When changing from lateral recumbent with the wider side of the nose downwards to lateral recumbent with the narrower side downwards, volume and minimum cross-sectional area of the wider sides and total volume showed that increased 41%, 24% and 12%. Our findings indicate that there is a complex response of the nasal cavity mucosa to posture changing. This factor should be taken into account in rhinological studies of environmental, clinical and pharmacological conditions.

[Prophylactic and therapeutic effects of beclomethasone inhalation on bronchial hyperresponsiveness and asthma: a randomized, double-blind, parallel-group controlled trial].

OBJECTIVE: To better understand the effects of corticosteroid inhalation on asthma and asymptomatic bronchial hyperresponsiveness (BHR). METHODS: A double blind, randomized study was conducted to compare the effects of beclomethasone dry powder (BDP, 600 micrograms/day) inhalation with placebo on BHR and asthmatic symptoms in 59 students (12-18 yrs) for one year. RESULTS: After one year of treatment, lgPD20-FEV1 increased significantly in asthmatics in the BDP group (0.385 +/- 0.424 vs 1.187 +/- 0.603 mumol histamine). Thirty percent of asthmatics in BDP group and 86% in the control group developed symptoms in this year (P = 0.076). There was no significant difference in lgPD20-FEV1 between the BDP and the control group in asymptomatic BHR students. However, none in the BDP group of asymptomatic BHR students developed asthmatic symptoms compared with 3 cases (15%) in control group. The accumulative symptom score showed significant difference between the BDP group and the control group in asymptomatic BHR students (1.50 +/- 2.54 vs 5.58 +/- 6.22, P < 0.01). CONCLUSION: The results suggested that BDP could significantly relieve the severity of BHR and symptoms in asthmatics, and could have some prophylactic effects on the development of asthma in those with asymptomatic BHR.

[Assessment of responsiveness of spontaneous baroreceptor-heart rate reflex under gravitational stress].

Beat by beat arterial blood pressure was measured using Finapres 2300 monitor. Sequences of 3 or more beats with systolic pressure and next beat interval changed in the same direction were identified as baroreflex sequences from 6-min data segments. A linear regression was applied to each sequence and the slope (correlation coefficient > or = 0.85) was taken as a measure of the baroreflex response slope (BRS). The changes in mean BRS during + 65 degrees head-up tilt (HUT), -6.67 kPa lower body negative pressure (LBNP) and 16 days -6 degrees head-down tilt (HDT) bed rest were observed. The results showed that there was a significant decrease in the mean BRS during LBNP or HUT as compared with that of the supine control, though the blood pressure responses differed. The mean BRS was also significantly decreased during bed rest. It suggests that the responsiveness of spontaneous baroreceptor-heart rate reflex can be evaluated in humans under gravitational stress.

[The role of acoustic rhinometry in the evaluation of the volume of nasopharynx before and after adenoidectomy].

OBJECTIVE: To determine the type of acoustic rhinometry curve in different kinds of nasal and nasopharyngeal diseases and the role of acoustic rhinometry in the evaluation of the changes of the volume of nasopharynx before and after adenoidectomy. METHODS: One hundred and fifty-one patients with nasal and nasopharyngeal diseases, including rhinitis, nasal polyps, obstructive sleep apnea syndrome, palatoschisis, atrophic rhinitis, adenoid vegetation, perforation of nasal septum, carcinoma of nasopharynx, and stricture of nasal limen, were measured with acoustic rhinometry. Meanwhile, 15 patients with adenoid vegetation were measured before and after adenoidectomy, the changes in volume recorded by acoustic rhinometry were compared with the adenoid volume obtained by the method of displacement. RESULTS: Acoustic rhinometry curve can be divided into normal and abnormal curves and the abnormal curves can be divided into four types according to the site of changes. Acoustic rhinometry curve altered with the changes in character, degree and site of nasal and nasopharyngeal diseases and could return to normal after surgery or medical treatment. The volume of nasopharynx of adenoid vegetation patients was significantly smaller than that of the normal control and it rose significantly and became closed to normal after adenoidectomy. The volume of adenoid and the calculated changes in volume of the nasopharynx was found to be interrelated and highly significant(r = 0.87, P < 0.001). CONCLUSION: The results indicate that acoustic rhinometry curve is a useful method in the assistant diagnosis and judgment of therapeutic effectiveness of many kinds of nasal and nasopharyngeal diseases and can provide reference for the evaluation of the volume of adenoids.
