A simplified method for the preparation of WGA-Sepharose 4B and its use in the purification of MN blood group antigens.

A simplified method for the preparation of wheat germ agglutinin(WGA)-Sepharose 4B by coupling highly purified WGA, prepared by improved affinity chromatography, with BrCN activated Sepharose 4B in a solution of high carbonate buffer is described. The amount of WGA linked to Sepharose 4B was 82.40% (3.07 mg WGA per ml Sepharose 4B). MN blood group antigens of human erythrocyte membranes purified with WGA-Sepharose 4B affinity chromatography showed a single band on sodium dodecyl sulfate/polyacrylamide gel electrophoresis (SDS/PAGE). The yield of the antigens from 400 mL fresh blood was 32-40 mg. The WGA-Sepharose 4B column could be used several times without loss of activity.

Anticancer drugs. II. Synthesis and biological evaluation of spin labeled derivatives of podophyllotoxin.

The spin labeled derivatives of podophyllotoxin, 4-[4''-(2'',2'',6'',6''-tetramethyl-l''-piperidinyloxy)amino] -4'-demethylepipodophyllotoxin(GP-7,3) and N-podophyllic acid-N''-[4-(2,2,6,6-tetramethyl-l-piperidinyloxy)] thiosemicarbazide(GP-4,5) were synthesized and tested for their anticancer activity against the mouse solid tumors S180 and HepA in vivo, and the mouse lymphocytic leukemia L1210 and human stomach carcinoma SGC-7901 cells in vitro. At equitoxic concentrations, the anticancer activity of GP-7(3) was found to be similar to that of the clinically used VP-16(2). The toxicity of GP-7(3) (LD50231.2 mg/Kg) is 3.3 times lower than that of VP-16 (LD50 69.5 mg/Kg). GP-7(3) exhibits low subchronic toxicity. The total chemical yield of GP-7 (26%) is 4 times higher than that of VP-16 (6%) (based on podophyllotoxin). Therefore, GP-7(3) seems to be a promising new entry into the podophyllotoxin class of anticancer drugs.

In situ benign growth after radical radiation therapy of head and neck cancers--report of eight patients.

Eight patients who developed benign growth at the primary site after radical radiotherapy of head and neck cancers (two laryngeal cancers, six nasopharyngeal carcinoma, NPC) are reported. Most of the lesions appeared as spheral or nodular growth with few symptoms. All were properly diagnosed and treated, but two of them had been wrongly diagnosed and treated as local malignant recurrence. The authors point out that repeated biopsies, before treatment, are very important for differentiating a malignant recurrence from a benign growth. Conservative treatment can cure these lesions.

[Studies on the quality of Gastrodia elata Bl. cultivated with various kinds of fungus-growing materials].

In this article, the chemical compositions of 14 kinds of fungus-growing materials, and their corresponding rhizomorph and G. elata are analysed with thin layer chromatography. The properties of each G. elata and the contents of gastrodin in each G. elata are also analysed. The result shows that their properties and chemical compositions are similar, thus providing a basis for a larger choice of fungus-growing materials in cultivating G. elata.

[On the types and distribution of crystals in medical plants].

Crystals in medical plants are classified into organic and inorganic according to their chemical components. This paper gives an account of the types, chemical compositions, appearances structures and existing ways in plant tissue and distribution patterns in plant kingdom of various kinds of crystals.

Anti-shock effects of synthetic effective compositions of fructus aurantii immaturus. Experimental study and clinical observation.

Satisfactory results have been obtained in treating infective shock with injection of natural Fructus Aurantii immaturus (nat-FAI). The results of animal experiments and clinical observations on the anti-shock effects of the synthetic effective compositions of Fructus Aurantii immaturus (syn-FAI) are reported. The cardiac output increased from 0.53 to 0.87 L/min (P less than 0.01), and the cardiac index increased from 0.99 to 1.63 L/m2/min (P less than 0.01) in the endotoxic shock dogs after the treatment with syn-FAI. At the same time the blood stream in bulbar conjunctiva became accelerated and the dilated microvessels began to get smaller in most dogs. Of fifty children with infective shock treated with syn-FAI, forty-eight showed curative effects, with a total effective rate of 96%. The anti-shock effective compositions in FAI have been proved to be synephrine and N-methyltyrosamine. Moreover, syn-FAI has shown a more stable property, less side-effects and better clinical results than nat-FAI.

[Morphology and immunohistochemistry of true histiocytic lymphoma--report of 4 cases].

True histiocytic lymphoma (THL) is a rare malignancy of histiocyte. It is difficult to diagnose by light microscopy alone. In this paper, 4 cases of THL were reported basing on light and electron microscopic examination, and immunohistochemical study. Histiocytic lymphoma was of Langerhans cell origin in one case and of interdigitating reticulum cell origin in the other. The other 2 patients probably belonged to neoplasm of macrophage or dendritic reticulum cell origin. The differential diagnosis between the true histiocytic lymphoma and other tumors or diseases was briefly discussed.

[Primary malignant melanoma of the vagina--report of 8 cases].

Eight patients with primary malignant melanoma of the vagina are reported. They were all localized in the vagina with typical histologic features. Fontana and S-100 protein stain were positive. The prognosis was poor. Seven of them died and one was lost in the follow-up. Most of them (5/7) died in 1-2 years after treatment, but two survived for more than 5 years. In this paper, one case of vaginal cellular blue nevus was also suggested and the differential diagnosis from malignant melanoma was emphasized. The histogenesis of this tumor is briefly discussed.

4-Bromo-2-octenoic acid specifically inactivates 3-ketoacyl-CoA thiolase and thereby fatty acid oxidation in rat liver mitochondria.

In an attempt to develop a compound which would specifically inhibit 3-ketoacyl-CoA thiolase (EC 2.3.1.16) in whole mitochondria, 4-bromo-2-octenoic acid was synthesized and studied. After rat liver mitochondria were preincubated with 4-bromo-2-octenoic acid for 3 min, respiration supported by either palmitoylcarnitine or pyruvate was completely abolished, whereas no inhibition was observed with rat heart mitochondria. Addition of carnitine stimulated respiration supported by pyruvate without relieving inhibition of palmitoylcarnitine-dependent respiration. Hence, this compound seems to be a specific inhibitor of beta-oxidation. When the enzymes of beta-oxidation were assayed in a soluble extract prepared from mitochondria preincubated with 4-bromo-2-octenoic acid, only 3-ketoacyl-CoA thiolase was found to be inactivated. 4-Bromo-2-octenoic acid is metabolized by mitochondrial beta-oxidation enzymes to 3-keto-4-bromooctanoyl-CoA which effectively and irreversibly inhibits 3-ketoacyl-CoA thiolase but not acetoacetyl-CoA thiolase (EC 2.3.1.9). Even though 3-keto-4-bromooctanoyl-CoA inhibits the latter enzyme reversibly, 4-bromo-2-octenoic acid does not inhibit ketogenesis in rat liver mitochondria with acetylcarnitine as a substrate. It is concluded that 4-bromo-2-octenoic acid specifically inhibits mitochondrial fatty acid oxidation by inactivating 3-ketoacyl-CoA thiolase in rat liver mitochondria.

The synthesis of protective fragments of alpha-human atrial natriuretic polypeptide (alpha-hANP).

In this paper the synthesis of alpha-human atrial natriuretic polypeptide fragments (positions, 1-4, 5-10, 11-16, 17-22, 23-28) by solution method is reported. In the synthesis of some peptides, the results from mixed anhydride procedure and active ester procedure are compared. In the synthesis of other peptides, the effects of reaction temperature, basicity of catalyst, and reaction time on the specific rotations and yields of these compounds are observed. The sequence of amino acids of all the protective polypeptides is confirmed by FAB-MS.

[Midline malignant reticulosis (MMR) and peripheral T-cell lymphoma--a clinicopathologic study of 78 cases].

Seventy-eight cases of MMR selected from 141 cases of so-called malignant granulomatosis (MG) in our hospital during the past 27 years (1960-1986) are reported. All were treated mainly by radiotherapy, and 50 cases had been followed for long period. The overall 5-year survival rate was 44%. Although most of the patients died within 2 years, two had survived for more than 20 years after treatment. The microscopic features of these lesions were consistent with the peripheral T-cell lymphomas (PTL), which occur in other organs or sites as well. It could be roughly divided into two groups: monomorphic and polymorphic. Monomorphic lesions could be subdivided into several types again, such as large cell, mixed cell, clear cell, intermediate round cell types, and so on. In these cases, 11 were assayed for immunophenotype, including kappa, lambda and ACT, showing that the tumor cells were negative. Furthermore, 3 other new cases were stained with B1, IgM, Kappa, lambda, and/or Leu-2, Leu-3, Leu-4. Two of them were positive for Leu-3+, Leu-4+ and one for Leu-2+, Leu-3+, Leu-4+. All the results conform well to the diagnostic criteria of PTL as agreed upon by some Chinese and foreign authors. That means most of the MMR may be a group of heterogeneous extranodal PTL, and should be treated as T-cell lymphomas.

[Prostatic carcinoid and neuroendocrine tumors--a case report].

This paper reports a rare primary prostatic adenocarcinoid tumor confirmed by electron microscopy as well as immunohistochemical method including prostate specific antigen and prostatic acid phosphatase. Electron microscopy showed the presence of neuroendocrine granules in the cytoplasm of some tumor cells and the latter method ruled out the possibility of secondary tumor from other organs. It is noted that prostatic adenocarcinoid tumor is one of the diffuse neuroendocrine system tumors. Although the histogenesis of these tumors is not entirely clear, it is believed to be of multiple origins, and may transform through metaplasia into neoplasm.

[Relation between human papillomavirus (HPV) and cancer of uterine cervix].

HPV-DNA fragments were detected in biopsy specimens (29 cases of cancer of uterine cervix, 2 cervical dysplasia and 9 normal cervix) using DNA hybridization technique. It was demonstrated that 52% of biopsy specimens of the cancer of uterine cervix was positive for HPV 16 DNA probe, while 9% was positive for HPV 18 DNA probe. 11% of non-cancerous biopsy specimens had a positive result for HPV 16 DNA probe. It was also demonstrated that the positive rate of HPV 16 DNA was 75% in grossly cauliflower and nodular type tumors but only 25% in erosion type. It seems that the positive rate of HPV 16 DNA is correlated to gross appearance of the tumor. The positive rates of HPV 16 DNA were different in 6 provinces in China. It was 64% in Shanxi Province, a high incidence area but 36% in Sichuan Province, a low incidence area. These results suggest that the carcinogenesis of cancer of the uterine cervix, be related to HPV infection.
