Low power density microwave radiation induced early changes in rabbit lens epithelial cells.

OBJECTIVE: To determine whether low power density microwave radiation can induce irreversible changes in rabbit lens epithelial cells (LECs) and the mechanisms of the changes. METHODS: One eye of each rabbit was exposed to 5 mW/cm2 or 10 mW/cm2 power density microwaves for 3 hours, while the contralateral eye served as a control. Annexin V-propidium iodide (PI) two-color flow cytometry (FCM) was used to detect the early changes in rabbit lens epithelial cells after radiation. RESULTS: Lots of rabbit LECs were in the initial phase of apoptosis in the 5 mW/cm2 microwave radiation group. A large number of cells became secondary necrotic cells, and severe damage could be found in the group exposed to 10 mW/cm2 microwave radiation. CONCLUSION: Low power densities of microwave radiation (5 mW/cm2 and 10 mW/cm2) can induce irreversible damage to rabbit LECs. This may be the non-thermal effect of microwave radiation.

Retinochoroidal changes after severe brain impact injury in rabbits.

OBJECTIVE: To investigate retinochoroidal changes and establish eye damage model after brain impact injury. METHODS: An eye damage model after brain impact injury was established by striking the frontoparietal zone in rabbits with BIM-II bioimpact machine. Seventeen rabbits were killed at 4 different intervals after injury. The pathological characteristics of the retinal and choroid damages were observed. RESULTS: All the rabbits had severe brain injury with subarachnoid hemorrhage and brain contusion. The eye damage occurred in all of the 17 rabbits. Hemorrhage in optic nerve sheaths was observed and retinal edema and bleeding was discovered with ophthalmoscope. Histopathologic study displayed subarachnoid hemorrhage in the retrobulbar portion of the retinal nerve, general choroid blood vessel dilatation, retinal nerve fibre swelling within 6 hours after injury, and flat retinal detachment with subretinal proteinoid exudation, and degeneration and disappearance of the outer segment of the optic cell over 6 hours after injury. CONCLUSIONS: The pathological characteristic of the eye damage at early stage following brain impact injury is local circulation disturbance. At late stage, it features in retinal detachment, and optic cellular degeneration and necrosis.

[Ultrastructural change of rabbit lens epithelial cells induced by low power level microwave radiation].

OBJECTIVE: To study the ultrastructural change of rabbit lens epithelial cells with transmission electron microscope and to observe whether low power level microwave can induce the injury of rabbit lens. METHODS: Eight rabbit eyes were exposed to 10 mW/cm(2) low power level microwave radiation for 30 hours. Transmission electron microscope was used to observe the ultrastructural changes of the lens epithelial cells. The other eye was served as self-control. RESULTS: The ultrastructure of exposed lens epithelial cells changed apparently. The characteristic apoptosis cell morphological changes, such as small cell size, the decreased number of mitochondria, balloon-shaped mitochondria, etc can be seen. No obvious changes can be found in the protected self-control eyes. CONCLUSIONS: The low power level microwave (10 mW/cm(2)) can induce the ultrastructural changes of rabbit lens epithelial cells, this may be the appearance of early irreversible microwave radiation injury. In regard to the pathogenic mechanism, it is necessary to perform further investigations.

[Homologous recombination in Streptomyces lincolnensis B48].

To study frequency and mechanism of homologous recombination in Streptomyces, an E. coli plasmid which cannot replicate in Streptomyces was transformed into Streptomyces lincolnensis B48. After homologous recombination between delta lincomycin biosynthetic genes inactivated by thiostrepton resistant gene (tsr) carried on pYYE04al and homologous sequences on the chromosome, S. lincolnensis YY1 and S. lincolnensis, YY2 were obtained on SMA with low thiostrepton concentration. Hybridization of chromosomal DNA samples of S. lincolnensis YY1, S. lincolnensis YY2, standard S. lincolnensis and S. lincolnensis YYc digested with SmaI with the probe of tsr gene gave signal corresponding to a fragment of 1.5 kb in the former two; Nevertheless, hybridization of chromosomal DNA digested with Hind III and Sma I using the probe of delta lacZ' gene resulted in positive fragment of 4.4 kb only in S. lincolnensis YY2. Southern hybridizations indicate that S. lincolnensis YY1 is the result of homologous exchange while S. lincolnensis YY2 comes from bomologous recombination. To prove the existence of E. coli replicon and ampicillin resistant gene on the chromosome of S. lincolnensis YY2, its DNA digested with SphI was ligated and then transformed into E. coli JM83 competent cell. Two transformants named pSLE1 grew on the plate containing ampicillin. It's confirmed that pSLE1 is a part of pYYE04a1 from its digestion with different enzymes.

[The cause and management of acute high intraocular pressure in phacoemulsification].

OBJECTIVE: To investigate the cause and management of acute high intraocular pressure and shallow anterior chamber in phacoemulsification. METHODS: A retrospective study was done on 1,500 eyes performed phacoemulsification. Eleven of 1,500 eyes showed higher intraocular pressure and shallowing of the anterior chamber during phacoemulsification. But phacoemulsification was successfully achieved after giving hyperosmotic agents, increasing infusion pressure and reducing aspiration rate as well as flow rate. RESULTS: Of 11 eyes, 10 eyes were treated by management of non vitrectomy. One of 11 eyes was unresponsive to medical management and require the use of pars plana vitrectomy. Phaco was performed in all patients. In 4 of 11 eyes, visual acuity postoperatively were 0.7 or better. Five of 11 eyes were 0.3 to 0.5. One eye was 0.05 and one was HM/30 cm. CONCLUSION: Aqueous misdirection, retrobulbar hemorrhage and intraoperative suprachoroidal hemorrhage are common causes of acute high intracular pressure in phaco. Most of patients could be performed phaco after medical treatment. If patients failed to improve after medical treatment, par plana vitrectomy is a useful method.

[Anatomic characteristics of lingual artery and midline glossectomy].

OBJECTIVE: To investigate the relationship between anatomic characteristics of lingual artery and midline glossectomy, for the purpose of increasing operative safety. METHODS: (1) 16 extraoral dissections of the submandibular region were performed on 8 human cadavers and to observe the origin and distribution of lingual artery. (3) The lingual artery was divided into three segments, and the length and adjacent organs were observed and measured. RESULTS: The whole length of lingual artery was (92.88 +/- 13.53) mm; At points of foramen cecum of the tongue, 5 mm and 10 mm prior to foramen cecum, 10 mm behind foramen cecum, and at adjacent part between root of the tongue and epiglotic vallecula, the distances between lingual artery and midline of the tongue were separately measured. The distances were (11.00 +/- 4.95) mm, (8.56 +/- 3.51) mm, (7.78 +/- 3.15) mm, (13.00 +/- 3.28) mm, (15.22 +/- 3.11) mm. The distances between lingual artery and the surface of tongue were (21.4 +/- 5.59) mm, (22.11 +/- 6.74) mm, (22.56 +/- 7.81) mm, (16.56 +/- 5.57) mm, (2.56 +/- 0.73) mm respectively. CONCLUSION: During the midline glossectomy, the lingual artery is easier to be damaged at the position where the root of tongue adjoining epiglottic vallecala. The operation should be done within 10 mm below the surface, 7 mm lateral to the midline, to avoid the damage of lingual artery.

[Clinical study of juvenile-onset recurrent respiratory papillomatosis].

OBJECTIVE: To study the clinical behavior of juvenile-onset recurrent respiratory papillomatosis in order to find some factors correlated to the development of this disease, and to sum up the significance and experience of CO2 laser surgery. METHOD: Sixty patients with juvenile-onset recurrent respiratory papillomatosis from September 1995 to December 1998 were retrospectively analyzed. RESULTS: The age of onset in 50 cases (83.3%) was below 4 years, and the peak-age was 2 years. The rates of recurrence were 72.0% and 45.7% (chi 2 = 4.71, P < 0.05) below and over 2 years, respectively. The rates of aggressive disease were 88.0% and 54.3% (chi 2 = 7.66, P < 0.01) below and over 2 years, respectively. The predominant sites of the disease were the vocal cords, the false vocal cords, the laryngeal ventricle, the laryngeal surface of the epiglottis and the subglottic region. Tracheostomy induced the development of tracheal papilloma, therefore should be avoided as possible. Laryngeal papilloma might be divided into four types on the basis of the growth manner and surface form corresponding to clinical behaviors. Five patients were followed-up for 1.5 years without recurrence, 18 patients had fewer recurrences following treatment, 33 patients were under treatment, and 3 patients died. Nineteen patients lost follow-up. The major complications included laryngeal and tracheal stenosis. CONCLUSION: Clinical behaviors of juvenile-onset recurrent respiratory papillomatosis were relevant to the age, growth form and tracheotomy. CO2 laser was an ideal instrument for ablation of the laryngeal papillomas with the following advantages: simple management, less bleeding, preservation of laryngeal structure and avoidance of tracheostomy.

[Determining the site of airway obstruction in obstructive sleep apnea with airway pressure measurements during sleep].

OBJECTIVE: Determine the site(s) of upper airway obstruction and the influence of sleep stage on the pattern of obstruction with upper airway (UA) pressure measurement. Analyses the clinical value of UA pressure measurement and it's influenced factors. METHODS: 30 OSAS patients underwent UA pressure measurement during polysomnography. Multisensory pressure catheters with five solid-state ultraminiature sensors were insert through patients' upper airway to esophagus, The sensors were located at the nasopharynx, oropharynx, tongue base, hypopharynx and esophagus and the lower limit of UA obstruction was determined relying on the observed pressure pattern. RESULTS: (1) During inspiration, obstruction occurred associated with a increased negative inspiratory pressure inferior to the site of obstruction and a disappeared negative inspiratory pressure above the site of obstruction. (2) Three patterns of obstruction were observed. 1. The site of obstruction was located at the level of the palate. 2. The level of the palate and tonguebase obstruction all present. 3. Nasopharyngeal obstruction soft plate obstruction and tongue-pharynx obstruction all present. (3) The factors which affected these measurement are 1. The catheter were placed by observation with a fiberoptic endoscope to keep the sensors located at the correct position. 2. Catheter plugging with secretions need to be prevented. CONCLUSION: UA pressure measurement can objectively identify the level of obstruction during sleep.

[Study on the interactions between a 3H-indole probe molecule and Triton X-100 micelles by spectral methods].

The interactions between a fluorescent molecule, [2-(p-hexylamino) phenyl-3,3-dimethyl-5-ethoxycarbonyl-3H-indole] methyldiioctadecylammonium iodide (A), with Triton X-100 micelle are investigated by absorption and fluorescence spectra. The relatively large binding constant Ks shows that strong interactions between A and micelles do exist. The polarity parameter confirms that the polarity of the environment of A in the micelle is similar to that of methanol. According to the spectral characteristics and pH effects, the various sites of different groups of A in micelles are suggested. Lifetime measurements indicate that A can recognize two sites with Triton X-100 micelle at lower surfactant concentration. However, in the Triton X-100 micelle at higher surfactant concentration, A can only recognize one site.

[Quantum chemistry Ab initio calculation of vibration spectrum of beta-elemene].

In this study, ab initio calculation of vibration spectrum of beta-elemene has been performed at RHF/6-31G* level. Comparison with the experimental results, we succeed in assignment of normal modes of the fundamental frequency of the molecule. We have also discussed the characteristic vibrations, and scaled theoretical frequencies empirically.

Activation of mitogen-activated protein kinase p38 and extracellular signal-regulated kinase is involved in glass fiber-induced tumor necrosis factor-alpha production in macrophages.

In a previous study, we demonstrated that the length of glass fibers was a critical determinant of fiber potency in induction of tumor necrosis factor (TNF)-alpha and that activation of NF-kappaB was an important factor in this response. In the present study, we analyzed the role of mitogen-activated protein (MAP) kinases in the induction of TNF-alpha by glass fibers. Glass fibers induced phosphorylation of MAP kinases, p38, and ERK in primary rat alveolar macrophages, and this phosphorylation was associated with TNF-alpha gene expression. Long fibers were more potent than short fibers in activation of MAP kinases. Results from mechanistic analysis support that MAP kinases activate transcription factor c-Jun. The activated c-Jun acts on the TNF-alpha gene promoter through two binding sites, the cyclic AMP response element and the activator protein 1-binding site. These results suggest that in addition to the NF-kappaB pathway for TNF-alpha production, glass fibers are able to activate c-Jun through MAP kinase pathways that lead to induction of TNF-alpha expression.

In vitro cellular tropism of human T cell leukemia virus type 2.

Human T cell leukemia virus type 1 (HTLV-1) and 2 (HTLV-2) are distinct oncogenic retroviruses that infect several cell types, but display their biologic/pathogenic activity only in T lymphocytes. HTLV-1 is associated with adult T cell leukemia, a malignancy of mature CD4(+) T cells, and a chronic neurological disorder termed HTLV-1-associated myelopathy/tropical spastic paraparesis (HAM/TSP). HTLV-2 is less pathogenic and has been associated with a few cases of a variant of hairy cell leukemia and neurological disease. Previous studies have indicated that in vivo HTLV-1 has a preferential tropism for CD4(+) T cells, whereas HTLV-2 in vivo tropism is less clear, but appears to favor CD8(+) T cells. The molecular mechanism that determines the cellular tropism of HTLV-1 and HTLV-2 has not been precisely determined. However, one study by our group has provided evidence that HTLV-1-enhanced viral transcription in CD4(+) T cells may be responsible for its tropism. In an effort to understand HTLV-2 tropism we tested the ability of HTLV-2 to infect, replicate in, and transform purified CD4(+) or CD8(+) T cells in cell culture. After cocultures of purified primary human CD4(+) and CD8(+) T cells with an HTLV-2-producer cell line we measured viral transcription by reverse transcription PCR analysis, virus production by p19(gag) ELISA, proviral integration by DNA slot-blot analysis, surface phenotype by FACS analysis, and cellular transformation. We also measured HTLV-2 long terminal repeat-directed transcription in the presence and absence of Tax in purified CD4(+) and CD8(+) T cells, using transient transfection assays. Our data indicate that CD4(+) and CD8(+) T cells are equally susceptible to HTLV-2 infection. We observed no significant difference in viral transcription based on mRNA and virus production in CD4(+) and CD8(+) T cell cocultures. Although LTR transcription was enhanced 12- to 16-fold in the presence of Tax, there was no significant difference in CD4(+) or CD8(+) T cells. Interestingly, we show that HTLV-2 preferentially transforms CD8(+) T cells in culture. Together, our data indicate that, unlike HTLV-1, HTLV-2 cell tropism is not due to inhibition of viral infection and inefficient gene expression in CD4(+) versus CD8(+) T cells, and likely involves unique interactions with viral and CD8(+) T cell-specific proteins.

Accuracy comparison of absolute optical frequency measurement between harmonic-generation synthesis and a frequency-division femtosecond comb

Using an iodine-stabilized He-Ne laser as a transfer oscillator, we compare absolute measurements of the optical frequency from a traditional frequency synthesis chain based on harmonic generation and from the frequency division technique of an ultrawide bandwidth femtosecond frequency comb. The agreement between these two measurements, both linked to the Cs standard, is 220+/-770 Hz, yielding a measurement accuracy of 1.6x10(-12). We report 473 612 353 604.8+/-1.2 kHz as a preliminary updated value of the absolute frequency of the " f" component for the He-Ne laser international standard at 633 nm.

Vanadate induces p53 transactivation through hydrogen peroxide and causes apoptosis.

Vanadium is a metal widely distributed in the environment. Although vanadate-containing compounds exert potent toxic effects on a wide variety of biological systems, the mechanisms controlling vanadate-induced adverse effects remain to be elucidated. The present study investigated the vanadate-induced p53 activation and involvement of reactive oxygen species (ROS) in p53 activation as well as the role of p53 in apoptosis induction by vanadate. Exposure of mouse epidermal JB6 cells to vanadate led to transactivation of p53 activity in a time- and dose-dependent manner. It also caused mitochondrial damage, apoptosis, and generated ROS. Scavenging of vanadate-induced H(2)O(2) by N-acetyl-l-cysteine (a general antioxidant) or catalase (a specific H(2)O(2) inhibitor), or the chelation of vanadate by deferoxamine, resulted in inhibition of p53 activation and cell mitochondrial damage. In contract, an increase in H(2)O(2) generation in response to superoxide dismutase or NADPH enhanced these effects caused by vanadate. Furthermore, vanadate-induced apoptosis occurred in cells expressing wild-type p53 (p53+/+) but was very weak in p53-deficient (p53-/-) cells. These results demonstrate that vanadate induces p53 activation mainly through H(2)O(2) generation, and this activation is required for vanadate-induced apoptosis.

Measurements of the mass, total width, and two-photon partial width of the eta(c) meson.

Using 13.4 fb(-1) of data collected with the CLEO detector at the Cornell Electron Storage Ring, we have observed 300 events for the two-photon production of ground-state pseudoscalar charmonium in the decay eta(c)-->K(0)(S)K-/+pi(+/-). We have measured the eta(c) mass to be [2980.4+/-2.3 (stat)+/-0.6 (syst)] MeV and its full width as [27.0+/-5.8 (stat)+/-1.4 (syst)] MeV. We have determined the two-photon partial width of the eta(c) meson to be [7.6+/-0.8 (stat)+/-0.4 (syst)+/-2.3 (br)] keV, with the last uncertainty associated with the decay branching fraction.

Study of charmless hadronic B meson decays to pseudoscalar-vector final states.

We report results of searches for charmless hadronic B meson decays to pseudoscalar( pi(+/-), K+/-, pi(0), or K(0)(S))-vector( rho, K(*), or omega) final states. By using 9.7x10(6) BB pairs collected with the CLEO detector, we report the first observation of B(-)--->pi(-)rho(0), B(0)-->pi(+/-)rho(-/+), and B(-)-->pi(-)omega, which are expected to be dominated by hadronic b-->u transitions. The measured branching fractions are (10.4(+3.3)(-3.4)+/-2.1)x10(-6), (27.6(+8.4)(-7.4)+/-4.2)x10(-6), and (11.3(+3.3)(-2.9)+/-1. 4)x10(-6), respectively. Branching fraction upper limits are set for all of the other decay modes investigated.

High-efficiency gene transfection of macrophages by lipoplexes.

Macrophage transfection studies are crucial for understanding gene regulation and expression. However, gene transfection in macrophages is difficult. We have shown here that macrophages are more resistant to gene transfection compared with other cell types. To further develop an efficient gene delivery system for macrophages, we evaluated various liposomal and non-liposomal agents including LipofectAMINE(R), Lipofectin(R), DOTAP, DEAE-dextran, and the DNA condensing agent protamine sulfate for their ability to promote gene transfection. CMV-luciferase was used as a reporter plasmid. Macrophage transfection was maximal at the DNA:LipofectAMINE:protamine ratio of 1:12:1 microg/ml. The LipofectAMINE formulation showed a 10-12-fold increase in transfection efficiency over DOTAP and a 4-5-fold increase over Lipofectin. This transfection method showed minimal toxicity at the concentrations tested and was at least 20-25-fold superior to the most frequently used DEAE-dextran method for macrophage transfection.
