Correlational patterns of cerebral glucose metabolism in never-medicated schizophrenics.

We studied 18 never-mediated schizophrenic patients and 22 normal control subjects with 18F-deoxyglucose (FDG) positron emission tomography. Patients and controls performed the continuous performance test during FDG uptake. Cortical and subcortical structures comprising two circuits selected on the basis of several theoretical models of schizophrenia were examined. The correlation of glucose metabolic rate (GMR) for each structure in each circuit with connected structures was calculated and tested for two-tailed significance. Schizophrenics showed significantly different patterns of intercorrelations for both circuits. The largest difference was in the correlation of GMR in the anterior thalamus with the frontal cortex, a key element in the thalamo-cortical-striatal circuit suggested to be abnormal in some models of schizophrenia. Correlations of the frontal lobe with other regions were also more positive in normal controls than schizophrenics; controls had three correlational paths from the frontal cortex (to temporal cortex, ventral anterior thalamus, and dorsal medial thalamus) with significantly more positive correlations than schizophrenics perhaps consistent with other findings of frontal cortical dysfunction in schizophrenia. Normal controls also had both more significant positive and more significant negative correlations between the occipital cortex and other brain areas than schizophrenics. Correlations between homologous areas in the right and left hemispheres were prominent in both groups.

PET and MRI of the thalamus in never-medicated patients with schizophrenia.

OBJECTIVE: This study reports the first paired measurements of glucose metabolism and size of thalamic regions in never-medicated schizophrenic patients using coregistered magnetic resonance imaging (MRI) templates. METHOD: Positron emission tomography with [18F]fluorodeoxyglucose and matching MRI scans were obtained in 20 never-medicated patients with schizophrenia and 15 normal volunteers. Methods for thalamic edge finding, statistical testing of shape differences with chi-square maps, and MRI localization of major thalamic subregions were developed. RESULTS: Patients with schizophrenia showed a diminished metabolic rate in the right thalamus, with a loss of the normal pattern of right greater than left asymmetry. Division into anterior/posterior segments revealed that the left anterior and right posterior showed the decrease. Differences were greater for metabolism in the weighted thalamic area (ratexarea) than for rate per unit area, a finding consistent with reported greater decreases in total neuron number than of neuron density in the thalami of schizophrenic patients. The area of the thalamus was smaller in the patients than in the volunteers, and this difference was greatest in the left anterior region. CONCLUSIONS: The reduced thalamic activity observed in this study lends further support to the concept of deficits in sensory filtering in schizophrenia.

Anti-inflammatory activities of 70% methanolic extract from Cinnamomi Cortex.

The effects of 70% methanolic extract (CC-ext) from Cinnamomi Cortex on acute and chronic inflammation were investigated. CC-ext inhibited the rise in vascular permeability induced by acetic acid and the increase of paw edema induced by carrageenin in mice. It was ineffective on edema derived by histamine or bradykinin, and exhibited only weak inhibitory effect on the edema derived by serotonin. CC-ext also showed inhibitory effects on the prekallikrein enzyme activity and ear edema induced by arachidonic acid. It also had an inhibitory effect on cotton pellet-induced granuloma but showed no atrophying action against the adrenal or thymus glands. Little effect was shown on secondary lesions in the development of adjuvant-induced arthritis (the arthritis reappeared from 11 to 27 d after injection of the adjuvant). These results suggest that some active component having an inhibitory effect on acute inflammation is contained in Cinnamomi Cortex.

The relationship of fluorosis and brick tea drinking in Chinese Tibetans.

Brick tea-drinking fluorosis is an unusual environmental problem. As a result of an investigation of tea-drinking habits, total fluoride intakes, dental fluorosis, and skeletal fluorosis, this disease has been found in the Sichuan Province of China in Tibetans with a long history of drinking brick tea. The dental fluorosis investigation of 375 Tibetan children (213 males, 162 females) and 161 Han children (86 males, 75 females), 8-15 years of age, was carried out in Daofu County, Sichuan Province. According to the standard of the Chinese Health Ministry, a skeletal fluorosis survey of 658 Tibetans (264 males, 394 females) and 41 Hans (20 males, 11 females), all over 16 years old, was performed. The total fluoride intake and fluorosis were determined from a question--calculation method in all participants. The morbidities of dental fluorosis in Tibetan and Han children are 51.2% and 11.05%, respectively, and the indexes of dental fluorosis are 1.33 and 0.17 (chi 2 = 75.7, p < 0.01) respectively. The morbidity of skeletal fluorosis is 32.83% for Tibetan children and zero for the Han children. The fluoride intakes of Tibetan children and adults were 5.49 mg/person/day and 10.43 mg/person/day, respectively, in this area. Of total everyday fluoride intake, 94.2% by children and 94.4% by adults was from brick tea and zanba (r = 0.99).

Protective effects of trolox C, vitamin C, and catalase on bromobenzene-induced damage to rat hepatocytes.

BACKGROUND/METHODS: The protective effects of trolox C (water-soluble vitamin E), vitamin C, and catalase on bromobenzene (BB)-induced toxicity to isolated rat hepatocytes were evaluated. The glutathione (GSH) content of the hepatocytes exposed to BB was measured. RESULTS: BB caused acute damage to the cells during 2 h of incubation (short) when BB was added directly to the culture wells, whereas a late-occurring and time-dependent increase in lactate dehydrogenase (LDH) leakage rate was observed during 24 h of incubation (long) when BB was dissolved in a different way. Incubation of the cells with trolox C (0.5-2.0 mM) prevented the hepatocellular damage induced by BB at 2.4 mM during the long-term incubation. Vitamin C (0.1-1.0 mM) had a protective effect on BB-induced toxicity during both the short- (BB, 1.6 mM) and the long- (BB, 2.4 mM) term incubations. Catalase (3200 U/ml) also showed a beneficial effect on the cells during the short-term BB exposure. Trolox C (2.0 mM) and vitamin C (0.5 mM) restored BB-induced GSH depletion in the cells. CONCLUSIONS: BB induced two patterns of LDH leakage from isolated hepatocytes on the basis of different ways of BB exposure and incubation periods. Trolox C, vitamin C, and catalase exerted protective effects on BB-induced toxicity during short- or/and long-term incubations. The effects were concentration-dependent. Restoration of GSH content in BB-exposed hepatocytes suggests that trolox C and vitamin C could reduce GSH consumption during BB metabolism and exert an antioxidant effect.

Immunolocalization of basic fibroblastic growth factor in avian tibial dyschondroplastic cartilage.

Recent research in our laboratory has demonstrated that basic fibroblast growth factor (bFGF) is a permissive mitogen for epiphyseal growth plate chondrocytes. Immunocytochemistry demonstrated the presence of bFGF in the proliferative and hypertrophic zones of normal epiphyseal growth plates of 4-wk-old broiler chickens. The purpose of this investigation was to extend this research to include examination of the status of bFGF in the cartilage lesion associated with tibial dyschondroplasia (TD). Immunocytochemistry revealed that the distribution of bFGF in the growth plate proximal to the TD lesion was similar to that observed with normal growth plate. However, the intensity of immunofluorescence was greatly diminished in the TD lesion. The number of chondrocytes staining positive for bFGF was also reduced. In the peripheral edges of the lesion where cartilage was being actively resorbed, the staining intensity was greatly increased when compared to the rest of the TD lesion. Similar patterns were observed in all TD tissues examined whether the lesions were spontaneous or induced by dietary treatments or genetic selection. It is hypothesized that the decrease in bFGF, a potent angiogenic factor, may be responsible for the poor vascularization of the TD lesion.

[Changes of brain potentials related to visual attention during simulated weightlessness].

To study the possible effect of simulated weightlessness on brain function state, the brain event-related potentials (ERPs) during a simple visual selective response task were compared between head down tilt (HDT) and head up tilt (HUT) in 9 normal subjects. The results were: both the target (T) and non-target (NT) flash signals induced significant slow positive potentials which were supposed to be related to the attention activity; the amplitude of the positive potentials in the frontal regions decreased significantly especially for NT-ERPs during HDT as compared with that during HUT. The data provide new evidence indicating that the ability of brain response declined during simulated weightlessness and more attention should be paid to the study of brain function during space flight.

[Evaluation of reserved hepatic function in patients with hepatobiliary tumor by 99mTc-GSA: effect of hyperbilirubinemia and usefulness of regional reserved hepatic functional imaging].

The evaluation of the reserved hepatic function was performed by 99mTc-galactosyl serum albumin (99mTc-GSA) in seventy patients with hepatobiliary tumor. The dynamic study was performed to evaluate global reserved hepatic function following the intravenous bolus injection of 99mTc-GSA, and the hepatic single photon emission computed tomography (SPECT) was obtained to assess the regional reserved hepatic function. The functional hepatic index (LHL15) was derived from liver time-activity data, and it was compared with serum total-bilirubin level, serum albumin level and plasma disappearance rate of indocyanine green (ICG15). In the patients with hepatocellular carcinoma, LHL15 value agreed well with ICG15 value, serum total-bilirubin level, and serum albumin level. Moderate or severe hepatic dysfunction was observed at 65.4% of these patients. In the patients with cholangiocellular carcinoma, a discrepancy of LHL15 value and ICG15 value was observed. Increment of the ICG15 value was correlated with that of the serum total-bilirubin level, whereas the correlations was not observed between the LHL15 value and the serum total-bilirubin level. These results indicate that 99mTc-GSA scintigraphy can evaluate the reserved hepatic function without the embellishment of jaundice. This method is useful for assessing the global and regional reserved hepatic function.

Dopamine-induced ionic currents in acutely dissociated rat neurons of CNS.

AIM: To determine whether or not the dopamine (DA) can induce ionic current in single neuron acutely dissociated from different central areas including striatum, ventral tegmental area (VTA), substantia nigra pars compacta (SNC) and hippocampal CA1 area. METHODS: Using a new patch-clamp whole-cell recording technic, namely nystatin-perforated whole-cell configuration under voltage-clamp mode. RESULTS: In 36 single neurons isolated from the striatum and VTA, DA 10-1000 mumol.L-1 was quite diverse to elicit ionic current responses. In 19 SNC neurons, 5 neurons (26%) response to 1 mmol.L-1 DA represented as a small outward current (11.3 +/- 2.4 pA) at a holding potential (V(H)) of -20 mV. In 25 of 69 (36%) examined hippocampal CA1 pyramidal neurons, however, application of DA induced 3 types of current responses: outward current (8 neurons) accompanied with an increase of membrane conductance, slow inward current (5 neurons) with an decrease of membrane conductance and outward-following inward current (12 neurons) at a V(H) of -20 mV. The concentration-response relationship of DA-induced currents showed the typical sigmoid shape with the threshold dose, being the maximum response dose are 3 mumol.L-1 and 1 mmol.L-1 respectively. The current-voltage (I-V) relationship of DA-induced responses did not show any voltage-dependent manner and the reversal potential (E(DA)) was close to the equilibrium potential of potassium (E(K)) calculated with the Nernst equation. TEA 5 mol.L-1 effectively inhibited the DA-induced response. CONCLUSION: These results suggest that DA-induced outward current is carried by K+ in single hippocampal CA1 pyramidal neuron.

Determination of serum and urinary aluminum by HPLC with fluorometric detection of Al-lumogallion complex.

We describe a simple and sensitive HPLC method for quantifying aluminum (Al) in biological fluids by measuring the fluorescence of the Al-lumogallion complex (excitation wavelength 500 nm, emission wavelength 575 nm). Serum samples are deproteinized with 0.83 mol/L perchloric acid and centrifuged; the supernates are mixed with lumogallion reagent. Urine samples are pretreated with sodium hydroxide (2 mol/L) and methanol, kept for 1 h at -20 degrees C, and then centrifuged; the precipitate is resuspended in perchloric acid and mixed with lumogallion reagent, as for serum. The maximal fluorescence complex is formed after 1 h at pH 5 +/- 0.5. The HPLC mobile phase consists of (per liter) 100 mL, of 0.2 mol/L potassium hydrogen phthalate, 220 mL of acetonitrile, and distilled deionized water. The flow rate is 1 mL/min, and the injection volume is 5 microliters. The major aluminum species is eluted at 3.5 min, the lowest detection limit being 0.45 pg. We validated the method with samples collected from normal subjects and from workers occupationally exposed to aluminum. Comparing the results with those by traditional atomic absorption spectrometry of urinary aluminum suggests that the proposed method is reliable.

Reversal of multidrug resistance by tacrolimus hydrate.

Tacrolimus hydrate, a potent immunosuppressant produced by Streptomyces tsukubaensis, was examined for its effect on epirubicin activity in multidrug-resistant P388 leukemia (P388/R) cells overexpressing P-glycoprotein and the parent (P388/S) cells. In the absence of modulator, the 50% inhibitory concentration for epirubicin after 48-h incubation, determined using a microculture tetrazolium assay, was 0.8 microgram/ml in P388/R cells and 0.009 microgram/ml in P388/S cells. P388/R cells demonstrated a 90-fold reduction in sensitivity to epirubicin. Tacrolimus hydrate (1 and 10 microM) markedly enhanced epirubicin cytotoxicity by 4.2- and 26.7-fold for P388/R cells. A significant increase in LDH release from cells by tacrolimus hydrate was also observed in P388/R cells treated with epirubicin. Tacrolimus hydrate had a marked effect on epirubicin-induced G2/M blockade in the resistant cells. Both tacrolimus hydrate and cyclosporin A dramatically increased the accumulation of epirubicin by the resistant cells, while these compounds had no effect on epirubicin accumulation in the parent cells. Thus, tacrolimus hydrate is able to down-modulate P-glycoprotein-associated resistance through inhibition of P-glycoprotein function, suggesting that the drug may be a candidate for killing drug-resistant tumor cells.

Presence of free radicals in pigment gallstone in vivo.

OBJECTIVE: To clarify whether free radical, which may play a role in pigment gallstone formation, is present in pigment gallstones in vivo. MATERIALS AND METHODS: Free radical signal of gallstones from 18 patients was detected by electron paramagnetic resonance spectroscopy at 77K under anaerobic condition and in air (control). As soon as the anaerobic determination was finished, the fresh anaerobic sample was exposed to air and stored in a freezer at -20 degrees C. RESULTS: Free radical signal (g = 2.0038) was detected in fresh anaerobic samples containing more than 2% bilirubin compound, and the signal intensity correlated linearly with the content of calcium bilirubinate (r = +0.95, P < 0.0005). During the storage at -20 degrees C and exposure to air, the signal intensity of each anaerobic sample and its control increased gradually, eventually reaching the same stable level. Fe(III) signal intensity was enhanced synchronously and related linearly with free radical signal (r = +0.99, P < 0.0005). CONCLUSIONS: Free radical exists originally in pigment gallstones in vivo, and it may play an important role in pigment gallstone formation. The free radical signal carried by gallstones may be strengthened by the action of oxygen in air on bilirubin. The transition metal ions probably take part in the formation of bilirubin free radical.

[Seven-year follow-up in pupils immunized with hepatitis B vaccine and its revaccination].

In order to evaluate immune persistence of plasma-derived hepatitis B vaccine (HBV), 344 children in a primary school were followed up for seven years by detection of serum HBsAg, anti-HBc and anti-HBs with radioimmunoassay (RIA) using Abbott reagent kits. In the seventh year after three-dose HBV immunization, level of anti-HBs in 55.4%-75.0% of the vaccinee was greater than 10 IU/L, with a geometric mean titer (GMT) of 20.9-65.3 IU/L. The higher the level of anti-HBs, the longer the duration of its decrease from more than to less than 10 IU/L. Seven years after immunization, 66 children with varied serum levels of anti-HBs were received a fourth dose of 10 micrograms HBV, and one month after revaccination, their levels of anti-HBs increased by 6.3-57.7 times. It suggests revaccination of HBV is not necessary for children vaccinated within seven years.

[Determinations of antiovarian antibodies and cellular immunity functions in patients with premature ovarian failure].

OBJECTIVE: To investigate the variations of cellular immunity function and its relationship with antiovarian antibodies (AOAb) in patients with premature ovarian failure (POF). METHODS: Serum AOAb, T-lymphocyte subsets and leukocyteprocoagulant activity (LPCA) to ovarian antigens of peripheral blood were examined in 30 normal women (control group) and 30 patients with POF (POF group). RESULTS: The mean serum AOAb level in POF group was significantly higher than that in control group (6.80 +/- 1.9 kU/L vs 1.39 +/- 0.72 kU/L, P < 0.01). The percentage of CD3+, CD4+ cells increased significantly (65.42 +/- 5.31% and 44.79 +/- 5.90%, respectively), the percentage of CD8- cells was significantly lower (25.63 +/- 4.26%) and the ratio of CD4+/CD8+ increased (1.66 +/- 0.27) in POF group when compared with control group (P < 0.01). The positive rate of AOAb in patients who had elevated ratio of CD4+/CD8+ was significantly higher than those with normal CD4+/CD8+ ratio (85.7%, 18/21 vs 3/9; P < 0.01). The level of LPCA was elevated in POF patients with positive AOAb, and was correlated positively with the level of AOAb (chi 2 = 8.378, P < 0.01). CONCLUSIONS: These data showed that cellular immunity and humoral immunity to ovarian antigens can be concurrently produced in POF patients, and suggested that pathogenesis of POF may be associated with immune factors.
