Regulation and reconstitution of human hematopoiesis.

Hematopoiesis is regulated by complex interactions between hematopoietic cells and stromal cells within the bone marrow microenvironment. The stromal cells of this microenvironment secrete hematopoietic factors, produce extracellular matrix and mediate direct cell-to-cell contact; each of these events provides a basis for regulatory control of hematopoiesis. With advances in cell surface phenotyping and cell separation technology, it has been shown that the CD34+CD38-CD45RAlowCD71low population contains hematopoietic stem cells. Most hematopoietic stem cells are in a quiescent, noncycling state (G0). In response to external signals, however, they can rapidly enter a functional state (G1) in preparation for DNA synthesis (S-phase). There have been high expectations, recently, that stimulatory hematopoietic factors will have an important impact on chemotherapy by reducing drug-induced neutropenia and, thereby, allowing dose-intensification of treatment. Similar clinical benefits, however, could also be realized by protecting hematopoietic cells from the cytotoxicity of cycle-specific chemotherapeutic agents by blocking the entrance of the hematopoietic stem cells into the cell cycle. Recent studies have suggested that the addition of a negative regulator can better maintain hematopoiesis in vitro and that cells capable of long-term engraftment are primarily comprised of a noncycling population. On the other hand, exposure to cycling-promoting cytokines ex vivo may produce an "engraftment defect". An hu-SCID murine model with a high degree of human cell engraftment will be useful for future studies of the in vivo effects of various agents, infections or gene therapy on human hematopoiesis.

Individual variation in recombination among human males.

Studies of recombination between the markers D6S291 and D6S109 in individuals by sperm typing provide direct evidence for significant variation in recombination among humans. A statistically significant difference in the recombination fraction (range 5.1%-11.2%) was detected among five donors. This variation could reflect polymorphisms in genes affecting recombination or in chromosome structure. Ignoring this variability in studies designed to examine the relationship between physical and genetic distances could lead to incorrect inferences. Individual variation in recombination makes it difficult to predict the recombination fraction for an interval in any particular individual. This could be important in certain genetic counseling situations.

Effect of subunit composition on GABAA receptor complex characteristics in a baculovirus expression system.

A baculovirus expression system was used to produce functional human recombinant GABAA receptors in Sf-9 insect cells in order to study the biochemistry, pharmacology and functional characteristics of this receptor complex. We have identified and characterized various factors which influence the level of receptor expression in multiple virus infections. We have shown that the level of expression of the GABAA receptor complex varies with the levels of expression of the individual subunits. We have also shown that the assembly process has a defined timecourse, and it is dependent upon the ratio of the number of infectious virus particles (MOI ratio) of each subunit in multi-virus infections. In multiple infections, the capacity for expression of the infected cell is shared proportionally by entering virus particles and, there is a direct correlation between the amounts of subunit mRNA and levels of subunit protein expression, and the amount of ligand binding to expressed protein. Finally, reinfection of previously infected cells does not result in subsequent protein expression. Knowledge of these various factors allows us to construct recombinant GABAA receptor complexes with reproducibility and flexibility with regard to subunit composition. By co-expression of alpha, beta, and gamma subunits, both the recognition site for GABA and the allosteric (benzodiazepine) modulatory site are formed and appear to reproduce the pharmacology of endogenously expressed receptors as measured in mammalian CNS. Only a single receptor is produced irrespective of the expression levels of the subunits, showing that GABAA receptor assembly is highly regulated.

Studies on the reaction model for the immobilized Streptomyces aureofaciens's protoplasts used to convert lincomycin.

A reaction kinetic model is proposed for immobilized S. aureofacien's protoplasts which can facilitate the conversion of lincomycin into clindamycin. The simulation result of the model has been given. Effects of various kinetic parameters of the reaction on the conversion rate are given in the formula. The formulas which can be used to calculate the maximum conversion rate and the optimal reaction time have been given. The results of model simulation are in good agreement with the experimental results.

Research on fermentation scale-up based on the OUR obtained from a shake flask.

On the basis of determination of oxygen permeability Pm of gauze layers covered on a specially designed shake flask by the method of sulfite oxidation, the variations of gas-phase and liquid-phase oxygen concentration were measured during the course of fermentation and then the oxygen uptake rate (OUR) and volumetric oxygen transfer coefficient (KLa) could be determined too. Fermentation process and fermentor scale-up were carried out based on the (OUR)max obtained from the shake flask. Scale-up process was studied in different air flow rates and agitation speeds in a 2.5 L fermentor with the analyses by mass spectrometer and manual sampling, it was found that OUR and biomass obtained from the fermentor were quite close to that obtained from the shake flask, but quite different in KLa and CL.

Structural alterations of resistance vessels of spontaneously hypertensive rats and protective effects of captopril.

OBJECTIVE: To observe the structural alterations of resistance vessels in spontaneously hypertensive rats (SHRs) and to evaluate the effects of captopril on them. METHODS: 15-week-old male SHRs were treated with oral captopril (100 mg/kg/day) for 12 weeks. Age- and sex-matched SHRs and Wistar-Kyoto rats (WKY) were fed under the same conditions without captopril administration for the same duration. All rats were killed at the age of 27 weeks. The structural parameters included media width, lumen diameter, media-lumen ratio, vascular collagen area (VCA) and vascular collagen density (VCD). The media width, lumen diameter and media-lumen ratio were measured in hhematoxylin and eosin staining sections under a direct light microscope and VCA and VCD in picrosirius red staining sections under a polarizing light microscope. All the values were processed with a computer image analyzer. RESULTS: There were thicker media width, smaller lumen diameter, higher media-lumen ratio and larger VCA in untreated SHRs than in WKY. Captopril inhibited media thickening, with lumen diameter decreasing, media-lumen ratio increasing and VCA enlarging. There was no significant difference in VCD among the three groups. CONCLUSIONS: Adverse structural alterations occur in resistance vessels of SHRs and captopril may prevent their progression.

[The combined subdermal vascular network skin flaps of the abdominal wall for the repair of soft tissue defects on the dorsum of hand].

Successful reconstructive surgery was performed on ten patients with skin and soft tissue defects on the dorsum of their hand and fingers using combined subdermal vascular network flaps of the abdominal wall. The pedicle of the flap was divided on 7-10 postoperative day when wound was closed, finger and web reconstruction were completed at the same stage. The operative results were satisfactory. The authors discussed the technique of the method, the blood supply of the flap, the principle of the design and the advantages of the operation.

[Analysis of clonal origin of concomitant chronic lymphocytic leukemia and multiple myeloma in a patient with advanced age].

Concomitant chronic lymphocytic leukemia (CLL) and multiple myeloma (MM) is a rare disease. Studies on the clonal origin of both lymphocyte and plasma cell in this disease has been few. So far it has not been reported in China. The paper reports a case of 92-year old patient with concomitant CLL and MM and the research results on the origin of lymphocyte and plasma cell by analyzing the morphologic characteristics, monoclonal antibodies, immunoglobulin Ig or TCR gene rearrangement (GR), Ig single-strand conformation polymorphsm (SSCP) fingerprint mapping and heteroduplex formation. The Ig H GR segments same in size, single strand conformation and sequence characteristics have been obtained from both peripheral blood and bone marrow DNA, even though the cells of CLL and MM had different characteristics in morphology and different secretion of IgM lambda and IgA lambda. The results suggested that the malignant cells of both CLL and MM in this patient had common origin from the B-cell.

[Study on relation of kidney-deficiency with hearing damage and serum trace elements in patients with chronic nephropathy].

The hearing and serum trace element of 73 cases of chronic nephropathy were measured. Results showed that the incidence of hearing damage in high frequency was 68.5% with a mean hypoacusis of 35.1 +/- 6.1 dB, while in 55 patients of Kidney Deficiency type, the incidence of hearing damage in high frequency was 83.6%, it was significantly higher than that in patients without Kidney Deficiency. The serum levels of trace elements (iron and zinc) in patients with Kidney Deficiency were lower than that of normal level and in patients without Kidney Deficiency, and the decrease of serum iron was directly proportional to the degree of hearing damage. There were close relationship between the Kidney Deficiency with the hearing damage as well as the serum levels of iron and zinc.

[A pathological study on phenotype differentiation and its significance in pulmonary large cell carcinoma].

This study was designed to find the differentiated phenotype and its clinicopathological significance in pulmonary large cell carcinoam (PLCC) by immunohistochemical methods using a panel of antibodies related to the phenotype differentiation: high molecular weight cytokeratin, low molecular weight cytokeratiin, secretory component, chromogranin A and synaptophysin in 60 cases of PLCC. The results demonstrated that all cases of PLCC possessed monophasic, biphasic and triphasic phenotype differentiation features respectively, including squamous (4 cases), adenomatous (20 cases), neuroendocrine (19 cases), adenosquamous (11 cases); and the the coexistence of adenomatous, squamous and neuroendocrine (6 cases). The median survival time was different between patients with various differentiated phenotypes of PLCC. There was a statistically significant difference (P < 0.05) in survival between neuroendocrine and adenomatous phenotypes. The results of this study implies that: (1) PLCC expressed different phenotypes of differentiation, (2) phenotypes of neuroendocrine differentiation has a poor prognosis, (3) it is necessary to classify PLCC according to the phenotype differentiation.

[Immunohistochemical studies of nonsecreting pancreatic islet cell tumors secreting many hormones].

To observe the distribution of multiple hormones in nonsecreting islet cell tumors of the pancreas and to study their histogenesis, 9 pancreas nonsecreting islet cell tumor cases were studies using 12 kinds of antisera. The results showed that 4 cases were positive for insulin, 6 for glucagon, 1 for gastrin, 6 for somatostatin, 1 for gastrin 5 for calcitonin, 7 for neurotensin, 4 for ACTH, 3 for TSH, 5 for FSH and 2 for LH. It is therefore confirmed that these tumors synthesize and secrete peptide hormones and glycoprotein hormones. We believe that these endocrine cells originate from primitive multipotential stem cells.

[Negative binomial distribution of eggs of Schistosoma japonicum in stool].

To understand the nature of distribution of schistosoma japonicum (S. japonicum) eggs in stool of the infected individuals, it is assumed that if regression coefficient b approaches two in the equation lg (v) = a + blg(m), with a larger mean of egg counts (m), they can be considered compatible with negative binomial distribution. Results of this study showed egg counts of S. japonicum in stool of the same infected individual on different days and in male and female population can be described by negative binomial distribution, with 95 percent confidence intervals of coefficient of regression 1.3006-2.2956, 1.6109-1.9703, 1.7060-2.1589, and 1.5507-2.2359, respectively. Goodness of fit tests for the observed data indicated that fitting for negative binomial distribution was good. It suggests that the method of moments (coefficient of regression) can be used to determine whether the observed distribution of frequencies is compatible with a negative binomial one.

[Chemical constituents from the whole plant of Epimedium fargesii Franch].

Six chemical constituents were isolated from the whole plant of Epimedium fargesii and identified as epimedoside A, ikarisosids C, epimedin B, epimedin C, icariin and hyperin by means of UV, IR, 1H-NMR, 13C-NMR and MS spectral analysis. They were isolated from this plant for the first time.

[Chemical constituents from the aerial part of Epimedium brevicornum Maxim].

Six flavonoids were isolated from the aerial part of Epimedium brevicornum and identified as baohuo side I, 2"' O-rhamnosyl icariside II, sagittatoside B, baohuoside II, ikarisoside F and ikarisoside C by means of UV, IR, 1H-NMR, 13C-NMR and FAB-MS spectral analysis, they were isolated from this plant for the first time.

[An experimental study on the relationship between BMP's activity and its carrier].

To find out the relationship between human bone morphogenetic protein's activity and its carrier and to document the clinical application of BMP, we investigated six kinds of BMP carrier's influence upon hBMP's osteogenic activity with small sample size at first. Then three kinds of carriers selected from the six, added with the seventh carrier, were investigated with large sample size. The result showed that the compound of hBMP and hDDBM had best osteogenic ability. The human bone matrix had been formerly demineralized and extracted without hBMP. The hDDBM showed porous structure under SEM, its mean pore's diameter was 127 +/- 34 microns. It was demonstrated that the function of demineralized bone matrix extracted without BMP (i.e. DDBM) was related not only to its pore's diameter, but to the fibrolike matter in it as well. With the method of BMP bioassay, BMP was composited with carrier, then the compound was injected into the calf of mouse. This made BMP's bioassay become simple and reliable.

[Management of subclavian artery aneurysm].

The subclavian artery aneurysm is not a commonly seen peripheral aneurysm. We treated 7 cases in recent 13 years. There were 4 tramatic pseudoaneurysms and 2 atherosclerotic aneurysms. Four aneurysms were resected and replaced by PTFE or Dacron vascular prostheses. Two were treated by arterial repair. Good long-term outcome was obtained.
