Antifibrotic and uveitogenic properties of gamma interferon in the rabbit eye.

The feasibility of gamma (gamma)-interferon injection for the treatment of ocular fibrotic conditions was studied in rabbits using recombinant rabbit gamma-interferon and the cell-injection model of tractional detachment. A toxicity study revealed that intravitreal injection of greater than 10(4) units gamma-interferon consistently produced panuveitis. For tractional detachment, 250,000 rabbit dermal fibroblasts were injected intravitreally into 20 eyes; 1 day later, 5 of these eyes received intravitreal injections of 10(4) units gamma-interferon, another 5 were given 10(6) units, and the remaining 10 received balanced salt solution. Slit-lamp examination and fundus photography were performed at regular intervals for 21 days and were graded by a masked observer. The eyes were then enucleated and processed for histology. Doses of 10(4) units gamma-interferon significantly reduced the severity of detachments, but injections of 10(6) units induced panuveitis.

Proton interactions in the resting form of cytochrome oxidase.

The effect of pH on the near-UV absorption spectrum of cytochrome oxidase has been examined. Several lines of evidence implicate a proton binding site that can modulate the optical properties of cytochrome alpha 3 in the resting enzyme. Changing the pH within the range 6.5-10.5 was found to reversibly shift the position of the Soret band over an 11-nm range. The lower pH values caused a progressive blue shift in the Soret band, whereas the high-pH range promoted a gradual red shift. Limiting band positions were approximately 416 and 427 nm. The incubation time required to reach a stable band position varied somewhat as did the actual extent of the shift. In most cases, the shift was associated with an isosbestic point. A pH titration profile for the apparent equilibrium position of the Soret band was obtained. Nonlinear least-squares fitting to a scatter plot, assuming a single acid/base group, showed an apparent pKa of 7.8. Magnetic circular dichroism (MCD) spectra of the low-pH form at 416 nm, the high-pH form at 427 nm, and the cyanide derivative at 428 nm were compared. No evidence of a high-pH-dependent low-spin transition or a change in the redox state of cytochrome a3 was found, confirming earlier work [Baker, G. M., Noguchi, M., & Palmer, G. (1987) J. Biol. Chem. 262, 595-604]. Subtraction of ferricytochrome a [spectrum taken from Vanneste, W. H. (1966) Biochemistry 5, 838-848] from a series of blue-shifting spectra showed a band at 414 nm that progressively gained amplitude and a band at 430 nm that correspondingly lost amplitude. A series of red-shifting spectra showed the opposite behavior with a clear isosbestic point being evident in both cases. The difference extinction change at 414 and 430 nm depended linearly on the position of the Soret band, both showing a reversible dependence on pH. The 430-nm band is noted to be unusually red-shifted for high-spin ferric heme a. An additional, pH-insensitive band was observed at 408-410 nm which was eliminated by treatment with cyanide. The kinetics of the pH-induced blue shift and red shift were obtained at 416 nm by using dual-wavelength method and found to be biphasic, despite the occurrence of an isosbestic point.(ABSTRACT TRUNCATED AT 400 WORDS)

[Studies on double-labelling method for determination of T lymphocyte subpopulations].

In order to determine the concurrent expression of DR and CD antigen of T lymphocyte surface markers, the fluorescence-rosette double-labelling technique was developed by employing monoclonal antibody against HLA-DR and sensitized sheep erythrocytes with OKT series of monoclonal antibodies against cell surface antigen. Its reliability was evaluated by comparison with two-step isolating technique for determining DR antigen of activated T lymphocyte subsets in 30 healthy volunteers. The result showed there was no statistic difference between results determined with two methods. The double-labelling technique was characterized by its samplity, no injury for cellular activity and ability for analysing the expression of the single antigen and double antigens on cell surface concurrently.

[Acrocallosal syndrome--an autopsy case report and literature review].

This is an autopsy report of a female neonate with acrocallosal syndrome, which shows developmental retardation, callosal dysgenesis and preaxial hemimelia of the upper limbs. Either dysgenesis or corpus callosum or congenital absence of radius is rare; their combination are even rarer and only eleven cases of acrocallosal syndrome have been reported in the English literature. Our case appears to be the first report in Taiwan. The family history and pregnant course were unremarkable except that her mother had taken Chinese herb medicine for common cold in the 4th pregnant month. Prenatal obstetric sonography showed "ventriculomegaly" at the 31st week. We reviewed the literature and discussed the incidence, embryology, pathogenesis, symptoms as well as etiology concerning this rare syndrome.

[Diagnostic evaluation of the determination of antibodies to purified protein derivative in tuberculosis].

The results with ELISA method showed the levels of the antibody to PPD in exudates of TB group were significantly higher than those of non-TB group (P less than 0.001). When OD value greater than 0.2 and greater than 0.35 were chosen as the positive criteria, for diagnosing tuberculous meningitis and other tuberculous, the sensitivity would be 79.3% and 86.6%, respectively; the specificity would be 93.3% and 86.0%, respectively. Meanwhile tuberculosis could be suggested when the ratio of the antibody level in exudate to that in serum is greater than 1. The authors think that the detection of the antibody to PPD in exudates might be a supplemental diagnostic tool for tuberculosis.

[A simple method for predicting equality of absorption and elimination rate constants].

A new method is proposed by which one can predict whether absorption rate constant (Ka) and elimination rate constant (Ke) are equal in the one-compartment open model with first-order processes. The method used only blood drug concentration-time data in the terminal segment of concentration versus time curve is described as follows. We defined Ai as Ai = 2/(tn + 1 + tn) + ln(cn/cn + 1)/(tn + 1 - tn) (i = 1,2,3,...) where n is the number of sampling sequence, cn and cn + 1, tn and tn + 1 are any two consecutive blood drug concentration (c)-time (t) data in the terminal segment of c-t curve, respectively. If Ai was equal or approximate, Ka and Ke might be equal or approach each other. In addition, both Ka and Ke were equivalent to the arithmetic mean of Ai. If Ai was reduced gradually while (tn + 1 + tn)/2 was increased, Ka might not be equal to Ke.

[Observation of immune function in 115 elderly patients with pulmonary tuberculosis].

This paper tested the indexes of the cellular immunity and the humoral immunity of initially treated 115 old patients with pulmonary tuberculosis, and the data were compared with these values of healthy elder and youth and middle aged patients with pulmonary tuberculosis. The results showed that the indexes of the cellular immunity elderly patients with pulmonary tuberculosis decreased (LTT Et-RFC Ea-RFC the discrepancy significantly, P less than 0.05), and the index of the humoral immunity increased (the varying of all the indexes P less than 0.01) compared with the health elders. All the immune indexes of the old pulmonary tuberculosis had no significant difference except for IgA compared with youth and middle aged patients.

Suppression of defective-sporulation phenotypes by mutations in the major sigma factor gene (rpoD) of Bacillus subtilis.

Mutations (crsA47 and crsA4) in the major sigma factor gene (rpoD) of Bacillus subtilis RNA polymerase have been found to be powerful intergenic suppressors of spoOB, spoOE, spoOF, spoOK and spoIIG mutations. The crsA47 suppressor restores sporulation of spoOE, spoOF, spoOK and spoIIG mutants to levels near those of wild type bacteria and substantially improves the sporulation of a spoOB strain. The crsA mutations are shown to prevent the induction by aliphatic alcohols of SpoO phenocopies in wild type B. subtilis cells.
