[The susceptible alleles on HLA-DRB 1 of type I diabetes in children in Harbin].

OBJECTIVE: To evaluate association between type I diabetes in children and HLA-DRB1 alleles in Harbin. METHODS: Case-control method was employed, and the PCR-SSP technique was used to identify HLA-DRB1 alleles. RESULTS: The frequencies of DRB1 * 0301 and DR3/4 heterozygosity were significantly higher but the frequency of DR2 was significantly lower in cases than that in controls. In DR4 subtypes, the frequency of DRB1 * 0405 was higher and the frequency of DRB1 * 0406 was lower in cases than that in the controls. The difference between cases and controls in DRB1 * 0901 was not significant. CONCLUSION: DRB1 * 0301 allele and DR3/4 heterozygosity were the important susceptibility and heterozygosity genes while DR2 was the most important protective gene in Harbin. The DR4 possessed double-tendency. DRB1 * 0405 conferred susceptibility and DRB1 * 0406 conferred little protection. The susceptibility conferred by DRB1 * 0901 was not certain in type I diabetes in childhood in China. This study provided useful genetic data on the study of etiology of type I diabetes mellitus.

[Studies on human streptococcal infectious syndrome caused by infected pigs].

OBJECTIVES: To demonstrate the clinical manifestations of S. suis infectious syndrome; to study the characteristics of causative organism and its source. METHODS: 25 cases of unknown causes of food poisoning were identified in the central area of Jiangsu province, China in July 1998. Biological specimens was collected in some patients, and the causative organism was isolated using a number of different culture medium. Isolated organisms were identified by serum antibody tests, API biochemical tests, drug sensitivity tests and animal model experiments. The human born strains and the pig born strains were compared by RAPD technique. Epidemiological methods were applied to trace the source of causative organisms. RESULTS: Cases were clinically categorized into two types: Streptococcal toxic shock syndrome (STSS) and streptococcal meningitis syndrome (SMS). Three strains of streptococci were isolated respectively from the blood and cerebrospinal fluid of 6 cases. All these 6 strains were confirmed to be S. suis type 2 by culture characteristics, morphology, biochemical characteristics, serum antibody tests and fatty acid profile analysis. Animal model experiment showed that these strains were sensitive to rabbits and pigs, but not to rats. RAPD fingerprint test revealed an identity between human born and pig born strains. CONCLUSIONS: The aetiological agent of these human cases was S. suis type 2. Human cases were contracted by direct contact with the infected pigs or the corpse of infected pigs.

[Relationship between blood lipid level and apolipoprotein E gene polymorphism in offspring from longevity and non-longevity families].

OBJECTIVE: To study relationship between genetic factor and level of blood lipid. METHODS: Levels of blood lipid and difference in apolipoprotein E (apo-E) allele frequencies were compared in 86 offspring in seven Uigur longevity families and 62 offspring in nine Uigur non-longevity families and their relationship was explored. RESULTS: After adjustment for age and occupation, blood indicators related to atherosclerosis were all lower in the offspring of longevity families than in non-longevity families, and those correlated inversely with it were higher in the former. Blood levels of triglyceride and apo-B were significant lower (P < 0.05) and the ratio of HDL-ch to LDL-ch was significantly higher (P < 0.05) in male offspring of longevity families than of non-longevity families. The ratio of apo-A to apo-B was significantly higher in female offspring of longevity families than of non-longevity families. Frequency of epsilon4 allele also was significantly fewer in the offspring of longevity families than of non-longevity ones (P < 0.01). Also, analysis showed that individuals with epsilon4 allele had higher levels of serum total cholesterol, lower density lipoprotein cholesterol and apo-B than those with other alleles. CONCLUSION: Average level of blood lipid was better in the offspring of longevity families than that of non-longevity families. apo-E allele polymorphism can in part explain its difference.

[A canine model of brain edema established in the early postburn stage].

OBJECTIVE: To establish a canine model of brain edema in the early stage of severe burn. METHODS: Mongrel dogs were inflicted by napalm with 50% TBSA III degree burn. The dogs were randomized into normal control (C), burn (B), burn with balanced salt solution (S) and burn with glucose (G) groups. The macropathological, histopathological and ultrastructural changes of brain tissue and brain water (%) were examined at 6, 12, 18 and 24 postburn hours (PBH), respectively, with simultaneous evaluation of blood brain barrier by (99) TC -- ECD. RESULTS: Pathological evidence of brain edema exhibited at as early as 6 PBH in B, S and G groups, which included cellular swelling of partial endothelia and pericapillary astrocytes, and ischemic necrosis of endothelia, neurons and axons to varying degrees. All the changes exhibited more and more obvious with the elapse of postburn time. The brain water content in each burn group, especially in G group at 24 PBH, was more than that in C group. It was indicated by nuclide imaging that there appeared gradual increasing of the concentration of (99) TC -- ECD in brain tissue in B and C groups since 6 PBH. CONCLUSION: This model was established for the convenience of burn scholars to study the pathogenesis and management of postburn brain edema.

[Study of the expression and function of c-kit receptor on the bone marrow mononuclear cells of the patients with myelodysplastic syndromes].

OBJECTIVE: To determine the expression and function of c-kit receptor on bone marrow mononuclear cells (BMMNC) of the patients with myelodysplastic syndromes. METHODS: c-kit protein (CD(117)) was detected by direct immunofluorescence assay, c-kit mRNA by reverse transcriptive polymerase chain reaction (RT-PCR) and the function of c-kit receptor by cell culture. RESULTS: c-kit protein expression in MDS was significantly higher than that in control [(8.58 +/- 5.28)% vs (3.04 +/- 1.49)%, P < 0.05]. c-kit protein expression in RA patients was significantly lower than that in RAEB/RAEB-t patients [(5.12 +/- 2.13)% vs (10.01 +/- 5.07)%, P < 0.05]. The rate of c-kit protein expression was (32.43 +/- 18.16)% in AML cases transformed from MDS (t-AML). c-kit mRNA expression in MDS was correlated with c-kit protein expression. Addition of interleukin-3 (IL-3), erythropoietin (Epo) and stem cell factor (SCF) to the semi-solid culture of BMMNC showed significant stimulating effects on CFU-GM and BFU-E formation in normal controls, but did not in MDS patients. CONCLUSION: The protein and mRNA expressions of c-kit receptor on BMMNC in MDS are higher than those in normal control, and the function of this receptor in MDS is abnormal.

[Autologous bone marrow transplantation for patients with chronic myelogenous leukemia after in vitro purging of the graft with bcr/abl antisense oligodeoxynucleotides].

OBJECTIVE: To investigate the efficacy of autologous bone marrow transplantation (ABMT) for patients with chronic myelogenous leukemia (CML) after in vitro purging of the graft with bcr/abl antisense oligodeoxynucleotides (AS-ODN). METHODS: Five CML patients, 2 in chronic phase (CP), 1 in accelerated phase (AP) and 2 in blast crisis (BC), all confirmed the presence of b3a2 bcr/abl mRNA by RT-PCR (reverse transcriptase-polymerase chain reaction). Patients had received 2 courses of intensive chemotherapy for "in vivo purging" before bone marrow harvesting. The autologous bone marrow cells were concentrated by CS3000plus and then incubated with 18-mer phosphorothioate bcr/abl AS-ODN (40 - 60 microg/ml, for 48 - 60 h). Conditioning regimens were TBI + Cy (total body irradiation + cyclophosphamide) or MAC + CCNU. RESULTS: After intensive chemotherapy, ph(+) cells in bone marrow reduced to 34% (24% - 46%) and the numbers of bcr/abl mRNA (+) CFU-GM to 45.6% (33% - 58%). After bcr/abl AS-ODN in vitro purging, two patients were bcr/abl mRNA negative and three positive with the number of bcr/abl mRNA (+) CFU-GM markedly reduced. Delayed hematopoietic reconstitutions were observed in all patients after ABMT. During a more than 2 years follow-up period, three patients obtained major cytogenetic remission (MCR) and lasted for 9 - 12 months and the durations of CP after ABMT were longer than that before ABMT. One of this three in BC before ABMT being survived disease-freely for 37 months with bcr/abl mRNA (-) after ABMT. Another patient in BC had a short duration of MCR and relapsed at +7 months. One patient died from severe infection and bleeding at day +74 because of delayed reconstitution. CONCLUSION: ABMT with bcr/abl AS-ODN purged graft can result in quite long duration of MCR and prolonged CP in some patients with CML.

[Ethological examination of Alzheimer's disease model rats treated with bushen yizhi decoction].

OBJECTIVE: To explore the ethological improving effect of Bushen Yizhi Decoction (BSYZD) in treating model rat of Alzheimer's disease (AD). METHODS: Ethologic examination was carried out with Morris water maze on young, aged, AD model, Huperzine A treated and BSYZD treated groups of rats. RESULTS: In the BSYZD treated group, the escape latency (EL) curve placed between that of the young, aged and AD model, Huperzine A treated group; the percentage of middle circle swim path, the 40 cm circle swimming time, and the platform-crossing times were not different from those in the normal aged group, but were different significantly as compared with the AD model group, P < 0.05. CONCLUSION: BSYZD could ameliorate the behavior deficits of AD model.

[Anatomic study on the nervous distribution of the human recurrent laryngeal nerve].

OBJECTIVE: In order to supply the anatomic data for the selective reinnervation operation of the recurrent laryngeal nerve, the nervous distribution of the recurrent laryngeal nerve was investigated. METHOD: 50 total fresh human larynx were systematically dissected by the method of microdissection. RESULT: In most cases, the recurrent laryngeal nerve diverges at the level of 1.0-2.5 cm under the cricothyroid articulation for forming the extralaryngeal division. There are three types of extralaryngeal division. Except the cricothyroid muscle, all the laryngeal intrinsic muscles are innervated by the anterior branch of the recurrent laryngeal nerve. In 88% of the cases, the posterior cricoarytenoid muscle is innervated by two nervous pedicle, the others are innervated by a single nervous pedicle. CONCLUSION: At the level of the posteroinferior part of the thyroid cartilage, by opening a small window, we can easily find the abducent branch of the recurrent laryngeal nerve, following this branch, we can see the adducent branch and the thyroarytenoid branch. The anatomic specifity of the nervous distribution can supply us the data for the operation of selective nervous reinnervation to overcome the possibility of syncinesis of the laryngeal muscle.

[The ejection of ascoporae of Cordyceps sinensis].

This paper has reported the effect of sunshine to stromata of Cordyceps senensis grown in Kanding, Sichuan and Naqu, Tibet. The ejective time, quantity and germination rate of ascoporaes between the two habitats are different with sunshine. To satisfy the needs of sunshine, moisure and temperature can be raised the ejection and germination of ascorporae.

[Trannasal-transsphenoidal endoscopic surgery of the sphenoid sinus and the sella turcica].

OBJECTIVE: The primary objective of this study is reporting our experience with transnasal endoscopic technique to management of the sphenoidal and the sellar lesions. METHOD: Forty one patients were management under endoscope who suffered from pituitary adenomas, craniopharyngioma and sphenoidal lesions, et al. RESULTS: In forty one cases of the sphenoidal and the sellar lesions, thirty two surgeries were successful. One patient died of bleeding, five developed the foramen of nasal septum, the adhesion of nasal cavity was complicated in three patients. CONCLUSION: The endoscope has the advantage of improved visualization, angled view, and a wide panoramic perspective. The transnasal-transspheniodal endoscopic technique is a effective, minimally invasive approach. But there are its disadvantages and occurred severe complication. Surgeons should be understood its feasibility, limits and indication.

Study on modified cold storage method of rat livers with self-made hyd solution.

OBJECTIVE: To investigate the cold preservation effect of rat livers by modified storage method with self-made HYD solution. METHODS: The modified method was that the vascular bed of rat livers was expanded with an additional 20 to 40ml self-made HYD solution/100g liver. After removing the liver, the extra HYD solution expressed as % liver weight was entrapped via portal infusion by tying off the supra-and infra hepatic inferior vena cava. According to the amount of extra HYD solution, 40 rats were randomly divided into four groups including: control group with conventional storage method, 20% group, 30% group and 40% group. The preservation effect of modified storage method with that of conventional storage method by using isolated perfused rat liver model was compared. RESULTS: Bile production and all the indices of hepatic microcirculation including portal perfusion pressure, endothelin-1 in the effluent, trypan blue distribution time and histology in modified method groups were significantly superior to those in control group (P < 0.05). The liver enzymes in 30% group were markedly lower than those in control group (P < 0.05). The preservation effect of rat liver in 30% group was the best among the modified method groups. CONCLUSION: The modified cold stage method is effective and may have potential for clinical application for liver preservation.

[Luminescence of zinc and europium composite alpha-thiophene carboxylate].

Zinc and europium composite alpha-thiophene carboxylate with very strong red luminescence was prepared from zine oxide, europium oxide, and alpha-thiophene carboxylic acid by a rheologic phase reaction method. The thermal stability and X-ray diffraction pattern were measured. The IR, UV, excitation and emission spectra were studied. The influence of zinc ion and thiophene on luminescence intensity was discussed. In zinc and europium composite alpha-thiophene carboxylate, zinc ion can reduce the concentration quenching and effectively enhance the luminescence of Eu3+ ion, thiophene ring can increase the luminescence efficiency.

[Studies on synthesis and IR spectrum of nickel salicylates complexes].

Nickel salicylates complexes were synthesized with the rheologic phase reaction method. The composition of these complexes were confirmed by elemental analysis, TG and IR, the principal infrared absorption peaks were assigned for the region 4,000-400 cm-1. The IR spectra indicated that a hydroxyl group on Ni atom was occurred and so absorption peaks of hydroxyl group were shifted obviously to higher frequency in nickel monosalicylates.

[Analysis of the proportion of salivary gland tumors in 528 elder patients].

OBJECTIVE: To analyse the clinical characteristic, evolution and prognosis of undifferentiated carcinoma with lymphoid stroma(malignant lymphoepithelial The purpose of this study was to investigate the present proportional ratio of salivary gland tumors in elder patients, to provide references for early diagnosis and treatment clinically. METHODS: The medical data of 528 elder patients with salivary gland tumors treated in our department in 1990s were reviewed and analyzed. RESULTS: The number of salivary gland tumors in elder patients had increased in recent years. These tumors were frequently found in patients of 60-74 years. The ratio of benign to malignant was 1.7:1. In benign tumors, the proportion of adenolymphoma approached pleomorphic adenoma. While in malignant tumors, adenoid cystic carcinoma accounted the most. These tumors were most commonly found in the parotid gland, followed by the palate. The prevalence of female had an increase trend. CONCLUSION: The proportion of salivary gland tumor in elder patients in 1990s was different from that in 1980s. This change should attract enough attention form the surgeons.

[Application of Continuous Wave Nd:YAG Laser Irradiation in the Management of Cavernous Hemangioma of Deep Oral-Maxillofacial Regions]

OBJECTIVE:Based on our previous clinical report of Nd:YAG laser irradiation as the means for deeling with cavernous hemangioma in deep oral-maxillofacial regions, this article detailed futher clinical application of this laser, trying to set up a non-surgical method for treatment of ang ioma. The relationship of laser dosage and therapeutic effect and possible surrounding tissue thermal damages were investigated.METHODS:Clinical study was carried out on 31 cases of deep hemangiomas involving different areas. Nd:YAG laser irradiation was delivered after surgical elevation of the flaps. All cases surveyed a follow up of 1.5 to 3 years.RESULTS:Posttherapeutic clinical and imaging examinations were taken to compare the findings before and after treatment. There are 29 cases with complete regression (93.55%), 2 cases with partial regression (6.45%). No complication occurred.CONCLUSION:Nd:YAG laser irradiation after flap elevation has been confirmed to be a valuable means to treat deep hemangiomas. It has the advantages of target tissue selection and normal tissue preservation, simplified manipulation and less bleeding, fewer complications and hihger successful rate.

Real-time measurements of DNA hybridization on microparticles with fluorescence resonance energy transfer.

When capture oligonucleotides are tethered on planar surfaces, mass transport limitations influence the kinetics of solid-phase nucleic acid hybridizations. By diffusion theory, however, hybridization of oligonucleotides on microparticles should be reaction-rate limited. In an initial effort to understand the kinetics of microparticle hybridization reactions, we developed a fluorescence resonance energy transfer method for monitoring oligonucleotide hybridization on microparticles. Microparticles were coated with a fluoresceinated oligomer at surface densities of 20, 40, and 80% saturation, hybridized to a complementary oligonucleotide labeled with tetramethylrhodamine, and monitored over time for quenching of the fluorescein signal as hybridization occurred on the particle surface. Association rate constants were compared for microparticle-based hybridization and solution-phase hybridization. Rate constants for hybridizations on the particle surface were about an order of magnitude less than those for hybridization in solution, but decreasing the surface density of the capture oligonucleotide to 20% saturation improved particle hybridization rates. Although a bimolecular reaction model adequately described solution-phase hybridization kinetics, oligonucleotide hybridization on microparticles did not fit this model but exhibited biphasic reaction kinetics. Based on two different lines of reasoning, we argue that microparticle-based oligonucleotide hybridization was indeed reaction-rate limited in our system and not diffusion-rate limited.

Evidence that apoptosis of activated T cells occurs in spontaneous tolerance of liver allografts and is blocked by manipulations which break tolerance.

BACKGROUND: Fully allogeneic liver grafts from piebald virol glaxo to dark agouti rats are spontaneously tolerated, whereas kidney transplants between these strains are rejected. Liver tolerance is broken by donor irradiation or peritransplant corticosteroid treatment of recipient rats, both of which interfere with the activation of recipient cells. METHODS: In this study we used a combination of immunohistochemical staining, reverse transcription-polymerase chain reaction, and terminal deoxynucleotide transferase-mediated dUTP nick end labeling and Annexin-V apoptosis assays to compare donor cell migration, cytokine profiles, and leukocyte apoptosis in grafts and lymphoid organs from tolerant liver and rejecting kidney recipients. We then examined the effect on apoptosis of treatments which abrogate liver tolerance. RESULTS: Liver transplantation in this tolerant strain combination is accompanied by rapid migration of many passenger leukocytes to the recipient spleen and lymph node, concurrent with a marked but transient increase in the amount of mRNA for the cytokines interleukin-2 and interferon-gamma. Apoptotic cells appear promptly in the spleen, their numbers reaching a peak 2 days earlier than has been previously shown for the graft infiltrate. Both CD4+ and CD8+ T cells undergo apoptosis and apoptotic cells are most concentrated among CD25+ T cells. In contrast, renal transplant rejection is associated with limited donor cell migration to lymphoid tissues and significantly less up-regulation of interleukin-2 and interferon-gamma in the spleen. Few apoptotic cells are detected in spleen or graft infiltrate during rejection, whereas apoptotic renal tubular and glomerular cells are found from day 5. Either recipient steroid treatment or donor irradiation significantly reduced the number of apoptotic cells in liver graft infiltrates and recipient spleen. CONCLUSIONS: Taken together, these findings suggest that a mechanism akin to activation-induced cell death, with apoptosis of alloreactive recipient cells may be responsible for the induction of spontaneous liver transplant tolerance.
