Suppression of depleted uranium-induced neoplastic transformation of human cells by the phenyl fatty acid, phenyl acetate: chemoprevention by targeting the p21RAS protein pathway.

Depleted uranium is a dense heavy metal used primarily in military applications. Published data from our laboratory have demonstrated that exposure to depleted uranium in vitro can transform immortalized human osteoblast (HOS) cells to the tumorigenic phenotype (associated with aberrant RAS oncogene expression and tumor suppressor protein production). Since depleted uranium is used in military applications, it would therefore be beneficial to identify and test potential antitumor-promoting agents. Chemopreventive interventions that target deregulated signal transduction pathways may be effective strategies to prevent carcinogenesis. Since the RAS protein plays a key role in signal transduction, disruption of its signaling pathway may be particularly effective. The phenyl fatty acid, phenyl acetate, a differentiation inducer that affects post-translational processing of RAS, was tested for its ability to prevent depleted uranium-induced neoplastic transformation using HOS cells. After a 24-h exposure to insoluble depleted uranium-uranium dioxide (1 mg/ml), cells were incubated for 1 day to 6 weeks with 2.5 mM phenyl acetate. Treatment with depleted uranium resulted in transformation to the tumorigenic phenotype. In contrast, HOS cells exposed to depleted uranium and then treated with phenyl acetate did not exhibit transformation to the tumorigenic phenotype. These data suggest that depleted uranium-induced neoplastic transformation in vitro can be prevented by targeting the RAS protein.

Molecular cloning of the mouse homologue of Rab3c.

Small GTP-binding proteins of the Rab subfamily are key regulators of intracellular vesicle transport. Here we report the isolation of a cDNA clone encoding the complete Rab3c isoform from mouse embryo using a degenerative PCR-based approach. Multiple sequence alignment revealed that the predicted amino acid sequence was identical to the previously identified rat Rab3c isoform and 98% identical to the published bovine Rab3c GTPase from brain. Furthermore by in situ hybridisation, Rab3c mRNA was detectable within various regions of the brain, cartilage and highly enriched within intestinal villi of foetal tissues. Chondrocytes in the hypertrophic zone, but not reserve or proliferative zones, expressed high levels of Rab3c. This pattern of expression corresponds with the genesis of matrix vesicles during endochondral ossification. In all, our results suggest that in addition to its functional role during regulated secretion in brain, Rab3c may play a part in matrix vesicle trafficking during skeletal development.

The prohormone convertase enzyme 2 (PC2) is essential for processing pro-islet amyloid polypeptide at the NH2-terminal cleavage site.

Impaired processing of pro-islet amyloid polypeptide (proIAPP), the precursor of the beta-cell peptide islet amyloid polypeptide (IAPP) (amylin), has been implicated in islet amyloid formation in type 2 diabetes. The prohormone convertase enzymes PC3 (also known as PC1) and PC2 are localized to beta-cell secretory granules with proIAPP and proinsulin and are responsible for proinsulin processing. To determine whether PC2 might be essential for proIAPP processing, we performed Western blot analysis of freshly isolated islets from normal mice and mice lacking active PC2. As expected, the primary species of IAPP immunoreactivity in islets from wild-type mice was fully processed (4-kDa) IAPP, with only small amounts of the 8-kDa precursor (unprocessed proIAPP) present. Islets from heterozygous PC2 null mice were identical to wild-type animals, suggesting that half the normal complement of PC2 is sufficient for normal proIAPP processing. By contrast, in islets from homozygous PC2 null mice, the predominant IAPP-immunoreactive form was of intermediate size (approximately 6 kDa), with no detectable mature IAPP and slightly elevated amounts of the 8-kDa precursor form present. Thus, in the absence of PC2, proIAPP processing appears to be blocked at the level of a proIAPP conversion intermediate. Immunofluorescence of pancreas sections and immunoblotting using antisera raised to the NH2- and COOH-terminal flanking regions of mouse proIAPP demonstrated that the 6-kDa intermediate form was an NH2-terminally extended proIAPP conversion intermediate (processed only at the COOH-terminus). These data indicate that PC2 is essential for processing of proIAPP at the NH2-terminal cleavage site in vivo and that PC3 is likely only capable of processing proIAPP at the COOH-terminal cleavage site.

Control of clonorchiasis by repeated treatments with praziquantel.

The present study aimed to evaluate control efficacy of clonorchiasis by two schemes of repeated treatment with praziquantel at two endemic villages in China. Residents of one village at Guangxi Autonomous Region were treated and examined 6-monthly and of another at Liaoning Province 12-monthly. In residents that took 25 mg/kg x 3 (total 75 mg/kg) of praziquantel every 6 months for one year the egg positive rate showed a significant drop from 69.0% to 17.1%. In contrast, a group of same praziquantel medication once showed a slight marginal decrease in the egg rate from 18.9% to 12.2% after one year. Of 39 subjects examined 3 times, 56.4% were cured, 7.7% persistently positive, one (2.6%) reinfected after cure or newly infected, but 25.6% were persistently negative. The present finding suggests that 6-monthly medication with 75 mg/kg of praziquantel should effectively lower the prevalence but incomplete for control of clonorchiasis in heavy endemic areas.

Clinical application of serial operations with preserving spleen.

AIM: To evaluate the clinical application of serial operations with preservation of spleen. METHODS: Serial operations with preserving spleen were performed on 211 cases in our hospital from 1980 to 2000. The patient's age ranged from 13 to 56 years, averaging 38 years. Diseases included splenic injury in 171 cases, portal hypertension in 9 cases, splenic cyst in 10 cases, and the lesion of pancreatic body and tail in 21 cases. RESULTS: All the cases were cured, and 129 patients were followed up from 3 months to 3 years with the leukocyte phagocytosis test, detection of immunoglubin, CT,(99m)Tc scanning and ultrasonography. The results were satisfactory. CONCLUSION: The operations with preserving spleen were safe, feasible, and worth of clinical application.

[Cloning of cNDAs involved in malignant transformation induced by glycidyl methacrylate].

The malignant transformation of human embryonic lung fibroblasts (HELF) induced by glycidyl methacrylate (GMA) in vitro was used as a model for comparing gene expression between the transformed cells and controls. Using mRNA differential display PCR(DD-PCR) technique, the three cDNA fragments up-regulated in the transformed cells were identified and highly homologous to human breast cancer transcription factor(ZABC1), Tbx3 and mouse ubiquitin conjugating enzyme(E2) gene. These results suggest that activation or inactivation of genes including ZABC1, Tbx3 and E2 may be involved in GMA-induced transformation.

[Effect of methionine supplementation on the selenium bioavailability in rats fed on grains from Keshan disease endemic area].

In order to study the effect of dietary methionine on tissue selenium and glutathione peroxidase activity(GPX) in rats fed on grains from Keshan Disease endemic area, a nonpurified selenium (Se) deficient diet (containing Se 0.007 mg/kg diet) taken from Keshan Disease endemic area was supplemented with selenomethionine (SeMet) to provide selenium content in diet for 0.007, 0.06 and 0.50 mg/kg and added or not added with DL-methionine (Met) 4 g/kg to make 6 kinds of diet. Fifty four male weanling Wistar rats were randomly divided into 6 groups, consuming each one of the 6 diets for 8 weeks. The Se content and GPX activity in tissues of animals with Met supplementation were compared with those without Met supplementation. The results showed that adding Met did not result in significant changes of tissue Se content and GPX activity in animals having dietary Se of 0.007 mg/kg, except for lower muscle Se content. However, in animals having dietary Se of 0.06 mg/kg and supplementing Met resulted in selenium redistribution in tissues-decrease of Se content in muscle, increase of Se content in liver and blood and significant elevation of GPX activity in all tissues. In animals having methionine-added and dietary Se of 0.50 mg/kg, tissue Se content declined to various extent, while GPX activity remained unchanged. The results suggest that SeMet (main chemical form of Se in cereals) is preferentially incorporated into body protein when dietary methionine is limited. Once Met is supplemented, dietary SeMet would provide more Se for the syntheses of GPX and other selenoproteins. The results further suggest that marginal deficiency in sulfur-containing amino acids in the diet from Keshan Disease endemic area might be an additional factor for the development of Keshan Disease under selenium deficiency.

Inducible nitric oxide synthase neutralizes carbamoylating potential of 1,3-bis(2-chloroethyl)-1-nitrosourea in c6 glioma cells.

Expression of iNOS in glioma and other tumors has been extensively documented but the effects of NO derived from iNOS on tumor-killing mechanisms of chemotherapy drugs remain to be fully defined. We note that increased NO synthesis by cytokine exposure or iNOS overexpression neutralized the cytotoxicity of 1,3-bis(2-chloroethyl)-1-nitrosourea (BCNU) and 1-(2-chloroethyl)-3-cyclohexyl-1-nitrosourea (CCNU), but not cisplatin, in rat C6 glioma cells. Suppression of BCNU cytotoxicity associated with iNOS overexpression could be abolished by pharmacological inhibition of NOS or coexpression of an antisense RNA against iNOS. Both BCNU and CCNU are chloroethylnitrosoureas that kill tumor cells via carbamoylating and alkylating actions. Further studies using compounds that each carry these different activities indicate that iNOS neutralized carbamoylating, but not alkylating, action of chloroethylnitrosoureas. Temozolomide, a novel chemotherapy drug recently available for treating brain tumors, carries only alkylating, but not carbamoylating, action. Overexpression of iNOS in C6 cells failed to neutralize temozolomide cytotoxicity. Results from the present study demonstrate the ability of iNOS-derived NO to confer chemoresistance against the carbamoylating potential of chloroethylnitrosoureas in vitro. Further investigation is needed to test whether iNOS expression, frequently noted in malignant brain tumors, also enhances chemoresistance against chloroethylnitrosoureas in vivo.

[Expression of human anti-HBsAg-interferon fusion protein in CHO cells].

OBJECTIVE: To study the possibility of expressing human anti-HBsAg-interferon fusion protein in CHO cells as a putative targeting drug for hepatitis B. METHODS: Both the heavy and light chain genes of human anti-HBsAg antibody derived from a phage display library were fused with alpha-2b interferon (IFN-alpha-2b) gene in vitro by polymerase chain reaction. The IFN alpha-2b gene was placed at the C terminal and a 15 amino acid linker was introduced at the fusion site. The light chain-IFN expression plasmid pLIC was constructed with a mammalian expression vector pcDNA3.1(+) and the heavy chain-IFN expression plasmid pFID was constructed with another mammalian expression vector pCdhfr1. These plasmids and the anti-HBsAg full-length light and heavy chain expression plasmids (pLIC and pHFD, respectively) were transfected into CHO (dhfr-) cells by the following three combinations:I, pLIC + pHFD;II, pLIC + pFID; III, pLFC + pFID. The cultured supernatant of the transfected cells was collected and assayed for IFN activity and HBsAg affinity. RESULTS: The supernatant of combination I and II displayed IFN activity but only combination I supernatant exhibited HBsAg affinity. CONCLUSIONS: The successful expression of a fusion protein with both HBsAg affinity and interferon activity may lead to a new way to make a targeting drug for hepatitis B.

[Anti-oxidation effect of water extract of Flos chrysanthemi on heart and brain in vivo and in vitro].

This paper reported the effect of water extract of Flos Chrysanthemi on lipid per-oxidation (LPO) of heart and cerebral homogenate from rats incubated in 37 +/- 0.5 degrees C, of heart and cerebral homogenate from mice induced by .OH, of heart and cerebral mitochondria member induced by .OH, which was compared with Radix Salviae Miltiorrhizae. The results showed that the inhibition effect of water extract of Flos Chrysanthemi on above-mentioned LPO was equal to Radix Salviae Miltiorrhizae. It was concluded that water extract of Flos Chrysanthemi could restrain the produce of free radicals and LPO induced by free radicals.

[Fluoride emission from different soil minerals at high temperatures].

The emission characteristics of fluoride pollutants from montmorillonite, kaolinite, vermiculite, geothite and allophane were studied to elucidate the mechanism of fluoride-releasing from soils during brick and tile making at high temperatures from 300 degrees C to 1000 degrees C. The rate of fluoride emission varied with temperature, mineral type, heating time, specific surface area and cations added to minerals. The escape of crystalline water resulting from crystal lattice collapse at a certain high temperature was found to affect the rate of fluoride emission. Calcium compounds could decrease fluoride emission rate from montmorillonite. At 800 degrees C, the rate of fluoride emission from Ca-treated montmorillonite decreased by 59.6% compared to untreated montmorillonite. The order for fluoride-fixing capacity of the 5 calcium compounds at 800 degrees C was as follows: CaCO3 > CaO > Ca3(PO4)2 > Ca(OH)2 > CaSO4.

[The apolipoprotein E gene polymorphism in children with steroid-resistant idiopathic nephrotic syndrome].

OBJECTIVE: To study apolipoprotein E gene polymorphism in children with steroid- resistant idiopathic nephrotic syndrome (SRINS). METHODS: Sixty children with SRINS (including 9 children with focalsclerosing glomerulonephritis) were observed in comparison with 80 age-sex-matched healthy children. Seven lipoprote in metabolism parameters including serum total-cholesterol(TC), triglyceride (TG), high-density lipoprotein cholesterol (HDL-C), low-density lipoprotein cholesterol (LDL-C), apolipoprotein AI(apoAI), apolipoprotein B (apoB), lipoprotein (a)[Lp (a)] were detected by enzyme method, and single-strand conformation polymorphism(PCR-SSCP) was used to determine the apoE genotypes in the two groups. RESULTS: Serum TC, TG, HDL-C, LDL-C, apoAI, apoB, Lp(a) in the observation group were significantly higher than those in the healthy controls (P<0.01). After 6 months, the levels of serum TC, TG, LDL-C,apoB,Lp(a) were 81.25%, 78.13%, 84.38%, 87.50% and 78.13% respectively in 32 SRINS cases of the observation group. The apo epsilon 2 allele gene frequence was higher in the observation group than in the controls (11.66% vs 5.00%, P<0.05). The apo apsilon 2 allele gene frequence in the cases of focalsclerosing glomerulonephritis was significantly higher than that in the controls (22.22% vs 5.00%, P<0.01). CONCLUSION: The above results indicate the obvious and long term abnormalities of serum lipoprotein metabolism in the children with SRINS. The apo apsilon 2 allele gene frequence is higher in the children with SRINS. There are risk factors for atherosclerosis and cardiovascular diseases in the children with SRINS. The antilipemic drugs should be considered in use for the children with SRINS.

[Removal of NH4(+)-N from polluted water resources by bioremediation of biofilm process with elastic packing & micropore aerator].

The effect of NH4(+)-N removal in the polluted raw water of water sources by the bioremediation action of the biocontact oxidation process with the elastic packing and micropore aerator was studied in this paper. The research results showed the bioremediation process can reach a removal ratio of 64%-95% for NH4(+)-N of water sources under the condition of ordinary temperature 20 degrees C-27 degrees C, CODMn 7-14 mg/L, NH4(+)-N 0.7-2.0 mg/L in the water sources and the running parameters HRT 1.4 h, gas and water ratio 0.5:1, and DO 7-9 mg/L for the bioremediation process and a removal ratio of 40%-63% for NH4(+)-N of water sources under the condition of low temperature 7 degrees C-12 degrees C, CODMn 6-11 mg/L, NH4(+)-N 1.2-8.0 mg/L in the water sources and the running parameters HRT 1.4 h, gas and water ratio 0.5:1, and DO 8-10 mg/L for the bioremediation process.

[Studies on cloning and expression of hepatitis B virus x gene].

OBJECTIVE: To express and purify the x gene of hepatitis B virus (HBV), and then the purity of the protein could specifically be analysed by the human antibodies against HBV x antigen (protein). METHODS: HBV x gene was amplified from the complete genome by PCR, and cloned in the pGEX-2T fused expression vector. After transforming the plasmid into E.coli, the recombinant with HBx gene was obtained. The fusion protein was generated with E.coli fused expression system and purified with affinity chromatography. RESULTS: The HBx protein could be applied to detecting specific antibody of hepatitis B virus patients. CONCLUSIONS: The purity of the HBx protein can specifically be recognized by the human antibodies against HBV x antigen.

[A retrospective analysis of therapeutic outcome of various types of severe virus hepatitis].

OBJECTIVE: To explore combined therapeutic schemes for severe virus hepatitis and to raise survival rate. METHODS: The therapeutic outcomes of different schemes were analyzed in 1020 cases of severe viral hepatitis treated in our hospital in recent twenty years. The treatments were classified as basic and combination of Chinese traditional and Western medicine therapies in which embryonic liver cell suspend liquid or hepatic growth factor, or/and artificial liver as blood cleaner were also used for the latter. RESULTS: The survival rate was 30.28% (33/109) treated by basic procedures and 60.15% (548/911) by combined procedures. There was obvious statistical difference in survival rate between the two groups (P<0.001). Among all the patients, 219 (21.47%) suffered from acute or sub-acute severe hepatitis, 801 (78.53%) chronic severe hepatitis; 446 started their treatment at the early stage of the disease and the survival rate was 77.13% (344/446); 404 at the mid stage and 170 at the late stage and the survival rates were 52.72% (213/404) and 14.12% (24/170), respectively. The survival rate for the patients treated at the early or the mid stage was much higher than that at the late stage (P<0.001), and had a positive correlation with the activity of prothrombin. CONCLUSION: The combined therapies used in present study are effective to raise the survival rate of the patients with severe virus hepatitis. Early treatment is extremely important for the patients' prognosis.

[EROD induction in liver microsome of carp(Cyprinus carpio) by nitroaromatic hydrocarbons in vitro].

Nine nitroaromatic hydrocarbons (NAHs) induction of EROD activity was investigated by an in vitro assay based on carp (Cyprinus carpio) hepatic microsome system. All nine NAHs tested were found to induce the activity of EROD. There was dose-effect relationship between EROD activity and concentrations of NAHs. It was found that EROD activity was different, when different groups substituted at the same position of benzene ring, or the same group was at different position of benzene ring.

[A research on the quantification of retinal blood vessels in cases of arteriovenous crossing pressure syndrome caused by hypertension].

The method for quantification of retinal blood vessels in cases of arteriovenous crossing pressure syndrome caused by hypertension was studied. The matched filtering pre-processing of image enhancement for local retinal blood vessels and closing of mathematical morphology for image smoothing, and the method of scale-up lineation for contour determination of retinal blood vessels and optimal seeking algorithm with reversible quantification were used to measure the pathological deformation of vessel, thus providing a computer method for auxiliary diagnosis of relative diseases.
