Identification of pelvic lymph nodes with chlorophyllin after injection into the uterine cervix: an experimental and clinical study.

We investigated the value of staining retroperitoneal lymph nodes with chlorophyllin in normal dogs and in women with malignant uterine tumors undergoing lymphadenectomy. In dogs, after 0.3% chlorophyllin (sodium copper chlorophyllin) was injected into the canine uterus, the concentration of dye in the bloodstream was measured with a spectrophotometer and sections of stained retroperitoneal lymph nodes were examined using light and electron microscopy. The highest blood levels were detected at 4 hrs and nearly all of the chlorophyllin was gone from the bloodstream by 18 hrs but was retained in nodal macrophages for at least 4 days. No morphological changes were found in the excised lymph nodes. Twenty-four patients with cervical carcinoma and 20 patients with endometrial carcinoma undergoing radical hysterectomy and lymphadenectomy were divided into a lymphatic coloration group (23 patients) and a non-coloration (control) group (21 patients). In the lymphatic coloration group (0.3% chlorophyllin) was injected into the cervix 5 days before elective lymphadenectomy. There were no complications attributed to injection of the chlorophyllin. The number of dissected lymph nodes in the coloration group were greater than the control group (p<0.01) and the time of operation was shorter (p<0.01). These results suggest that chlorophyllin is safe and facilitates identification of retroperitoneal lymph nodes, allows more complete nodal excision and shortens the time of operation in patients undergoing radical hysterectomy with lymphadenectomy.

[Studies on chemical constituents of mycelium of fungus Cephalosporium sp. AL031(I)].

Five compounds were isolated from the mycelium of the fungus Cephalosporium sp. AL031 whose metabolites have been proven to possess antifungal and antibacterial activities. Based on the spectral data and elemental analysis, they were identified as ergosterol(I), 2,4,6-octatrienoic acid(II), succinic acid(III), uracil(IV), and D-mannitol(V). All of these compounds were obtained from the culture of this fungus for the first time.

[Physiology and development of two Roegneria kamoji populations stressed by copper].

Roegneria kamoji grown copper tailings had weak stem, short height, and thick leave color, while its counterpart on normal soil was the opposite. Exposed to copper, R. kamoji had a decreased chlorophyll content. A certain concentration of copper could promote the seed germination of R. kamoji, and the extract of copper tailings did not decrease the seed germination rate, but deferred the germination. Root growth test indicated that when the concentration of copper was 0.125 mg.L-1, the rooting of R. kamoji was promoted. If the concentration was more than 0.25 mg.L-1, the rooting was inhibited significantly. R. kamoji colonized on normal soil wase more sensitive to copper than that on Cu-polluted soil.

[Set up brain compartment model and simplify CBF-quantification].

In order to simplify the non-invasive quantitative evaluation of cerebral blood flow (CBF), we used computer simulation and adopted the pulse function as the input function of compartment model for calculting the mean retention fraction of 99mTc-ECD in brain from the time activity curves (n = 24). We found that there is a significant correlation between R2 (the retention fraction) from the 2-compartment model and R3 from the 3 compartment model with a correlation coefficient of 0.9836. The CBF by R2 is 29.48 +/- 1.51 ml/min per 100 g, the CBF by R3 is 29.10 +/- 1.48 ml/min per 100 g, and no significant difference between them is seen (P > 0.05, n = 24). These data indicate that the simplified two-compartment model for the mechanism of the uptake of 99mTc-ECD in brain is better. And we found that, compared with the entricular function, the pulse function may be used as a more appropriate input function for the model.

[Studies on lignocellulolytic enzymes production and biomass degradation of Pleurotus sp2 and Trametes gallica in wheat straw cultures].

Pleurotus sp2 and Trametes gallica were selected in this assay because of their high activities of lignocellulolytic enzymes and the enzyme peaks appeared at the early stage of liquid state fermentation. Solid state fermentation was also investigated for their abilities and behaviors of enzyme-production. The capabilities and characteristics of the two strains in degrading biomass were studied. When Pleurotus sp2 was incubated in wheat straw powder containing the liquid medium of low-nitrogen, no-carbon and high inorganic salt, the activities of MnP and Lac reached the peaks on the tenth day, but the activities of hemicellulases reached the peak on the 40th day. Pleurotus sp2 caused 17.6% of biomass loss. When T. gallica was incubated in wheat straw powder containing the liquid medium of hlig-nitrogen, or low-nitrogen, no-carbon and high inorganic salt, the activities of MnP reached the peaks on the tenth day, the lac and hemicelluloses on the 40th day, and the lignin peroxidases reached the peaks on the 50th day, and it caused more than 64% of biomass loss. Among them the hemicellulose was degraded by 71.96%, and the cellulose 66.21%. T. gallica was very capable of degrading lignin of wheat straw and caused 34.37% loss during 20 days, 46. 71% loss during 30 days and 70.14% loss during 60 days. It was interesting that T. gallica degraded lignin preferentially with respect to cellulose, which was very beneficial to biopulping of paper industry.

[Effects of copper mine tailings on growth and development and physiological function of wheat].

Through pot culture experiments, the effects of Tonling copper mine tailings on growth and development and physiological function of wheat were investigated in this paper. The results showed that the seeds of wheat could germinate in copper mine tailings, but seed germination percentage and germination rate were lower than that in control. High ratio tailings decreased seedling height. Tailings decreased chlorophyll contents, but didn't effect on chlorophyll a/b. Compared with control, tailings significantly inhibited root growth and decreased root number. Tailings increased root and shoot biomass and significantly increased root/shoot ratio of wheat. The permeability of cell membrane and the free proline content in leaves of wheat increased with the increase of the ratio of tailings, they were relative tolerance index of wheat to copper tailings stress.

[Selection of optimal ultrasonic extraction process of Elaeagnus angustifolia L. by uniform design].

The influences of ultrasonic frequency, ultrasonic time and solvent content on the extraction rate of fat oil in Elaeagnus angustifolia L. were studied. The optimum extraction condition was obtained. The results showed that the ultrasonic extraction method of fat oil in Elaeagnus angustifolia L. could save time, improve extraction rate and need not be hot compared with traditional extraction method.

[The examination of DNA repair capacity in the patients with oral precancerous lesion].

OBJECTIVE: The occurrence of oral precancerous lesion is related with not only carcinogenic environmental factors but also some self-regulating mechanisms. The genetic heredity factors determine individual's DNA repair capacity. The study is attempted to investigate the change of DNA repair capacity of the patients with the precancerous lesion. METHODS: The study is conducted in 16 pairs of oral precancerous lesion and normal controls by using host cell reactivation assay, while some dangerous factors such as smoking, alcohol intaking and family history of cancers are also recorded. RESULTS: The host cell reactivation in the patients with precancerous lesion is lower than that of normal controls (P < 0.05) and is shown to be an independent risk factor in the occurrence of oral precancerous lesion (OR: 3.25 95% CI = 1.01-10.49). CONCLUSION: The host cell reactivation is likely to be a helpful indicator for predict prognosis of cancer for the individuals with highly genetic sensitivity to cancer.

[High concentration ethanol continuous fermentation using yeast flocs].

Continuous ethanol fermentation using yeast flocs was carried out in 4 air-lift suspended-bed bioreactors operated in series. Drafted by CO2, with complete recycle of ethanol distilled effluent broth and at the dilution rate of 0.2/h, the average ethanol concentration of the fermentation broth was 96.6 g/L, while the average concentration of residual total sugar was 4.1 g/L and residual reducing sugar was 1.2 g/L.

[Cloning and sequence analysis of the phytase phyA gene of Aspergillus niger N25].

The phyA encoding phytase of Aspergillus niger N25 was amplified by the polymerase chain reaction (PCR) with primers designed according to the sequences of the phyA in GenBank. The amplified fragment was cloned and sequenced. The results show that: the coding region is 1506 bp in size, includes a 102 bp intron, and encodes a peptide of 476 amino acid residues, in which there is a signal peptide with 19 amino acids and a mature peptide of 448 amino acids. Comparison of this sequence with the phyA of the natural A. niger NRRL3135 (GenBank Accession: M94550), the most highly secreting-phytase strain, shows that the nucleotide homology is as high as 96.746%, and the amino acid homology comes up to 97.64%. The phyA of A. niger N25 strain in this paper is appropriate to be used to construct the phytase gene-engineering bacteria.

[Study on calcium-cholate-phosphate ternary complexes by FTIR spectroscopy].

The aim of this paper was to examine the calcium-cholate-phosphate tenary complexes by FTIR spectroscopy. The results indicated that the mole ratio of cholate to phosphate were 1:3 and 1:4 in ternary complexes respectively when the initial mole ratio of cholate to phosphate were 3:1 and 1.5:1 in reaction. The FTIR spectroscopic results demonstrated that in the ternary complexes Ca2+ was coordinated with O in both COO- and PO(4)3- groups.

[The clinical significance of detection of urinary ceruloplasmin in type 2 diabetes].

OBJECTIVE: To evaluate the clinical significance of detection of urinary ceruloplasmin (Cp) in type 2 diabetes. METHODS: To measure albumin/creatinine(Alb/Cr) and Cp/Cr in morning urinary specimens, from 134 normal controls and 3 960 diabetes, with RIA and ELISA separately. RESULTS: (1) The range of urinary Cp/Cr for normal controls was 0.01 - 1.12 ng/mmol, median 0.36 ng/mmol. Cp/Cr > 0.924 ng/mmol was considered as abnormal, and there was no significant difference between males and females, as well as among subjects of different age. (2) The sensitivity and specificity of Cp/Cr were 90% and 66% respectively with Alb/Cr > 2.5 mg/mmol as a golden standard for diabetic nephropathy, and Kappa value 0.48. Urinary Cp/Cr correlated well with Alb/Cr. (3)After sub-maximal exercise, Cp/Cr elevated markedly with no significant change of Alb/Cr. CONCLUSION: Urinary Cp/Cr could be considered as a marker of diabetic nephropathy and might be more sensitive than Alb/Cr.

Effect of subthalamic nucleus stimulation on levodopa-induced dyskinesia in Parkinson's disease.

Article abstract-The authors studied the effect of bilateral subthalamic nucleus stimulation on levodopa-induced dyskinesias in 24 consecutive parkinsonian patients with disabling dyskinesias. The improvement in the three subtypes of levodopa-induced dyskinesias was significant from the third postoperative month and was mainly due to the decrease in the daily dose of levodopa allowed by the stimulation-induced improvement in the motor score.

The pachytene checkpoint in Saccharomyces cerevisiae requires the Sum1 transcriptional repressor.

Saccharomyces cerevisiae mutants that fail to complete meiotic recombination are blocked by the RAD17/RAD24/MEC1 checkpoint signaling pathway in pachytene when early sporulation genes are expressed. Middle genes are not activated in checkpoint-arrested cells because the Ndt80 transcription factor is inhibited. We find that the pachytene checkpoint requires Sum1, a transcriptional repressor that recognizes a subset of Ndt80-binding sites. Mutants lacking Sum1 or Rad17 partially bypass the block to the nuclear divisions but do not form spores, while mutants lacking both Sum1 and Rad17 completely bypass the block and form morphologically normal spores. The level of Sum1 protein decreases as middle genes are expressed, and this decrease is blocked in checkpoint-arrested cells. These data suggest that Sum1 levels are regulated by the checkpoint and that progression of the meiotic divisions and spore differentiation can be differentially controlled by competition of the Sum1 repressor and Ndt80 activator for occupancy at key middle promoters.

Adeno-associated virus production of soluble tumor necrosis factor receptor neutralizes tumor necrosis factor alpha and reduces arthritis.

The major limitation of adenovirus is its association with induction of an inflammatory response and relatively short-term production of the gene therapy transgene product. Adeno-associated virus (AAV) is a 4.68-kb single-strand DNA virus that contains ITRs for viral replication and a packaging signal, and also has been engineered to contain therapeutic genes up to 5 kb in length. Transduction of recombinant AAV (rAAV) results in low inflammatory response and long-term expression. We have cloned a low-immunogenic form of human sTNFRI (sTNFRI2.6D) into AAV (rAAVsTNFRI). This vector was analyzed for its ability to transfect and neutralize the effect of TNF-alpha on primary rheumatoid arthritis synovial fibroblast (RASFs). The rAAVsTNFRI was transduced into the cells at 1.8 x 10(1), 1.8 x 10(2), and 1.8 x 10(3) viral particles per cell. There was greater than 90% neutralization of TNF-alpha at 1.8 x 10(3) viral particles/cell. There was a significant decrease in the synovial cell hyperplasia and cartilage and bone destruction in human TNF-alpha transgenic mice treated intraarticularly with rAAVsTNFRI. These results indicate that the low-immunogenic and long-term expressing vector, rAAVsTNFRI, can be used to deliver the soluble TNF-alpha in vitro and in vivo and effectively reduce the severity of arthritis.

The mammalian sodium channel BNC1 is required for normal touch sensation.

Of the vertebrate senses, touch is the least understood at the molecular level The ion channels that form the core of the mechanosensory complex and confer touch sensitivity remain unknown. However, the similarity of the brain sodium channel 1 (BNC1) to nematode proteins involved in mechanotransduction indicated that it might be a part of such a mechanosensor. Here we show that disrupting the mouse BNC1 gene markedly reduces the sensitivity of a specific component of mechanosensation: low-threshold rapidly adapting mechanoreceptors. In rodent hairy skin these mechanoreceptors are excited by hair movement. Consistent with this function, we found BNC1 in the lanceolate nerve endings that lie adjacent to and surround the hair follicle. Although BNC1 has been proposed to have a role in pH sensing, the acid-evoked current in cultured sensory neurons and the response of acid-stimulated nociceptors were normal in BNC1 null mice. These data identify the BNC1 channel as essential for the normal detection of light touch and indicate that BNC1 may be a central component of a mechanosensory complex.
