3-anilino-L-alanine, structural determination of UV-5, a contaminant in EMS-associated L-tryptophan samples.

An aniline derivative which corresponds to UV-5 in the preceding paper was isolated from the case L-tryptophan sample associated with eosinophilia-myalgia syndrome (EMS). By spectroscopic analyses, the structure was identified as 3-anilinoalanine. The compound was optically active, and the stereochemistry of alanine moiety was determined as L, by comparing the specific rotation with a synthesized 3-anilino-L-alanine.

Characteristics of ventricular activation and recovery patterns in the rat.

Bipolar or unipolar epicardial and intraseptum electrograms were recorded from the in situ rat hearts with spontaneous sinus rhythm or myocardial electrical stimulation of 400 bpm in order to investigate the sequence and duration of cardiac activation in the rat. The ventricular epicardial isochronous maps in the rat showed the activation sequence comparable to those in humans and dogs, although the activation time (6 to 13 msec) of epicardium was approximately one-third of those of such species. The delay of activation between septum and ventricular surface in the rat was 5 to 8 msec, which was also shorter as compared with those in humans and dogs. However, the conduction velocity of the ventricular epicardium in the rat was approximately 40 cm/sec, mostly equal to that of humans. These findings might be accounted for by the possible presence of the bundle branch-purkinje system, at least functionally, similar to humans. The local QT interval in the septum were relatively longer (69 to 123 msec), compared with that of the ventricular surface (50 to 98 msec), and such durations of ventricular epicardium and septum were considerably less than those of dogs and humans. These results are consistent with the configuration of QRS-T on the limb lead ECG.

Clinical analysis of ten cases with surgically treated brain stem cavernous angiomas.

Ten cases of surgically treated brain stem cavernous angiomas are reported in this paper, and their clinical and radiological features are reported. There were 9 males and 1 female with ages ranging from 9 to 69 years (average 36 years). Four of the cavernous angiomas were located in the dorsal pons, two in the middle cerebellar peduncle, two in the hypothalamus, and one each in the cerebral peduncle and the superior colliculus. All the ten patients came to medical attention because of the sudden onset of neurological abnormalities following brain stem hemorrhage. Six of the 10 patients showed neurological deterioration due to rebleeding at various intervals ranging from 10 days to 2 years. After intravenous injection of iodinated contrast materials, negative contrast enhancement was commonly seen. In a few cases, there was faint enhancement of the nodule at the periphery of the hematoma. Serial computed tomography (CT) scans over a period revealed an unchanged hyperdense mass lesion, which was thought to be a most characteristic and indicative CT feature of the cavernous angioma. On magnetic resonance imaging (MRI), the combination of a mixed signal intensity with a surrounding rim of decreased signal intensity (hemosiderin rim) suggested the presence of a cavernous angioma. Enhanced MRI, using gadopentate dimeglumine, was thought to be more sensitive than contrast enhanced CT in detecting cavernous angiomas. All these cases underwent surgery with good results. Abnormal vascular tissue in the wall of the hematoma cavity was recognized at operation and pathological confirmation was obtained in all cases. These data indicate that brain stem cavernous angiomas can be surgically removed with acceptable morbidity.

Experimental investigation of a new liquid embolization method: enhancing effect of 25% ethanol on conjugated estrogen.

A new liquid embolization material applicable to intracranial lesions was evaluated by selective renal artery embolization in mongrel dogs. Absolute and diluted ethanol, estrogen, and estrogen in 25% ethanol (ethanol-estrogen) were injected. The embolization and side effects were investigated angiographically and histologically. Ethanol-estrogen caused similar embolization to absolute ethanol, but no direct damage to perivascular tissue, and is considered a promising embolization agent in intravascular surgery.

Pyrimidine nucleoside monophosphate kinase isolated from pig brain homogenate catalyzes disproportionation of phosphate between two CDP molecules.

An enzyme fraction that catalyzes CTP formation from CDP was purified from pig brain homogenate to a single band on SDS-PAGE. The preparation had a molecular weight of about 36,000 and showed a high activity of phosphorylating CMP and UMP by ATP and turned out to be one of pyrimidine nucleoside monophosphate kinases. It also catalyzes UTP formation from UDP, although rather weakly. These characteristics show that the enzyme species from pig brain homogenate has a rather broad specificity toward phosphate donor in phosphorylating nucleoside monophosphate.

[13-week subchronic toxicity study of 1,1-bis(t-butylperoxy) 3,3,5-trimethyl cyclohexane in mice].

A 13-week subchronic toxicity study of 1,1-bis(t-butylperoxy)3,3,5-trimethyl cyclohexane (TMCH) was performed in male and female B6C3F1 mice by feeding a CRF-1 powder diet containing 0, 0.5, 1.0, 2.0 and 4.0% TMCH, to determine the maximum tolerable dose (MTD) for subsequent carcinogenicity study. Mice were randomly divided into 5 groups, each consisting of 10 males and 10 females. Eight animals of both sexes in the 4.0% TMCH group died during the early administration period. Suppression of body weight gains was marked in both sexes of the 4.0% and 2.0% TMCH groups as compared with the control group. The mean food consumption in all groups treated with TMCH was lower than that in the control group in a dose-related manner. Hematological examination revealed decreases in the number of erythrocytes, volume of hemoglobin, and values for hematocrit and mean corpuscular volume (MCV) in both sexes of the 4.0% and 2.0% TMCH groups. The relative liver weights of all groups treated with TMCH increased in a dose-related manner, whereas both absolute and relative spleen weights decreased in a dose-dependent manner. Histopathologically, swelling of the hepatocytes was found centrilobularly or diffusely in both sexes of the 1.0, 2.0 and 4.0% TMCH groups, and decrease of the hematopoietic cells in the bone marrow was observed in both sexes of the 2.0 and 4.0% TMCH groups. In addition, atrophy of red pulp and white pulp of the spleen was found in both sexes of the 2% and 4% TMCH groups.(ABSTRACT TRUNCATED AT 250 WORDS)

Characterization of murine hemopoietic-supportive (MS-1 and MS-5) and non-supportive (MS-K) cell lines.

Characteristics of hemopoietic-supportive (MS-1 and MS-5) and non-supportive (MS-K) cell lines were compared. Supportive cells adhered to hemopoietic stem cells and produced granulocyte-macrophage colony-stimulating factor (GM-CSF), whereas non-supportive cells did not adhere to hemopoietic cells and only produced macrophage colony-stimulating factor. Both cell lines produced substantial levels of IL-6 and steel factor (SLF) which is reportedly a stem-cell factor. Northern blot analysis revealed that SLF but neither c-kit nor interleukin 3 (IL-3) mRNA was detectable in these cell lines, although IL-3-like activity was found in the supernatant of MS-5 cell culture. These observations suggest that the hemopoietic-supportive function of stromal cells may reside in adherence of stem cells, and production of GM-CSF probably in combination with SLF. SLF may be transferred from stromal cells directly to stem cells through adhesion of stem cells to supportive stromal cells.

[Gene rearrangement analysis of conjunctival malignant lymphomas].

Specific DNA probes for genes encoding immunoglobulins (Ig) and the T cell receptor (TCR) are useful diagnostic tools in lymphoproliferative disorders. Gene rearrangement analysis was carried out in 2 cases of conjunctival lymphoid lesions. A 36-year-old man (case 1) had a 1-year history of left conjunctival tumor. A biopsy was performed and histopathological findings showed diffuse proliferation of small lymphocytes, but monoclonality was not revealed by immunophenotypic analysis. A right conjunctival lesion developed and five months later a biopsy of the left conjunctiva was performed again. A frozen sample was analyzed and immunoglobulin heavy chain gene rearrangement was found. A 53-year-old woman (case 2) had a 6-month history of bilateral conjunctival tumor. The first biopsy did not reveal monoclonality immunophenotypically. A second biopsy with a frozen specimen was analyzed and immunoglobulin heavy chain gene rearrangement was found. We diagnosed these two cases as B-cell lymphoma. We discuss the clinical value of gene rearrangement analysis as a diagnostic method for lymphoproliferative disorders.

Apolipoprotein(a) is present in urine and its excretion is decreased in patients with renal failure.

To investigate the relation between renal function and concentrations of lipoprotein(a) [Lp(a)] in serum, we measured Lp(a) in samples of serum and urine from patients with diabetes mellitus and in samples sent to a laboratory center for measurements of creatinine clearance. Serum Lp(a) concentrations were significantly increased in subjects with obvious renal dysfunction (serum creatinine > or = 176.8 mumol/L) compared with normal control subjects. Urinary Lp(a) excretion was decreased in subjects with obvious renal dysfunction compared with subjects without obvious renal dysfunction (serum creatinine < or = 88.4 mumol/L) and was negatively and positively correlated with serum creatinine and creatinine clearance, respectively. More than 80% of urinary Lp(a) was recovered in the d > 1.21 kg/L fraction. At least six bands for apolipoprotein(a) [apo(a)] fragments, which were smaller than native apo(a) in serum, were observed in urine by immunoblotting, and some of these were also detected in serum. Degraded apo(a) fragments are probably present in urine, and their excretion decreases in parallel with decreases in the glomerular filtration rate.

[A case of superior vena cava syndrome due to extensive venous thrombosis of an idiopathic nature].

A 37-year-old man was admitted with facial edema and right arm swelling. Venography, computed tomography and magnetic resonance imaging showed massive organizing thrombi in the superior vena cava and bilateral internal jugular as well as subclavian veins, and showed no mass lesions occluding the veins in the mediastinum. Angioscopy demonstrated a white thrombus at the entrance of the right subclavian vein. All results of blood coagulation tests were normal. The patient was diagnosed as having superior vena cava syndrome caused by idiopathic venous thrombosis. Anti-coagulant therapy with intravenous tissue plasminogen activator injection and continuous urokinase and heparin infusion into the thrombi through a catheter were not effective in lysing the thrombi. Collateral circulation gradually developed and his symptoms decreased. We decided to follow this patient on warfarin medication because of the difficulty in removing the thrombi surgically.

[A fatal case of acute mediastinitis caused by periodontal infection].

A 57-year-old man had suffered from poorly controlled diabetes mellitus and liver cirrhosis due to alcohol and hepatitis C for about 10 years. He developed fever and swelling of the right cheek and neck due to periodontal infection. The symptoms worsened in spite of antibiotic therapy and were accompanied by dyspnea. He was therefore referred to our hospital. Chest radiographs and computed tomographs revealed widening of the superior mediastinum, pulmonary infiltrates and right pleural effusion. He was diagnosed as having mediastinitis, right pyothorax and pneumonia caused by periodontal infection. Tracheotomy and mechanical ventilation were performed. Antibiotic therapy resulted in improvement of the mediastinitis and pyothorax. However, renal and liver dysfunction developed and the patient died of multiorgan failure after 35 days of hospitalization. Death due to periodontal infection is rare. We give a review of the literature.

[Practical use of mortality statistics and mass examination results by health workers in the local authority of the community].

In Kochi prefecture, the prefectural health service bureau tabulates both mortality statistics and mass examination results for each community and the tabulated report is sent annually to public health services staff in each local authority. While recognizing its limitations, methods of how to use the annual report are demonstrated. For example comparison of the mortality statistics and the mass examination results of the male population of A town, which is administered by Susaki Health Center, to that of the combined male population of all the towns administered by the health center shows that although the mortality rate for cerebrovascular disease for the A town-male population had been much higher than that of the combined male population in the early 1980's, the difference disappeared in the late 1980's. On the other hand, prevalence of systolic hypertension in the A town-male population continued to be higher than that for the combined male population in the late 1980's. Therefore hypertension prevention programs appears to still have significance in the A town-male population. Certain weaknesses exist in this annual report. In the comparison of mass examination results among different populations, the presence of selection bias of those receiving examinations should be considered. Standardization of mass examination procedures and quality controls of the examinations should also be considered. To get health workers to utilize the annual report further, the reported items should be improved. Particularly useful would be tabulation by residential subdivision and occupation.

Assay of urinary free fucose by fluorescence labeling and high-performance liquid chromatography.

The concentrations of free fucose and other sugars in urine of cancer patients and healthy subjects were analyzed by high-performance liquid chromatography. After the urine samples were dried, we coupled the sugars in the residue with 2-aminopyridine to be detected as fluorescent derivatives. We then analyzed the pyridylamino derivatives of the sugars with an anion-exchange column and borate buffer. The difference between cancer patients and healthy subjects for the mean concentrations of fucose corrected for creatinine was significant (P less than 0.001). We checked the relationship between the concentrations of other sugars and the presence of cancer. This method is highly sensitive, and neither a cleanup procedure before labeling nor purification before injection into the column is needed. Not only fucose but also other sugars can be detected simultaneously, so this method should be useful for studying any changes in sugars in urine in various diseases.

Effect of thyroid hormone on in vivo contractility of the canine diaphragm.

This study was designed to examine the effects of long-term (4 wk) administration of thyroid hormone on the in vivo contractility of the canine diaphragm. We implanted a pair of piezoelectric crystals chronically in the left crural and costal parts of the diaphragm by a midline laparotomy. Contractility was assessed by changes in the shortening of muscle fibers after twitch stimulation of both the crural and the costal parts of the diaphragm and in the transdiaphragmatic pressure (Pdi) after tetanic stimulation (10 to 100 Hz). As a reference, we also studied the response of the quadriceps femoris. Pretreatment measurements were taken 2 wk after surgery. Then, dogs assigned to the hyperthyroid group were given thyroid powder, 0.6 g/kg/day, orally for 4 wk. The control group was fed a diet without thyroid powder for 4 wk. Serum free-T4 level (RIA) in the hyperthyroid group (n = 9) increased from 0.68 +/- 0.07 to 5.72 +/- 0.95 ng/dl (mean +/- SE) (p less than 0.01). Pdi decreased 30 to 40% at all frequencies (p less than 0.05) except 10 Hz. Twitch shortening of the crural and costal parts, compared with pretreatment state, decreased significantly by 47.7 +/- 13.1 and 48.1 +/- 15.0%, respectively (p less than 0.05). The maximal rate of relaxation became significantly faster, by 63.5% (p less than 0.05), in the crural part, whereas that of the costal part tended to become faster but not to a significant extent. In the quadriceps femoris, although twitch force showed no change, both the maximal rate of contraction and maximal rate of relaxation became faster, and tetanic force decreased. Histologic examination of hyperthyroid dogs showed vacuolization and loss of fiber area of the diaphragm. These observations suggest that thyroid hormone impairs contractility of both the crural and costal parts of the diaphragm similarly, and that the decrease in contractility may be due to a loss of muscle mass and summation impairment of twitch contraction, which differs from that in other skeletal muscles.

Molecular cloning of a porcine gene syk that encodes a 72-kDa protein-tyrosine kinase showing high susceptibility to proteolysis.

By using antibodies raised against a portion of N terminus of 40-kDa kinase (Kobayashi, T., Nakamura, S., Taniguchi, T., and Yamamura, H. (1990) Eur. J. Biochem. 188, 535-540), not only 40-kDa protein but also 72-kDa protein were detected on immunoblot analysis of porcine spleen homogenate. In splenocytes preparation, the antibodies could immunoprecipitate protein-tyrosine kinase activity of the 72-kDa protein but not detect the 40-kDa protein even on immunoblot. After incubation of crude spleen homogenate at 37 degrees C with or without various protease inhibitors, immunoblot analysis revealed proteolytic breakdown of the 72-kDa protein to 40-kDa fragment. Next, using oligonucleotides designed according to partially sequenced information of the 40-kDa kinase as a probe, we have isolated a clone containing entire coding sequence for the 40-kDa kinase from a porcine spleen cDNA library. This clone had a 1884-base-pair-long open reading frame encoding 628-amino-acid polypeptide with a calculated molecular weight of 71,618. The deduced amino acid sequence did not contain a ligand binding or membrane spanning region but did a well-conserved protein-tyrosine kinase domain and two src homology region 2 domains. The sequences of these domains showed 30-40% identity to those of other protein-tyrosine kinases, but those of remaining parts were quite unique. From these results, we concluded that the 40-kDa kinase was generated by proteolysis from the 72-kDa holoprotein which was a new member of nonreceptor protein-tyrosine kinase so far reported. We therefore designated this gene as syk after spleen tyrosine kinase.
