The 5' stem-loop from human U4 snRNA adopts a novel conformation stabilized by G-C and G-U base pairs.

1H NMR and molecular modeling studies of the 5' stem-loop from human U4 snRNA were undertaken to determine the conformation of this stem-loop that is essential for spliceosome formation and pre-mRNA splicing. Sixteen of the 35 nucleotides of this stem-loop are in the loop region and inspection of the loop sequence revealed no decomposition into elements of secondary structure commonly found in other RNA stem-loops. An analysis of possible base pairing interactions for this stem-loop using the methods of Zuker revealed the lowest energy secondary structure for the 16 nucleotide loop consisted of four base pairs at the base of a non-canonical tetraloop (UUUA). This shorter stem-loop was joined to the nine base pair stem by two A residues on the 5' side and a single bulged A on the 3' side. Both stems also had bulged A residues. 1H NMR experiments performed on solutions of the 35 mer stem-loop, the stem region, and the loop region confirmed the 35 mer adopted this secondary structure in solution. A 3D molecular model of this structure consistent with the NMR data was generated to assist in visualization of this novel structure.

Distribution of integrins and the extracellular matrix proteins in the baboon endometrium during the menstrual cycle and early pregnancy.

Integrins are heterodimeric glycoproteins that have been found to undergo dynamic temporal and spatial changes in distribution in the endometrium during the menstrual cycle in women. Likewise the extracellular matrix (ECM) ligands for these receptors are likely to play a role in the establishment of a receptive endometrium. To develop primate models to study the role of these molecules in the cascade of molecular events leading to implantation, integrin expression and associated changes in ECM were investigated during the menstrual cycle and in early pregnancy in the baboon. Antibodies specific for the integrins (alpha(1-6) and alpha(v); beta1, beta3, and beta4) and ECM (laminin, collagen IV, fibronectin) were utilized. In addition, cytokeratin and alpha-smooth muscle actin were used as epithelial, stromal, and smooth muscle cell markers, respectively. Endometrium was obtained in duplicate or triplicate during the menstrual cycle and early pregnancy. Changes observed during the natural menstrual cycle were confirmed using ovariectomized, steroid-treated animals. Constitutively expressed integrins on the endometrial epithelium included the collagen/laminin receptors: alpha2, alpha3, alpha6, and beta4. The pattern of expression correlated well with the distribution of ECM in this tissue. Collagen IV was confined to the basement membrane of glandular epithelium and blood vessels. Laminin immunostaining was found in the basement membrane, mostly in the stroma of the basal region, in the glandular endometrium and vasculature. Fibronectin was present throughout the stroma but not in the basement membrane. The collagen receptor alpha1 beta1 and fibronectin receptor alpha4 beta1 appeared in the glandular epithelium in the luteal phase. As in the human, alpha1 and alpha4 disappeared from the glandular epithelium with the establishment of pregnancy. In contrast, the alpha4 beta3 vitronectin receptor appeared in the glandular epithelium only in pregnancy or following long-term steroid treatment with estrogen and progesterone but not during the time of uterine receptivity associated with the initial period of embryo attachment. Osteopontin, an ECM ligand for alpha(v) beta3, was coexpressed with this integrin in invading cytotrophoblasts, glandular epithelium, and decidualizing stromal cells. Decidualization in the baboon was associated with changes in integrin expression similar to those found in humans: there was an increase in alpha1, alpha3, alpha6, beta1, and alpha(v) beta3 in the decidualized stromal cells. Laminin and collagen IV expression also increased at the implantation site and throughout the endometrium. In contrast, fibronectin expression was most evident at the implantation site and corresponded to alpha5 expression on the invading cytotrophoblasts. In summary, marked similarities were found in the expression of ECM and the integrin receptors between the baboon and the human endometrium throughout the menstrual cycle and in pregnancy. Cycle-specific integrins, alpha1, and alpha4, were present on epithelial cells during the secretory phase. Delayed expression of alpha(v) beta3 in baboon endometrial glands correlated closely with the time of enhanced glandular secretory activity in this primate. The baboon appears to be an excellent model for the investigation of the role of integrins and ECM leading to successful implantation.

Biochemical and molecular biological characterization of CAC2, the Arabidopsis thaliana gene coding for the biotin carboxylase subunit of the plastidic acetyl-coenzyme A carboxylase.

The biotin carboxylase subunit of the heteromeric chloroplastic acetyl-coenzyme A carboxylase (ACCase) of Arabidopsis thaliana is coded by a single gene (CAC2), which is interrupted by 15 introns. The cDNA encodes a deduced protein of 537 amino acids with an apparent N-terminal chloroplast-targeting transit peptide. Antibodies generated to a glutathione S-transferase-CAC2 fusion protein react solely with a 51-kD polypeptide of Arabidopsis; these antibodies also inhibit ACCase activity in extracts of Arabidopsis. The entire CAC2 cDNA sequence was expressed in Escherichia coli and the resulting recombinant biotin carboxylase was enzymatically active in carboxylating free biotin. The catalytic properties of the recombinant biotin carboxylase indicate that the activity of the heteromeric ACCase may be regulated by light-/dark-induced changes in stromal pH. The CAC2 gene is maximally expressed in organs and tissues that are actively synthesizing fatty acids for membrane lipids or oil deposition. The observed expression pattern of CAC2 mirrors that previously reported for the CAC1 gene (J.-K. Choi, F. Yu, E.S. Wurtele, B.J. Nikolau [1995] Plant Physiol 109: 619-625; J. Ke, J.-K. Choi, M. Smith, H.T. Horner, B.J. Nikolau, E.S. Wurtele [1997] Plant Physiol 113: 357-365), which codes for the biotin carboxyl carrier subunit of the heteromeric ACCase. This coordination is probably partially established by coordinate transcription of the two genes. This hypothesis is consistent with the finding that the CAC2 and CAC1 gene promoters share a common set of sequence motifs that may be important in guiding the transcription of these genes.

Detoxified lipooligosaccharide from nontypeable Haemophilus influenzae conjugated to proteins confers protection against otitis media in chinchillas.

Detoxified-lipooligosaccharide (dLOS)-protein conjugates from nontypeable Haemophilus influenzae (NTHi) elicited a significant rise of anti-LOS antibodies with bactericidal activity in rabbits (X.-X. Gu, C.-M. Tsai, T. Ueyama, S. J. Barenkamp, J. B. Robbins, and D. J. Lim, Infect. Immun. 64:4047-4053, 1996). In this study, we evaluated whether vaccination with the conjugates would protect against NTHi otitis media in chinchillas. Fifty-eight chinchillas received three subcutaneous or intramuscular injections of dLOS-conjugated tetanus toxoid, dLOS-conjugated high-molecular-weight proteins from NTHi, or saline (control) in Freund's adjuvant and then were challenged by intrabullar inoculation with 140 CFU of NTHi. All vaccinated animals responded with elevated serum titers of anti-LOS antibody, and 49% (19 of 39) demonstrated bactericidal activity against the homologous strain. Otitis media with culture-positive NTHi effusions developed in all 19 controls and 56% (22 of 39) of the vaccinated animals during a period of 21 days (P < 0.001). Bacterial counts of the middle ear effusions were lower in the vaccine groups than in the controls (P < 0.01). The incidences of infection in the unchallenged ear or inner ear were 26 or 28% in the vaccine groups and 53 or 58% in the controls (P < 0.05). The signs of infection observed by otoscopy were less severe in the vaccine groups than in the controls. There was no significant difference between the two vaccine groups. These data indicate that active immunization with LOS-based conjugates reduces the incidence of NTHi-induced otitis media.

Molecular characterization and transcriptional analysis of type 8 capsule genes in Staphylococcus aureus.

A 20.5-kb contiguous DNA fragment from Staphylococcus aureus Becker affecting type 8 capsule (CP8) biosynthesis was previously cloned. Sequencing analysis indicated that 16 open reading frames (ORFs) encoded within this fragment might be involved in CP8 synthesis. Using various plasmids containing DNA inserts derived from the 20.5-kb region, we showed by complementation of chemical mutants that 8 of the 16 ORFs were required for CP8 synthesis. To determine the involvement of the remaining eight ORFs, nonpolar gene-specific chromosomal mutations located in each of these ORFs were constructed. We found that three additional ORFs were also involved in the CP8 synthesis. Thus, 11 of the 16 ORFs were shown to affect CP8 synthesis. Complementation analyses of these 11 type 8 capsule (cap8) genes affecting CP8 production showed several promoters within the cap8 gene cluster. However, by Northern hybridization using either the entire cap8 gene cluster or the internal fragments of individual ORFs as probes, one 17-kb cap8-specific transcript was detected. Using xylE as the reporter gene, we found that the promoter at the beginning of the cap8 operon was much stronger than any of the internal promoters. These results suggest that the cap8 genes are transcribed mainly as a single large transcript. In addition, Southern hybridization analyses showed that cap8H, cap8I, cap8J, and cap8K, located in the central region of the cap8 gene cluster, were CP8 specific.

Interaction of ozone and allergen challenges on bronchial responsiveness and inflammation in sensitised guinea pigs.

BACKGROUND: Environmental pollutants such as ozone may interact with airway responses to allergen in sensitised individuals. METHODS: We examined the effects of a single exposure to ozone (1 ppm for 1 h) on bronchial responsiveness to acetylcholine (ACh) aerosol 3 and 24 h after single ovalbumin (OA) challenge in OA-sensitised anaesthetised guinea pigs. Broncho-alveolar lavage (BAL) was performed and protein content and differential cell counts were determined. RESULTS: Ozone increased bronchial responsiveness at 3 h but not at 24 h, while OA alone had no effect. At 3 h after ozone, OA induced further, but non-significant increases in bronchial responsiveness to ACh, but at 24 h after ozone, there was enhanced responsiveness. Neutrophil counts in BAL fluid increased at 3 and 24 h after exposure to ozone alone, but there were no further increases with ozone followed by OA exposure. Protein concentration was significantly increased in the ozone and OA groups at 3 and 24 h (163.4 +/- 25.6 and 128.7 +/- 7.4 mg/ml, respectively) compared to 54.0 +/- 18.1 mg/ml in the control group (p < 0.02 and p < 0.01, respectively). CONCLUSION: Our data demonstrate an interaction of OA with ozone exposure on bronchial responsiveness; one underlying mechanism could be through damage at the endothelial-epithelial barrier.

Characterization of integrin expression in a well differentiated endometrial adenocarcinoma cell line (Ishikawa).

The pattern of constitutive and cycle-dependent integrins in normal endometrium has recently been established, suggesting a role for cell adhesion molecules in endometrial receptivity and implantation. Currently few, if any, models exist for the study of human endometrial integrins and their role in establishment of the receptive endometrial phenotype. The Ishikawa cell line maintains functional estrogen receptors and progesterone receptors. The progesterone receptors in these cells are inducible by priming with estradiol and down-regulated by treatment with progesterone. In the present study the pattern of integrin expression in this well differentiated endometrial cell line is compared to that in normal endometrial epithelium using immunohistochemistry and flow cytometry and is confirmed by immunoprecipitation, Western immunoblot, and PCR. Like normal endometrial epithelium, Ishikawa cells maintain constitutive expression of alpha 2 beta 1, alpha 3 beta 1, alpha 6 beta 4. PCR demonstrates the expected size fragments of each, although evidence for alternatively spliced forms of the alpha 2-subunit was noted. Progesterone treatment of estradiol-primed cells resulted in increased expression of the alpha 1 beta 1 collagen-laminin receptor and suppression of the alpha v beta 3 vitronectin receptor, two of the cycle-dependent integrins expressed by normal endometrial epithelium. These data support the use of Ishikawa cells as an excellent model to study the regulation endometrial integrins and advance our understanding of hormonally mediated events surrounding implantation.

[Effects of acetyl-DL-leucine on the recovery of the locomotor equilibrium and spontaneous discharge of lateral vestibular nucleus neurons in unilateral vestibular neurectomized cats].

The effects of acetyl-DL-leucine (AL) treatment (28 mg/kg.day, p.o.) on the vestibular compensation were studied over period of 6 weeks in unilateral vestibular neurectomized cats. This investigation was performed by measuring the recovery time of locomotor equilibrium in the rotating beam test and recording the unitary extracellular activity of single neurons of lateral vestibular nucleus (LVN) in totally awake cats. The results showed that AL treatment strongly accelerated the recovery of locomotor equilibrium and significantly inhibited the recovery of resting spontaneous firing rate within the deafferented LVN neurons (n = 506) but improved the sensitivity and recovery of relative amount and proportion of neurons (n = 454) in response to head roll tilts in frontal plane. The neurophysiological mechanisms of AL role in the vestibular compensation were discussed.

[Operative treatment of displaced acetabular fractures through the ilioinguinal approach].

To reduce the incidence of ectopic ossification and improve hip joint function after the operative treatment of displaced acetabulum fractures. We surgically treated 12 acetabular fractures which involved anterior column or both anterior and posterior column fractures through the ilioinguinal approach. 10 patients were judged postoperatively to have an anatomic reduction, and 2 had a satisfactory reduction. An average of 3-year follow-up showed excellent results in 8 patients (67%) and good in 4 (33%). Radiographic results indicated excellent results in 8 (67%), patients good in 3 (25%), fair in 1 (8%). The results suggested that the ilioinguinal approach appears to diminish many of the problems associated with utilization of an extrapelvic approach, including the disturbances of the hip abductor system, heterotopic ossification, sciatic palsy, the slow recovery of hip mobility. We conclude that it is not only good for an operative treatment of anterior column fractures but also good for some of posterior column fractures.

[Early diagnosis of pancreatic infection by CT-guided needle-aspiration].

35 CT-guided percutaneous needle-aspirations with bacteriological sampling were performed in 21 patients suspected of pancreatic infection out of 122 patients with severe acute pancreatitis. Thirteen patients were found by smear and staining culture to have pancreatic infection, and diagnosis was confirmed by surgery. The pathogens found were E coli, pseudomonas and fungi. All pancreatic infections were in grade D or E according to the grading system of Balthazar. Seven of 8 patients with sterile aspirates were cured by nonsurgical therapy. We conclude that CT-guided aspiration is a safe, accurate method for identifying infection of the pancreas at the early stage of severe acute pancreatitis.

[High-malar plasty through an intraoral approach].

Request for high-malar plasty, a purely aesthetic surgery, is fairly common in the Oriental. For an optimal result, consideration should be given to integrating the malar reconstruction with the facial features. A new method for correction of the high-malar deformity was introduced, in which the intraoral approach through an L-shaped incision and osteotomy were employed. The technique is simple, safe and can be done with local anesthesia on an outpatient basis. From December 1994 to February 1996, we have successfully treated 25 cases with this method. Postoperative follow-up for 6 months to 1 year revealed aesthetically satisfactory results. Complications are minor, including swelling, ecchymosis, etc. We conclude that high-malar plasty through the intraoral approach is a safe, effective and relatively simple technique for mild and moderate high-malar deformity.

[Craniofacial growth changes following premature closure of the bilateral coronal sutures in rabbits].

The study was designed to examine the amount and the direction of craniofacial growth changes following bilateral coronal sutures (BCS) closure in young rabbits. At 2 weeks of age, the BCS were fused by the application of enamel adhesive. These animals and a control group also received amalgam implants on the frontonasal, coronal, and lambdoid sutures to monitor suture growth. At 4 or 8 weeks after suture fusion, separate groups of animals underwent release of fused sutures. Immobilization of BCS resulted in growth cessation of the coronal suture. The normal sutures underwent very rapid growth from 2 to 8 weeks of age. BCS release at 8 weeks after fusion resulted in growth restriction. Significant compensatory growth was also noted in the frontal and the lambdoid sutures from 2 to 8 weeks. Nasal bone length was increased significantly. Vault length and craniofacial length were reduced. The results of the present study give experimental documentation to emphasize that surgical correction of craniosynostosis should be performed at a young age to achieve better functional and cosmetic results.

[Evaluation of parotid sialography and labial salivary gland biopsy to the diagnosis of Sjögren syndrome].

OBJECTIVE: To investigate the relationship between the change of parotid sialograms and the results of labial gland biopsies in the patients with Sjögren syndrome (SS). METHODS: Parotid sialography and labial salivary gland biopsy were performed on 100 patients with Sjögren syndrome. RESULTS: The sialectasis of parotid terminal ducts was found in 87 patients, the widening only of main ducts was found in 8 patients, and 5 cases showed normal sialograms. The results of labial salivary gland biopsies showed, that 90 cases had focal lymphocytic sialadenitis (Zhen Linfan's grade score of 2), 59 cases had a score of 4 accoding to Chisholm's grading score; moderate scattered lymphocitic infiltration was shown in 10 patients. CONCLUSION: Our results suggested that the lesions of parotid gland and labial salivary gland in SS were concerted closely. Therefore a new way of combining the changes of parotid gland and labial salivary gland for the diagnose of SS was proposed.

[The uncinate process: ultrastructural and CT studies].

To provide further understanding of uncinate process, 15 uncinate processes were examined under scanning electron microscope, and 32 high-resolution coronal CT scans were studied in patients with septal deformity or sinusitis. It was found that(1)pathological ultrastructural changes of uncinate process were noted in patients with chronic sinusitis; (2) medially deviated, laterally deviated, pneumatization and hypertrophy of uncinate process were demonstrated. The following conclutions were drawn(1)the anatomic variations in ostiomeatal complex is responsible for the formation of chronic sinusitis; (2)for patients with chronic sinusitis, uncinatectomy is the treatment of choice; (3)preoperative and intraoperative studies of CT scans are mandatory for avoidance of complications.

[Deletion of MTS1/p16 gene in endometrial carcinoma].

OBJECTIVE: To investigate the possible relationship between deletion of MTS1/p16 gene and progression of endometrial carcinoma. METHODS: Thirty two primary endometrial carcinoma, 7 tumor-adjacent endometrial tissue and 10 normal endometrial tissue specimens were examined for homozygous deletion of MTS1/p16 gene by polymerase chain reaction-based analysis. RESULTS: Of 32 endometrial cancer specimens, 6(18.8%) showed homozygous deletion of MTS1/p16 gene. No deletion was detected in the tumor-adjacent and normal endometrial tussues. Nor was it detected in well differentiated endometrial carcinoma. CONCLUSIONS: Deletion of MTS1/p16 gene might contribute to the progression of endometrial carcinoma and could be served as indicator for predicting prognosis.

[A computer-aided design assessment system for recovery life support technology options].

A specific computer-aided decision support system was designed and implemented for design computation and decision assessment of environmental control and life support system of manned space station. An advanced multiobjective decision methodology and a hierarchic structure model of assessment index of recovery life support system was developed. The program incorporates a database for each technology option, metabolic design loads associated with crew activity, mission model variables to accommodate evolving mission requirements, and algorithms to produce products criteria in order to provide recommendations relative to candidate technology selection and development. A specific structure was developed for the decision system which consists of a database, a methodology base and a model base as well as their management systems. Moreover, a centre control system with friendly user interface plays a very important role in the man-computer interaction.

[Dynamic analysis of 24 h blood pressure in pilots and hypertensive patients].

Blood pressure of 63 healthy male pilots and 20 primary hypertensive patients was measured with the "Smart LINK Ambulatory Monitoring System". The BP readings were taken every 30 min in daytime and 60 min at night for 24 h. The results showed that two peaks appeared at 7:00-9:00 A.M. and 16:00-19:00 P.M. and two valleys at 12:00-14:00 P.M. and 24:00-5:00 A.M. in healthy pilots, but the hypertensive subjects showed higher systolic or/and diastolic blood pressure continuously.
