[Effect of catecholaminergic agonists and antagonists on adaptive gastric mucosal protection in rats].

The gastric mucosal damage caused by 70% ethanol significantly decreased in rats pretreated with chronic mild restraint stress (CMRS, P < 0.001). This adaptive mucosal protection induced by CMRS was absent in sympathectomized rats, while the administration of isoprenaline or dopamine could partially, and norepinephrine could not, restore the protection. In rats with rntact sympathetic nerve, preinjection of haloperidol or propranolol inhibited CMRS-induced protection which, however, was not affected by phentolamine. No change in plasma somatostatin level was observed either in stress or sympathectomized rats alone or in stress plus sympathectomized rats. The results suggest that the sympathetic nerve or its mimetic agent ang this is are involved in the adaptive protection of the gastric mucosa possibly mediated through beta and dopamine receptors.

[Protective effects of 15-methyl-prostaglandin F2 alpha on primary cultured rat hepatocyte against CCl4-induced injury].

The effect of 15-Mt-PGF2 alpha on CCl4-induced injury of primary cultured hepatocytes was studied. 15-Mt-PGF2 alpha treatment (2 mg/L) significantly decreased CCl4 (10 mmol/L)-induced damages of primary cultured rat heptocytes as indicated by decreases GPT and GOT leakage and LPO production. 15-Mt-PGF2 alpha significantly promoted 3H-uridine incorporation into RNA and [3H]-thymidine incorporation into DNA of the rat hepatocytes. Cytopathology study showed that 15-Mt-PGF2 alpha attenuated damages of mitochondria, endoplasmic reticulum and ribosome caused by CCl4. 15-Mt-PGF2 alpha appeared to maintain the stability of rat hepatocytes by inhibiting lipid peroxidation. These results indicated that 15-Mt-PGF2 alpha has notable protective effect on primary cultured rat hepatocytes against CCl4-induced damage by reducing lipid peroxidation and promoting synthesis of RNA and DNA.

[Antagonistic effect of calcitonin gene related peptide on arrhythmia in rats].

Premature ventricular contraction (PVC), ventricular tachycardia (VT) and ventricular fibrillation (VF) were developed by endothelin-1 (ET-1) injected into the coronary ostia at the dose of 900 pmol/kg in anesthetized rats. No arrhythmia was elicited but blood pressure fell temporarily by calcitonin gene related peptide (CGRP), the injection into the coronary ostia. After pretreatment with CGRP incidence and severity of arrhythmia decreased at the same ET-1 dose. Arrhythmia score in CGRP 1200 pmol/kg + ET-1 group was lower than that in ET-1 group (P < 0.01). The results revealed that antiarrhythmic effect of CGRP may be partially brought about by its antagonistic effect against ET-1 induced arrhythmia.

[Effect of traditional Chinese medicine treatment on levels of blocking antibody, beta-hCG, progesterone and estradiol in spontaneous abortion].

OBJECTIVE: To explore the effect of traditional Chinese medicine (TCM) on abnormal maternal-fetal immune and endocrine in 24 cases of threatened abortion (TA) and 68 cases of recurrent spontaneous abortion (RSA). METHODS: The levels of blocking effect (BE), antiidiotype antibody (AIA), cytotoxin antibody (CTA), beta-human chorionic gonadotropin (beta-hCG), progesterone (P), estradiol (E2) were measured in all 92 patients. RESULTS: Incidence rates of the lack of BE, AIA and CTA in 92 cases were 60.87%, 57.61% and 72.83% respectively. The lower levels of BE, AIA and CTA were elevated significantly (P < 0.05-0.01), and the serial levels of beta-hCG, P and E2 were markedly increasing (P < 0.01) in all successful pregnant patients. The rate of the successful pregnancy of 92 cases was 89.13% (to TA 91.67%, to RSA 88.24% respectively). CONCLUSIONS: It suggests that the lack of blocking antibody (BA) and endocrine hormone deficiency may result in spontaneous abortion during early pregnancy. The therapeutic mechanism of those herbs in treating spontaneous abortion was associated with the growing levels of BA and hormone by regulating the maternal-fetal immunity and endocrine.

[The effect of GM-CSF on endothelin-1 and endothelin converting enzyme gene expression in human airway smooth muscle cells with or without theophylline incubation].

OBJECTIVE: To explore the role of airway smooth muscle cells (ASMC) stimulated by inflammatory factors in the development of asthma and bronchial hyperresponsiveness (BHR) and the possible mechanism of low dose theophylline in the treatment of airway inflammation, the effects of GM-CSF on endothelin-1 and endothelin converting enzyme (ECE) gene expression in human ASMC and theophylline on their expressions were investigated. METHOD: The cultured human ASMCs were treated with GM-CSF (1,000 micrograms/ml), GM-CSF plus theophylline (125 ng/ml) respectively for 8 hours. Endothelin-1 and ECE gene expresion were measured by RT-PCR. RESULT: There was no ET-1 gene expression in control group, but strong expression in GM-CSF treated group; theophylline entirely inhibited GM-CSF induced ET-1 gene expression in the ASMC; no significant difference of ECE gene expression was found in three groups. CONCLUSION: GM-CSF causes a strong ET-1 gene expression but not ECE gene expression in cultured human airway smooth muscle cells; theophyllin inhibites this abnormal expression which may be one of the anti-inflammatory mechanisms of low dose theophylline in the management of chronic airway inflammation.

[OKT3 treatment of refractory renal allograft rejection].

Between January 1993 and June 1996, 44 episodes of refractory renal allograft rejection were treated with OKT3 to understand the curative effect of OKT3. Six of 7 accelerated and 31 of 37 acute rejection episodes were reversed. The reverse rate was 84.6%. Immediate response to OKT3 was in 25 and delayed response in 12 patients. The mean reverse time was 7 +/- 4 days in immediate response and 34 +/- 3 days in delayed response (P < 0.01). It was claimed that OKT3 had high effect in the treatment of refractory renal allograft rejection. Cytomegalovirus infection and cytokins release syndrome were the main side effect of treatment with OKT3. The delayed response would be associated with cytokins release syndrome and damage of rejection. Patients with lower titre of anti-OKT3 antibodies could be retreated with OKT3.

[Treatment of patients with dislocation of acromio-clavicular joint using kuntsher nail with steel wire: report of 27 cases].

There has been much controversy about ways of operation of the traumatic dislocation of acromio-clavicular joint. Our studies revealed that internal fixation of acromio-clavicular joint using kuntsher nail with steel wire is superior to traditional internal fixation of crossing kuntsher nail. We treated 27 cases in this way who were followed-up for 22 months to 9 years. Excellent result was achieved in 25 cases (92.6%) and good in 2 (7.4%). No complication was observed. We conclude that the way of internal fixation using kuntsher nail with steel wire to treat the dislocation of acromio-clavicular joint has following advantages: (1) simple operation; (2) minimum injury to articular surface; (3) stable internal fixation; and (4) no side effect.

[Early corneal limbal autograft transplantation after alkali burns].

OBJECTIVES: To understand the clinical significance of corneal limbus transplantation on alkali burns. METHODS: 14 New Zealand rabbits were used, 1.0 mol/L NaOH solution was placed on the anterior surface of the eyes for 1 minute. Limbal autograft transplantation was performed on 7 rabbits immediately after the burn. The other 7 rabbits were used as controls. The follow-up period lasted for 1 month and then the histopathological changes of corneas were observed under light and scanning electron microscopes. RESULTS: Faster corneal epithelial healing, less corneal neovascularization and inflammatory cell infiltration occurred in the operative group. CONCLUSIONS: Early limbal autograft transplantation can accelerate corneal epithelial healing, restrain corneal neovascularization and inflammatory response, therefore it can prevent corneal ulceration.

[A comparative study of autologous facial nerve graft in situ with the vein tubulation: the observation of neurons changes].

For further study of the effects of the repairment of facial nerve with vein tubulation, we used horseradish peroxidase (HRP) retrograde mark method to observe the changes of the labeled cells in facial neucleus after the tubulation of rat facial nerve. And, we compared the changes with nerve graft in situ and the normal group. The results showed: 1. The vein tubulatiton can repair the short defect of facial nerve as well as the nerve graft does. 2. Irregular outgrowth of axon sprouts could be found in both methods. 3. There was few chance for homolateral target organ to be reinnervated by contralateral motoneurons in both methods. 4. Using the vein tubulation method, a more natural relationship between the peripheral target organ and central nucleus may be easily achieved.

[Observations on before and after employ of screw of implant denture with SEM].

The loss of screw is one of the clinic complication in MDIC implant denture cases. The purpose of this study is to find the cause of loss of screw. The authors has studied MDIC implant denture with SEM before and after employ of the screw and found that the causes of screw loss were crevice corrosion and friction pressure. Crevice corrosion may promote the loss of screw. Base the above condition, it is necessary to select Ti-alloy screw and associat with anticorrosive measure in MDIC implant denture construction.

[Video image analysis of AgNORs distribution in the testicular tumor].

For the purpose of exploring the relationship between AgNORs and the biological behaviour of the testicular tumor, the value of enumeration and image analysis of AgNORs has been investigated in 10 normal testicular tissues and 49 cases of testicular tumors. The major findings were: AgNORs value for number, area, mean area, large partical number (LPN) and roundness in normal, benign and malignant tumors had significant difference (P < 0.01). The former four parameters turned larger when the tumor became more malignant, and they were significantly higher in the good prognosis group than in the bad one. LPN was higher in stage III than in stage I and II (P < 0.05), no difference was found between stage I and II. Other parameters had no significant difference in each stage. These results indicate that AgNORs is helpfull in differentiating the benign and malignant testicular tumors, in determining the degree of malignancy and offering an additional prognostic indicator in testicular tumors. Partical numbers, area and LPN were more helpfull. It had no important value in differenciating each stage.

[Nucleolar organizer regions enumeration in testicular teratoma].

To differenciate the benign and malignant testis teratoma. We investigated the value of AgNORs enumeration in 10 normal testis tissues, 6 benign teratomas and 7 malignant teratomas. The shape of the AgNORs in normal and benign teratoma was round, regular, with clear boundary and even size, setting in the center or the margin of the nucleus. In the malignant teratoma, the shape was irregular with unclear boundary and uneven size, setting in the near center or scattered in the nucleus. AgNORs enumeration: normal group: 1.56 +/- 0.17, benign teratoma: 2.40 +/- 0.26, malignant teratoma: 5.24 +/- 0.36. There was significant difference between the two groups (P < 0.01). The range of AgNORs enumeration in benign and malignant teratoma was separate. These results indicate that the mathod is helpful in differentiating benign and malignant testicular teratoma, and may provide an objective parameter.

[The effects of Tripterygium wilfordii polyglycosidium on neuromuscular junctions of adult toad].

The effects of tripterygium wilfordii polyglycosidium (TWP) on the transmission at the neuromuscular junctions (NMJ) of sciatic nerve-sartorius muscle preparations of 20 adult toads (Bufo bufo gargarizans) in vitro were studied by means of intracellular microelectrode recording of endplate potentials (EPP). The results revealed that TWP could cause changes of EPP amplitude so as to influence the transmission at NMJ, and the effects apparently depended on the concentration of the drug. A proper concentration of TWP could produce a significant increase in EPP amplitude and therefore facilitate the transmission at the NMJ.

[Predictive chemotherapy of breast cancer directed by MTT assay in vitro].

OBJECTIVE: The predictive value of in vitro MTT assay for chemotherapy to breast cancer patients was studied. METHODS: Of the 156 advanced breast cancer patients, 83 had had MTT assay before chemotherapy. According to the result of the MTT assay, 73 patients in MTT sensitive group received chemotherapy. The other 10 patients in MTT resistant group and 73 patients in the control group were given chemotherapy according to clinician' experience. RESULTS: The response rate in MTT sensitive group was 76.7% (56/73). There was statistically significant difference as compared with 0(0/10) in the resistant group and 43.8% (32/73) in the control group. Between MTT sensitive group and control group, similar results were obtained at different therapeutic phases, in the subgroups of most lesions and chemotherapeutic schemes. Both the relapse rate and mortality rate were reduced remarkably in the MTT sensitive group. But there was no difference in the median remission and survival of relapsed patients in the two groups. Between in vitro and in vivo, the overall coincident rate was 79.5% [(56 + 10)/83]. CONCLUSION: The in vitro MTT assay can predict chemotherapy for breast cancer giving satisfactory results.
