Using an adiabatic approximation method, which searches for Tomlinson model-like instabilities for a simple but still realistic model for two crystalline surfaces, with mobile molecules present at the interface, sliding relative to each other, we are able to account for the virtually universal occurrence of "dry friction" at zero temperature. A modified version of this method allows us to calculate the kinetic friction at nonzero temperature as well. We have also considered the static friction, and have demonstrated that the model is able to account for static friction being larger than kinetic friction.
We report the first observation of the charmless vector-vector decay process B-/+-->rho(-/+)rho(0). The measurement uses a 78 fb(-1) data sample collected with the Belle detector at the KEKB asymmetric e(+)e(-) collider operating at the Upsilon(4S) resonance. We obtain a branching fraction of B(B-/+-->rho(-/+)rho(0))=[31.7+/-7.1(stat)+3.8-6.7(syst)]x10(-6). An analysis of the rho helicity-angle distributions gives a longitudinal polarization fraction of Gamma(L)/Gamma=0.95+/-0.11(stat)+/-0.02(syst). We also measure the direct-CP-violating asymmetry A(CP)(B-/+-->rho(-/+)rho(0))=0.00+/-0.22(stat)+/-0.03(syst).
Polymorphisms at positions -491, -427 and -219 in the promoter region of the Apolipoprotein E APOE gene have been variously reported to confer an increased risk of developing Alzheimer's disease (AD) independent of the effect of epsilon 2, 3 or 4 alleles in exon 4. In order to assess APOE promoter polymorphisms as independent risk factors in AD we have compared results in 183 definite or probable AD cases with 133 controls. We assayed markers at sites -491, -427, -219, and +113 in APOE gene and a polymorphic Hha1 site in the nearby APOC1 gene. We found that APOE promoter polymorphisms and APOC1 insertion alleles were significantly associated with AD. However, after stratification for epsilon 4 allele, only the A allele at -491 in APOE remained significantly associated with AD. The effects of the other markers depended almost entirely upon linkage disequilibrium with epsilon 4 allele, and only trends remained when cases and controls were stratified for the presence or absence of epsilon 4 allele. This occurred irrespective of whether markers were examined separately or together as haplotypes. So in the Chinese population only APOE -491 promoter alleles confer significant risk of AD independent of epsilon 4 status.
We noted previously that after unilateral cochlear ablation (UCA) in young adult guinea pigs, plastic changes in glutamatergic transmitter release in several brain stem auditory nuclei depended on protein kinase C. In this study, we assessed whether such changes depended on protein kinase A (PKA) and calcium/calmodulin-dependent protein kinase II (CaMKII). The electrically-evoked release of D-[3H]aspartate (D-[3H]Asp) was quantified in vitro as an index of glutamatergic transmitter release in the major subdivisions of the cochlear nucleus (CN) and the main nuclei of the superior olivary complex (SOC). In tissues from intact animals, dibutyryl-cyclic adenosine monophosphate (DBcAMP), a PKA activator, elevated D-[3H]Asp release by 1.9-3.7-fold. The PKA inhibitor, H-89 (2 microM), did not alter the evoked release but blocked the stimulatory effects of DBcAMP. These findings suggested that PKA could positively regulate glutamatergic transmitter release. Seven days after the ablation of one cochlea and its cochlear nerve, the stimulatory effect of DBcAMP remained evident. After 145 postablation days, H-89 blocked the plastic elevations of D-[3H]Asp release in the ipsilateral CN and lateral (LSO) and medial (MSO) superior olive. A CaMKII inhibitor, KN-93, produced similar blocks, suggesting that the postablation plasticities in these nuclei depended on PKA or CaMKII. Both H-89 and KN-93 elevated release in the medial nucleus of the trapezoid body (MNTB) and the contralateral MSO, suggesting that either kinase could be used by endogenous mechanisms in these nuclei to downregulate glutamatergic release.
The objectives of the present study were to examine the effects of acute and chronic nicotine on dopamine (DA) release in the ventral tegmental area (VTA) of Long-Evans rats using in vivo microdialysis. Systemic application of acute nicotine (0.1-0.3 mg/kg, s.c.) significantly increased (145% of baseline) DA release in the VTA. Chronic exposure to nicotine (0.3 mg/kg, s.c.) for 5 days followed by a challenge dose of nicotine (0.3 mg/kg, s.c.) also produced significant enhancement (136% of baseline) of DA release in the VTA. The results suggest that both acute and chronic nicotine treatment exert stimulatory effects on somatodendritic DA release in the VTA. The enhancement of DA release to subsequent challenge nicotine may be susceptible to mild desensitization.
Ion emission is investigated using particle-in-cell simulations where a Gaussian laser pulse with duration 50 fs and intensity 1.37x10(19) W/cm(2) is incident obliquely onto ultrathin solid foils. When the foil is thicker than 0.1 microm, it is opaque to the laser light and the highest ion energy drops exponentially with target thickness. Optimization of ion acceleration occurs for a target with a thickness of 0.04 microm when it becomes transparent to the laser light. The behaviors of the high-energy electrons oscillating in the charge separation potential at the front and the rear of the target, as well as the enhanced electron acceleration in the laser pulse, play dominant roles for the observed features of ion emission. The relation of the optimal target thickness with parameters of the incident laser pulse and foil targets is also discussed.
We have measured transverse momentum distributions of charged hadrons produced in d+Au collisions at sqrt[s(NN)]=200 GeV. The spectra were obtained for transverse momenta 0.25<p(T)<6.0 GeV/c, in a pseudorapidity range of 0.2<eta<1.4 in the deuteron direction. The evolution of the spectra with collision centrality is presented in comparison to p+pmacr; collisions at the same collision energy. With increasing centrality, the yield at high transverse momenta increases more rapidly than the overall particle density, leading to a strong modification of the spectral shape. This change in spectral shape is qualitatively different from observations in Au+Au collisions at the same energy. The results provide important information for discriminating between different models for the suppression of high-p(T) hadrons observed in Au+Au collisions.
OBJECTIVE: To investigate the potential for exposure to polycyclic aromatic hydrocarbons (PAHs) to induce oxidative DNA damage, we conducted a cross-sectional study in coke-oven workers employed at an iron-steel factory. METHODS: The study population contained 119 coke-oven workers from different work areas of the oven and 38 controls. Personal information on age, employment duration, smoking habit and alcohol consumption was obtained at an interview. Leukocyte 8-hydroxydeoxyguanosine (8-OHdG) was measured by high performance liquid chromatography with electrochemical detection. Leukocyte aromatic DNA adducts as effective dose, and urinary 1-hydroxypyren as internal dose, were also measured, and used to analyze the relationship of 8-OHdG with other biomarkers for PAH exposure, tobacco smoke and alcohol consumption. RESULTS: The leukocyte 8-OHdG revealed a wide inter-individual variation. The highest 8-OHdG level was detected in bottom-workers of the coke-oven. There were significant differences among the four different work areas ( P=0.02). We could not find significant correlation between 8-OHdG levels and urinary 1-hydroxypyrene, but a weakly positive correlation was found between 8-OHdG and leukocyte aromatic DNA adducts among all subjects (r=0.19 P=0.03). We could not observe any effect of smoking and alcohol drinking on 8-OHdG production. CONCLUSION: We could not find clear evidence that PAH exposure induces oxidative DNA damage.
Using an adiabatic approximation method, which searches for Tomlinson-model-like instabilities for a simple but still realistic model for two crystalline surfaces, with mobile molecules present at the interface, sliding relative to each other, we are able to account for the virtually universal occurrence of "dry friction." The model makes important predictions for the dependence of friction on the strength of the interaction of each surface with the mobile molecules.
The behaviors of the blast waves produced by femtosecond laser-water interactions, and the blast waves induced by laser self-focusing in air, have been investigated using optical shadowgraphy at a maximum intensity of 1 x 10(16) W/cm(2). The temporal evolution of the blast wave launched by the water plasma can be described by a planar blast wave model including source mass. An aneurismlike structure, due to the quick propagation inside a hollow channel formed by laser self-focusing, is observed. The expansion of the channel in air is found to agree with a cylindrical self-similar blast wave solution.
BACKGROUND: Our aim was to evaluate the effect of FTY720 in discordant islet xenotransplantation. METHODS: Fetal porcine islet-like cell clusters (ICCs) were transplanted into normoglycemic rats that were either left untreated or treated with FTY720 only, with FTY720 plus cyclosporine A (CsA) or with CsA only. Twelve or 24 days after transplantation, graft morphology was evaluated immunohistochemically. Furthermore, adult porcine islets (APIs) were transplanted into diabetic rats immunosuppressed with FTY720 plus CsA. Blood glucose and porcine C-peptide levels were monitored. RESULTS: In untreated rats, the ICC xenografts were completely rejected after 12 days. Treatment with CsA had only a marginal effect on the rejection. In animals given FTY720, only the number of infiltrating cells was somewhat reduced. However, at 12 days, no intact ICCs remained. Immunosuppression with FTY720 plus CsA had a marked inhibitory effect on islet xenograft rejection and plentiful morphologically intact ICCs remained. Twelve days after transplantation, only occasional macrophages and T cells could be detected. At 24 days after transplantation, the findings were similar. Furthermore, diabetic rats transplanted with APIs and immunosuppressed with FTY720 plus CsA remained normoglycemic for 53.0+/-15.8 days. In fact, one animal remained normoglycemic for more than 100 days. Serum levels of porcine C-peptide remained at levels similar to those for human C-peptide in healthy individuals. CONCLUSIONS: Immunosuppression with FTY720 plus CsA inhibited almost all morphological signs of pig-to-rat islet xenograft rejection for up to 24 days after transplantation. Diabetic rats transplanted with APIs and immunosuppressed with FTY720 plus CsA remained normoglycemic for 53.0+/-15.8 days.
High energy electrons emitted by water plasmas produced by a single or a multiple laser pulse are investigated. The multipulse mode greatly enhances the generation and the temperature of hot electrons. Directional emission of high energy electrons over 25 keV is observed in two symmetric directions with respect to the laser axis and at 46 degrees from the directions of the laser electric field. Two-dimensional particle-in-cell simulations reproduce well the experimental results and indicate that the acceleration mechanism of the high energy electrons is due mainly to the resonance absorption at the edge of the spherical droplets formed by the leading pulse.
KcURA3 was cloned from Kluyveromyces cicerisporus CBS4857 by complementation of the ura3 mutation in Saccharomyces cerevisiae. KcURA3 encodes a 267-amino-acid protein with 80% sequence identity to that of S. cerevisiae. An ura3 mutant strain from K. cicerisporus CBS4857, named Y179U, was obtained by selection on 5-fluoroorotic acid plates. Sequence analysis of this mutated gene revealed that it contained a point mutation at nucleotide position +277. Two vectors, pUK1 and pUKD, bearing KcURA3 were constructed. Either the lithium acetate method or electroporation could be used to transform pUK1 and pUKD intoY179U. The transformation efficiency using electroporation was higher than that using the lithium acetate method.
AP-2 transcription factors are key regulators of mouse embryonic development. Aberrant expression of these genes has also been linked to the progression of human breast cancer. Here, we have investigated the role of the AP-2 gene family in the postnatal maturation of the mouse mammary gland. Analysis of AP-2 RNA and protein levels demonstrates that these genes are expressed in the mammary glands of virgin and pregnant mice. Subsequently, AP-2 expression declines during lactation and then is reactivated during involution. The AP-2alpha and AP-2gamma proteins are localized in the ductal epithelium, as well as in the terminal end buds, suggesting that they may influence growth of the ductal network. We have tested this hypothesis by targeting AP-2alpha expression to the mouse mammary gland using the MMTV promoter. Our studies indicate that overexpression of AP-2alpha inhibits mammary gland growth and morphogenesis, and this coincides with a rise in PTHrP expression. Alveolar budding is severely curtailed in transgenic virgin mice, while lobuloalveolar development and functional differentiation are inhibited during pregnancy and lactation, respectively. Our studies strongly support a role for the AP-2 proteins in regulating the proliferation and differentiation of mammary gland epithelial cells in both mouse and human.
