Expression of progesterone receptor in human keratinocytes.

Despite the various responses of human skin to female sex hormones, cellular and subcellular targets and the mechanisms of action of estrogen and progesterone in human skin are not well understood. The detection of estrogen receptor (ER) and progesterone receptor (PR) in the skin is of great importance to understand the effect of estrogen and progesterone. In primary cultures of human keratinocytes, expression of ER and PR was monitored by immunocytochemistry and reverse transcriptase polymerase chain reaction (RT-PCR). Paraffin embedded skin tissues were stained with monoclonal antibodies to human ER and PR by immunohistochemistry. Cultured human keratinocytes expressed cytoplasmic PR protein and PR mRNA transcripts. By contrast, ER was detected only at the mRNA level. Suprabasal keratinocytes from samples of pruritic urticarial papules, plaques of pregnancy (PUPPP) and psoriasis were stained positively only for PR, while those from samples of erythema nodosum were negative for both ER and PR. Lesional epidermis of PUPPP showed positive PR immunoreactivity, while nonlesional epidermis did not. No other cells in the normal human skin were stained with ER and PR. The present study suggests that by expressing PR human keratinocytes act as targets for progesterone action.

Construction of a bacterial artificial chromosome (BAC) library for potato molecular cytogenetics research.

Lack of reliable techniques for chromosome identification is the major obstacle for cytogenetics research in plant species with large numbers of small chromosomes. To promote molecular cytogenetics research of potato (Solanum tuberosum, 2n = 4x = 48) we developed a bacterial artificial chromosome (BAC) library of a diploid potato species S. bulbocastanum. The library consists of 23,808 clones with an average insert size of 155 kb, and represents approximately 3.7 equivalents to the potato genome. The majority of the clones in the BAC library generated distinct signals on specific potato chromosomes using fluorescence in situ hybridization (FISH). The hybridization signals provide excellent cytological markers to tag individual potato chromosomes. We also demonstrated that the BAC clones can be mapped to specific positions on meiotic pachytene chromosomes. The excellent resolution of pachytene FISH can be used to construct a physical map of potato by mapping molecular marker-targeted BAC clones on pachytene chromosomes.

Pretreatment prostate-specific antigen and Gleason score predict the risk of extracapsular extension and the risk of failure following radiotherapy in patients with clinically localized prostate cancer.

The purpose of this study is to evaluate the use of a relatively simple equation for predicting the risk of extracapsular extension (ECE) based on the pretreatment prostate-specific antigen (PSA) and Gleason score (GS) in patients with clinically localized prostate cancer. Three hundred and seventy-four patients who underwent radical prostatectomy between 1988 and 1994 and 521 men undergoing definitive radiotherapy during a similar time period were eligible for this analysis. Surgically treated patients were considered eligible if the pathological stage, preoperative PSA, and GS were available. Among these patients, the median preoperative PSA was 8.1 ng/mL (range, 0 to 195 ng/mL), and the median preoperative GS was 6 (range, 2 to 10). The empirically derived equation tested was (1.5 x PSA + [GS - 3] x 10). For this equation, the range of calculated risk was limited to 0% to 100%. Using the empirically derived equation, patients with a low calculated risk (CR) of < or = 33% had an average calculated risk (ACR) of 21.9% and an observed incidence (OI) of ECE was 17.8%. Patients with a moderate CR of 34% to 66% had an ACR of 46.3%, and an OI of ECE was 46.7%. Patients with a CR of 67% to 100% had an ACR of 83.7% and an OI of ECE of 66.7%. Of the 21 patients who had a PSA < or = 4 and a GS < or = 4, only 1 patient (4.8%) was found to have ECE. Men with an estimated risk of ECE of <33%, 33% to 67%, and >67% had a 4-year risk of biochemical failure following radiotherapy of 29%, 56%, and 78% (P < .00001). This empirically derived data appears to be reasonably accurate at estimating the incidence of ECE in patients with at low or intermediate risk before surgery. The risk of biochemical failure following radiotherapy also correlated the risk of ECE. Future staging systems for prostate cancer should use similar approach for defining risk groups.

Plasma nitric oxide concentrations and nitric oxide synthase gene polymorphisms in coronary artery disease.

BACKGROUND: Plasma NOx (nitrate and nitrite) is a stable end product of the vasodilator NO. Several polymorphisms in the endothelial constitutive NO synthase (ecNOS) gene have been reported, including the 4a/4b VNTR polymorphism in intron 4, the E298D mutation in exon 7, and the G10-T polymorphism in intron 23. The aims of this study were to examine plasma NOx in patients with coronary artery disease (CAD) and to assess the association between plasma NOx concentrations and the three ecNOS gene polymorphisms. METHODS: Plasma NOx was measured in samples from 128 healthy controls and from 110 CAD patients at least 2 months after myocardial infarction. Three genetic polymorphisms that are known or have been suggested to be associated with plasma NOx concentration were also analyzed by PCR-restriction fragment length polymorphism. RESULTS: Median plasma NOx was significantly higher (P <0.001) in CAD patients (95.9 micromol/L) than in controls (73.8 micromol/L). Furthermore, the median plasma NOx was significantly higher (P <0.001) in hypertensive CAD patients (116.0 micromol/L) than in controls and normotensive CAD patients (86.0 micromol/L). The G-allele frequency of the G10-T polymorphism in intron 23 was significantly higher in CAD patients than in controls. Other polymorphisms showed no differences in allelic frequencies among the control and CAD groups. In controls, individuals with the E298D mutation in exon 7 (136.1 micromol/L) showed significantly higher (P = 0.001) median plasma NOx than those without this mutation (64.5 micromol/L). CONCLUSIONS: Plasma NOx was higher in hypertensive CAD patients than in normotensive CAD patients and controls. The E298D polymorphism of the ecNOS gene was associated with increased plasma NOx. Further study is needed to understand the gene expression and enzyme activity of ecNOS and their association with genotypes.

[The second dorsal metacarpal artery flap with distant pivot point].

OBJECTIVE: To introduce a modified flap pedicled with the second dorsal metacarpal artery. METHOD: Seven thumb wounds were repaired with the second dorsal metacarpal artery flap. The size of the flap ranged from 1 cm x 3 cm to 2 cm x 4 cm, and the length of the vascular pedicle was 6 to 7 cm. RESULT: All flaps survived and the results were satisfactory. CONCLUSION: This method is an option for repairing thumb defects when the local skin flap or island flap on the index and middle finger is not available.

[Precise determine the position of the opposite upper lid after correcting the congenital unilateral ptosis and their relationship].

OBJECTIVE: The good results should be obtained after correction of the congenital unilateral ptosis for bilateral eyes. METHODS: A formula was made and determined the position of the opposite upper lid according to Hering's law. RESULTS: Thirty-two patients with congenital unilateral ptosis were operated. The correct rate is 94.12% according to the formula and the control is 66.67%. CONCLUSION: The position of the opposite upper lid should be determined after the ptosis eye corrected before operation in order to obtain a good results.

[Clinical application of the first metacarpal fasciocutaneous flap].

OBJECTIVE: To report the application of the first metacarpal fasciocutaneous flap in repairing digital soft tissue defects. METHODS: The first metacarpal fasciocutaneous flap was designed to repair the lesions of the thumb, index, middle and ring fingers in 14 cases. The blood supply of the flap originates from the branches of the radial pollicis artery at the metacarpophalangeal joint and the venous drainage is to the cephalic vein through the deep pollicis vein and dorsal pollicis vein. RESULTS: All the flaps survived successfully except that one survived partly, two had cyanosis and blister. Followed-up for 3 to 12 months showed the flaps had sensation with good texture and thickness. CONCLUSIONS: The first metacarpal fasciocutaneous flap can be used to repair the lesions of the thumb, index, middle and ring fingers. It has following advantages: 1. reliable blood supply; 2. convenient dissection and 3. less damage of the donor site.

Mechanism of ligustrazini against thrombosis.

OBJECTIVE: To investigate the mechanism of Chinese medicine ligustrazini against thrombosis, and the effects of ligustrazini on plasminogen activator inhibitor (PAI-1) expression in normal endothelial cells and lipopolysaccharide (LPS) stimulated endothelial cells. METHODS: Human umbilical vein endothelial cells (HUVECs) were cultured by trypsin digestion method. PAI-1 protein in HUVEC conditioned medium was measured by Sandwich enzyme-linked immunosorbent assay (ELISA), and PAI-1 mRNA expression was determined by Northern blot analysis. Using electrophoretic mobility shift assay (EMSA), we observed HUVEC nuclear factor-kappa B (NF-kappa B) nuclear translocation. RESULTS: LPS treatment of cultured HUVECs resulted in a significant increase in PAI-1 protein and mRNA expression by these cells. However, when HUVECs were incubated with LPS plus ligustrazini, the upregulation of PAI-1 by LPS was abated. Moreover, ligustrazini could decrease the basal level of PAI-1 protein and mRNA in HUVECs as compared with control. Nuclear extracts prepared from HUVECs stimulated by LPS demonstrated that binding to the NF-kB oligo nucleotide increased as compared with the unstimulated cells, but ligustrazini did not change those binding in the absence or presence of LPS. CONCLUSIONS: Ligustrazini inhibited both basal and LPS-induced PAI-1 protein and mRNA expression in endothelial cells, and the modulation of PAI-1 in HUVECs by ligustrazini might have other mechanisms rather than NF-kB pathway.

Association of coagulation factor VII with the risk of myocardial infarction in the Chinese.

OBJECTIVE: To elucidate the association of plasma factor VII coagulant activity (FVIIc) with the risk of myocardial infarction (MI) and to assess the influence of factor VII gene MspI polymorphism and lipid metabolism on FVIIc in the Chinese. METHODS: A total of 137 patients with angiographically confirmed MI and 125 healthy individuals were evaluated retrospectively. Plasma FVIIc was measured by one-stage prothrombin time, and FVII genotype was determined after MspI digestion of polymerase chain reaction-amplified genomic DNA. Serum lipid levels were assessed by routine methods. RESULTS: MI patients had significantly higher levels of FVIIc (119.5% +/- 22.7% vs 99.9% +/- 21.8%, P < 0.01) and total serum cholesterol (5.80 +/- 1.06 mmol/L vs 5.53 +/- 1.08 mmol/L, P < 0.05) than controls, but only FVIIc independently correlated with the risk of MI (OR = 1.04, P < 0.01). There were no significant differences in FVII genotype or allele frequency between patients and controls (P > 0.05). Subjects with the Gln353 allele were associated with significantly lower FVIIc levels than Arg353 homozygotes (99.7% +/- 19.3% vs 111.4% +/- 24.6%, P < 0.05). Serum triglyceride was positively correlated with plasma FVIIc in both control (r = 0.25, P < 0.01) and case (r = 0.87, P < 0.01) groups, but this correlation was restricted to Arg/Arg genotype (r = 0.68, P < 0.01). A significant correlation of total serum cholesterol with FVIIc only appeared in Arg/Arg homozygotes (r = 0.17, P < 0.01). CONCLUSIONS: Our findings support the role of plasma FVIIc as a risk factor for MI in Chinese. Plasma triglyceride and FVII gene MspI polymorphism are two independent determinants of FVIIc. Assay of this polymorphism will be helpful in determining who will benefit most from lipid-lowing therapy.

[A study on the mechanism of anisodamine against thrombosis].

OBJECTIVE: To investigate the effects and the mechanism of Chinese herb anisodamine on PAI-1 expression in LPS stimulated endothelial cells (ECs). METHODS: Human umbilical vein endothelial cells (HUVECs) were cultured by trypsin digestion method. PAI-1 protein in HUVEC conditioned medium was measured by sandwich enzyme-linked immunosorbent assay (ELISA), and PAI-1 mRNA expression in HUVEC was determined by Northern blot analysis. HUVEC nuclear factor-kappa B (NF-kappaB) nuclear translocation was assessed by immunocytochemistry technique. RESULTS: LPS treatment of cultured HUVECs resulted in a significant increase of PAI-1 protein as well as mRNA expression by these cells. However, when HUVECs were incubated with LPS plus anisodamine, the upregulation of PAI-1 by LPS was significantly abated. The expression of PAI-1 protein and mRNA was decreased by 30% and 24% respectively compared with LPS treatment alone. Moreover, anisodamine was able to decrease the basal level of PAI-1 protein and mRNA by 74% and 70% respectively. Furthermore, anisodamine could block LPS induced NF-kappaB of HUVECs nuclear translocation. CONCLUSION: Anisodamine inhibits both basal and LPS-induced PAI-1 protein as well as mRNA expression in ECs. The inhibition of anisodamine on LPS-induced PAI-1 in ECs may take place via the NF-kappaB pathway.

[The preliminary analysis on the changes of the oral functional space in Class II division 1 malocclusion after triggered by functional appliance].

OBJECTIVE: The purpose of this paper is to study the area changes of the oral functional space in the patients with Angle's Class II division 1 malocclusion after triggered by FR-I functional appliances. METHODS: Twenty patients with Angle's Class II division 1 malocclusion were selected, and the patients were selected by the standards of an horizontal growth pattern with normal maxilla, retracted mandible and short lower facial height. All patients were triggered by FR-I functional appliances and achieved the Angle's Class I occlusion after treatments. The average periods of the treatments were about 9 months. The assessments were made from the lateral cephalograms taken in centric occlusion before and after treatments. The upper and behind borders of the oral functional space are hard palate and soft palate, the forward border is the lingual side of upper and lower incisors, the lower border is the line between apex of lower incisor and lower first molar. The graphics from the lateral cephalogram were painted and input into the computer by scaner, which were measured by special software. Self-controls of individuals before and after treatments were used in statistic analysis. RESULTS: The paired-samples t test confirmed the oral functional space was enlarged after triggered by FR-I functional appliance, which was induced by obviously increased mandibular body and ramus, anterior lower facial height and posterior facial height. At the same time, the angle of Y axis increased and the upper incisor inclined lingually, but the inclining angle of lower incisor was not obvious. CONCLUSION: Enlarged oral functional space facilitates the development of the maxilla, mandible and alveolar bones and consummates the functions of oral and jaw system. This study aids the design and application of FR-I functional appliance.

[A study on the intrinsic compensative mechanism of craniofacial soft and hard tissue structures of Uygur adults with normal occlusion].

OBJECTIVE: To study the intrinsic compensative mechanism of craniofacial soft and hard tissue structures of Uygur adults with normal occlusion and establish standard cephalometric values to evaluate craniofacial soft and hard tissue structures of normal Uygur adults. METHODS: Lateral cephalometric radiographs of 93 Uygur adults with normal occlusion (43 men and 50 women) were evaluated with the same standard. Their age ranged from 18 to 25 years old, and they had normal and well-balanced faces without receiving any orthodontic treatment. Based on the study of craniofacial soft and hard tissue structures with normal occlusion, the horizontal tendency, the vertical tendency pattern and the medial pattern of craniofacial soft and hard tissue structures were compared, and the intrinsic compensative mechanism in the inner structures of the horizontal and the vertical tendency patterns was investigated. Finally 21 indexes were analysed by mean squar deviation and Newman-Keuls test. RESULTS: The obvious compensative changes in the horizontal and vertical tendency patterns were found in teeth, skeleton and soft-tissues. 1. When the horizontal tendency was compared with the medial pattern, the value of ODI and angles of FH-NA and FH-NB increased (P < 0.01), while the angle of SN-PP decreased (P < 0.01). It showed that the anterior maxilla rotated in the counter-clockwise direction, and the anterior mandible rotated in the clockwise direction. The angle of U1-L1 increased (P < 0.01), which meant that the inclination of anterior teeth decreased. 2. When the vertical tendency was compared with the medial pattern, the value of ODI decreased (P < 0.01), while the height of ANS-Me, U1-PP, L1-MP, and the thickness of LL-SnPg increased (P < 0.01); The angle of L1-MP decreased (P < 0.05), while the length of Sn-Stms increased (P < 0.05). These results showed that there was an obvious positive relationship between hard and soft tissues. The soft tissue thickness could compensate the changes of hard tissues to keep the facial patterns harmony. CONCLUSION: Normal occlusion depends on the homeostasis of teeth, skeleton and soft tissues, but doesn't only depend on the normal craniofacial soft and hard tissue structures respectively. The intrinsic compensative mechanism plays an important role in forming normal occlusion, therefore, doctors should choose the best treatment to obtain optimal individual intrinsic compensative mechanism and achieve an optimal balance between the craniofacial structures and the occlusion.

[An cephalomectric study on craniofacial structures of Uygur adults with normal occlusion in Xinjiang autonomous region].

OBJECTIVE: To study craniofacial structures of Uygur adults with normal occlusion and establish the standard cephalometric values to evaluate the craniofacial soft and hard tissue structures of normal Uygur adults. METHODS: Lateral cephalometric radiographs of 93 Uygur adults with normal occlusion (43 men and 50 women) were evaluated with the same standard. Their age ranged from 18 to 25 years old, and they had normal and well-balanced faces without receiving any orthodontical treatment. The lateral cephalogram of each normal adult was traced digitized and measured by the same computer system, then the indexes of craniofacial structures were analyzed by multifactor cluster analysis. 18 typical items were chosen to analyze the characters of the craniofacial structures of Uygur adults with normal occlusion. At the same time, Bayers discriminant analysis was used to identify and evaluate the cluster results. RESULTS: Uygur adults with normal occlusion were divided into three patterns according to the multifactor cluster analytic results: horizontal tendency pattern (40%), medial pattern (43%) and vertical tendency pattern (17%). When the horizontal tendency pattern was compared with the medial pattern, it showed that the height of S-Go increased (P < 0.05), while the height of ANS-Xi-Pm decreased (P < 0.05); The proportion of S-Go/N-Me increased (P < 0.01). The angle of Ar-Go-Me and Y axis decreased (P < 0.05), which meant that the mandible rotated in the counter-clockwise direction. SN-MP, PP-MP, OP-MP, MP-FH all decreased (P < 0.01), which meant that the SN plane, the PP plane, the occlusion plane and the MP plane all had the trend of hypodivergence. When the vertical tendency pattern was compared with the medial pattern, the height of Ar-Fo decreased (P < 0.01), while the height of ANS-Me increased (P < 0.01); The angle of Ar-Go-Me, Y axis, SN-MP, PP-MP, OP-MP and MP-FH all increased (P < 0.01). These results showed that the mandible rotated in the clockwise direction and the SN plane, the PP plane, the occlusion plane, and the MP plane all had the trend of hyperdivergence. At last, according to the different skeletal patterns, the normal cephalometric values of Uygur adults with normal occlusion were established. CONCLUSION: Significantly craniofacial structure difference exist in the adults with normal occlusion. Therefore, it is necessary to establish the normal cephalometric values of Uygur adults with normal occlusion according to different skeletal patterns.

[Biological effects of supplements on soil properties and the growth of Panax quinquefolium].

This paper suggests that the physical and chemical characteristics of soil in American ginseng field were greatly improved after utilization of supplement on soil. Compared with the contrast, the soil gravity ratio decreased from 0.812 g/cm3 to 0.715 g/cm3, while the soil porosity increased from 69.36% to 73.03%. These improvements are benefit for the growth of Panax quinquefolium. Biological observations show that the fresh weight of ginseng root enhanced 53.8%, and the content of total saponins in root enhanced 0.5%-1% (get to 8.28%).

[Partial laryngectomy for T3 glottic carcinoma].

OBJECTIVE: To explore the possibility of partial laryngectomy for T3 glottic carcinoma and selection of reconstruction methods. METHODS: From January 1986 to January 1994, 78 patients with T3 glottic carcinomas were treated by partial laryngectomy. The operative techniques and the reconstruction methods were retrospectively analyzed. RESULTS: The overall 3 and 5 years survival rates were 88.5% and 75.6% respectively. The local recurrence rate was 15.4%. The reconstruction methods for laryngeal defect repariring following partial laryngectomy were the use of pyriform sinus mucosa with thyroid perichondrium, epiglottic cartilage and bi-pedicaled and bi-muscular flap of strap muscle. The function of respiration, phonation and deglutition recovered well. CONCLUSION: The partial laryngectomy is suitable for T3 glottic carcinoma. The selection of reconstruction method is based on the size of laryngeal defect.
