Mutations in BMPR-IB and BMP-15 genes are associated with litter size in Small Tailed Han sheep (Ovis aries).

The Small Tailed Han is a prolific local sheep breed in China. The bone morphogenetic protein receptor IB (BMPR-IB) gene, which affects the fecundity of Booroola Merino sheep, and the bone morphogenetic protein 15 (BMP-15) gene, which affects the fecundity of Inverdale, Hanna, Belclare, Cambridge, and Lacaune sheep, were studied as candidate genes associated with the prolificacy of Small Tailed Han sheep. Single nucleotide polymorphisms of BMPR-IB and BMP-15 genes were detected in Small Tailed Han ewes (n = 188) by PCR-RFLP. The combined effect of the 2 genes on the prolificacy of Small Tailed Han sheep was studied. The results indicated that the same FecB mutation (Q249R) occurred in the BMPR-IB gene in Small Tailed Han ewes as found in Booroola Merino ewes. The Small Tailed Han ewes with genotypes FecB(B)/FecB(B) and FecB(B)/FecB(+) had 1.40 (P < 0.01) and 1.11 (P < 0.01) more lambs, respectively, than those with genotype FecB(+)/FecB(+). The same FecX(G) mutation (Q239Ter) of the BMP-15 gene was found in Small Tailed Han ewes as in Belclare and Cambridge ewes. The Small Tailed Han ewes with the heterozygous mutant FecX(G)/FecX(+) had 0.55 (P < 0.01) more lambs than those with the wild-type FecX(+)/FecX(+). The Small Tailed Han ewes carrying mutations in both BMPR-IB and BMP-15 genes had greater litter size than those with either mutation alone. In view of our results, marker-assisted selection using both BMPR-IB and BMP-15 genes is warranted to increase litter size in sheep and will be of considerable economic value to sheep producers.

Association between expression of reproductive seasonality and alleles of melatonin receptor 1A in goats.

To determine whether a link exists between reproductive seasonality and the structure of the melatonin receptor 1A (MTNR1A) gene, the latter was studied in year-round estrous breeds (Jining Grey and Boer goats) and seasonal estrous breeds (Liaoning Cashmere, Inner Mongolia Cashmere, Wendeng milk and Beijing native goats). A large fragment of exon 2 of MTNR1A gene was amplified by PCR using sheep sense and antisense primers in 260 does of six breeds. The uniform 824 bp PCR product was digested with restriction endonucleases MnII and RsaI, and checked for the presence of restriction sites. No polymorphism at the MnII cleavage sites was detected in all six goat breeds and no relationship could be established between the MnII cleavage sites of MTNR1A gene and reproductive seasonality in goats. For polymorphic RsaI cleavage site at base position 53, only genotype RR (267 bp/267 bp) was detected in Jining Grey goats, both genotype RR and genotype Rr (267 bp/320 bp) were found in all other goat breeds, no genotype rr (320 bp/320 bp) was detected in all six goat breeds. Frequency of genotype RR was obviously higher, and frequency of genotype Rr was obviously lower in year-round estrous goat breeds than in seasonal estrous goat breeds. Sequencing revealed one mutation (G52A) in genotype Rr compared with genotype RR. For polymorphic RsaI cleavage site, the differences of genotype distributions were significant (P<0.05) between year-round estrous goat breeds and seasonal estrous goat breeds. These results preliminarily showed an association between genotype RR and year-round estrus in goats, and an association between genotype Rr and seasonal estrus in goats.

Quantitative analysis of electron field-emission characteristics of individual carbon nanotubes: the importance of the tip structure.

Electron field-emission measurements on individual carbon nanotubes (CNTs) were performed inside the transmission electron microscope (TEM). The field-emission characteristics of CNTs with different tip structures were compared, and their field conversion factor and emission area were studied systematically. It was found that the field-emission characteristics of a CNT depend sensitively on its tip structure, and in particular an opened CNT was shown to be superior to a capped CNT. High-resolution TEM observations revealed that the tip of an opened CNT may, in general, be regarded as being composed of irregular shaped graphitic sheets, and these graphitic sheets have been found to improve dramatically the field-emission characteristics, but the sharp edge may result in larger error in the calculated emission area. The influence of uncertainty in the work function of the CNTs on the field conversion factor and emission area calculation was also investigated.

In situ fabrication and graphitization of amorphous carbon nanowires and their electrical properties.

Individual amorphous carbon nanowires (a-CNWs) were fabricated inside a transmission electron microscope (TEM) by the electron beam induced deposition (EBID) method, and the a-CNWs were graphitized in situ by introducing Fe particles into these a-CNWs and controlled movement of the Fe particles in these CNWs. Detailed structural characterizations and electrical measurements were carried out, and it was found that the current-induced movement of Fe particles has significant effects in purifying the as-fabricated a-CNW, transforming the a-CNW into a graphitized-CNW (g-CNW). Two-terminal current voltage characteristics measurements showed that the g-CNW has a very good electrical conductivity with a resistivity of about 5.3 x 10(-4) Omega cm, a current carrying capacity of at least 4.35 mA, and a current density of 4.6 x 10(8) A/cm(2), and these values are comparable to those of multiwalled carbon nanotubes. Field emission characteristics of both a-CNWs and g-CNWs were also measured, and their respective Fowler-Nordheim plots were found to have basically a linear form.

The roles of different subtypes of opioid receptors in mediating the nucleus submedius opioid-evoked antiallodynia in a neuropathic pain model of rats.

Previous studies have indicated that thalamic nucleus submedius is involved in opioid-mediated antinociception in tail flick test and formalin test. The current study examined the effects of opioids microinjected into the thalamic nucleus submedius on the allodynia developed in neuropathic pain model rats, and determined the roles of different subtypes of opioid receptors in the thalamic nucleus submedius opioid-evoked antiallodynia. The allodynic behaviors induced by L5/L6 spinal nerve ligation were assessed by mechanical (von Frey filaments) and cold (4 degrees C plate) stimuli. Morphine (1.0, 2.5, and 5.0 microg) microinjected into the thalamic nucleus submedius contralateral to the nerve injury paw produced a dose-dependent inhibition of the mechanical and cold allodynia, and these effects were reversed by microinjection of the non-selective opioid receptor antagonist naloxone (1.0 microg) into the same site. Microinjection of endomorphin-1 (5.0 microg), a highly selective mu-opioid receptor agonist, and [D-Ala2, D-Leu5]-enkephalin (10 microg), a delta-/mu-opioid receptor agonist, also inhibited the allodynic behaviors, and these effects were blocked by selective mu-opioid receptor antagonist beta-funaltrexamine hydrochloride (3.75 microg). However, the [D-Ala2, D-Leu5]-enkephalin-evoked antiallodynic effects were not influenced by the selective delta-opioid receptor antagonist naltrindole (5.0 microg). Microinjection of the selective kappa-receptor agonist spiradoline mesylate salt (100 microg) into the thalamic nucleus submedius failed to alter the allodynia induced by spinal nerve ligation. These results suggest that the thalamic nucleus submedius is involved in opioid-evoked antiallodynia which is mediated by mu- but not delta- and kappa-opioid receptor in the neuropathic pain model rats.

MHC haplotypic association in Chinese Han patients with vitiligo.

BACKGROUND: Serological typing of the human leucocyte antigen (HLA) has shown discrepancies in HLA associations with vitiligo in different ethnic populations. OBJECTIVE: To evaluate the distributions of HLA at class I and II loci that may contribute to the genetic susceptibility of vitiligo patients in Chinese Hans population. METHODS: We analysed the allelic frequencies of HLA class I and II by using the polymerase chain reaction sequence-specific primer (PCR-SSP) method in 187 patients with vitiligo and 273 controls in Chinese Hans. The linkage disequilibrium was calculated from a 2 x 2 table. RESULTS: Two-locus haplotypes including HLA-A25-B13, HLA-A25-B27, HLA-A25-Cw*0602, HLA-A25-DQA1*0302, HLA-A25-DQA1*0601, HLA-A25-DQB1*0303, HLA-B13-Cw*0602, HLA-B13-DQA1*0302, HLA-B13-DQA1*0601, HLA-B27-Cw*0602, HLA-B27-DQA1*0302, HLA-B27-DQA1*0601, HLA-B27-DQB1*0303, HLA-B27-DQB1*0503, HLA-Cw*0602-DQA1*0302, HLA-Cw*0602-DQA1*0601, HLA-Cw*0602-DQB1*0303, HLA-Cw*0602-DQB1*0503 and HLA-DQA1*0302-DQB1*0503 were associated with all types of vitiligo in Chinese Hans. The extended haplotypes HLA-A25-B13-Cw*0602, HLA-A25-B27-Cw*0602, HLA-DQA1*0302-DQB1*0303-Cw*0602 and HLA-B13-DQB1*0303-Cw*0602 were found to be associated with all types of vitiligo in Chinese Hans, whereas the frequency of HLA-A25-Cw*0602-DQA1*0302 was significantly increased in generalized vitiligo but not in localized vitiligo. The frequencies of HLA-A25-DQA1*0302-DQB1*0503 and HLA-A30-DQA1*0302-DQB1*0303 were higher in childhood vitiligo than in adult vitiligo, and the frequency of HLA-A25-B13-DQB1*0303-Cw*0602 was shown to be associated with adult vitiligo but not childhood vitiligo. CONCLUSION: This study demonstrates not only the differential association between HLA markers and types of vitiligo according to distribution or age at onset but also newly found high-risk haplotypes in Chinese vitiligo patients.

Molecular detection of disseminated tumor cells in the peripheral blood of patients with gastric cancer: evaluation of their prognostic significance.

Early detection of disseminated tumor cells in the peripheral blood of patients with early stage gastric cancer could help to improve the outcome after tumor resection. The aim of this study is to evaluate the prognostic significance of tumor-related mRNA for the detection of circulating tumor cells in gastric cancer patients by a reverse-transcriptase polymerase chain reaction (RT-PCR) method. We simultaneously analyzed human telomerase reverse transcriptase (hTERT), cytokeratin-19 (CK-19), cytokeratin-20 (CK-20) and carcinoembryonic antigen (CEA) mRNA (messenger RNA) expression in the peripheral blood of 42 gastric cancer patients and 30 healthy individuals. Additionally, analyses were carried out for the correlation of these four molecular markers with patients' clinicopathologic features, as well as the occurrence of postoperative recurrence/metastasis. Among 42 gastric cancer patients, the prevalence of mRNA for hTERT, CK-19, CK-20, and CEA was 61.9% (26/42), 69% (29/42), 61.9% (26/42), and 78.6% (33/42), respectively. All 30 healthy individuals were negative for hTERT and CEA mRNA, while two were positive for either CK-19 mRNA or CK-20 mRNA. Positive CEA mRNA was significantly correlated with tumor size p=0.008), vessel invasion (p=0.001), depth of tumor invasion (p=0.007), lymph node metastasis (p< 0.001), and TNM stage (p<0.001). In addition, the multivariate logistic regression demonstrated that CEA mRNA expression was an independent and significant predictor for postoperative recurrence/metastasis (p=0.032). Our findings suggest that CEA mRNA may be a more reliable marker than hTERT, CK-19 and CK-20 for the detection of circulating cancer cells in gastric cancer patients' peripheral blood. Patients with positive CEA mRNA expression in peripheral blood have a significantly higher risk of postoperative recurrence/metastasis.

Recognising tuberculosis in the lower lung field: an age- and sex-matched controlled study.

OBJECTIVE: Tuberculosis (TB) can sometimes present with consolidation in the lower lung field. This study was conducted to compare the manifestations of lower-lung-field TB (LLFTB) and other pulmonary TB. DESIGN: All new culture-proven TB patients with lower-lung-field consolidation from July 2001 through December 2002 were included. Age- and sex-matched TB patients with upper lung involvement were selected as controls. RESULTS: A total of 79 patients with LLFTB were included. Their mean age was 58.8 years; 46 were male. The clinical, radiographic and laboratory findings were similar in the LLFTB and the control groups, except that the LLFTB patients had less cavitation (P = 0.005). Patients with LLFTB were diagnosed (P = 0.051) and treated (P = 0.001) later than the control patients. The calibres of the trachea and both main bronchi were significantly smaller in the LLFTB group (P < 0.001). More patients with LLFTB developed segmental or lobar atelectasis during follow-up (P = 0.028). CONCLUSIONS: The manifestations of LLFTB are non-specific. The lower-lung involvement, the lower incidence of cavitation and the higher probability of segmental or lobar atelectasis implied that LLFTB was primary TB. A small bronchial calibre probably contributed to its development.

Efficient diode-pumped actively Q-switched Nd:YAG/BaWO4 intracavity Raman laser.

Barium tungstate (BaWO4) is employed to achieve efficient stimulated Raman scattering conversion in a compact diode-pumped actively Q-switched Nd:YAG laser. With an incident pump power of 9.2 W, 1.56 W of 1181 nm first-Stokes average output power was generated at a pulse repetition rate of 20 kHz, corresponding to an optical-to-optical conversion efficiency of 16.9%.

Natural attenuation of MTBE at two petroleum-hydrocarbon spill sites.

Methyl tert-butyl ether (MTBE) has been used as a gasoline additive to improve the combustion efficiency and to replace lead since 1978. Because it is widely used and it has been disposed inappropriately, MTBE has become a prevalent groundwater contaminant worldwide. In this study, two petroleum-hydrocarbon contaminated sites (Sites A and B) were selected to evaluate the occurrence and effectiveness of natural attenuation of MTBE at these two sites. Field investigation results indicate that the natural attenuation mechanisms of MTBE at both sites were occurring with the first-order attenuation rates of 0.0021 and 0.0048 1day(-1) at Sites A and B, respectively. Results also reveal that the intrinsic biodegradation pattern was the most important mechanism among the natural attenuation processes at both sites. Results from BIOSCREEN simulation suggest that biodegradation was responsible for 78 and 59% of MTBE mass reduction at Sites A and B, respectively. Investigation results show that MTBE plume at Site B could be effectively controlled via natural attenuation processes. However, MTBE plume at Site A has migrated to a farther downgradient area and passed the boundary line of the site. Thus, more active groundwater remedial technologies should be applied at Site A to protect the downgradient environment. Results from this study suggest that natural attenuation might be feasible to be used as a remedial option for the remediation of MTBE-contaminated site on the premise that (1) detailed site characterization has been conducted and (2) the occurrence and effectiveness of natural attenuation processes have been confirmed.

Electron field emission characteristics and field evaporation of a single carbon nanotube.

Direct transmission electron microscope (TEM) observations of the field emission and evaporation process of emitting carbon nanotubes (CNTs) shown that the tip structure of the CNT is in general composed of irregular shaped graphitic sheets which extend typically more than 10 nm from the end of the CNT. It is found that the irregular shaped graphitic sheets at the tip of the CNT may greatly enhance the field emission characteristics of the CNT when compared with that having an ideal circular edge. The field evaporation of the CNT proceeds typically via the removal of the irregular shaped graphitic sheets from the tip of the CNT, and field emission characteristics of a CNT depend far more sensitively on the tip structure than on the geometric length of the CNT.

Factors changing the manifestation of pulmonary tuberculosis.

OBJECTIVE: To investigate factors altering the manifestation and outcome of pulmonary tuberculosis (PTB). DESIGN: The medical records of culture-proven TB patients from July 2001 through December 2002 were reviewed. RESULTS: A total of 461 patients were identified. Diabetes (21.5%) and malignancy (15.2%) were the most common underlying comorbidities. Sixteen cancer patients were initially misdiagnosed as pulmonary metastasis or recurrence. Younger patients frequently had constitutional symptoms and haemoptysis, while older patients frequently had respiratory symptoms and pleural effusion. Male predominance was noted, except in the 21-40-year-old non-diabetic group. Diabetic patients were more likely to present with cavitary nodules. Lower lung field TB was noted in 96 (20.8%) patients, and was associated with female sex and consolidative pattern. Factors associated with poor prognosis included underlying malignancy, chronic renal failure, serum albumin <35 g/l, and need for intensive care. CONCLUSION: Age influenced the manifestation of PTB. Diabetes mellitus was associated with a higher probability of cavitary nodules. TB should be considered in 21-40-year-old women with lower lung field consolidation. In cancer patients, pulmonary lesions should not be straightforwardly considered as metastasis or recurrence. A hybrid anaerobic solid-liquid system for food waste digestion.

A hybrid anaerobic solid - liquid (HASL) system was developed to enhance food waste bioconversion in comparison with the conventional two-phase anaerobic digester. The advantages of the HASL system were the higher efficiency of methane production and smaller volume of effluent from the system. The biogas, which was generated from the methanogenic phase, had an average methane content of 71-72%. Total removal of volatile solids consisted of 78-80%. The HASL system can be operated in both batch and semi-continuous modes with satisfactory performance. The addition of a submerged biofilter for ammonia removal to the HASL system further enhanced the performance of anaerobic digestion. Methane production in the enhanced HASL system was increased by 26% in comparison with the HASL system without submerged filter. This paper describes the development of the enhanced HASL system for anaerobic treatment of food waste.

Transient in vivo gene delivery to the silkworm Bombyx mori by EGT-null recombinant AcNPV using EGFP as a reporter.

Several strains of silkworm Bombyx mori were tested for the gene delivery feasibility of Autographa californica nucleopolyhedrovirus (AcNPV) in vivo. In contrast to the general belief that silkworms were non-permissive to AcNPV, we found that 3 of 7 tested strains were AcNPV permissive. To dispel the physiological influence of the ecdysteroid UDP-glucosyltransferase (EGT) on the silkworm, we modified the AcNPV bacmid by disruption of that gene. Expression pattern of EGFP in tissues of silkworm larvae after injection of EGT-null AcNPV vector carrying EGFP cassette was revealed by green fluorescence and Western blot analysis. Viral DNA was detected and semi-quantified in various kinds of tissues by dot blot assay. Active recombinant virus from larval hemolymph was detectable by TCID(50). Our results indicate that some strains of silkworm were permissive to AcNPV, which could serve as a novel gene deliver tool to silkworm in vivo.

Performing probe experiments in the SEM.

A four nanoprobe system has been installed inside a FEI XL30 F scanning electron microscope (SEM), and shown to be fully compatible with the normal functions of the SEM and also a Gatan cold stage (model C1003, -185-400 degrees C). With some selected examples of applications, we have shown that this nanoprobe system may be used effectively for gripping, moving and manipulating nanoobjects, e.g. carbon nanotubes, setting up electric contacts for electronic measurements, tailoring the structure of the nanoobject by cutting, etc. and even for making unexpected nanostructures, e.g. a nanohook. Applications in other areas have also been speculated, limitations or disadvantages of the current design of the probe system were discussed, and methods for possible improvement were suggested.

Association between PCR-SSCP of growth differentiation factor 9 gene and high prolificacy in Small Tail Han sheep.

Small Tail Han sheep that has significant characteristics of high prolificacy and nonseasonal ovulatory activity is an excellent local sheep breed in P.R. China. The lambing percentage averaged 260% in Small Tail Han sheep. Growth differentiation factor 9 (GDF9) gene, which was essential for growth and differentiation of early ovarian follicles, was considered as a possible candidate gene for litter size in Small Tail Han sheep. The genetic polymorphism of a part of the GDF9 gene was detected in 130 ewes of Small Tail Han sheep by PCR-SSCP. The results indicated that there were two genotypes (AA and AB) detected by two primer pairs. In both exon 1 and exon 2 of the GDF9 gene in Small Tail Han sheep, frequencies of AA genotype were 0.846 and 0.908, frequencies of AB genotype were 0.154 and 0.092, frequencies of A allele were 0.923 and 0.954, and frequencies of B allele were 0.077 and 0.046, respectively. The results of chi2 fitness test indicated that both exon 1 and exon 2 of the GDF9 gene were in Hardy-Weinberg equilibrium (p > 0.05) in Small Tail Han sheep. Least squares means of litter size in the first and the second parity for genotype AA were 0.30 (p <0.05) and 0.77 (p <0.0001) more than those for genotype AB detected in exon 1 of the GDF9 gene in Small Tail Han sheep, respectively. Fragments detected in exon 2 of the GDF9 gene had no significant effect (p > 0.05) on litter size in both the first and the second parity in Small Tail Han sheep. Litter size in sheep is lowly heritable, expressed only in females, and manifested relatively late in life. Access to genetic markers would thus be advantageous in selection programs.

Sodium chloride-induced acute and chronic pulmonary hypertension syndrome in broiler chickens.

Two hundred forty 1-d-old Arbor Acres commercial broiler chicks were divided into control and experimental (T1 and T2) groups that, between 8 and 42 d of age, were provided drinking water containing 0, 600, or 1,200 mg/L sodium from sodium chloride, respectively. The pulmonary hypertension syndrome (PHS) incidence and the right to total ventricle weight ratio (RV/ TV) were calculated weekly, and blood samples and lung tissues were collected weekly from 10 birds per group to evaluate the structural and hemodynamic characteristics of pulmonary vessels. Saline drinking water significantly increased the incidence of PHS and RV/TV ratios. In the T2 group the PHS mortality exhibited 2 peaks, including an acute peak from 14 to 21 d of age and a chronic peak from 35 to 42 d of age. During the acute peak of PHS mortality the blood volume (BV), filtration index (FI), and packed cell volume (PCV) increased in groups T1 and T2 when compared with the control group. During the acute peak there were no differences among groups in the ratio of wall to total area (WA/TA), medial thickness of pulmonary arteriole walls (mMTPA), the percentage of thick-walled peripheral lung vessels (%TWPV), the percentage of muscular arterioles (%MA), or the percentage of nonmuscular arterioles (%NMA) in pulmonary arterioles. During the chronic peak of PHS mortality, group T2 exhibited the highest values for %TWPV, %MA, WA/TA, and mMTPA and the lowest values for %NMA when compared with the T1 and control groups. Also during the chronic peak the groups did not differ in BV or FI, whereas PCV remained elevated above control values in groups T1 and T2. These observations indicate that hemodynamic changes related to viscous resistance to blood flow (BV, FI, PCV) predominated throughout the acute peak of PHS mortality, whereas, during the chronic stages of PHS mortality, increased vascular resistance to blood flow also was imposed by remodeling of the pulmonary vasculature.

Iron-mediated removal of ammonium from strong nitrogenous wastewater from food processing.

The combination of microbial reduction and further microbial oxidation of iron was applied to the treatment of food-processing wastewater and recovery of ammonium. Fe2+ ions were formed by iron-reducing bacteria under anaerobic conditions. Ammonium was recovered by co-precipitation with negatively charged iron hydroxides produced during oxidation of Fe2+ by iron-oxidizing bacteria under microaerophilic conditions. The value-added by-product of this process can be used as a slowly released ammonium fertilizer.
