Noradrenergic inputs enhance the response of neurosecretory cells to osmotic stimulation.

Extracellular recordings from neurosecretory cells in the hypothalamic paraventricular nucleus (PVN) displayed either an excitation or no response following electrical stimulation (5 Hz) of the A1 noradrenergic region of the ventrolateral medulla, and exhibited either an increase or no effect in neuronal activity in response to intracarotid injections of hypertonic saline (0.2 M, 0.05 ml). The neurons responded to both the electrical and osmotic stimuli, A1 region stimulation at the subthreshold current (80-90% of the threshold required to evoke the response) significantly enhancing the facilitatory response caused by the osmotic stimulation. These results suggest that the A1 region may serve to potentiate the responsiveness of PVN neurosecretory cells in response to changes in plasma osmolality.

Increased monoamine turnover in the subfornical organ area following body fluid depletion.

To clarify whether monoaminergic inputs to the subfornical organ (SFO) area participate in fluid regulatory systems, we examined the effects of body fluid depletion on monoamine turnover in the region of the SFO using microdialysis techniques in rats. An iso-osmotic reduction of fluid volume following subcutaneous treatment with polyethylene glycol (PEG) significantly increased dopamine (DA), 3,4-dihydroxyphenylacetic acid (DOPAC), and 5-hydroxyindoleacetic acid (5-HIAA) concentrations in the SFO area. 5-hydroxytryptamine (5-HT) in the SFO area could be detected after the PEG treatment, while 5-HT was undetectable before the treatment. The data imply that both dopaminergic and serotonergic systems in the SFO area may be involved in controlling body fluid balance.

Noradrenergic excitatory inputs to median preoptic neurones in rats.

Extracellular single-unit activity was recorded from 21 median preoptic nucleus (MnPO) neurones, antidromically identified as projecting to the hypothalamic paraventricular nucleus (PVN), in urethane-anaesthetized male rats. Of these identified MnPO neurones, 14 displayed an excitatory response in neuronal excitability following electrical stimulation (5 Hz, 600 microA) of the A1 noradrenergic region of the ventrolateral medulla, while the remaining neurones were unresponsive. The excitatory response of MnPO neurones was blocked by microiontophoretically applied phentolamine, an alpha-adrenoceptor antagonist, but not by timolol, a beta-adrenoceptor antagonist. These results suggest that the A1 region acts to enhance the activity of MnPO neurones projecting to the PVN via an alpha-adrenoceptor mechanism.

A proposal of ovarian ERAANPS (endothelin-renin-angiotensin-atrial natriuretic peptide system) and effects of tokishakuyakusan, keishibukuryogan, shakuyakukanzoto and unkeito on the ERAANPS.

We have previously shown the presence in a high concentration of endothelin-1 (ET) in the corpus luteum and renin-angiotensin system (RAS) and binding sites for atrial natriuretic peptide (ANP) in the ovarian follicle. The present study was undertaken to identify the existence of ET, renin, angiotensin II and the binding site for ANP in the ovary at proestrus and examine in vivo the effects of herbal medicines [Tokishakuyakusan (TS), Keishibukuryogan (KB), Shakuyakukanzoto (SK) and Unkeito (UT)] on them. ET, all components of RAS and binding sites for ANP were found at high levels in the ovary. TS, KB, SK and UT decreased the ET levels in ovary, while components of RAS and binding sites for ANP have the propensity to increase. However, ET, renin, angiotensin II and ANP levels in plasma were not at all affected before and after treatment with TS, KB, SK or UT. Taken together with previous observations showing the existence of ET, RAS and the binding site for ANP in the ovary, we propose here the ERAANPS (endothelin-renin-angiotensin-ANP system) in the ovary as a functional regulator. Further, these results suggest that TS, KB, SK or UT may regulate the ovarian ERAANPS.

Effects of tokishakuyakusan, keishibukuryogan, shakuyakukanzoto and unkeito on ovarian endothelin, renin and angiotensin II in pregnant mare's serum gonadotropin-treated immature rats.

We have previously proposed the ovarian ERAANPS (endothelin-rein- angiotensin-atrial natriuretic peptide system). The present study was undertaken to examine in vivo the effects of herbal medicines [Tokishakuyakusan (TS), Keishibukuryogan (KB), Shakuyakukanzoto (SK) and Unkeito (UT)] on endothelin-1 (ET), renin and angiotensin II (A II) in the ovaries, of immature rats treated with 10 IU PMS for 48 h. ET and all components of renin-angiotensin system (RAS) were found at high levels in the ovary. Concomitant treatment with PMS plus TS, KB, SK or UT, especially TS and UT, tended to decrease the ET levels in ovary, while components of RAS tended to increase. However, ET, renin and A II levels in plasma were not at all affected after treatment with TS, KB, SK or UT. These results suggest that TS, KB, SK or UT may regulate the ovarian ERAANPS.

Cell kinetics of regenerating liver after 70% hepatectomy in rats--2-color flow cytometric analysis.

Two-color flow cytometric (FCM) analysis using anti-bromodeoxyuridine (BrdU) monoclonal antibody (MoAb) was used to investigate the cell kinetics of regenerating liver after 70% partial hepatectomy in rats. Three peaks were seen in DNA histograms of rat hepatocyte nuclei, corresponding to diploid(2c), tetraploid(4c), and octaploid(8c). These proportions changed in the course of regeneration which were clearly demonstrated by DNA histograms using flow cytometry. The proportion of diploid, tetraploid, and octaploid nuclei in control liver were 49.3 +/- 1.6%, 45.0 +/- 7.4%, and 1.7 +/- 0.7%, respectively. A significant change occurred at 24 hours after hepatectomy, as FCM revealed 25.9 +/- 1.1% diploid, 54.5 +/- 1.2% tetraploid, and 9.0 +/- 0.9% octaploid. This shift to polyploid nuclei persisted until 72 hours, and then gradually returned to the pattern of control liver. The S-phase nuclei which incorporated BrdU increased rapidly at 24 hours to a peak of 11.3 +/- 0.9%, and gradually decreased to 5.8 +/- 0.8%, 5.3 +/- 0.8%, 2.4 +/- 0.6%, 2.9 +/- 1.1%, and 1.2 +/- 0.6%, at 48, 72, 96, 120, and 168 hours, respectively. This 2-color FCM analysis made a detailed analysis of the cell kinetics in regenerating hepatocytes possible, and may be applied in investigations of various aspects of liver regeneration.

Synthesis and antitumor activities of mitomycin C (1----3)-beta-D-glucan conjugate.

The conjugate of mitomycin C (MMC) with linear (1----3)-beta-D-glucan from Alcaligenes faecalis var. myxogenes IFO 13140 was synthesized and its antitumor activities investigated. The conjugate (MMC-carboxymethylated linear (1----3)-beta-D-glucan (CMPS)) was obtained by treatment of CMPS with MMC in the presence of 1-ethyl-3-(3-dimethylaminopropyl)-carbodiimide. In vitro cytotoxicity of MMC-CMPS against L1210 leukemia cells was similar to that of MMC. In i.p.-i.p. system in vivo against P388 leukemia in mice, the maximum increase of MMC-CMPS conjugate in life span (ILSmax) was higher than that of MMC but the therapeutic index was reduced. However, the antitumor activity of MMC-CMPS conjugate against subcutaneously implanted sarcoma 180 solid tumor in mice by i.p. administration was similar to that of MMC at a dose of 1.5 mg eq MMC/kg/d x 7 and the reduction of the number of leukocytes caused by MMC was suppressed by attaching MMC to CMPS. In addition, on assay using serum of sarcoma 180 solid tumor-bearing mice with injection of MMC-CMPS conjugate, a drastic loss of tumor cells and an increase in polymorphonuclear leukocytes (PMN) were observed. This result suggested that MMC-CMPS conjugate induced tumor-regressing factor similar to CMPS.

Characterization of the subunits and sugar moiety of human placental and leukemic beta-glucuronidase.

beta-Glucuronidase purified from human placenta and chronic myelogenous leukemic cells was composed of three components of 18, 64 and 80 kDa, though the relative contents of the components were different between the sources. Analysis of their N-terminal amino-acid sequences showed that the 18-kDa and 64-kDa components were derived from the 80-kDa component by cleavage between Val159 and Gly160. Furthermore, the enzyme was found to be glycosylated at Asn173 and Asn420 with high mannose-type oligosaccharides, based on the electrophoretic mobility of the components as well as the endopeptidic peptides before and after endoglycosidase treatment. The enzyme purified from leukemic cells was poorly phosphorylated by N-acetylglucosamine 1-phosphotransferase as compared to the placental enzyme.

Identical 24-hour intragastric pH response to low continuous infusion doses of famotidine in active gastric ulcer patients.

UNLABELLED: In order to establish the therapeutic plasma concentration of famotidine, the 24-h intragastric pH response to low-dose intravenous (i.v.) continuous infusion of 40 (group A) or 20 mg/day (group B), administered over more than two days, was studied in 10 adult patients with active gastric ulcers (GU). In group A, the mean age was 64.8 years and in group B, 54.2 years. On the second day, 4 blood samples were collected for famotidine assay and 24 h intragastric pH monitoring was performed. RESULTS: the mean 24 h pH did not differ significantly (P less than 0.01) between group A at 6.9 and group B at 7.2. In group A, the percentage of time with pH above 6.0 and above 7.0 were 94.8 and 59.0, respectively, and the mean famotidine dose and plasma concentration were 0.81 mg/kg/day and 140.89 ng/ml. In group B, the percentage of time with pH above 6.0 and 7.0 were 97.6 and 65.4, and the mean dose and famotidine concentration were 0.34 mg/kg/day and 45.42 ng/ml. In conclusion, in fasting patients with active gastric body ulcers, continuous infusions of low-dose famotidine maintain both the therapeutic plasmatic concentration and the intragastric pH to near anacidity level.

Does oxygen supply improve graft viability in liver preservation?

While the clinical results of orthotopic liver transplantation have greatly improved, the viability of liver grafts and extension of the safe time for preservation are necessary factors in need of improvement. The liver is one of the organs most sensitive to anoxia. The addition of an oxygen carrying agent to the preservation solution was evaluated. Pyridoxalated hemoglobin-polyoxyethylene conjugate (PHP) is used as an oxygen carrier. Viaspan (UW) served as a control solution. Test solution (PHP+UW) composition was composed of a 1:1 mixture of PHP and UW solutions with hemoglobin 4.0g%, hydroxyethyl starch 2.5g%, osmolality 320 mOsm/kg H2O, and colloidal osmotic pressure 33 mmHg. The oxygen carrying capacity of PHP+UW solution is about 10 times higher than UW solution at 4 degrees C. Male Lewis rats (BW: 250-300 g) were divided into five groups. After flushing the solution via the portal vein, rat livers were harvested. Two preservation methods, simple storage and perfusion (0.1 ml/min/g liver), were studied at 4 degrees C for 24 or 48 hours. OxyHb, MetHb, pO2, pH, Na, K, GOT, and GPT of perfusate, hepatic mitochondrial functions after preservation, and tissue adenine nucleotides by HPLC were measured. Light microscopy on the tissue was also performed. No significant differences were noted in perfusate biochemical parameters. Oxygen consumption during the perfusion was significantly higher in the PHP+UW than in the UW group. Hepatic mitochondrial functions and tissue ATP levels were better preserved in perfusion than in simple storage, and in PHP+UW than in UW at 48 hours. The oxygen carrying agent, PHP, can provide significantly higher levels of oxygen to liver grafts and improve graft viability.

[Clinical characteristics and prognosis of renal cell carcinoma. Statistical evaluation of possible determinants for distant metastasis, venous tumor thrombi, and lymph node metastasis].

To clarify the recent clinical characteristics of renal cell carcinomas and to evaluate possible determinants for metastasis and venous tumor thrombi, the authors reviewed data from 99 renal cell carcinoma patients treated at Nagoya University Hospital between 1980 and 1989. According to Robson's classification, stage I tumors were found in 48 patients, stage II in 9, stage III in 16, and stage IV in 26. Incidentally detected tumors appeared to be on the increase in recent years. Grade 1 tumors were significantly associated with low-stage tumors and expansive growth. Univariate and multivariate analyses using a logistic regression model demonstrated that venous tumor thrombi and histological grade were significantly related to distant metastasis. Univariate analysis revealed relative risks of 4.7 for venous tumor thrombus presence (pV1b-pV2 vs. pV0-pV1a, p = 0.005) and 8.5 for histological grade (grades 2 and 3 vs. grade 1, p = 0.04). Local invasion (pT3 vs. pT2a-pTb: a relative risk of 7.5, p = 0.0009) and infiltration pattern (INF beta and INF gamma vs. INF alpha: a relative risk of 11.5, p = 0.002). were associated with venous tumor thrombi. Local invasion (pT3 vs. pT2a-pT2b: a relative risk of 6.6, p = 0.03) was the only significant determinant for lymph node metastasis. The 5-year actuarial survival rate was 60.0% for all 99 patients. The 5-year survival rates for stage I and II tumors were, respectively, 91.8% and 64.8%.(ABSTRACT TRUNCATED AT 250 WORDS)

Expression of genes encoding type IV collagen-degrading metalloproteinases and tissue inhibitors of metalloproteinases in various human tumor cells.

Uncontrolled expression of matrix metalloproteinases 2, 3 and 9 (MMP-2, -3 and -9) is believed to be a critical part of the invasive potential of tumor cells because of their ability to degrade type IV collagen, a major structural component of basement membranes. Availability of proteolytic activity in the vicinity of the cell surface is further affected by a local balance between the enzymes and their inhibitors produced by the cell. To determine how frequently deregulated expression of the MMPs and tissue inhibitors of metalloproteinases (TIMPs) is associated with tumor cells, 26 human tumor cell lines were examined by Northern blotting. Transcripts for MMP-2 and MMP-9 were more frequently expressed in mesenchymal tumor cells (9/9 for MMP-2 and 6/9 for MMP-9) than in epithelial tumor cells (4/17 for MMP-2 and 2/17 for MMP-9). Although expression of MMP-2 mRNA was clearly cell type-specific, MMP-9 mRNA expression in mesenchymal cells correlated well with the reported tumorigenicity of the cells. Enhanced expression of MMP-9 mRNA was also associated with the tumorigenic transformation of cells by an activated c-H-ras gene in human embryonic fibroblasts. Only 3 of the 26 tumor cells expressed MMP-3 mRNA, and 2 of the 3 were epithelial tumor cells which coordinately expressed MMP-9 and TIMP-1 mRNAs. TIMP-1 mRNA was almost undetectable in 50% of the tumor cells, but TIMP-2 mRNA was expressed in the majority of the cells. These findings provide comprehensive information about mRNA expression of the MMPs and TIMPs in tumor cells, the deregulation of which is thought to be an integral part of the invasive potential of tumor cells.

[Tyrosine hydroxylase-immunohistochemical study in the midbrain of experimental MPTP parkinsonism].

We have performed tyrosine hydroxylase (TH)-immunohistochemical study in the midbrain of experimental MPTP parkinsonism. Eight female adult crab-eating monkeys (macaca fascicularis) were subjected to the experiment and divided into four groups. In the acute experimental parkinsonism (group A), a daily dose of 1.3 mg/kg MPTP was given intravenously for 7 days. Monkeys in the subacute group B were injected every other day a dose of 1.0 mg/kg MPTP intravenously for 7-14 days. Chronic experimental parkinsonism group C was induced by administration of a dose of 0.5 mg/kg twice a week intravenously for 120 days. Group D served as a control. TH-immunoreactive cells were counted in substantia nigra pars compacta (SNc: consisting of lateral, intermediate, medial regions) and paramedian region of midbrain. The remaining TH-immunoreactive cells in group A,B and C were approximately 65.1%, 46.6% and 32.1% of the control group D. The ratio of TH-immunoreactive cells in the lateral region of group A, B and C was lower than in group D. The TH-immunoreactive ratio in the intermediate and medial region of group B and C was lower than in group D. In the medial region of group C, the TH-immunoreactive ratio was increased than in group B. In the paramedian region of all groups, there was no significant differences in the ratio of TH-immunoreactive cells, but many more TH-immunoreactive nerve fibers were observed in group B and C than in group A and D. From the 90th day after MPTP treatment, monkeys in group C began to show a gradual recovery from the flexed posture and hypokinesia.(ABSTRACT TRUNCATED AT 250 WORDS)

[A case of verrucous carcinoma of the tongue, effectively treated with preoperative chemotherapy (UFT, CDDP, PEP) and irradiation].

A 70-year-old male complained of a mass on the left side of his tongue. A biopsy specimen was diagnosed as verrucous carcinoma. He was treated with preoperative chemotherapy (UFT, CDDP, PEP) and irradiation, continuous partial glossectomy. The tumor response to chemotherapy was excellent, and no tumor cells were found in the surgical specimen. There was no recurrence of the tumor 1 year and 7 months after therapy. For advanced tumors, preoperative chemotherapy appears to be significantly effective.

[Long-term follow-up of clinical features and ECG changes in 50 patients with HCM].

We studied the relation of ECG changes during the clinical course in patients with hypertrophic cardiomyopathy (HCM). Fifty patients with HCM were categorized in two groups; 20 patients with, and 30 patients without signs of deterioration of clinical state. The changes between the first and the final ECG were compared in these two groups. The average follow-up period was 7.6 years. Twenty patients with clinical deterioration presented increase in QRS intervals (0.06----0.071 sec, p less than 0.025), decrease in voltage of RV5 (2.81----2.38 sec, p less than 0.025), increase in newly developed abnormal Q waves, and P wave changes. In contrast to these cases, 30 patients without clinical deterioration presented no significant ECG changes. Amplitudes of the negative T wave were unchanged in both groups. We conclude that electrocardiographic observations proved to be useful for predicting clinical features in patients with HCM.
