Kinetics and products of OH radical addition to baicalin.

The reaction of OH radicals with baicalin was studied by pulse and gamma radiolysis in N2O-saturated aqueous solutions. The addition of an OH radical to baicalin produced an intermediate species that shows a maximum UV absorbance band at 420 nm and decays with a unimolecular rate constant of 1.25 x 10(4) s-1 and a bimolecular rate constant of 6.1 x 10(8) mol-1 dm3 s-1. Four major final products were obtained by gamma irradiation, and their structures were identified. Two of them (P1 and P2) are suggested to originate from the unimolecular reaction and the other two (P3 and P4) from the bimolecular reaction of the radicals.

Resting asynchronous left ventricular contraction abnormality analyzed by a phase method in spastic angina pectoris.

UNLABELLED: Quantitative phase analysis of equilibrium ventriculography was performed to study the character of left ventricular (LV) wall motion abnormalities in patients with spastic angina pectoris, who may have clinically and electrocardiographically silent ischemia combined with myocardial stunning, during rest and hyperventilation stress testing. METHODS: Phase analysis of the left ventricle at rest was performed by equilibrium radionuclide ventriculography in 13 control subjects and 36 patients with spastic angina pectoris. First-pass methodology along with hyperventilation stress testing was performed to assess spasm occurrences. Phase analysis of equilibrium multigated blood-pool scintigrams was performed to evaluate LV asynchrony at rest. RESULTS: The mean s.d. of LV phase distribution in the patients with variant and vasospastic angina was greater than that in the healthy control subjects (11.28 +/- 1.79 and 10.02 +/- 1.57 degrees versus 6.16 +/- 1.07 degrees). In addition, the mean s.d. of LV phase distribution in the variant angina group was greater than that in the vasospastic angina group. Furthermore, a linear correlation was found between the s.d. of LV phase distribution at rest and the percent decrease in ejection fraction during hyperventilation stress. CONCLUSION: Asynchronous LV contraction without significant hypokinesis was detected at rest in spastic angina pectoris. The severity of this asynchronous contraction corresponded well with decreases in ejection fraction during hyperventilation stress testing. Thus, analysis of the s.d. of LV phase distribution at rest is expected to provide useful information regarding LV asynchrony in spastic angina pectoris.

[Immunohistochemical observations on production and distribution of endothelin in atherosclerosis plaques].

An analysis of endothelin in atherosclerotic plaques using immunohistochemical methods showed that endothelin not only existed in endothelial cells, large amounts were also found in the proliferating intimal vascular smooth muscle cells (VSMC). In the de-endothelized thoracic aorta of rats, significant amounts of endothelin could be produced by proliferating intimal VSMC. Radio-immunological studies demonstrated that the extent of VSMC proliferation was in direct ratio to endothelin content, which suggests that production of endothelin is related to proliferation of VSMC in atherosclerotic plaques.

Smooth muscle myosin: a high force-generating molecular motor.

Smooth muscle generates as much force per cross sectional area of muscle as skeletal muscle with only one-fifth the myosin content. Although this apparent difference could be explained at the tissue or cellular level, it is possible that at the molecular level smooth muscle cross-bridges generate greater average force than skeletal muscle cross-bridges. To test this hypothesis, we used an in vitro motility assay (VanBuren et al., 1994) in which either chicken thiophosphorylated gizzard smooth or pectoralis skeletal muscle monomeric myosin is adhered to a nitrocellulose surface. A fluorescently labeled actin filament, attached to an ultracompliant (50-200 nm/pN) glass microneedle, is brought in contact with the myosin surface. Isometric force, being generated by myosin cross-bridges pulling on the attached actin filament, is calculated from the extent to which the calibrated microneedle is deflected. By measuring the density of myosin adhered to the surface, we estimated the number of myosin cross-bridges that are able to interact with a length of actin filament in contact with the myosin surface. In a direct comparison between smooth and skeletal muscle myosin, the average force per cross-bridge was 0.8 and 0.2 pN, respectively. Surprisingly, smooth muscle myosin generates approximately 4 times greater average force per cross-bridge head than skeletal muscle myosin. Because average isometric force is the product of the cross-bridge unitary force and duty cycle, we are presently using a laser optical trap in an attempt to measure unitary events from single myosin molecules. This approach should allow us to determine whether an increase in unitary force, duty cycle, or both contribute to smooth muscle myosin's enhanced force-generating capacity compared with skeletal muscle myosin.

[The effect of prostaglandin E1 on myocardial ischemia reperfusion injury].

The effect of prostaglandin E1 (PGE1) on ischemia reperfusion injury (IRI) was investigated. The changes of leucocytes in venous blood and infarcted myocardial tissue and the level of lipid peroxide (LPO), superroxide dismutase (SOD) and catalase (CAT) were also studied in these animal models at the same time. The experimental results were as follows: the count and activity of leucocytes and the level of LPO in IRI were obviously increased. The activity of SOD and CAT in the untreated group was much lower than that in the treated group (P < 0.01). The infarcted size (IS) was larger in the untreated group. The count of leucocytes was positively related with the level of LPO and IS, but negatively related with the activity of SOD and CAT. However, the relationship disappeared after the administration of PGE1. These data suggested that leucocyte plays an important role in IRI. PGE1 can inhibit leucocyte activity, decrease peroxidation, reduce LPO production and increase the activity of SOD and CAT. It can decrease the infarct size. PGE1 takes the main role in protecting IRI.

[A fundamental study on accumulation of [125I]IBZM in the rat striatum and on effect of non-labeled ligand].

Iodo-125-labeled iodobenzamide ([125I]IBZM) is used as a specific binding radioligand to dopamine D2 receptors with high affinity and selectivity. The radioligand was homogenously distributed in the whole brain initially after administration, and rapidly washed out from the dopamine receptor-poor area followed by persistent retention in the striatum. Regression curve generated from striatum/cortex PSL ratio indicated the constant washout rate from striatum and cortex respectively. In the pretreated rat by cold benzamide (2 mg/kg), the accumulation of the radioligand was significantly suppressed in the striatum (48.8%). Iodine-125-labeled iodo-benzamide has the promise for investigation of dopamine D2 receptors in the living brain.

[Study on CCK-induced gallbladder emptying with real-time ultrasonography].

In the present study, we adopted real-time ultrasonography to investigate the effect of atropine on the gallbladder emptying induced by CCK-OP in the normal subjects, and the difference of gallbladder emptying induced by CCK-OP between the "silent" gallstone patients and the controls. The results showed that: (a) CCK-induced gallbladder emptying in normal subjects was inhibited from 88.7% +/- 5.5% without atropine to 43.4% +/- 9.4% with atropine (P < 0.001). (b) The fasting gallbladder volume in the "silent" gallstone patients (26.7 +/- 10.9cm3) was significantly larger than that in the control (19.2 +/- 7.3cm3) (P < 0.05) and gallbladder emptying in the "silent" gallstones patients (44.0% +/- 8.7%) was significantly lower than that in the control (53.4% +/- 6.4%) (P < 0.01). We concluded that, significantly inhibited CCK-induced gallbladder emptying and there was significantly impaired gallbladder emptying in the patients with gallstones.

Ultrastructural cytochemistry of human gastric cancer: electron microscopic observations of five organellae marker enzymes.

The distribution of ALPase, ACPase, G6Pase TPPase and CCOase of gastric cancer and normal gastric epithelium were studied ultrastructurally. The results showed that normal gastric epithelium had no ALPase reaction. The reactions of ACPase, G6Pase, TPPase and CCOase were found in the corresponding organellae which were consistent with their functions. In tubular adenocarcinoma cells, their reactions were more apparent in the corresponding organellae. Some cells of tubular adenocarcinomas showed ALPase reaction. The mucinous adenocarcinoma cells had higher ACPase and TPPase reactions. In poorly differentiated adenocarcinoma cells, the five marker enzymes showed negative or faint reactions. The biological significance and mechanisms of distribution of the five marker enzymes were discussed.

[Analysis of causes of accidental deaths in several districts of Sichuan province].

The causes of accidental deaths from 1981 to 1988, including drowning, traffic accident, poisonning, child asphyxia, hitting, electric shock, fire and explosion in two cities and six districts of Sichuan Province were analysed. The average mortality from accidental death was 35. 58/10(5). Drowning and traffic death stood first and constituted 65.55% of the accidental deaths. Drowning, traffic death, poisoning and hitting death were more frequent in the male (P < 0.05). The mortality from traffic accident in the city was higher than that in the countryside. The mortality from drowning, child asphyxia and hitting in the city was lower than that in the countryside (P < 0.05).

[Inhibition of colorectal carcinoma induced by 1, 2-dimethylhydrazine in mice with tea polyphenols].

Colorectal cancer was induced by subcutaneous injection of 1,2-dimethylhydrazine (DMH) in mice, and the animals were administered orally with 0.4% of tea polyphenols (TP) simultaneously for 20 weeks to study its preventive effects. Results showed incidence of colorectal cancer in mice administered with TP was significantly lower than in positive controls (P < 0.05), and average number of tumor foci and proportion of adenocarcinoma were significantly fewer in TP group than in positive controls (P < 0.01). Level of cytochrome P450 (CP450) in liver microsome of the mice administered with TP was lower than that both in negative and positive controls (P < 0.01), and activities of superoxide dismutase (SOD) in liver tissue were higher than those in positive controls (P < 0.01), but lower than in negative controls (P < 0.01). Proliferative index of epithelial cells in mice was not obviously influenced by TP. It suggested that TP could have preventive effects on experimental colorectal cancer with a possible mechanism of lowering CP450 and increasing the activity of SOD in the liver.

[Significance of CEA and CA242 in the diagnosis of colorectal carcinoma].

Carcinoembryonic antigen (CEA) is frequently used in the diagnosis of the colorectal carcinoma. CA242 is a novel unique tumor-associated antigen characterized by higher tumor specificity and sensitivity for colorectal cancer, as compared with other mucin antigens. In this study, preoperative levels of serum CEA and CA242 were measured in 63 cases of colorectal carcinoma. It was disclosed that the positive rate of CA242 was higher than that of CEA, particularly in patients with colon cancer. The combined determination of CEA and CA242 significantly increased the sensitivity and accuracy in the detection of colorectal cancer as compared with the use of CEA alone (P < 0.01). In patients with advanced disease the positive rate was markedly elevated, especially in patients with liver metastasis. The results indicate that the combined use of CEA and CA242 assays is an useful adjunct diagnostic measure for colorectal carcinoma, and is helpful in the assessment of the stage of the disease as well as in making treatment plan.

[Experimental study on the effect of abstinence with herbal preparation composite dong yuan gao].

The effect of herbal preparation "Composite Dong Yuan Gao" (CDYG) on the animal model of drug dependence was studied. After the morphinistic models of white rats and mice were made by ever increasing doses of morphine each time, the animals were divided into large dose CDYG group, small dose CDYG group, sustained morphine group, and control group. The withdrawal symptoms were observed after naloxone was given to the animals intraperitoneally. The results showed that in the CDYG groups withdrawal symptoms of the addicted white rats were alleviated, the number of jumps of the addicted mice was reduced, the scores of the principal symptoms of the animals were decreased, while the body weights of the animals were gained. Comparing with the control group, the difference was significant (P < 0.01). The effects of the CDYG enhanced in the large dose group, which suggests that the CDYG has abstinence effects.

[An experimental study of corneal allograft rejection with cytotoxin IL-2-PE40].

We used a new immunosuppressive agents interleukin (IL)-2-Pseudomonas exotoxin 40 (PE40) to treat corneal allograft rejection, using the Wistar rat model of heterograft-heterotopic graft rejection. 36 rats were divided randomly into the treatment and the control groups. On 2, 5, 7, 10 and 14 days after the transplantation, the transplanted corneas were removed for pathological examinations. The peripheral blood was collected for the analysis of T cell subgroups and T lymphocyte colony forming unit (T-CFU) assay. These results indicate that IL-2-PE40 can delay the development of corneal graft rejection and significantly reduce the percentage of T-helper cells. On the other hand, IL-2-PE40 can weaken the gathering capacity of the peripheral T-cells.

[A randomized control study on the treatment of 123 cases of bacterial infections with cefteram and cefaclor].

To evaluate the efficacy and the safety of cefteram in bacterial infections, a randomized control study of cefteram and cefaclor on the treatment of 123 patients with respiratory and urinary tract infections was carried out. The result showed that the effective and bacterial eradication rates were 92.1% and 91.4% for cefteram. 83.3% and 85.2% for cefalor. Adverse reactions were mainly gastrointestinal reactions, occurring in 4.6% of the cefteram group and 9.4% of the cefaclor group. Study of minimum inhibitory concentration displayed high antibacterial activity of cefteram for enterobacteriaceae and other Gram-negative organisms and its activity was higher than that of gentamyicin and ciprofloxacin for E. coli. It is concluded that cefteram was effective and safe in the treatment of respiratory and urinary tract infections.

[Immunohistochemical study of 5-HT on gunshot wound of human skin].

Fourty antemortem and 30 postmortem gunshot wound samples of human skin were studied by immunohistochemical method. The 5-HT was seen mainly in wound edge, papillary layer, hypodermis and surrounding tissue of all antemortem gunshot wounds. The 5-HT was also discovered on the postmortem gunshot wounds which occurred within 8 minutes after death. The result showed that immunohistochemical staining of 5-HT be useful for diagnosing the antemortem gunshot wound. It also demonstrated that the mast cells of human do not contain 5-HT. We also studied the rate of mast cell degranulation of gunshot wounds of human skin by toluidine blue staining. The rate of mast cell degranulation of antemortem gunshot wounds increased (50%), and it was higher than that of postmortem gunshot wounds, suggesting that the rising of degranulation rate is a sign of antemortem injury.

[Research on the relationship between estrogen receptor, progesterone receptor, cell proliferation associated antigen in uterine leiomyoma and nuclear body density of myoma, serum reproductive hormone concentrations].

OBJECTIVE: To study the role of sex hormones in the pathogenesis of uterine leiomyoma in order to provide theoretical basis for medical treatment. METHODS: Estrogen Receptor (ER), progesterone receptor (PR) and cell proliferation associated antigen (Ki-67) contents of both normal uterine myometrium and leiomyoma were quantitatively analysed by ABC method and image analyser system (IAS) in 30 and 19 specimens obtained during hysterectomy respectively. Nuclear body density were also measured in 8 of them under electronic microscope. Serum luteinizing hormone (LH), follicular stimulating hormone (FSH), estradiol (E2) and progesterone (P) concentrations were determined by radioimmunoassay in 22 cases on the operation day. 21 age-matched normal cycling women served controls. RESULTS: Contents of ER, PR, Ki-67 and the number, volume densities of nuclear body were significantly higher in leiomyoma than those in normal myometrium (P < 0.01, P < 0.05). ER,PR contents of the two tissues in proliferative phase were greater than those in secretory phase, (P < 0.05), while Ki-67 contents were the opposite (P < 0.01). ER, PR contents of leiomyoma did not correlate with serum E2 and P concentrations, but correlate negatively with serum LH and FSH levels (P < 0.050). Ki-67 contents correlated positively with serum P levels and also showed inverse correlation with serum LH. FSH concentrations (P < 0.05). ER of leiomyoma positively correlated with density of nuclear body (P < 0.05). CONCLUSION: High ER, PR and Ki-67 contents in leiomyoma, may be the result of local high E2, P concentrations, and cause growth and development of leiomyoma. High Ki-67 content in secretory phase may indicate that progesterone has a synergistic effect in the pathogenesis of leiomyoma.
