Electron microscopic observation of bFGF immunoreactivity in the hippocampus.

Light and electron microscopic observation showed two types of neuronal immunostaining for basic fibroblast growth factor in the hippocampal CA2 subfield, where the densest immunoreactive neurons were localized in the brain. One neuronal type showed intense nuclear (eu- and heterochromatin) immunostaining but weak cytoplasmic immunostaining (N-type), and the other showed intense cytoplasmic but no or only faint nuclear immunoreactivity (C-type). The N-type also showed weak immunoreactivity in the perinuclear rough endoplasmic reticulum and contained bFGF mRNA as observed by in situ hybridization histochemistry, showed that this type can produces the bFGF protein. The N-type localized exclusively in the CA2 subfield. The C-type showed strong immunoreactivity on the rER, free ribosomes, and Golgi apparatus, although no clear evidence for bFGF production was observed. The multivesicular bodies, a pathway of endocytosis in hippocampal neurons showed apparent immunoreactivity under EM observation of both of types neurons (Parton et al. J. Cell Biol. 119: 123-137, 1992) suggesting a receptor-mediated type of incorporation of the bFGF.

[The etiology of peptic ulceration in patients with chronic pulmonary emphysema].

The purpose of this study was to determine the etiology of peptic ulceration in patients with pulmonary emphysema. We performed endoscopy in 50 patients with chronic pulmonary emphysema, these were patients with and without peptic ulcer. There was no significant differences between the patients with and without ulcer, with respect to past history of pneumonia, degree of dyspnea, current drugs, nutritional status, the extent of gastric mucosal atrophy, gastric mucosal blood flow, gastric mucosal hexosamine content and serum pepsinogen levels. But patients with peptic ulcer were significantly younger than patients without peptic ulcer. Cigarettes per day in patients with peptic ulcer 29.3 +/- 11.6 cigarettes/day were significantly higher than those without peptic ulcer 22.5 +/- 10.0 cigarettes/day. We concluded that increased incidence of peptic ulceration in patients with chronic pulmonary emphysema is related to the quantity of cigarette smoking.

[Pigmentation on the anterior chamber angle in retinal detachment associated with atopic dermatitis].

We reported pigmentation on the anterior chamber angle in twenty-two patients suffering from atopic dermatitis. Dense pigmentation on the anterior chamber angle was observed in all eight eyes with retinal detachment and was evident before clinical recognition of detached retina. Minimal or slight pigmentation was also evident in eyes without retinal detachment. Increased pigmentation on the anterior chamber angle is a characteristic and predictive sign of retinal detachment associated with atopic dermatitis. This pigment appeared to migrate from latent breaks because of inflammation of the peripheral retina and ciliary body.

[A case of bronchiectasis accompanied by ulcerative colitis (UC) and HTLV-1 associated myelopathy (HAM)].

We report a case of bronchiectasis with marked thickening of the respiratory tract wall occurring in a 37-year-old man with UC and HAM. He was diagnosed as UC at age 20. HTLV-1 was presumably transmitted to this patient by means of a blood transfusion he received at around age 30. On admission, chest X-ray films, tomography and CT-scan revealed dilated lumens and thickened airway walls extending from the trachea to subsegmental bronchi. Pulmonary function tests showed hypoxemia and mixed ventilatory disturbance with a predominantly obstructive component. HAM was diagnosed on the basis of neurological examination and cerebrospinal fluid analysis. A biopsy specimen from the carinal mucosa showed marked T cell infiltration. In these T cells, we detected polyclonal integration of HTLV-1 proviral DNA. Some of the infiltrating T cells showed atypia. In recent times, respiratory diseases other than infiltration of adult T-cell leukemia cells or opportunistic infection have been reported in HTLV-1 carriers and new clinical entities designated as HABA (HTLV-1 associated bronchiolo-alveolar disorder) and HBA (HTLV-1 associated bronchopneumopathy) have been proposed. This case is classified among these new entities, in a broad sense, and is a rare case in that the respiratory disorder is apparently related to UC.

Sound evaluation of partially implantable piezoelectric middle ear implant: comparative study of frequency responses.

Two comparative studies were performed in six subjects using partially implantable middle ear implants (MEI). The sound quality was evaluated by speech discrimination tests and sound evaluation tests. In the first experiment, the performance of the output transducer of the MEI and an ear phone of a conventional hearing aid was compared. The MEI showed better results than the hearing aid in both the speech discrimination tests and sound quality rating. In the second experiment, four kinds of frequency responses for the external component of the MEI were compared. Sound quality evaluation in a wide frequency response with a peak at 4000 Hz were superior to those with the frequency response of the present MEI model. In conclusion, improvement of the external component in the frequency response can provide better performance of the MEI.

[A 13-week subchronic toxicity study of bisphenol A in B6C3F1 mice].

A 13-week subchronic toxicity study of bisphenol A (BPA) was performed in male and female B6C3F1 mice at dose levels of 0, 0.2, 0.5, 1.0, 2.0 and 4.0% in the diet, to facilitate dose selection for a subsequent carcinogenicity study. Mice were randomly allocated to 6 groups, each consisting of 10 males and 10 females. Two 0.2% group males and two 4.0% group females died during the experimental period. Suppression of body weight gain and increase in food consumption were observed in males and females of the 4.0% groups. Hematological examination revealed decrease in number of erythrocytes, volume of hemoglobin and value of hematocrit in males and females of the groups receiving 1.0% or above, and an increase in number of platelets in males of 4.0% group. Decrease in number of erythrocytes and hematocrit value was also noticed in females of 0.5% group. Histopathologically, cystic dilatation, degeneration or regeneration of renal tublues were found in males and females of 1.0% or higher groups, multinucleated hepatocytes were increased in mice of both sexes treated with any dose of BPA, and fibrous osteodystrophy was observed in males and females of the 4.0% groups. Based on the results of the present study, it was concluded that the maximum tolerance dose (MTD) of BPA is 0.2% in diet, because the dose level of 0.5% proved to exert significant hematological toxicity.

Presence of carnitine acetyltransferase in peroxisomes and in mitochondria of oleic acid-grown Saccharomyces cerevisiae.

Activity of carnitine acetyltransferase was detected in glucose- and oleic acid-grown Saccharomyces cerevisiae. Oleic acid-grown cells showed a ten-fold higher activity than glucose-grown cells. Subcellular fractionation of oleic acid-grown cells showed that carnitine acetyltransferase was present in peroxisomes, mitochondria, and cytosol. The results suggested the plausible presence of an 'acetylcarnitine shuttle' in this yeast, as in the case of an n-alkane-assimilating yeast, Candida tropicalis.

Epileptic seizure of El mouse initiates at the parietal cortex: depth EEG observation in freely moving condition using buffer amplifier.

The initiation site of seizure discharges and the relationship between behavioral manifestations and electroencephalography were investigated in the El mouse, a hereditary epilepsy model. The chronic depth electrodes were implanted stereotaxically into the frontal cortex, parietal cortex, temporal cortex, hippocampus, striatum, amygdaloid complex, non-specific nuclei of thalamus and substantia nigra. Electrical activities were recorded in freely moving condition with use of the buffer amplifier devised in the laboratory and behaviors were monitored simultaneously. Seizure spike discharges started in the parietal cortex and spread out into other brain areas. When the hippocampus was involved, the tonic convulsion occurred behaviorally. The paper describes the first direct evidence of the initiation and propagation of seizure discharges in the brain of El mouse.

Purification and characterization of an N-acetyllactosamine-specific lectin from larvae of a moth, Phalera flavescens.

A lectin (Phalera flavescens agglutinin, PFA) of a moth (P. flavescens) has been isolated from hemolymph by DEAE-Toyopearl followed by Cellulofine GCL-1000 chromatography. On size-exclusion chromatography, PFA showed a molecular mass of about 74 kDa, and on sodium dodecyl sulfate-polyacrylamide gel electrophoresis it was separated into two different subunits, glycosylated 18-kDa and unglycosylated 17-kDa subunits, suggesting that PFA occurs as a heterotetrameric protein. N-Acetyllactosamine and laminin inhibited the hemagglutinating activity of PFA. Studies involving Synsorbs showed that PFA interacts with the Gal beta 1-->4GlcNAc group and that nonsubstituted hydroxyl groups at the C-2 and C-4 positions of the galactose residue were essential for interaction with PFA, whereas substitution at the C-3 position of the galactose did not inhibit the interaction between PFA and sugar chains associated with N-acetyllactosamine. PFA is not mitogenic for native or desialylated mouse thymocytes or splenocytes.

Role of the amino-terminal amino acid sequences determining the in vitro refolding process of prochymosin polypeptide.

Prochymosin, a zymogen of an aspartic proteinase chymosin, is produced as inclusion bodies in the recombinant Escherichia coli cells. Solubilization of the inclusion bodies with 8 M urea followed by dialysis at pH 10.5 achieves correctly refolded prochymosin to some extent, which is then activated by self-processing at acidic pHs. Analyses of the alkaline dialysates by anion exchange chromatography revealed broad distribution of prochymosin polypeptides with different conformations. Stepwise dialysis with a slower decreasing rate of urea resulted in marked improvement of the yield of correctly refolded molecules. A hybrid prochymosin (CR601) possessing a short NH2-terminal replacement with the trp-leader peptide was not refolded into the correct conformation by one-step dialysis, but it was by stepwise dialysis. Replacement of Lys at the NH2-terminal second position of CR601 with Asp or Glu caused marked enhancement of correct refolding. These findings suggest that the amino acid sequence in the NH2-terminal region of prochymosin plays a crucial role in determining the whole refolding process of the polypeptide.

New subtype of apical hypertrophic cardiomyopathy identified with nuclear magnetic resonance imaging as an underlying cause of markedly inverted T waves.

OBJECTIVES: The aim of this study was to elucidate the clinical importance of a new subtype of apical hypertrophic cardiomyopathy that could not be diagnosed with the classical diagnostic criteria. BACKGROUND: Apical hypertrophic cardiomyopathy is recognized by a characteristic spade-shaped intraventricular cavity on the end-diastolic left ventriculogram in the right anterior oblique projection, often associated with giant negative T waves [negativity > or = 1.0 mV (10 mm)]. As an underlying cause of giant negative T waves, an additional new subtype of apical hypertrophic cardiomyopathy has been identified. METHODS: In 40 patients with inverted T waves (negativity > or = 0.5 mV), including 26 patients with giant negative T waves, nuclear magnetic resonance (NMR) long-axis images corresponding to the left ventriculogram in the right anterior oblique projection and short-axis images at various levels, including the apical level, were obtained to define the site of hypertrophied myocardium. RESULTS: Long-axis images indicated a spadelike configuration in 17 patients, whereas this diagnostic configuration was not present in the other 23 patients. Nine of these 23 patients had significantly hypertrophied myocardium at the basal level. In the 14 remaining patients, short-axis images indicated no hypertrophy at the basal level and proved that the area of hypertrophied myocardium was confined to a narrow region of the septum or the anterior or lateral wall at the apical level (nonspade apical hypertrophic cardiomyopathy). The hypertrophied myocardium of the nonspade type was so narrowly confined that the mass did not form a spadelike configuration or could not be detected on the long-axis image. CONCLUSIONS: Nonspade apical hypertrophic cardiomyopathy was newly identified on NMR short-axis images, and this could be an additional, important underlying cause of moderately to severely inverted T waves.

Tension inhibitory effect of lead ions and lead uptake in ileal longitudinal muscle of the guinea-pig.

1. In ileal longitudinal smooth muscle of the guinea-pig, lead ions (Pb2+) inhibited the phasic response to high-K+ (60 mM) solution more strongly than the tonic response. 2. Pb2+ displaced Ca2+ concentration-response curves to the right. Moreover, Pb2+ inhibited the 45Ca binding at high affinity sites determined by the La3+ method than low affinity sites during K(+)-induced contraction. 3. After treatment with Pb2+, the tonic tension caused by high-K+ was fairly restored by a wash with normal medium. The lead bound to the muscle cells was almost eliminated after 20-30 min with a normal medium. 4. We suggest that Pb2+ binds loosely to the surface membrane of ileal longitudinal muscle. Pb2+ probably reduced the tension in response to the high-K+ mainly by inhibiting Ca2+ release rather than by affecting Ca2+ influx.
