Differentiation of the nucleotide-binding sites on nucleotide-depleted mitochondrial F1-ATPase by means of a fluorescent ADP analogue.

The interaction of the fluorescent ADP analogue lin-benzo-ADP (containing a linearly extended version of adenine, in which a benzene ring is inserted between pyrimidine and imidazole ring) with nucleotide-depleted mitochondrial F1 was investigated. It was found that lin-benzo-ADP is able to occupy all six nucleotide-binding sites present on the enzyme. Two sites exhibit a very high affinity for the analogue (dissociation constant, Kd, less than 10 nM) and bind it rapidly (association rate constant, k+1, about 1.10(6) M-1 S-1). A third site shows a lower affinity for the analogue (Kd = 1-2 microM) and is occupied relatively fast (k+1 approximately 10(4) M-1 S-1). Binding of lin-benzo-ADP to these three sites is prevented not only in the presence of excess ADP and ATP, but also by IDP and ITP, thus indicating that these sites are the catalytic ones. As it will be discussed, this conclusion is further corroborated by the finding that release of the analogue from the two high affinity sites can be promoted by binding of nucleoside di- and triphosphates to the third site. The remaining three sites were found to bind lin-benzo-ADP with identical affinity (Kd = 1-2 microM) and with a rather low association rate (k+1 = 300-600 M-1 S-1). Binding of the analogue to them is only prevented by ADP and ATP, but not by IDP and ITP, which confirms that these sites are the noncatalytic ones. The analogue could be displaced by excess ADP also from these sites; however, in contrast to the catalytic sites, no promotive effect was observed here. The obvious changes in the nucleotide binding behavior of the non-catalytic sites after depletion of endogenous nucleotides will be discussed.

Poly(gamma-glutamic acid)s are the major constituents of nematocysts in Hydra (Hydrozoa, Cnidaria).

The 2.80 +/- 0.20 mumol of anions found/mg of isolated and purified dry nematocysts (capsular secretory products of stinging cells) from Hydra make up the majority of the soluble capsular content. They are, in cooperation with corresponding cations, responsible for the generation and regulation of an internal osmotic pressure that amounts up to 150 bar (Weber, J. (1989) Eur. J. Biochem. 184, 465-476). The anions are organized as linear homopolymers of L-glutamic acids which are linked by gamma-carboxyl-alpha-amino amide bonds; the degree of polymerization is heterogeneous and dependent on the particular type of nematocyst. In situ the intracapsular glutamic acid monomer concentration is as high as 2 M. This is the first time that poly(gamma-glutamic acid)s, which are known to occur in some selected bacteria, are reported for eucaryotes. It is suggested that they may also be present as predominant components in nematocysts of other cnidarian species and thus might represent a class of compounds which is characteristic for a whole phylum of the animal kingdom.

Organization of intrahippocampal projections originating from CA3 pyramidal cells in the rat.

The distribution of intrahippocampal projections arising from the CA3 region of the rat hippocampus was investigated using in vitro and in vivo methods. In the in vitro hippocampal slice preparation, single CA3 pyramidal cells were intracellularly labeled with horseradish peroxidase (HRP), and the three-dimensional organization of the axonal plexus was analyzed by using a computer-aided digitizing system. As many as eight primary collaterals originated from the principal axon of CA3 pyramidal cells and these commonly bifurcated further and innervated stratum oriens and stratum radiatum of CA3 and CA1. Within the 400 microns slice, the summed length of all visible collaterals per neuron ranged from 2.6 mm to approximately 12.5 mm. While the CA3 principal axon tended to be relatively smooth, the axonal collaterals bore numerous varicosities that electron microscopy confirmed to be presynaptic boutons. These varicosities occurred, on average, once every 7 microns of collateral length. The distribution of axonal collaterals differed depending on the location of the parent pyramidal cell. Only rarely could CA3 collaterals be followed in the slice to their terminations within CA1. To study the topographic organization of CA3 projections both to other levels of CA3 and to CA1, the anterograde tracer, Phaseolus vulgaris leucoagglutinin (PHA-L) was injected into various transverse and septotemporal levels of CA3. Immunohistochemical visualization of the lectin was conducted in dissected and "extended" hippocampi to facilitate analysis of the topographic distribution of projections along the long or septotemporal axis. Projections from all portions of CA3 reached widespread regions of CA3, CA2, and CA1, but only a few fibers entered the subicular complex and there were no projections to the entorhinal cortex. There were also some CA3 and CA2 projections to the hilus of the dentate gyrus, but these did not enter the granule cell or molecular layers. The CA3 projections to CA1 were organized according to several distinctive and consistent gradients that can generally be summarized as follows. 1. CA3 cells located close to the dentate gyrus (proximal CA3), while projecting both septally and temporally, tended to project more heavily to levels of CA1 located septal to the injection site. CA3 cells located closer to CA1, in contrast, projected more heavily to levels of CA1 located temporally to the injection site. 2. At, or close to, the septotemporal level of the injection, cells located proximally in CA3 gave rise to collaterals that tended to terminate more superficially in stratum radiatum than did those arising from mid and distal levels of CA3.(ABSTRACT TRUNCATED AT 400 WORDS)

[Piezoelectric lithotripsy in the treatment of symptomatic gallstone patients].

From January to May 1989, 50 patients with symptomatic gallstones were treated by means of extracorporeal piezoelectric lithotripsy combined with oral bile acid therapy. In 98% of all patients stone fragmentation was achieved. The mean number of treatment sessions per patients was 3.24. We observed as complications one gallbladder- and one liver hematoma each besides colicy pain and asymptomatic laboratory changes. Two patients developed mild pancreatitis and two a cholecystitis; three patients had symptomatic common bile duct stones during the follow-up. ERCP with endoscopic papillotomy was performed four times, one female patient had to undergo surgery due to an acute gallbladder. Three months after the first treatment, 18 patients (36%) were stonefree, six months later 54% (21/39). Patients with solitary stones up to 2 cm in size were in 71.5% stonefree respectively after three months. Thus, piezoelectric lithotripsy represents in our opinion a possible therapeutic modality for the treatment of symptomatic gall stones in selected patients.

[Clinical experiences with early colonoscopic decompression in acute pseudo-obstruction of the colon].

The prognosis of an acute colonic pseudoobstruction has improved significantly since the introduction of colonoscopic decompression. Our experience confirms this technique as a safe and effective therapy even for critically ill patients. In most of our cases several endoscopic decompressions were necessary before a lasting improvement of the clinical symptoms could be achieved. The additional use of new procinetic substances and the endoscopic placement of thick decompression tubes may reduce the number of endoscopic procedures necessary for each patient.

Microdissection and microcloning of human chromosome 7q22-32 region.

Genetic information contributing to cystic fibrosis in addition to the CF gene is suggested to reside on the long arm of the human chromosome 7. In our attempt to analyze this genomic region in detail, we generated a region-specific DNA probe library by microdissection and microcloning of the midpiece of the chromosome 7q arm. Microdissection was performed in unstained metaphase spreads from a human x mouse hybrid cell line containing chromosome 7 as the only human chromosome. We obtained 593 clones from 75 dissected chromosomal fragments. At least 88% of the microclones were true recombinants; 40% of the clones contained repetitive sequences as determined by plaque hybridization with genomic DNA as probe. The overall mean fragment size of insert fragments was 3.2 kb, the median size was 3.5 kb. Regional mapping of 30 DNA fragments was performed by the aid of hybrid cell lines containing different segments of human chromosome 7; 50% of the microcloned inserts were found to map to 7q22-32.

Anorectal manometric anomalies in seven patients with frontal lobe brain damage.

We studied the anorectal manometric and urodynamic pattern of seven patients who had right and/or left frontal lobe injury to investigate the possibility that the frontal lobe area plays an important role in control of anorectal motility as it does for the urinary bladder. We found that the disturbances of anorectal manometric recordings were similar to those of urodynamic recordings: (1) lack of urinary bladder filling sensation (two cases), increased perception threshold of rectal distension (two cases); (2) uninhibited detrusor contractions during filling (five cases), spontaneous rectal contractions during the resting step (four cases); (3) absence of micturition (one case), absence of rectoanal inhibitory reflex (one case). Furthermore, in all but one patient, we observed a decrease in the amplitude and duration of the voluntary contraction of the anal canal (six cases). However, in two of the seven patients we found hypertonia in the anal canal (upper part: one case, lower part: one case), but never in the urethral canal. Our results suggest that frontal lobe may be involved in the nervous control of anorectal motility as it is for urinary bladder function. However, the lack of correlation between urinary and anorectal anomalies in individual cases suggests that these functions depend on distinct areas of the frontal lobe.

The nematocyst extract of Hydra attenuata causes single channel events in lipid bilayers.

The nematocyst extract of Hydra attenuata causes single conductance events in reconstituted planar lipid membranes as well as in inside-out patches derived from liposomes. The smallest single channel conductance level of the toxins is 110 pS. The conductance levels increase stepwise with time up to 2000 pS. These large conductance jumps indicate channel cooperativity. If the membrane-voltage is changed from positive to negative values, the single channel events become undefined and noisy, indicating major reorganizations of the proteins which form the channels. The molecular properties of the ionophoric component(s) of the nematocyst extract may help explain the observed macroscopic effects, such as hemolysis of human erythrocytes, after addition of the nematocyst extract.

Colonic transit time and anorectal manometric anomalies in 19 patients with complete transection of the spinal cord.

In order to determine the relative importance of sympathetic and parasympathetic centers in the control of colorectal motility, colonic transit and anorectal motility were studied in 19 patients with complete spinal cord transection: group 1 (n = 5) where transection was above T9; group 2 (n = 6) where transection was between T9 and L2; group 3 (n = 8) where transection involved S2-S4. Colonic transit time was calculated by the radiopaque markers. Methods and results were compared with those of eight controls, all bedridden for non-digestive surgery. Anorectal motility was investigated by anorectal manometry, and results were compared with those of 17 healthy controls. Both mean right and left colonic transit times were not different in the three groups of patients and the bedridden control group. Mean rectosigmoid and total transit time increased in groups 2 and 3. In five patients of group 3 the mean anal canal (lower part) pressure was higher than in controls. Recto-anal inhibitory reflex was present in all patients, but their was no correlation in group 3 between the volume of rectal distension and both amplitude and duration of the recto-anal inhibitory reflex, and in group 1 between rectal distension and the duration of the recto-anal inhibitory reflex. Rectosigmoid transit time increased when sympathetic and parasympathetic spinal centers are injured, suggesting the importance of this extrinsic nervous control; right and left colonic transit are not affected by spinal cord lesion suggesting that the distal colon but not the proximal colon, is under spinal cord nervous control.(ABSTRACT TRUNCATED AT 250 WORDS)

[Pancreatitis-induced pseudocysts of the spleen: a rare complication].

Secondary splenic cysts usually occur following blunt abdominal trauma. In rare cases, the spleen can be affected in pancreatitis. Since the clinical findings are fairly uncharacteristic, this was a difficult diagnosis to make before imaging examination techniques became available. These cysts only became apparent as a result of their complications. Today, thanks to sonography and abdominal CT-scan, these cysts can be detected in their asymptomatic stage. Furthermore, interventional sonography allows the puncture and drainage of these cysts, and this technique is thus an alternative to surgery. Two cases of pancreatitis with splenic involvement are presented.

Empyema after pneumonectomy--empyema window or thoracoplasty?

369 pneumonectomies carried out within the years 1981 to 1988 and their empyema complications form the basis for a retrospective analysis to assess the outcome of treating pneumonectomy cavity empyema by window healing. During the same period 31 empyema after 322 pneumonectomies to treat bronchial carcinoma are investigated in more detail as far as their therapeutic modalities are concerned. 20 patients received a thoracic window. 18 of 31 patients (58%) could be cured, and 9 of these were in the thoracic window group. The hospital mortality of empyema patients after pneumonectomy amounted to 42%. In an historical comparison no progress could be recognized in the treatment of this septic complication.

Antibodies to HIV-1 nef(p27): prevalence, significance, and relationship to seroconversion.

A sensitive and specific enzyme-linked immunoassay for antibodies to the human immunodeficiency virus type 1 (HIV-1) nef gene product, p27, has been developed using recombinant Escherichia coli-derived protein from the LAV-1-Bru sequence. Of 92 HIV-1 infected hemophiliacs, 72 (78%) produced anti-nef antibodies in this assay; the early appearance of anti-nef prior to full seroconversion was a rare event in this population, occurring in only one subject (approximately 1%). Anti-nef antibodies were not detected in any of 500 sera from 98 repeatedly HIV seronegative subjects who had been exposed to sexually transmitted modes of HIV infection (45 subjects) or through blood products (53 subjects). There was no significant association of titer or anti-nef antibody with protection from disease in HIV infection (p = 0.1). Although the nef protein is relatively immunogenic in natural infection, this study cannot confirm the previously reported high prevalence of anti-nef antibodies prior to seroconversion, nor the finding of anti-nef antibodies in HIV seronegative but exposed subjects.

Antibodies to a putative HIV gp41 immunosuppressive peptide, pHIVIS (583-599), do not correlate significantly with outcome in HIV infection.

We have studied the prevalence of antibodies to peptides derived from the transmembrane protein of HIV, gp41. Previous work has suggested that the presence of antibodies to the gp41 peptide known as pHIVIS (env 583-599) is associated with protection from immunosuppression in HIV infection. We studied 171 sequential sera from 55 HIV-1-infected people in various clinical stages of disease. There was no significant association between antibodies to pHIVIS and clinical status in this study. Although pHIVIS has sequence similarity to the putative immunosuppressive region of the C-type oncornaviruses (p15E), antibodies to this peptide do not appear to be associated with protection from immunosuppression in natural HIV infection.

Maturation in Larch : II. Effects of Age on Photosynthesis and Gene Expression in Developing Foliage.

The effect of maturation on the morphological and photosynthetic characteristics, as well as the expression of two genes involved in photosynthesis in the developing, current year foliage of Eastern larch (Larix laricina [Du Roi]) is described. These effects were observed on foliage during the third growing season after grafting of scions from trees of different ages onto 2 year old rootstock. Specific leaf weight (gram dry weight per square meter), leaf cross-sectional area (per square millimeter), and chlorophyll content (milligram per gram dry weight) all increase with increasing age in long shoot foliage from both indoor- and outdoor-grown trees. Net photosynthesis (NPS) (mole of CO(2) per square millimeter per second) increases with age on indoor- but not outdoor-grown trees. NPS also increases with increased chlorophyll content, but outdoor-grown scions of all ages had higher chlorophyll content, and chlorophyll does not appear to be limiting for NPS outdoors. To extend these studies of maturation-related differences in foliar morphology and physiology to the molecular genetic level, sequences were cloned from the cab and rbsS gene families of larch. Both cab and rbcS gene families are expressed in foliage but not in roots, and they are expressed in light-grown seedlings of larch but only at very low levels in dark-grown seedlings (~2% of light-grown seedlings). Steady-state cab mRNA levels are relatively higher (~40%) in newly expanding short shoot foliage from juvenile plants compared to mature plants. Unlike cab, the expression of the rbcS gene family did not seem to vary with age. These data show that the maturation-related changes in morphological and physiological phenotypes are associated with changes in gene expression. No causal relationship has been established, however. Indeed, we conclude that the faster growth of juvenile scions reported previously (MS Greenwood, CA Hopper, KW Hutchison [1989] Plant Physiol 90: 406-412) is not due to increased NPS or cab expression. Long shoot foliage is the dominant foliar type on young trees and its lower specific leaf weight will permit production of more photosynthetic surface area per unit of leaf biomass.
