Production of stem cells with embryonic characteristics from human umbilical cord blood.

When will embryonic stem cells reach the clinic? The answer is simple -- not soon! To produce large quantities of homogeneous tissue for transplantation, without feeder layers, and with the appropriate recipient's immunological phenotype, is a significant scientific hindrance, although adult stem (ADS) cells provide an alternative, more ethically acceptable, source. The annual global 100 million human birth rate proposes umbilical cord blood (UCB) as the largest untouched stem cell source, with advantages of naive immune status and relatively unshortened telomere length. Here, we report the world's first reproducible production of cells expressing embryonic stem cell markers, - cord-blood-derived embryonic-like stem cells (CBEs). UCB, after elective birth by Caesarean section, has been separated by sequential immunomagnetic removal of nucleate granulocytes, erythrocytes and haemopoietic myeloid/lymphoid progenitors. After 7 days of high density culture in microflasks, (10(5) cells/ml, IMDM, FCS 10%, thrombopoietin 10 ng/ml, flt3-ligand 50 ng/ml, c-kit ligand 20 ng/ml). CBE colonies formed adherent to the substrata; these were maintained for 6 weeks, then were subcultured and continued for a minimum 13 weeks. CBEs were positive for TRA-1-60, TRA-1-81, SSEA-4, SSEA-3 and Oct-4, but not SSEA-1, indicative of restriction in the human stem cell compartment. The CBEs were also microgravity--bioreactor cultured with hepatocyte growth medium (IMDM, FCS 10%, HGF 20 ng/ml, bFGF 10 ng/ml, EGF 10 ng/ml, c-kit ligand 10 ng/ml). After 4 weeks the cells were found to express characteristic hepatic markers, cytokeratin-18, alpha-foetoprotein and albumin. Thus, such CBEs are a viable human alternative from embryonic stem cells for stem cell research, without ethical constraint and with potential for clinical applications.

Tension and stress in the rat and rabbit stomach are location- and direction-dependent.

Distension studies in the stomach are very common. It is assumed in pressure-volume (barostat) studies of tone and tension in the gastric fundus that the fundus is a sphere, i.e. that the tension in all directions is identical. However, the complex geometry of the stomach indicates a more complex mechanical behaviour. The aim of this study was to determine uniaxial stress-strain properties of gastric strips obtained from rats (n=12) and rabbits (n=10). Furthermore, we aimed to study the gastric zero-stress state since the stomach is one of the remaining parts of the gastrointestinal tract where residual strain studies have not been conducted. Longitudinal strips (in parallel with the lesser curvature) and circumferential strips (perpendicular to the lesser curvature) were cut from the gastric fundus (glandular part) and forestomach (non-glandular part). The residual stress was evaluated as bending angles (unit: degree per unit length and negative when bending outwards). The residual strain was computed from the change in length between the zero-stress state and no-load state. The stress-strain test was performed using a tensile test machine. The thickness and width of each strip were measured from digital images. The strips data were compared with data obtained in the intact stomach in vitro. Most residual stresses and strains were bigger in the glandular part than in the forestomach, and in general the rat stomach had higher values than the rabbit stomach. The glandular strips were stiffer than the forestomach strips and the longitudinal glandular strips were stiffer than the circumferential glandular strips (P<0.05). The gastric strips were stiffer in rats than in rabbits (P<0.01). The data obtained in the intact rat stomach confirmed the strips data and indicated that those were obtained in the physiological range. In conclusion, the biomechanical properties of the gastric strips from the rat and rabbit are location-dependent, direction-dependent and species-dependent. The assumption in physiological pressure-volume studies that the stomach is a sphere with uniform tension is not valid. Three-dimensional geometric data obtained using imaging technology and mechanical data are needed for evaluation of the stomach function.

Biosynthesis and purification of a hydrophobic peptide from transmembrane domains of G-protein-coupled CB2 receptor.

A major challenge for the structural study of the seven-transmembrane G-protein-coupled receptors is to obtain a sufficient amount of purified protein at the milligram level, which is required for either nuclear magnetic resonance (NMR) spectroscopy or X-ray crystallography. In order to develop a high-yield and cost-effective method, and also to obtain preliminary structural information for the computer modeling of the three-dimensional receptor structural model, a highly hydrophobic peptide from human cannabinoid subtype 2 receptor CB2(65-101), was chosen to develop high-yield membrane protein expression and purification methods. The peptide included the second transmembrane helix with the associated loop regions of the CB2 receptor. It was over-expressed in Escherichia coli, with a modified TrpDelta LE1413 (TrpLE) leading fusion sequence and a nine-histidine tag, and was then separated and purified from the tag in a preparative scale. An experimental protocol for the chemical cleavage of membrane protein fragment was developed using cyanogen bromide to remove the TrpLE tag from the hydrophobic fusion protein. In addition, protein uniformly labeled with isotopic 15N was obtained by expression in 15N-enriched minimum media. The developed and optimized preparation scheme of expression, cleavage, and purification provided a sufficient amount of peptide for NMR structure analysis and other biophysical studies that will be reported elsewhere. The process of fusion protein cleavage following purification was monitored by high-performance liquid chromatography (HPLC) and mass spectrometry (MS), and the final sample was validated by MS and circular dichroism experiments.

Radiologically placed temporary hepatic artery catheter for treatment of liver cancer.

Summary Hepatic artery infusion (HAI) chemotherapy is associated with higher response rates compared to systemic chemotherapy in those patients with unresectable liver malignancies. Operative hepatic artery catheter (HAC) insertion has significant morbidity and mortality, especially in patients with high-volume disease, some of whom may not respond to HAI chemotherapy. We report our experience in 45 patients with high-volume liver disease who were initially treated with HAI chemotherapy via a radiologically placed temporary HAC to try to select the responders who then went on to have an operative HAC. In these 45 patients who had 62 radiologically placed HAC, we found very few major complications, and certainly no complications such as cholecystitis, vascular or malperfusion problems.

Estrogen, but not testosterone, down-regulates cytokine production in nicotine-induced murine macrophage.

We have investigated the effects of 17beta-estradiol (E2) and testosterone (Te) on the production of proinflammatory factors by nicotine-induced murine macrophage (Ana-1). Ana-1 pretreated with or without E2/Te was challenged with different doses of nicotine at various time points. TNF-alpha, IL-1beta IFN-gamma, and IL-8 in the supernatant of Ana-1 were detected using radioimmunoassay (RIA) or enzyme-linked immunosorbent assay (ELISA). The presence of estrogen and androgen receptors in Ana-1 was detected by reverse transcription polymerase chain reaction (RT-PCR) and Western blotting. E2 could obviously suppress the expression of TNF-alpha and IL-1beta whereas Te could not. There were no obvious changes in the levels of either IL-8 or IFN-gamma in Ana-1 stimulated with nicotine or sex hormones (p>0.05). The antagonist tamoxifen of E2 abolished the effects of E2 on TNF-alpha and IL-1beta whereas the antagonist cyproterone of Te did not alter the effects of Te on Ana-1. Furthermore, RT-PCR and Western blotting showed that both the estrogen receptors (ER-alpha and ER-beta existed in Ana-1; however, androgen receptor (AR) was not detectable. Our data suggested that E2 could regulate the immunoreaction in the nicotine-induced inflammatory process possibly through classic intercellular ER, whereas Te had no significant effect in this process.

Measurement of the exclusive 3He(e,e'p) reaction below the quasielastic peak.

New, high-precision measurements of the 3He(e,e(')p) reaction using the A1 Collaboration spectrometers at the Mainz microtron MAMI are presented. These were performed in antiparallel kinematics at energy transfers below the quasielastic peak, and at a central momentum transfer of 685 MeV/c. Cross sections and distorted momentum distributions were extracted and compared to theoretical predictions and existing data. The longitudinal and transverse behavior of the cross section was also studied. Sizable differences in the cross-section behavior from theoretical predictions based on the plane wave impulse approximation were observed in both the two- and three-body breakup channels. Full Faddeev-type calculations account for some of the observed excess cross-section, but significant differences remain.

Taurine protected myocardial mitochondria injury induced by hyperhomocysteinemia in rats.

Taurine can protect against cardiovascular diseases, whereas elevated levels of plasma homocysteine are associated with atherosclerotic and thromboembolic cardiovascular diseases. To illustrate the effects of taurine on hyperhomocysteinemia, we observed the myocardial mitochondria dysfunction in the rats with hyperhomocysteinemia induced by diet methionine loading, and the therapeutic effect of taurine. A methionine diet increased plasma homocysteine concentration (133.51 +/- 27.91 micromol/L vs 12.31 +/- 2.58 micromol/L in control, P < 0.01), stimulated the production of reactive oxygen species (ROS) in the myocardial mitochondria, and inhibited the activities of mitochondrial Mn-superoxide dismutase and catalase. The 45Ca uptake and Ca2+-ATPase activity in the myocardial mitochondria were significantly lowered in rats with hyperhomocysteinemia. Taurine supplements effectively attenuated the hyperhomocysteinemia-induced ROS production and inhibition of Mn-superoxide dismutase and catalase activities in the myocardial mitochondria, and increased its 45Ca uptake and Ca2+-ATPase activity. Thus, taurine antagonizes the oxidative stress injury in the myocardial mitochondria induced by the hyperhomocysteinemia.

New view of the high-pressure behaviour of GdFeO(3)-type perovskites.

Recent determinations of the structures of several GdFeO(3)-type orthorhombic perovskites (ABO(3)) show that the octahedra in some become more tilted with increasing pressure. In others the octahedra become less tilted and the structure evolves towards a higher-symmetry configuration. This variety of behaviour can be explained in terms of the relative compressibilities of the octahedral and dodecahedral cation sites in the perovskite structure. If the BO(6) octahedra are less compressible than the AO(12) sites then the perovskite will become more distorted with pressure, but the perovskite will become less distorted if the BO(6) site is more compressible than the AO(12) site. In this contribution we use the bond-valence concept to develop a model that predicts the relative compressibilities of the cation sites in oxide perovskites. We introduce the site parameter M(i) defined in terms of the coordination number N(i), average bond length at room pressure R(i), and the bond-valence parameters R(0) and B,M(i) = ([R(i)N(i)/B)exp](R(0) - R(i))/B].M(i) represents the variation in the bond-valence sum at the central cation in a polyhedral site because of the change of the average bond distance. Experimental data suggest that the pressure-induced changes in the bond-valence sums at the two cation sites within any given perovskite are equal. With this condition we show that the ratio of cation-site compressibilities is given by betaB/beta(A) = M(A)/M(B). This model, based only upon room-pressure bond lengths and bond-valence parameters, correctly predicts the structural behaviour and some physical properties of the oxide perovskites that have been measured at high pressure.

Part II. Chromatography using ultra-stable metal oxide-based stationary phases for HPLC.

In this part of the review authors discuss methods used for modification of metal oxide surfaces. On the basis of literature data it is shown, that silanization of the surfaces do not form stable supports for chromatography. On the other hand, the success of polymer modified surfaces such as polybutadiene (PBD) and polystyrene (PS) is emphasized. Permanent modification of metal oxide surfaces with Lewis bases is also widely discussed. Chromatographic properties of polymer modified surfaces of zirconia are discussed in details. The perspectives of carbon-coated metal oxide surfaces in HPLC and high temperature separations are described.

Combined family trio and case-control analysis of the COMT Val158Met polymorphism in European patients with anorexia nervosa.

The high activity Val158 (H) allele of the dopamine-metabolizing enzyme catechol-O-methyltransferase (COMT) was associated with anorexia nervosa (AN) in a recent family trio-based study of patients from Israel. In an attempt to replicate this finding, we performed a combined family trio and case-control study in an European population from seven centers in six different countries (Austria, Germany, Great Britain, Italy [Milan], Italy [Florence], Slovenia, and Spain), together contributing a total of 372 family trios, 684 controls and 266 cases. TDT analyses of high (H) and low (L) alleles in family trios showed that H allele and L allele were each transmitted 101 times (chi(2) = 0, ns). Allele-wise case-control analysis using separate samples simply combined from the centers was also not significant, with the frequencies of the H allele 50% in cases and same in controls. Stratified analysis of data from all centers gave an odds ratio of 0.98 (Cornfield 95% confidence limits 0.78-1.24). Analysis by genotype was likewise not significant (overall chi(2) = 0.42). Because we were not able to support the primary hypothesis that Val158Met is a risk factor for AN, we did not perform secondary analysis of minimum body mass index (mBMI), age at onset or illness subtype (restricting or binge purging anorexia). Overall we found no support for the hypothesis that the Val158 allele of COMT gene is associated with AN in our combined European sample.

Percutaneous radiofrequency ablation of pulmonary metastases in patients with colorectal cancer.

INTRODUCTION: This study aimed to assess the safety and efficacy of imaging-guided percutaneous radiofrequency ablation (RFA) for local control of lung metastases from colorectal cancer (CRC). METHODS: Twenty patients with lung metastases from CRC were treated with a RITA Starburst XL electrode and RITA 1500 generator using temperature control and impedance monitoring. Patients received intravenous sedation and analgesia, or local anaesthetic, and stayed in hospital for at least 24 h after treatment. RFA was assessed with computed tomography (CT) at 1 week and 1 month, and then every 3 months. RESULTS: Forty-four CRC lung metastases in 19 patients were treated successfully at 25 treatment sessions. Five of 19 patients were retreated for new lesions. There were 13 pneumothoraces following the 25 treatments, and six patients required drainage. The median length of follow-up was 730 (range 148-924) days. Six months after treatment CT demonstrated that three lesions had progressed, 25 metastases were stable or smaller, and 11 were no longer visible. At 12 months five metastases had progressed, 11 were smaller or stable, and nine were not visible. CONCLUSION: Percutaneous imaging-guided RFA was associated with modest morbidity. RFA can achieve local control of CRC lung metastases: nine of 25 metastases were not visible on CT at 12 months after treatment.

A novel microscope system for time-lapse observation of corneal cells in a living mouse.

A novel microscope system is presented for observation of corneal cells in a living mouse. It enables tracking of individual cells in all layers of the cornea at various times, thus allowing the generation of time-lapse recordings. The system consists of three major components: an upright fluorescence microscope for visualization of corneal cells, a mouse-holding unit for immobilization of the animal and the eye, and a set of gimbals which permit observation of a wide area of corneal surface without refocusing. The same cells could be observed at different limes with the help of fiducial marks in the cornea, allowing their changes in position to be determined under natural and experimental conditions. This technique should prove useful in investigation of the cell movement in normal and diseased corneas, including the study of wound healing after an injury or surgery.

Association between apolipoprotein CI HpaI polymorphism and sporadic Alzheimer's disease in Chinese.

OBJECTIVE: To investigate into the relationship of apolipoprotein CI (ApoCI) polymorphism with sporadic Alzheimer's disease (AD) in Chinese. SUBJECTS AND METHODS: A total of 257 AD patients and 242 age-matched elderly individuals were genotyped for the ApoCI HpaI and apolipoprotein E (ApoE) HhaI polymorphisms. RESULTS: The ApoCI A allele was associated with AD of moderate to severe dementia when patients were divided into two subgroups according to Clinical Dementia Rating scale, and the AA genotype was strongly associated with moderate to severe AD in ApoE epsilon4 allele carriers [odds ratio (OR) = 8.19, 95% confidential interval: 1.28-52.30, after adjusting for age and gender by logistic regression analysis], although in total no significant differences of allele or genotype frequency between patients and controls were found. CONCLUSION: The present study partially confirmed the previous findings, suggesting that the ApoCI A allele might contribute to the susceptibility to moderate to severe sporadic AD in Chinese.

Protothecosis successfully treated with amikacin combined with tetracyclines.

Summary We report a case of protothecosis in an 18-year-old female student caused by Prototheca zopfii successfully treated with amikacin combined with tetracyclines. Zusammenfassung Es wird über eine Protothecose, verursacht durch Prototheca zopfii, bei einer 18-jährigen Studentin berichtet, die erfolgreich mit Amikacin in Kombination mit Tetracyclinen behandelt wurde.

A comparative study of quantitative structure-activity relationship methods based on gallic acid derivatives.

By using hologram quantitative structure-activity relationship (HQSAR) and comparative molecular field analysis (CoMFA) methods, the relationships between the structures of 49 gallic acid derivatives and their analgesic activity have been investigated to yield statistically reliable models with considerable predictive power. The best HQSAR model was generated using atoms, bond and connectivity as fragment distinction parameters and fragment size 5-7 from a hologram length of 307 with 3 components. High conventional r2 (r2 = 0.825) and cross-validation r2 (r2(cv) = 0.726) values were obtained. CoMFA analyses varying lattice size and location, grid spacing, probe charges and using, Tripos standard and Indicator force field were performed. The best model was developed with 4 components using sp3-hybridized carbon atom with +1.0 charge as probe, grid spacing (2 A), lattice offset (1.0, 3.0, -2.5). The CoMFA model showed a conventional correlation coefficient r2 of 0.889 and across-validation r2(cv) equals to 0.633. The robustness and predictive ability of the HQSAR and CoMFA models have been validated by means of an external test set. The results indicate that both models possess high statistical quality in the prediction of analgesic potency of novel gallic acid analogs.

Preferential HER-2/neu overexpression and/or amplification in aggressive histological subtypes of invasive breast cancer.

AIMS: To investigate whether alterations of the HER2 gene occur more frequently in histologically unfavourable subtypes of invasive breast cancer. METHODS: The study was composed of nine invasive apocrine, six lipid-rich, 12 glycogen-rich, 11 micropapillary and 33 pleomorphic lobular breast carcinomas. Lymph node involvement was represented in all subgroups. HER2 status was confirmed in all cases by using immunohistochemistry (CB11, Herceptest) and fluorescent in-situ hybridization (FISH) analysis (Vysis). RESULTS: Micropapillary and apocrine carcinomas showed the highest rate of protein overexpression (72% and 66%) and gene amplification (45% and 44%). Protein overexpression was common in poorly differentiated pleomorphic lobular carcinomas (56%); however, this subgroup failed to show an increased number of gene copies by FISH (31%). The incidence of HER2 overexpression (33% and 50%, respectively) and gene amplification (25% and 33%, respectively) among glycogen-rich and lipid-rich carcinomas was not higher than that observed in breast cancer generally. CONCLUSION: Our data suggest that preferential involvement of the HER2 gene in micropapillary and apocrine breast carcinomas may contribute to their aggressive behaviour.

Biomechanical properties of the rat oesophagus in experimental type-1 diabetes.

The gastrointestinal tract remodels its morphology and circumferential stress-strain properties in diabetes mellitus. This study adds one more piece of mechanical knowledge, namely the oesophageal shear modulus and its dependence on the circumferential and longitudinal stresses and strains of oesophagus in diabetic rats and control rats. Diabetes was induced by a single intraperitoneal injection of streptozotocin (50 mg kg(-1) body weight). The diabetic rats lived up to 28 days after the induction of diabetes. The oesophagus was studied in vitro using a triaxial machine. Stepwise elongation and inflation plus continuous twist were carried out with measurement of the resultant forces. Circumferential and longitudinal stresses and strains were computed from steady-state values of longitudinal force, outer diameter and the applied pressure. The shear modulus was computed from the twist angle-torque relationship. The circumferential and longitudinal stress-strain relationships were exponential. The circumferential stiffness increased 1 week after induction of diabetes (P < 0.001). The longitudinal stiffness increased in the 4-week diabetic rats (P < 0.001). The shear modulus varied as function of longitudinal strain and pressure. The oesophagus became stiffer in shear 4 weeks after the induction of diabetes (P < 0.001).

Genotype of Candida species associated with different conditions of vulvovaginal candidosis.

It is unknown whether strains isolated from recurrent vulvovaginal candidosis (RVVC), vulvovaginal candidosis (VVC) and asymptomatic carriers are varying or not in genotyping. Candida isolates were genotyped to determine whether specific types were associated with RVVC and VVC. A total of 97 Candida isolates from RVVC patients (n = 43), from VVC patients (n = 47) and asymptomatic carriers (n = 7) were identified by germ tube test, chlamydospore test, CHROMagar Candida and API20 system and were genotyped by polymerase chain reaction (PCR) fingerprinting employing random amplification of polymorphic DNA (RAPD) assay with nine random primers screened. The RAPD identified distinct genotypes that were shared among isolates from most different individuals. Most of the 85 C. albicans strains failed to corroborate any obvious differences in the genotypic variability in the VVC, RVVC patient groups and carriers by clustering analysis. Only a few individuals were found to harbor a distinct genotype of C. albicans isolates associated with a specific disease condition.
