[Photodynamic effect of HPD on DNA functions].

After treatment of the DNA by HPD and light the number of transformants of the M13mp8 DNA obviously decreased during the period of transforming the treated DNA into E coli. Meanwhile some of the mutation sequences appeared in the DNA which recovered from the DNA/HPD/Light transformant. It showed that the wrong replication happened because of the mutated template. Also the in vitro run-off transcription of IL-2R alpha gene was affected by HPD and light. It means that the DNA functions are influenced by the photodynamic effects.

[The deterioration of refractive accommodative esotropia].

137 patients with refractive accommodative esotropia who were followed for at least 5 years were investigated. The results showed that in 23 of the 137 patients (16.8%) occurred the deterioration of esotropia that means esotropia can no longer be controlled by wearing a pair of complete corrective spectacles. The deterioration develops most likely in patients with delay of anti-accommodative therapy or with malfunction of binocular vision. The age of onset, refractive status and the visual acuity difference between bilateral eyes are not etiologic factors in the process of deterioration. We consider that early diagnosis of accommodative esotropia, timely prescription of optical correction and maintenance of normal binocular vision play important roles in preventing the deterioration.

[The quantitative and morphological analysis of placenta and brain in rabbits with intrauterine fetal growth retardation].

OBJECTIVE: To observe the changes of quantitative and morphological structure in placenta and fetal brain of rabbits with intrauterine fetal growth retardation (IUGR) resulted from exposure to cigarette smoking. METHODS: The pure species of 23 pregnant New Zealand rabbits were used and divided into three groups. (1) group A: 9 rabbits were exposed to cigarette smoking everyday in the whole period of pregnancy; (2) group B: 7 rabbits were exposed to cigarette smoking everyday in the late half period of pregnancy; (3) group C: no smoking, as control group. The villus area of placenta, the capillary area in villus and the thickness of granulosa layer in fetal brain were studied by histomorphometric parameter. RESULTS: It was showed that the placental villus area was decreased significantly in group A than that of the control group. The capillary area in villus of both group A and group B was less than that of the group C. The thickness of granulosa layer in cerebrum and cerebellum of group B is less than that of the control group. Nevertheless, only the thickness of granulosa layer in cerebrum of group B decreased significantly than that of the control group. CONCLUSIONS: The development of cerebrum in fetal rabbits were influenced significantly by smoking. The development of cerebellum was influenced by smoking mainly in the early half period of pregnancy. It means that the most important time for cerebellum development is the early period of pregnancy and both the early and late half period of pregnancy were important for the cerebrum development.

Study on motor functions of antrum after highly selective vagotomy (HSV) plus mucosal antrectomy.

To elucidate the effects of highly selective vagotomy plus mucosal antrectomy (HSV-MA) antral functions, eighteen dogs were randomly divided into control, HSV and HSV-MA group for determining gastric emptying, antral myoelectric activity and antral pressure. The tests were performed 4 months after operation in the HSV-MA and HSV group. By roentgenoscopy, the gastric and duodenal shape were normal, and gastric peristalsis was clearly seen in all the dogs in the three groups. The beginning and ending time of gastric emptying in the HSV-MA group were 5.0 +/- 0.06 minutes and 4.0 +/- 0.4 hours respectively, which were similar to those in the control and HSV group (P > 0.05). The frequency of antral myoelectric action potential in the HSV-MA group (3.11 +/- 0.65 cycles/minutes) was significantly lower than those in the control and HSV group (P < 0.05). Intramuscular injection of pentagastrin significantly increased the frequency of antral myoelectric action potential (P < 0.01) in all groups, and eliminated the difference among them before the injection. The antral pressure wave occurred immediately following the antral myoelectric action potential waves. The antral pressure values in the control and HSV-MA group were similar (P > 0.05), and were significantly lower than that in the HSV group (P < 0.05). Intramuscular injection of pentagastrin significantly increased the antral pressure (P < 0.001) in all groups and eliminated the difference among them before the injection. These evidences suggest that the motor function of the reconstructed antrum following HSV-MA nearly reached normal level despite a lower antral myoelectric frequency. Combined with other reports, HSV-MA has a good clinical prospect as an operation for the treatment of chronic duodenal ulcer.

Early histologic response to titanium implants complexed with bovine bone morphogenetic protein.

This study established a method for combining bovine bone morphogenetic protein (bBMP) with titanium and evaluated the early bone formation induced by the bBMP/titanium complex in edentulous dogs. Results showed newly formed bone within the interface bone 2 weeks after implantation and nearly complete osseointegration 4 weeks after implantation. Bone formed with the apical opening of the implant within 1 month. This study indicates that osseointegration can be enhanced by bBMP-bone induction. The apical opening of the implant may provide a site in which osteogenesis can occur with protection from implant stress before and after loading.

[Bleeding and complications related to hemorrhage during transsphenoidal removal of pituitary tumor].

450 cases who undertook the transsphenoidal removal of pituitary tumors from 1982 to 1992 were reviewed. The bleeding during operation and the complications related to the bleeding were reported in 19 cases. The intercavernous sinuses were traumatized during operation in 9 cases. The possibility of abnormal dural arteries existed in 3 cases. The bleeding happened in the course of dissociation of nasal septum in 2 cases. The cause of bleeding in 3 cases were unclear. There was no much bleeding or bleeding record during operation, but postoperative bleeding complications appeared in other 2 cases. Heavy bleeding came from nose repeatedly in 2 cases, one of which was confirmed as pseudoaneurysm. The postoperative subarachnoid haemorrhage in 2 cases, one blindness and one paralysis of abductor nerve happened. The causes of bleeding and complication were discussed.

Molecular cloning and expression of the catalytic subunit of bovine pyruvate dehydrogenase phosphatase and sequence similarity with protein phosphatase 2C.

After many unsuccessful attempts to detect cDNA encoding the catalytic subunit of bovine pyruvate dehydrogenase phosphatase (PDPc) in bovine cDNA libraries, an approach based on the polymerase chain reaction (PCR) was undertaken. Overlapping DNA fragments were generated by PCR from bovine genomic DNA and from cDNA synthesized from total RNA with synthetic oligonucleotide primers on the basis of experimentally determined amino acid sequences. The DNA fragments were subcloned and sequenced. The complete cDNA is 1900 base pairs in length and contains an open reading frame of 1614 nucleotides encoding a putative presequence of 71 amino acid residues and a mature protein of 467 residues with a calculated M(r) of 52,625. Hybridization analysis showed a single mRNA transcript of about 2.0 kilobases. Comparison of the deduced amino acid sequences of the mitochondrial PDPc and the rat cytosolic protein phosphatase 2C indicates that these protein serine/threonine phosphatases evolved from a common ancestor. The mature form of PDPc was coexpressed in Escherichia coli with the chaperonin proteins groEL and groES. The recombinant protein (rPDPc) was purified to near homogeneity. Its activity toward the bovine 32P-labeled pyruvate dehydrogenase complex was Mg(2+)-dependent and Ca(2+)-stimulated and comparable to that of native bovine PDP. An active, truncated form of rPDPc, with M(r) approximately 45,000, was produced in variable amounts during growth of cells and/or during the purification procedure.

An immunohistochemical study of bone morphogenetic protein in pleomorphic adenoma of the salivary gland.

Bone morphogenetic protein (BMP) is a potent induction factor for new bone formation including heterotopic chondro-ossification in soft tissues. The immunohistochemical reaction for BMP was studied in 23 cases of pleomorphic adenoma of salivary gland by using a monoclonal antibody produced by hybridoma technique. Positive BMP immunoreactivity was seen in 87% of tumours. Immunohistochemical expression of BMP was observed in modified myoepithelial cells (88% cases), luminal tumour cells of tubulo-ductal structures (78% cases) and chondroid cells in hyaline tissue (22% cases). The authors concluded that the simultaneous presence of glycosaminoglycans as matrix substance with BMP-mediated cellular activity of modified myoepithelial cells in the formation of chondroid structures in pleomorphic adenomas of the salivary glands.

The effect of bone morphogenetic protein on osseointegration of titanium implants.

This study evaluated the osteointegration of threaded cylindrical uncoated endosseous implants in combination with either bovine bone morphogenetic protein (bBMP) (group 1) or bovine serum albumin (BSA) (group 2) in edentulous dogs. Histologic observations showed that new bone formation was markedly increased by bBMP. The screw-surfaced implants in combination with bBMP exhibited apparent direct bone apposition 4 weeks postimplantation with no evidence of a connective tissue capsule. Thick fibrous connective tissue still encapsulated the control implants, although some new bone was observed slowly growing from the margins of the host bed. Eight weeks after implantation, there was an exact fit between the implant and the bone in group 1, and the surrounding bone was arranged in a regular manner. In group 2, there were still a few areas of thin fibrous tissue surrounding the implants. The results indicate that the rate of osteointegration was increased by bBMP.

Immunohistochemical localization of bone morphogenetic protein (BMP) in calcifying fibrous epulis.

Immunohistochemical detection of bone morphogenetic protein (BMP) in calcifying fibrous epulis was performed to elucidate the biological process of ossification and cemento-ossification. In a total 25 cases, 15 (60%) showed positive BMP staining in bone forming areas. Histopathological features of developing hard tissues were varied, consisting of structures such as woven bone and cemento-osseous formations. BMP immunostaining was limited to osteoblasts and fibrous connective tissue surrounding the bone matrix. BMP was concentrated in the periodontal fibres and in dense fibrous structures in the cemento-osseous masses. On the basis of histopathological and immunohistochemical features, the histogenesis of ossifying and cemento-ossifying processes appear to be of two possible origins; the excessive proliferation of periodontal ligament and a metaplastic process occurring in the connective tissue fibres (non-periodontal in origin), with the former being more common.

Insect tolerance of transgenic Populus nigra plants transformed with Bacillus thuringiensis toxin gene.

Leaves and stem segments of Populus nigra were transformed with A. tumefaciens LBA4404 harboring a binary vector containing chimeric genes of NPT and 35S-omega-B.t. toxin-Nos. Nineteen regenerated kanamycin resistant plants were analyzed by DNA hybridization, out of which 10 were shown to be the candidates of transgenic plants. Insect tolerance tests showed that the transgenic plants were toxic to two lepidopteran pests, Lymantria dispar L. and Apocheimia cinerarius Erschoff. Based on the results of Southern blot of PCR products and the cluster analysis of their growth and insect resistance, three independent transgenic plants were selected, which were then propagated in the nursery field. This field test is currently in progress.

Cytochrome P-450 arachidonic acid epoxygenase. Regulatory control of the renal epoxygenase by dietary salt loading.

The rat kidney microsomal epoxygenase catalyzed the asymmetric epoxidation of arachidonic acid to generate as major products: 8(R),9(S)-, 11(R),12(S)- and 14(S),15(R)-epoxyeicosatrienoic acids with optical purities of 97, 88, and 70%, respectively. Inhibition studies utilizing a panel of polyclonal antibodies to several rat liver cytochrome P-450 isoforms, indicated that the renal epoxygenase(s) belongs to the cytochrome P-450 2C gene family. Dietary salt, administered either as a 2-2.5% (w/v) solution in the drinking water or as a modified solid diet containing 8% NaCl (w/w), resulted in marked and selective increases in the renal microsomal epoxygenase activity (416 and 260% of controls, for the liquid and solid forms of NaCl, respectively) with no significant changes in the microsomal omega/omega-1 oxygenase or in the hepatic arachidonic acid monooxygenase reaction. Immunoblotting studies demonstrated that dietary salt induced marked increases in the concentration of a cytochrome P-450 isoform(s) recognized by polyclonal antibodies raised against human liver cytochrome P-450 2C10 or rat liver cytochrome P-450 2C11. Comparisons of the stereochemical selectivity of the induced and non-induced microsomal epoxygenase(s) with that of purified rat liver cytochrome P-450 2C11 suggest that the salt-induced protein(s) is catalytically and structurally different from liver cytochrome P-450 2C11. The in vivo significance of dietary salt in regulating the activities of the kidney endogenous arachidonic acid epoxygenase was established by the demonstration of a salt-induced 10-20-fold increase in the urinary output of epoxygenase metabolites. These results, in conjunction with published evidence demonstrating the potent biological activities of its metabolites, suggest a role for the epoxygenase in the renal response to dietary salt.

Rates of spontaneous exchange of synthetic radiolabeled sterols between lipid vesicles.

14C-labeled sterols with structural variation in the polar function [3 alpha-OH, 3-O(CH2)2O-(CH2)2O(CH2)2OH, 3 alpha-NH2, 3 beta-NH2, and 3-OC(O)CHN = N] and at the 7 position (7-oxo, 7 alpha-OH, and 7 beta-OH) were synthesized and incorporated into unilamellar vesicles for studies of the rates of transfer to an excess of acceptor vesicles. Cholesterol, cholestanol, and epicholesterol underwent full exchange in a single kinetic pool, and 90% of the 3 alpha-triethoxycholesterol was exchangeable in one pool. Biphasic kinetics with full exchangeability were observed for cholesterylamines, which bear a positive charge at the 3 position; the slow phase reflects the high activation energy for inner-to-outer leaflet movement of the charged lipid. Biphasic kinetics were also found for cholesteryl diazoacetate, indicating that this photoaffinity probe and cholesterol have different mechanisms of transfer. Sterols that are more hydrophilic than cholesterol as estimated by reversed-phase high-performance chromatography (elution with acetonitrile-2-propanol, 4:1 v/v, with varying proportions of water) gave faster exchange rates than cholesterol, whereas sterols that are more hydrophobic gave slower exchange rates. However, the rates of [14C]sterol desorption from the lipid-water interface are not correlated with the relative sterol hydrophobicity as estimated by the logarithm of the capacity factors using acetonitrile-2-propanol-water as the mobile phase. These studies suggest that the interaction of sterols with phospholipids provides the principal physical-chemical basis for determining the rates of spontaneous exchange of sterols between bilayers.

Conserved terminal sequences of rice ragged stunt virus genomic RNA.

The 5'- and 3'-terminal nucleotide sequences of the dsRNA genome segments of rice ragged stunt virus (RRSV), a member of the plant Reoviridae, were determined and compared with those published for other viruses in this family. The 5'- and 3'-terminal regions of the RRSV plus strand RNA from all genome segments were found to have the same conserved hexanucleotide (5' GAUAAA---) and tetranucleotide (---GUGC 3') sequences, respectively. These conserved terminal sequences were different from those found in viruses in the Phytoreovirus and Fijivirus genera. This result confirms that, as already suggested, RRSV should be placed in a third genus.
