Isolation and characterization of subacute sclerosing panencephalitis virus nucleocapsids.

Nucleocapsids from subacute sclerosing panencephalitis (SSPE) virus-infected CV-1 cells were concentrated by differential centrifugation employing sucrose cushion techniques and further purified by centrifugation through a linear CsCl density gradient. The bouyant density of (3)H-uridine-labeled nucleocapsids in CsCl was found to be 1.31 g/cm(3). Ultraviolet absorption spectra of the purified SSPE nucleocapsid showed an absorption maximum at 260 to 265 nm and a 280/260 ratio that corresponded to a nucleic acid content of approximately 4.3%. Negatively stained preparations of SSPE nucleocapsids were found to have a width of 18 +/- 1 nm, a periodicity of 5 to 6 nm, and a length between 1-1.4 mum, with the greatest number at 1.3 mum.

Contact inhibition in colony formation.

Contact inhibition of replication of the established mammalian cell line 3T3 was examined during growth of the colony and compared with that of the Chinese-hamster cell line CHL-1. The growth curves of cells in the colonies conformed to the predicted exponential and linear rates for CHL-1 and 3T3 respectively. Autoradiographs of colonies in which DNA was labeled with tritiated thymidine showed that in 3T3 colonies, only peripheral cells were labeled, while CHL-1 colonies were labeled throughout.

Apoptosis as a basic mechanism in the ovarian cycle: follicular atresia and luteal regression.

OBJECTIVE: We summarized general principles related to apoptosis and reviewed the current evidence regarding apoptosis in the normal ovary during follicular atresia, ovulation, and corpus luteum formation and regression, and in the polycystic ovary. DATA SOURCES: Published articles and abstracts from national scientific meetings were reviewed. DATA SYNTHESIS: Apoptosis is a physiologic type of cell death that may regulate tissue remodeling. In the ovary, apoptosis occurs in atretic follicles, in the ovarian epithelium at the time of ovulation, and in the corpus luteum. Certain apoptosis-related genes are expressed during follicular atresia, which suggests a role for genes in the regulation of apoptosis in the ovary. In a rat model of polycystic ovaries, apoptosis occurs in both atretric follicles and cysts. CONCLUSIONS: Apoptosis is an important physiologic process in the ovary, and abnormalities in this process may lead to pathologic conditions. The study of ovarian apoptosis is still at an early stage, and further research will uncover the steps in this process.

Results of in vitro fertilization pregnancies: experience at Boston's Beth Israel Hospital.

The outcomes of 65 consecutive pregnancies achieved by in vitro fertilization (IVF) at Boston's Beth Israel Hospital (BIH) were studied. A total of 38 infants were delivered from the 65 pregnancies. The rate of multiple gestation was 17%. Premature delivery occurred in 24% of the IVF patients, and the cesarean section rate was 45% (neither significant versus controls); there was one IVF maternal death. Tocolytic agents were used more often (P less than 0.25) in IVF pregnancies than in controls, but no other pregnancy complication was statistically more frequent. The IVF perinatal mortality rate was 26.3 per 1,000. Of the BIH IVF infants, 55% had problems in the immediate neonatal period (P less than .01 compared with controls), and 37% of the IVF neonates required transfer to the neonatal intensive care unit. Congenital defects, including minor ones, were present in 8.6% of the IVF newborns. We concluded that IVF pregnancies and deliveries require careful monitoring.
