[Hydroxyproline in the rat lung following exposure to industrial ashes and surface dusts].

Experimental animals (female white rats of Wistar strain) were given intratracheally a suspension of soil dusts and electroenergetic ashes. 3 months later, hydroxyproline contents in lung tissue was determined with PROKOP'S and UNDENFRIEND'S method modified by TOMASZEWSKI and HANZLIK. The highest hydroxyproline levels were found in animals from experimental groups, which had been administered electroenergetic ashes.

Liver histology in chronic metribuzin poisoning of the guinea-pig.

Forty male guinea-pigs weighing 400-600 g, 8 months old, were given metribuzin directly into the gastric lumen over a period of 30 days (20 animals) or 90 days (20 animals), 6 times a week. The intoxicated animals showed congestion and obliteration of the trabecular structure of the liver. Acidophilic and vacuolar degeneration of the liver cells with signs of necrosis were also observed. Histochemical tests revealed a poorer staining for glycogen and succinic dehydrogenase and intensification of the alkaline and acid phosphatases reaction. The above findings indicate a progressive degeneration due to metribuzin of different organelles of the liver cells including nuclei, mitochondria, lysosomes, microvilli and the cytoplasm.

Immunologic studies of the prediabetic stage in the spontaneous autoimmune diabetes mellitus of the BB rat.

Twenty-eight BB diabetes-prone rats underwent prospective analysis to determine levels of circulating Ia antigen-bearing "activated" T lymphocytes prior to 50 days of age using mouse antirat monoclonal antibodies OX6 (anti-Ia) and W3/13 (anti-T-cell) and a fluorescence-activated cell sorter. Pancreatic biopsies were obtained within 48 hrs of lymphocyte sampling and histologically examined for the presence of a lymphocytic infiltrate. Elevated levels of "activated" T lymphocytes (above 4.00%) accurately identified a prediabetic stage and predicted which BB rats would subsequently become diabetic (93% sensitivity). The insulitis lesion occurs in the latter portion of this immunologically defined prediabetic stage just prior to the development of hyperglycemia. This observation suggests that islet cell destruction represents the final focus of a chronic immune response.

Carbohydrate, protein and glycoprotein metabolism by the guinea-pig liver in chronic metribuzin poisoning.

Male guinea-pigs weighing 400-600 g, 8 months old, were given metribuzin directly into the gastric lumen over a period of 30 days (20 animals) or 90 days (20 animals) 6 times a week. In the liver of the poisoned animals, the glycogen level and the AspAT and AlAT activities, while in the serum the total protein and the fractions albumin, alpha 1-globulin and gamma-globulin significantly decreased; serum glucose and the serum fractions alpha 2-globulin and beta-globulin, each showed an increase. The glycogen level in the liver, total protein, glucose as well as the alpha 1 and alpha 2 globulin fractions in the serum showed not appreciable difference between 30 and 90 days of intoxication. After 90 days of metribuzin treatment AspAT and AlAT dropped in the liver and rose in the serum, in comparison to the 30-day values. As to the parameters of glycoprotein metabolism, the intoxicated animals showed a significant decrease and increase in concentration of hexosamines and sialic acids in the liver and serum, respectively. Metribuzin intoxication also cause a significant decrease in activity of glucosamine phosphate isomerase and significant increase in activity of glycosidases in the liver. The results suggest that metribuzin disturbs the metabolism of carbohydrates, proteins and glycoproteins in the guinea-pig liver.

[Medical examinations of workers in the artificial leather plant in 1975-1980].

The studies of general morbidity rate and that related to circulatory, respiratory and alimentary systems covered altogether 232 subjects, in this: 151 men and 81 women. The population was evaluated taking into account: exposure factors, age, employment duration, magnitude of exposure to chemical hazards, expressed by MAC excess. The percentage indices of analysed parameters registered in the first and fourth examinations, carried out at an interval of five years, were compared. Increased general morbidity rate in men occupationally exposed to chemical hazards was found, which was particularly due to increased cardiovascular diseases rate. Women exposed to chemical hazards revealed increased morbidity rate due to alimentary tract diseases.

Glycoprotein metabolism in the guinea-pig gastric mucosa in chronic metribuzin poisoning.

Forty male guinea pigs weighting 400--600 g, 8 months old, were given metribuzin directly into the gastric lumen over a period of 30 days (20 animals) or 90 days (20 animals), 6 times a week. The intoxicated animals showed in the gastric mucosa a significant decrease in glucosamine isomerase activity and a significant increased in beta-glucosidase and beta-galactosidase activity. The results suggest that the biosynthesis of the sugar moiety of glycoproteins is depressed, the degradation of glycoproteins is stimulated by metribuzin.

Amino acid composition of the mu chains of IgM of normal serum, monoclonal cryoglobulin and Waldenstrom macroglobulin.

The purified IgM of normal human serum, monoclonal cryoglobulin and Waldenstrom macroglobulin were reduced, alkylated and subjected to Sephadex G-100 filtration. The mu chains separated were hydrolysed and analysed for amino acid content. In both monoclonal proteins the content of hydroxyproline in mu chains was lower than in the IgM of normal serum. The mu chain of monoclonal cryoglobulin contained less serine, methionine and tyrosine, whereas mu chains of Waldenstrom macroglobulin showed a lower level of cystine as compared with the IgM of normal serum. On the other hand, the content of valine and basic amino acids was higher in both monoclonal proteins.
