[A preliminary study on the inhibitory action of target-primaqine on the exoerythrocytic forms of Plasmodium yoelii yoelii].

Target-primaquine (TPQ)--the primaquine entrapped by galactosylneoglycoalbumin (G-NGA), was given intravenously (iv) to Wistar rats 2 h after sporozoite inoculation. The sporozoite viability (SPV) of TPQ 20 mg/kg group was revealed to be 1.2% 42 h after inoculation, which showed a decline of about 6% as compared to the control; besides, degeneration or retardation of development was present in 90% of EEF. In TPQ 10 mg/kg and PQ 20 mg/kg groups, the SPV were 2.8% and 5.0%, respectively, showing drops by 15% and 26% as compared with that of the control. Degeneration or retardation in development was also visible in about half of the EEF. In mice which were treated iv with TPQ 10 mg/kg or PQ 20 mg/kg 2 h after sporozoite inoculation, the prepatent periods were delayed markedly as compared with that of the control. Parasitemia did not appear in 2 mice in 10 mg/kg TPQ group further indicated the effectiveness of the drug. The results suggest that the anti-EEF effect of TPQ is apparently more promising than that of PQ.

[Repair of soft tissue defect on the hand using antebrachial posterior interosseous artery retrograde island flap].

Eight cases of hand defects have been repaired with the antebrachial posterior interosseous artery retrograde island flap. The vessel of the flap is constant and rich in cutaneous branches. The flap has good blood supply, larger surface, appropriate thickness and good skin texture with well-matched color. The operation is simple with high rate of success and does not need to sacrifice a major artery in the forearm.

[Effects of injury to endothelial cells on the binding of t-PA to cultured human umbilical vein endothelial cells].

Tissue plasminogen activator (t-PA) plays an important role in fibrinolysis. When t-PA is bound to endothelial cells (EC), its catalytic efficiency is increased and is prevented from being inhibited by its inhibitor PAI. In this paper, diamine was used to cause injury to cultured human umbilical vein endothelial cells by lipoperoxidation, and selenium was used as an anti-oxidant. Radioassay and autoradiography were used to detect the effects of injury on the binding ability of t-PA to EC. The results showed that the binding ability of t-PA to EC was reduced significantly after injury to EC by lipid peroxidation. Selenium could inhibit this reduction to a certain extent. It suggests that the decrease of fibrinolysis was related with the decrease of the binding ability of t-PA to EC after EC was injured.

Study on the mutagenicity of diesel exhaust particles.

The mutagenicity of diesel exhaust particles (DEP) was studied by using Salmonella typhimurium strains TA98 and TA100 in vitro and mice micronucleus in vivo test. DEP from six kinds of medium and heavy-duty diesel vehicles, which were made in China and imported, were tested. The vehicles were operated under free accelerating condition. The results showed that the DEP contained mutagenic activity. An increase in the number of the Salmonella TA98 was observed in the presence and especially in the absence of S9 mix. Positive results were also obtained from mice micronucleus assay. The frequency of mice bone marrow micronucleated polychromatic erythrocytes (M PCE) was increased and it showed a definite dose-response relationship. Comparing the different types of the vehicles, we found that the mutagenicity of DEP from domestic made vehicles was stronger than that from the imported ones.

Crown gall culture and production of tanshinone in Salvia miltiorrhiza.

Crown galls were induced by direct infection of sterile seedlings with Agrobacterium tumefaciens C58, and the transformation was proved by opine identification. The crown galls grew well in a hormone-free medium. B5 and MS basic media were good for the growth of crown galls (strain Ca), which increased up to 102 and 90 times in a month, respectively. However, 67-V and WP basic media are good for tanshinone production. It has been shown that crown galls can be utilized as a culture system to produce secondary metabolites.

Socio-economic status and micro-environmental factors in relation to the risk of Japanese encephalitis: a case-control study.

In a population-based case-control study in Southern Henan Province, central China, children suffering from Japanese encephalitis (JE) were compared with neighborhood controls matched by age and sex in terms of several social and environmental variables. Factors found by crude analysis to be associated with an increased risk of JE included lower family income, lower parental education, living in houses near the periphery of villages and poor quality of houses. When adjustment was made for confounding variables, only the association of house location within village remained of borderline significance (OR = 0.51, 95% CI = 0.15 approximately 1.03). It is suggested that the beneficial effect of higher family income and parental education was partly due to the fact that those parents might be more conscious about having their children vaccinated in the situation where there was a shortage of JE vaccine in the study area.

[Clinical use of the C-shaped cross finger flap].

The C-shaped cross finger flap is an axial flap based on the digital vascular bundle either distally or proximally. We have successfully used this flap to repair different defects of the finger in 12 cases. The flap can be made rather big and the operation is easy. The surgical technique and the advantages and disadvantages are described.

[Gamma interferon-like immunoreactivity in adrenal gland and cortical tumor].

Interferon-gamma (IFN-gamma) has many important immunoregulatory functions. It has previously been presumed to be produced by activated lymphoid cells alone. Recently, IFN-gamma like immunoreactivity has been described in certain parts of the nervous system and the skeletal or cardiac muscle system in the rat. We wanted to know whether this molecule could be expressed in the human adrenal gland and cortical tumors. Adrenal tissue samples were taken adjacent to 14 tumor cases, from 17 adrenal cortical adenomas, and 7 cortical carcinoma cases. Deparaffinized 5 microns sections were stained with IFN-gamma monoclonal DB-1 antiserum. The avidin-biotin complex method was used. IFN-gamma appeared in the cytoplasm of tumor cells in 7/7 cases of adrenal cortical carcinoma, 8/17 cases of cortical adenoma and endothelium in 2/7 cases of adrenal cortical carcinoma and 5/17 cases of cortical adenoma. No immunoreactivity was observed in tumor adjacent adrenal gland. Therefore, IFN-gamma is associated with cell differentiation, the higher the immunoreactivity, the lower the differentiation. The capillary endothelium in adrenal cortical tumor tissue may possess receptor for IFN-gamma or can produce IFN-gamma.

Demonstration of 5'-nucleotidase activity in unfixed cryostat sections of rat liver using a combined light- and electron-microscope procedure.

In the present study a technique was developed to demonstrate 5'-nucleotidase activity in unfixed cryostat sections of rat liver at the light- and electron-microscope level using a semipermeable membrane. In order to retain the ultrastructure of the unfixed material as much as possible, incubations were also performed at 4 degrees C rather than at 37 degrees C. The optimized incubation medium contained 300 mM Tris-maleate buffer, pH 7.2, 5 mM adenosine monophosphate as substrate, 30 mM cerium chloride as capturing agent for liberated phosphate, 10 mM magnesium chloride as activator and 1.5% agar. At the light-microscope level, similar localizations of 5'-nucleotidase activity were obtained when incubations were performed at 37 degrees C and 4 degrees C. Enzyme activity was present mainly at bile canalicular membranes and at sinusoidal membranes of hepatocytes; total activity was higher in pericentral than in periportal areas. Cytophotometric analyses revealed that specific formation of final reaction product (FRP) (test minus control reaction) at 37 degrees C followed a hyperbolic curve with time. A linear relationship was found between specific amounts of FRP and section thickness up to 8 micrograms. 5'-Nucleotidase activity was about three-fold higher after incubation for 30 min at 37 degrees C than at 4 degrees C. At the electron-microscope level, it was demonstrated that the ultrastructure of rat liver was rather well-preserved after incubating unfixed cryostat sections attached to a semipermeable membrane and electron-dense FRP was found at bile canalicular and sinusoidal plasma membrane of hepatocytes. The most distinct changes in ultrastructure after incubation at 37 degrees C, in comparison with that at 4 degrees C, were the appearance of multi-lamellar structures at bile canaliculi at 37 degrees C. We conclude that the present method is valid for the demonstration of 5'-nucleotidase activity in unfixed cryostat sections of rat liver at both the light- and electron-microscope levels and that hypothermic incubations improve ultrastructural morphology substantially.

Prognostic factors of early sequelae and fatal outcome of Japanese encephalitis.

A clinical case control study to identify prognostic factors present at hospital admission associated with early sequelae and fatal outcome of acute Japanese encephalitis (JE) was carried out in Gusi county, Henan Province, central China from June to September 1991. A total of 70 patients with laboratory-confirmed acute JE were studied, of whom 3 cases died and 33 cases had neurological or psychiatric sequelae at the end of three months follow-up. The results showed that acute JE at younger age, with higher body temperature, high white cell count in CSF, and deep coma present at hospital admission were markers for unfavorable outcomes (sequelae or fatal). A history of the vaccination was not correlated with the early sequelae and fatal outcome of the disease. The paper suggests that early diagnosis and treatment and universal JE vaccination for all susceptible populations are keys for decreasing incidence of sequelae and fatal outcome of acute JE.

Phosphatidylcholine-specific phospholipase D activity is elevated in v-Fps-transformed cells.

Activating the protein-tyrosine kinase of v-Fps results in a rapid increase in diglyceride (DG) in rat fibroblasts. The v-Fps-induced increases in DG were detected only when phospholipids were prelabeled with [3H]-myristate, which is incorporated primarily into phosphatidylcholine (PC). Inhibition of phosphatidic acid (PA) phosphatase (PAP), which converts PA to DG, blocked v-Fps-induced DG production. PA is a primary metabolite of type D phospholipases (PLD). Consistent with these observations, PLD activity was activated in response to the kinase activity of v-Fps. The increased PLD activity was detected only when the cells were prelabeled with the PC-specific [3H]-myristate. These data support the hypothesis that v-Fps-induced DG is derived from PC via the PLD/PAP pathway.

Molecular cloning and characterization of the cDNA coding for the biotin-containing subunit of 3-methylcrotonoyl-CoA carboxylase: identification of the biotin carboxylase and biotin-carrier domains.

Soybean genomic clones were isolated based on hybridization to probes that code for the conserved biotinylation domain of biotin-containing enzymes. The corresponding cDNA was isolated and expressed in Escherichia coli through fusion to the bacterial trpE gene. The resulting chimeric protein was biotinylated in E. coli. Antibodies raised against the chimeric protein reacted specifically with an 85-kDa biotin-containing polypeptide from soybean and inhibited 3-methylcrotonoyl-CoA carboxylase (EC 6.4.1.4) activity in cell-free extracts of soybean leaves. Thus, the isolated soybean gene and corresponding cDNA code for the 85-kDa biotin-containing subunit of 3-methylcrotonoyl-CoA carboxylase. The nucleotide sequence of the cDNA and portions of the genomic clones was determined. Comparison of the deduced amino acid sequence of the biotin-containing subunit of 3-methylcrotonoyl-CoA carboxylase with sequences of other biotin enzymes suggests that this subunit contains the functional domains for the first half-reaction catalyzed by all biotin-dependent carboxylases--namely, the carboxylation of biotin. These domains are arranged serially on the polypeptide, with the biotin carboxylase domain at the amino terminus and the biotin-carboxyl carrier domain at the carboxyl terminus.

Pseudomonas aeruginosa possesses homologues of mammalian phenylalanine hydroxylase and 4 alpha-carbinolamine dehydratase/DCoH as part of a three-component gene cluster.

Pseudomonas aeruginosa possesses a multigene operon that includes phenylalanine hydroxylase (PhhA; phenylalanine 4-monooxygenase, EC 1.14.16.1). phhA encodes PhhA (M(r) = 30,288), phhB (M(r) = 13,333) encodes a homologue of mammalian 4 alpha-carbinolamine dehydratase/homeodomain protein transregulator, and phhC encodes an aromatic aminotransferase (M(r) = 43,237). The reading frames specifying phhB and phhC overlap by 2 bases. The P. aeruginosa PhhA appears to contain iron and is pterin dependent. Unlike the multimeric mammalian hydroxylase, the native P. aeruginosa enzyme is a monomer. The P. aeruginosa PhhA is homologous with mammalian PhhA, tryptophan hydroxylase, and tyrosine hydroxylase. Expression of PhhA from its native promoter required phhB. This may suggest a positive regulatory role for phhB, consistent with the dual catalytic and regulatory roles of the corresponding mammalian homologue.

A chimeric gene (orf256) is expressed as protein only in cytoplasmic male-sterile lines of wheat.

Mitochondria derived from Triticum timopheevi have a chimeric gene, orf256, immediately upstream from coxI. Antibodies to a peptide corresponding to a part of the encoded amino acid sequence of orf256 detect a 7 kDa protein on western blots of mitochondrial proteins from cytoplasmic male-sterile (cms) wheat (T. aestivum nucleus, T. timopheevi mitochondria) but not in mitochondrial proteins from T. aestivum, T. timopheevi, or cms plants restored to fertility by introduction of nuclear genes for fertility restoration. The 7 kDa protein appears to serve as a marker for cms wheat. Its occurrence as an integral protein of the inner membrane may indicate a cms effect through an influence on mitochondrial membrane function.

Spatial and temporal expression of basic fibroblast growth factor (FGF-2) mRNA and protein in early Xenopus development.

We have analyzed the expression pattern of bFGF (FGF-2) mRNA and protein in early Xenopus development using RNAse protections, in situ hybridization and immunocytochemical methods. We find that the maternal bFGF mRNA content is at its highest in the previtellogenic oocytes (stage I-II) but decreases during further oogenesis. This low maternal level persists into the early embryonic stages and is uniformly distributed on an RNA basis. Zygotic transcription is turned on both from anterior and posterior regions but not from the middle region in the mid-neurula stage, and the expression greatly increases during the late neurula and tailbud stages. In the tadpole stage, the expression is detected in the brain, eyes, ears and neural crest-derived mesenchyme of the head. Also, it is expressed along the mesodermal axis, the level falling as the myotomes differentiate. Immunocytochemical study shows abundant bFGF protein in early oocytes and much less in later oocytes. The localization is mainly to the nuclei of the early oocytes and to the cytoplasm of the later oocytes. Localization of maternal bFGF protein in the animal hemisphere is observed in the early embryonic stages and some reuptake into nuclei occurs by the early blastula stage. The zygotically synthesized protein starts to be expressed in the anterior region of the mid-neurula stage and soon also becomes detectable in the posterior region. By tailbud and pre-larval stages, this zygotic protein appears to be present along the entire neural and mesodermal axis. When these cases are sectioned it can be seen that bFGF protein is detected in most parts of the head. In the posterior domain, it is present in the mesoderm and clearly becomes localized in the muscle cells to the nuclei and to the cell termini adjoining the intermyotomal septa.

Toward cataloguing all rice genes: large-scale sequencing of randomly chosen rice cDNAs from a callus cDNA library.

A large-scale sequence analysis of rice cDNA was performed for a library from rice callus cultured in a medium containing 1 p.p.m. of 2,4-dichlorophenoxyacetic acid. Random sequencing of 2778 cDNA clones generated 2259 non-redundant expressed sequence tags (ESTs). The strategy of sequencing cDNAs can yield quickly a large number of novel genes. After translation, 690 sequences showed a significant amino acid sequence similarity to sequences already known from PIR. The source of known proteins ranged from bacteria to human. In this report, the non-redundant set of 280 identified ESTs is analyzed in detail.
