Detection of mutated K12-ras in histologically negative lymph nodes as an indicator of poor prognosis in stage II colorectal cancer.

Stage II colorectal carcinoma is characterized by negative lymph node pathology as determined by conventional microscopic examination. These patients generally do not receive adjuvant therapy although 20%-30% will die from metastatic disease. To determine whether K-ras mutations at codon 12 could be used as a sensitive indicator of occult lymph node metastasis in stage II colon carcinoma, a retrospective study was performed using restriction endonuclease-mediated selective polymerase chain reaction (REMS-PCR) amplification. Of 106 colonic tumors analyzed, 46 were identified as positive for a K12-ras mutation in the primary tumor. Multiple lymph node samples from 38 of these 46 patients were examined by a sensitive nested PCR protocol for the presence of a K12-ras mutation. Of these 38 patients, 14 had 1 or more positive lymph nodes by PCR (37%) and 24 were negative for the mutation (63%). Of the 14 patients with a K12-ras mutation detected in lymph nodes, 8 died of the disease within 5 years (57%) compared to only 4 of the 24 patients with ras-negative lymph nodes (17%). The difference in time to death from disease, stratified using K12-ras status of lymph nodes, was statistically significant (P = 0.036; log-rank test). These results suggest K-ras mutation status of lymph nodes in patients with stage II colon cancer might identify a subgroup of patients who are more likely to develop recurrent and/or metastatic disease and benefit from adjuvant therapy. Larger studies are indicated to determine whether detection of K-ras mutation positivity in histologically negative lymph nodes portends a poor prognosis and to determine whether more aggressive use of adjuvant therapy is warranted.

[Fasting insulin level and hypertensive family history in overweighted children].

In order to analyze the relationship between fasting insulin level and body mess index (BMI), blood pressure and lipids, and to discuss the effect of hypertensive family history, 317 healthy children aged 8-11 years including 74 overweighted and 243 non-overweighted were selected randomly from 4 primary schools in Beijing. Anthropometry, fasting insulin and lipids were measured, and the family history of hypertension (FHH) was recorded. After adjusting of age and sex, the levels of blood fasting insulin, cholesterol and triglyceride were significantly higher (P < 0.01) and the ratio of HDL/LDL and apoA/apoB were significantly lower (P < 0.01) in the overweighted than that in non-overweighted children. After controlling BMI, the partial correlation coefficients between fasting insulin and TG, systolic pressure was 0.20 (P < 0.01) and 0.12 (P < 0.01) respectively. Moreover, among the overweighted children, but not among non-overweighted children, those with FHH had a significantly higher fasting insulin than those without FHH (P < 0.01). It is concluded that hypertensive family history may play an important role in contributing to the higher level of serum insulin of overweighted children, and higher serum insulin may concerted with some metabolic disorders in overweighted children.

[Investigation of different dosages of inserted Bt genes and their insect-resistance in transgenic Bt cotton].

The plants with various dosages of inserted Bt genes were developed by intercrossing among transgenic Bt lines in which Bt genes were inserted in different chromosomes, and commercial non-transgenic Bt varieties. Laboratory bioassay of insect-resistance and Bt toxin protein assays with ELISA were used to detect efficacy to bollworm in different cotton growing stages. As transgenic Bt insect-resistant cotton, temporal difference of resistance existed in plants converging on different dosages of inserted Bt genes, i.e., there was a declining level of efficacy with plant age as the mortality (%) of Helicoverpa and Bt toxin protein expressional level decreased gradually, when analyzed with leaves from the main stem. Our research also revealed that the insect-resistance of converging 4 Bt genes plants was decreased due to homozygous suppression.

[Removing the nitrogen from the ammonium sulfite method paper mill wastewater by anoxic/aerobic SBR].

The experiment results showed that the best technology of removing the nitrogen was anoxic time to aerobic time of 1:1.5, a cycle of 8 hours; the SRT should be more than 12 d and the loading of NH3-N should be less than 0.063 g/(g.d); when the concentration of CODCr, NH3-N and NOx-N was 1200-1800 mg/L, 135-200 mg/L and 7-10 mg/L respectively, without external carbon sources, the removal rate of NH3-N and TN could respectively achieve 95% and 66%; after adding sodium acetate the removal rate of TN could get to 85%. The addition of sodium acetate at the concentration equivalent to theoretical CODCr values of 125 mg/L appeared to be the most economical and reliable option.

[Investigation of water sorption and solubility of three denture soft lining materials].

This assay was made to investigate the water sorption and solubility of autopolymerizing hydrogel, acrylic resin and silicone denture soft lining materials. The test discs with 50 mm in diameter and 1 mm in thickness were prepared according to powder and liquid ratio. The weight of sample was measured after 24 h drying (m1). The sample was put into artificial saliva and distilled water for 7 d and 28 d, the moisture weight was measured after soaking (m2). After drying again, the last data of weight (m3) was acquired till the constant mass was recorded. Finally the values of water sorption and water solubility were calculated respectively with statistical analysis. The results showed the means of percentage about absorption at 7 d and 28 d were 28.62% and 24.96% for hydrogel, 1.89% and 1.65% for acrylic resin, 0.25% and 0.29% for silicone in artificial saliva. The percentage solubility at two stages were 0.56% and 0.51% for hydrogel, 0.74% and 0.83% for acrylic resin, 0.10% and 0.12% for silicone. In distilled water, the means of percentage about absorption at 7 d and 28 d were 30.15% and 27.85% for hydrogel, 3.64% and 6.17% for acrylic resin, 0.83% and 0.98% for silicone. The percentage solubility at two stages were 0.79% and 1.16% for hydrogel, 0.86% and 1.01% for acrylic resin, 0.10% and 0.14% for silicone respectively. The conclusion is that the order of percentage about absorption for these three denture soft lining materials were hydrogel > acrylic resin > silicone. The percentage solubility of acrylic resin was higher than that of the other two materials because of the plasticizer existing in acrylic resin material. In some degree, the ionic component increased may affect water sorption and solubility. Further clinical demonstration is needed for this hydrogel with higher percentage absorption.

[The association of methylenetetrahydrofolate reductase gene polymorphism with nephropathy in type 2 diabetes mellitus in Chinese].

OBJECTIVE: To evaluate the role of methylenetetrahydrofolate reductase (MTHFR) gene polymorphisms in Chinese patients with type 2 diabetes mellitus and diabetic nephropathy(DN). METHODS: 85 type 2 diabetes mellitus patients and 57 controls were recruited in the study. MTHFR genetic C677T polymorphism was determined with PCR-restriction fragment length polymorphisms (RFLP). Plasma total homocysteine level was measured using high-performance liquid chromatography (HPLC) with fluorescence detection. RESULTS: The frequency of MTHFR TT homogenetic type and CT heterogenetic type and allele T(28.21%, 51.28%, 53.85%) was significantly higher in type 2 diabetes mellitus with diabetic nephropathy group than those without nephropathy (19.57%, 28.26%, 33.70%) or normal controls (17.54%, 28.07%, 31.58%). However, there was no significant difference of MTHFR genotype and allele frequency between type 2 diabetes mellitus without nephropathy and normal controls. The presence of T allele appeared to have a stronger association with the development of diabetic nephropathy. The odds ratio was 2.30 and the 95% confidence interval was 1.24-4.26. Moreover, plasma homocysteine levels were markedly higher in patients with TT or CT genotype than those in patients with CC genotype. CONCLUSIONS: MTHFR gene C677T mutation associated with a predisposition to increased plasma homocysteine levels may represent a genetic risk factor for diabetic nephropathy in Chinese type 2 diabetes mellitus.

Regulation of growth-regulated oncogene alpha expression by estrogen in human endothelial cells.

AIM: To study the effect of estrogen on expression of growth-regulated oncogene alpha (GROalpha) in human umbilical vein endothelial cells (HUVEC) in vitro. METHODS: Expressions of CXC chemokine GROalpha mRNA and protein were measured by Northern blotting assay and ELISA, respectively. The physiological significance of GROalpha expression was tested by static cell adhesion assay. RESULTS: Both the GROalpha mRNA and protein levels decreased markedly after HUVEC were exposured to 17beta-estradiol (2 0.05 micromol/L. Moreover, the inhibition of the protein was depended on the concentration of 17beta-estradiol. Tamoxifen (0.1 micromol/L), an estrogen receptor alpha antagonist, alone did not affect GROalpha protein expression, but can reverse the E2 -induced inhibition of GROalpha protein expression (by up to 50 %) and the binding of U937 cells to E2-treated HUVEC (by up to 40 %). CONCLUSION: Estrogen might functionally down-regulates GROalpha expression through estrogen receptor alpha on endothelial cells.

[Effects of volatiles from different trophic level on foraging behavior of Aphidius avenae].

The responses of female Aphidius avenae to volatiles from undamaged wheat plant, Sitobion avenae and Rhopalosiphum padi, aphids-plant complexes and aphids damaged plant were investigated in wind tunnel and Y-tube olfactometer. The volatiles from undamaged wheat plant, S. avenae and R. padi were much less attractive to A. avenae than those from aphids-plant complexes and aphids-damaged plant. Although the parasitic rate was much lower, the R. padi-plant complexes and R. padi damaged plant were also attractive to A. avenae. Identification of volatiles by GC-MS showed that the main volatiles induced by aphids feeding were 2-camphene, 6-methyl-5-hepten-2-one, 6-methyl-5-hepten-2-ol, Z-3-hexenyl-acetate, and methyl salicylate, among which, 6-methyl-5-hepten-2-one and 6-methyl-5-hepten-2-ol were more attractive to A. avenae, while methyl salicylate was not attractive.

[Microsurgical treatment of multiple intracranial aneurysms].

OBJECTIVE: To summarize the experience in microsurgical treatment of multiple intracranial aneurysms. METHOD: Eight patients with 17 aneurysms were analysed retrospectively. RESULTS: Seven patients were followed up from 0.5 month to 10 years. According to Sundt's standard, the results in 5 patients were excellent and 2 good. One patients with bilateral PCom aneurysm died 1 week after operation. CONCLUSIONS: Multiple intracranial aneurysms should be removed as soon as possible, especially bilateral ones. The best way is first to clip the bled aneurysm and then the contralateral one by using coronal incision and bilateral pterional approach. This approach has the advantage of minimal invasion, good exposure, and less time-consuming.

Modified cold storage of rat livers with self-made HYD solution.

OBJECTIVE: To investigate the cold preservation effect on rat livers of a modified storage method with self-made HYD solution. METHODS: The vascular bed of rat livers was expanded with an additional 20 to 40 ml self-made HYD solution/100 g liver. After resection of the liver, the extra HYD solution (expressed as % liver weight) was entrapped via portal infusion by tying off the supra- and infra-hepatic inferior vena cava. Forty rats were randomly divided into four groups including control group with conventional storage method, and 20%, 30% and 40% groups according to the amount of extra HYD solution. We compared the preservation effect of the modified storage method with that of the conventional storage method using an isolated perfused rat liver model. RESULTS: Bile production and all the indices of hepatic microcirculation including portal perfusion pressure, endothelin-1 in the effluent, trypan blue distribution time and histology were significantly superior in the modified method groups compared to those in the control group (P < 0.05). The contents of dihydroxybenzoic acid (DHBA) in the modified method groups were significantly lower than those in the control group (P < 0.05). Liver enzymes activities in the 30% group were markedly lower than those in the control group (P < 0.05). The preservation effect on rat liver in the 30% group was the best among the modified method groups. CONCLUSION: The modified cold storage method is effective and may have potential for clinical application in liver preservation.

Expression and function of c-kit receptor in bone marrow mononuclear cells of patients with myelodysplastic syndromes.

OBJECTIVE: To determine the expression and function of the c-kit receptor in bone marrow mononuclear cells (BMMNC) of patients with myelodysplastic syndromes (MDS). METHODS: Direct immunofluorescence assay and reverse transcriptase-polymerase chain reaction (RT-PCR) were used to detect c-kit protein and c-kit mRNA expressions in the BMMNC of 29 MDS patients and 10 normal controls. Cell culture was used to detect the function of the c-kit receptor. RESULTS: c-kit protein expression in the MDS group was significantly higher than that in the control group (8.58% +/- 5.28% vs 3.04% +/- 1.49%, P < 0.05). c-kit protein expression in the refractory anemia (RA) group was significantly lower than that in the RA with an excess of blasts (RAEB)/RAEB in transformation (RAEB-t) group (5.12% +/- 2.13% vs 10.01% +/- 5.07%, P < 0.05). The rate of c-kit protein expression was 32.43% in acute myeloblastic leukemia (AML) cases transformed from MDS (t-AML). c-kit mRNA expression in the MDS group was correlated with c-kit protein expression. Interleukin-3 (IL-3) and erythropoietin (Epo), with or without stem cell factor (SCF), upregulated c-kit protein and its mRNA expression. In the presence of IL-3 and Epo, SCF showed significant stimulating effects on the formation of CFU-GM and BFU-E in semi-solid cultures of normal BMMNC, but had no effects on those of the MDS patients. CONCLUSION: The protein and mRNA expression of the c-kit receptor in the BMMNC of MDS patients were higher than those of normal controls, and the function of this receptor in MDS BMMNC was abnormal.

In vitro study on the morphology of human blood dendritic cells and LPAK cells inducing apoptosis of the hepatoma cell line.

OBJECTIVE: To observe in vitro effects and morphological changes of human peripheral blood dendritic cells (DCs) on the ability of lymphokine and phytohaemagglutininum (PHA) activated killer (LPAK) cells to induce apoptosis of the human hepatoma cell line (BEL-7402, B). METHODS: Experimental groups were divided into LD group (DCs + L + B), L group (L + B), D group (DCs + B) and B group. The methods of neutral red uptake, ordinary light microscopy, electron microscopy, TDT mediated X-dUTP nick end labeling (TUNEL) were used. RESULTS: The difference between the D group and the B group was not distinct (P > 0.05). The difference between the LD group and the L group was distinct, with DCs + LPAK > LPAK (P < 0.01) in cytotoxicity. Apoptotic cells were TUNEL positive in light microscopy, and apoptotic nuclei were stained yellow brown and dark brown, with size and shape varying from cell to cell. Ultrastructural change in apoptotic tumor cells comprised of compaction and condensation of nuclear chromatin, and condensation of cytoplasm and apoptotic bodies. At the same time, LPAK cells manifested the characteristics of autophagic apoptosis, and there were some autophagic bodies in it. CONCLUSIONS: The combination of human blood DCs and LPAK cells could induce apoptosis of BEL-7402 cells effectively, with some LPAK cells manifesting the characteristics of autophagic apoptosis.

[Bax gene expression in endometrial carcinoma].

OBJECTIVE: To study the relationship between bax gene expression and endometrial carcinoma. METHODS: Bax mRNA expression was evaluated by RT-PCR in a series of 8 normal proliferative endometrium (NPE), 6 simple and complex hyperplasia (SCH), 6 atypical hyperplasia (AH) and 42 endometrial carcinoma (EC). RESULTS: All of the 8 NPE showed moderate bax mRNA expression. In 1 of 6 SCH, 2 of 6 AH and 27 of the 42 EC (64.3%), there was overexpression of bax mRNA. In endometrial carcinoma, with the progression of clinical stage, increase in histologic grade and in depth of invasion to muscle layer, and lymph node metastasis, the positive rate of bax overexpression was correspondingly increased. CONCLUSION: The results suggest that bax expression affects the balance between the endometrial cell proliferation and apoptosis. Bax might play an important role in carcinogenesis and tumor progression of endometrial carcinoma. As a high risk factor it can be used to evaluate the biological behavior of endometrial carcinoma.

[The effect of tamoxifen of endometrium].

OBJECTIVE: To evaluate the effect of tamoxifen on the endometrium. METHODS: Hysteroscopy and endometrial biopsies were performed in 26 patients with breast cancer receiving tamoxifen and having vaginal bleeding or endometrial thickness detected by ultrasound. In addition 78 women with postmenopausal bleeding without receiving tamoxifen underwent the same examination. RESULTS: Thirteen (50.0%) patients receiving tamoxifen had endometrial or endocervical polyps, whereas 14 cases(17.9%) not receiving tamoxifen developed the polyps (P < 0.05). Nine (34.6%) patients receiving tamoxifen had thyperplasias, which was significantly higher than that of women not receiving tamoxifen (P < 0.05). CONCLUSIONS: The incidence of endometria disease in the patient with breast cancer receiving tamoxifen was increased. These patients should be closely monitored by transvaginal sonography and hysteroscopy.

[Telomerase activity in cervical intraepithelial neoplasia].

OBJECTIVE: To investigate the relationship between telomerase expression and the carcinogenesis of cervix, and the possibility of using telomerase as a marker to evaluate the natural history of cervical intraepithelial neoplasia (CIN). METHODS: Use the telomere repeat amplification protocol (TRAP) assay to detect cervical lesion in cervical scraping samples and cervical biopsy tissues. RESULTS: Sixty two point five percent, 60.0 percent, 82.4 percent and 61.9 percent cervical scraping samples exhibited telomerase activity in CINI, CINII, CINIII and cervical cancer, respectively, compared 50.0 percent, 45.0 percent, 96.4 percent and 95.2 percent in cervical biopsy tissue. In contrast, 20.0 percent and 26.7 percent in cervical scraping samples and cervical biopsy tissues of normal cervix. With the development of cervical lesion, the expression of tolemerase activation was increasing (P < 0.05). There was no significant difference between CINI and CINII (P > 0.05). There were significant differences between CINIII and CINI, CINII (P < 0.05) Telomerase activity was coincidence between the cervical scraping samples and cervical biopsy tissues. CONCLUSIONS: These results seem to suggest that the Telomerase expression may associate with the carcinogenesis of cervix. The telomerase activity assay of cervical scraping sample could be used as a marker to monitor and predicate the transit of CIN in clinical practice.

[Radiotherapy in the treatment of uterine sarcomas].

OBJECTIVE: To study radiotherapy in the treatment of uterine sarcomas and response to radiation in primary or recurrent uterine sarcomas. METHODS: A total of 102 uterine sarcomas treated in the Cancer Hospital from May 1960 to June 1999. A retrospective review of 45 patients receiving surgery alone, 29 patients for postoperative radiotherapy, 4 radiotherapy alone, 24 recurrent sarcomas receiving radiotherapy. RESULTS: The 5-year overall survival for 45 patients treated with surgery alone and 29 patients managed by postoperative radiotherapy were 56.7% and 58.6% respectively (P > 0.05). The addition of radiotherapy significantly reduced the local recurrence rate from 42.2% to 20.7% (P < 0.05). The tumor was disappeared completely by treatment of radiotherapy alone in 3 patients of primary uterine cervix sarcomas. 1 patient was free of disease at 36 months. Other 2 patients died 9 and 11 months later because of distant metastases. The complete and partial response rate of radiotherapy was 50.0% for recurrent tumors. The radiotherapeutic sensibility of endometrial stromal sarcomas, mixed mesodermal sarcomas and leiomyosarcomas was downward tendency in turn. CONCLUSIONS: The postoperation adjuvant radiotherapy reduced the pelvic recurrent significantly. Radiotherapy should be used in the treatment of uterine sarcomas as an important part of combination therapy.

Comparative observation with MRI and pathology of brain edema at the early stage of severe burn.

OBJECTIVE: To investigate the correlation between MRI features and pathology in brain edema at the early stage of severe burn (5 0% TBSA III degree) in dogs. METHODS: Fifty-two dogs were randomized into control, simple b urn (SB), burn plus sodium lactate (BSL), and burn plus glucose solution groups (BGS). The manifestation of the brain of control group was compared with that of burn groups at 6, 12, 18 and 24 hours postburn with MRI and pathological examination (gross appearance, electron microscopy and light microscopy). RESULTS: The earliest findings of brain edema were seen at 12 h ours after burn in BGS group, in which brain swelling was the main feature of MR I. The decrease of SIR on T(1)WI was not observed until it was exceeded 10%. Sig nal of T(2)WI increased by 8.29% at 24 hours after burn. It was difficult to distinguish the gray matter from the white matter at the boundary line, which became blurred later. Histological changes of brain edema were observed as early as 6 hours after burn, being accompanied by swelling of endothelial cells and peri-vescular astrocytes, and vacuolation took place in neurons at 12 hours after burn, with different degrees of necrosis of capillary endothelium, neurons, and axons. These changes became more marked with elapse of time. The BGS group showed the most obvious changes mentioned above at 24 hours after burn. CONCLUSIONS: The model of the brain edema after severe burn has the feature of both vasogenic edema and cytotoxic edema on the MRI and pathology. Positive MRI findings lagged behind that of the pathomorphological changes.
