The role of magnesium in regulating CCK-8-evoked secretory responses in the exocrine rat pancreas.

This study investigates the effect of magnesium (Mg2+) on the secretory responses and the mobilization of calcium (Ca2+) and Mg2+ evoked by cholecystokinin-octapeptide (CCK-8) in the exocrine rat pancreas. In the isolated intact perfused pancreas CCK-8 (10(-10) M) produced marked increases in juice flow and total protein output in zero and normal (1.1 mM) extracellular Mg2+ [Mg2+]o compared to a much reduced secretory response in elevated (5 mM and 10 mM) [Mg2+]o. Similar effects of perturbation of [Mg2+]o on amylase secretion and 45 Ca2+ uptake (influx) were obtained in isolated pancreatic segments. In pancreatic acinar cells loaded with the fluorescent bioprobe fura-2 acetomethylester (AM), CCK-8 evoked marked increases in cytosolic free Ca2+ concentration [Ca2+]i in zero and normal [Mg2+]o compared to a much reduced response in elevated [Mg2+]o. Pretreatment of acinar cells with either dibutyryl cyclic AMP (DB2 cAMP) or forskolin had no effect on the CCK-8 induced changes in [Ca2+]i. In magfura-2-loaded acinar cells CCK-8 (10(-8) M) stimulated an initial transient rise in intracellular free Mg2+ concentration [Mg2+]i followed by a more prolonged and sustained decrease. This response was abolished when sodium (Na+) was replaced with N-methyl-D-glucamine (NMDG). Incubation of acinar cells with 10 mM Mg2+ resulted in an elevation in [Mg2+]i. Upon stimulation with CCK-8, [Mg2+]i decreased only slightly compared with the response obtained in normal [Mg2+]o. CCK-8 caused a net efflux of Mg2+ in pancreatic segments; this effect was abolished when extracellular sodium [Na+]o was replaced with either NMDG or choline. The results indicate that Mg2+ can regulate CCK-8-evoked secretory responses in the exocrine pancreas possibly via Ca2+ mobilization. Moreover, the movement of Mg2+ in pancreatic acinar cells is dependent upon extracellular Na+.

Piperine impairs cytochrome P4501A1 activity by direct interaction with the enzyme and not by down regulation of CYP1A1 gene expression in the rat hepatoma 5L cell line.

Modulation of benzo(a)pyrene (BP) metabolism and regulation of CYP1A1 gene expression by piperine in 5L cells in culture was studied. Treatment of cultures with 60 microM piperine for different time periods inhibited metabolism of BP by 50% within 4-8 h. Piperine uptake in 5L cells attained saturation plateau at 8 h and this related with the maximal impairment of BP metabolism. Exposure of cell cultures to piperine for 24 h indicated activation of CYP1A1 gene transcription. There was a 10-fold increase in CYP1A1mRNA and an approximately 7-fold increase in arylhydrocarbon hydroxylase (AHH) activity, while treatment with benzanthacene (BA) increased CYP1A1mRNA by 86- and AHH by 56-fold. Combined treatment with BA plus piperine further increased CYP1A1mRNA contents by about 25%. The BA-inducible AHH activity, however, registered a decrease of about 45%. Piperine neither destroyed CYP1A1-protein nor affected the total cellular protein contents. Exposure of cultures to 0.01 to 3.0 microM trans-7,8-dihydroxy-7,8-dihydrobenzo(a)pyrene for 24 h reduced maximum cellular growth by about 50%. Piperine at 60 microM offered full protection against the diol cytotoxicity. The results, suggest that piperine mediated inhibition of the AHH activity and consequent suppression of the procarcinogen activation is the result of direct interaction of piperine with CYP1A1-protein and not because of down regulation of the CYP1A1 gene expression.

Requirement of a CCGAC cis-acting element for cold induction of the BN115 gene from winter Brassica napus.

Mutation of the core pentamer, CCGAC, of two putative low temperature responsive elements (LTREs) in the 5'-proximal region for the winter Brassica napus cold-induced gene BN115 was carried out. Analyses of transient expression of the resultant mutated BN115 promoter-GUS fusions revealed the loss of low-temperature regulation by the promoter. This indicates that the CCGAC sequence is critical to the low-temperature response in the BN115 gene. In contrast, mutation of two G-boxes, CACGTG, staggered between the LTREs in the same region of the promoter did not alter cold-inducible gene expression. Replacement of a possible enhancer region of the BN115 promoter with the enhancer from the CaMV 35S promoter resulted in a several-fold increase in low temperature-induced GUS activity.

Clinico-pathological studies on the effect of different anti-neoplastic chemotherapy regimens on transmissible venereal tumours in dogs.

Thirty-two dogs affected with transmissible venereal tumour (TVT) were divided into three treatment groups. In group I vincristine sulphate at 0.025 mg/kg body weight, in group II vinblastine sulphate at 0.150 mg/kg body weight, and in group III vinblastine sulphate at 0.100 mg/kg body weight plus methotrexate at 0.35 mg/kg body weight were given intravenously at weekly intervals. Biopsies were performed on days 0, 3, 7 and 14. The tissues were preserved in 10% neutral buffered formalin and processed routinely for haematoxylin and eosin staining. Histopathologically, the untreated TVT was characterized by sheets or bundles of mostly rounded cells having a large, highly basophilic nucleus with a prominent, highly basophilic necleolus. Both vincristine and vinblastine primarily affected the nuclei of neoplastic cells, causing condensation, karyorrhexis and karyolysis within 3 days of chemotherapy. The regressing tumour mass showed marked infiltration by lymphocytes, lymphoblasts and macrophages by day 7. There was nearly complete regression of the tumour by day 14, as shown by the almost complete loss of neoplastic cells, with fibrous tissue substitution. However, in group III, the changes occurred more slowly and more injections were needed for complete regression. In both groups I and II, 11/12 of the animals responded completely to the chemotherapy within 3 weeks, while in group III, 6/8 of the dogs responded to the treatment by 21-28 days.

Immunohistochemical study of leucine-enkephalin and its secretory effects in the isolated pig lacrimal gland.

BACKGROUND: In our previous studies immunohistochemical studies have demonstrated the presence of leucine-enkephalin (Leu-Enk) in the intrinsic nerves of the pig lacrimal gland, which are discernible in the interlobular and interacinar areas from where branches that innervate the acinar tissues are sent. Since the intrinsic nerves have been shown to contain Leu-Enk, this study aimed to investigate the secretory effects of this neuropeptide in isolated segments of the pig lacrimal gland and to reconfirm its presence in the neuronal tissue of the lacrimal gland. METHODS: Leu-Enk was identified using immunohistochemical techniques, while total protein output was measured in the effluent samples by an automated on-line colorimetric method. The Ca2+ and Mg2+ concentrations in the effluent samples were determined using an atomic absorbance spectrophotometer. RESULTS: Leu-Enk (10(-12)-10(-7) M) evoked marked increases in total protein output from superfused lacrimal gland segments. The secretory effect of Leu-Enk was not blocked by pretreatment of the tissue with atropine but was substantially reduced by a combination of phentolamine and propranolol. The competitive antagonist, naloxone, has no effect on basal protein output, but when combined with Leu-Enk it caused a significant reduction in total protein output. Combining theophylline (10(-3) M) with Leu-Enk resulted in a marked potentiation of total protein output. In superfused lacrimal gland segments Leu-Enk (10(-8) M) evoked a net efflux of magnesium (Mg2+ release) and a net influx of calcium (Ca2+ intake). CONCLUSION: The results indicated a physiological role for Leu-Enk in the regulation of protein secretion in the pig lacrimal gland.

Comparison of two methods for determining in vitro intestinal absorption of nutrients using rats fed different diets.

Absorption of sucrose, glucose, leucine and aspartate was studied using intestinal everted sac of rats fed on french bean diets namely PDR-14, HUR-137 and HUR-15 using casein as a control. Absorption of nutrients was monitored spectrophotometrically and by 14C radio assay of metabolites using scientillation counting. The absorption pattern of amino acids was found to be similar but of glucose and sucrose differed. Glucose was found to be more absorbed than sucrose in spectrophotometer assay and the pattern reversed in radio assay. Absorption of sucrose and leucine were higher by rats fed on HUR-137 diet and similarly, more aspartate was absorbed when fed on HUR-15 diet as demonstrated by both the methods. Rats fed on HUR-137 diet exhibited higher glucose absorption as shown by spectrophotometric assay, but rats on HUR-15 diet by radio assay. Absorption of nutrients differed significantly between casein and french beans.

Immunohistochemical identification and effects of atrial natriuretic peptide, pancreastatin, leucine-enkephalin, and galanin in the porcine pancreas.

This study demonstrates the presence and distribution of atrial natriuretic peptide (ANP) pancreastatin (PST), leucineenkephalin (Leu-ENK), galanin (GAL), and insulin in the pig pancreas. The effects of PST, ANP, Leu-ENK, and GAL on protein and amylase secretion were also investigated to determine their functional role in the control of pancreatic secretion. PST-immunoreactive cells were observed in the islet of Langerhans and in the wall of the ducts. Leu-ENK-immunopositive cells were observed in both the endo-and exocrine pancreas. It is colocalized with insulin in the islet of Langerhans. ANP immunoreactivity was discernible in nerve fibers and cells of the exocrine pancreas. GAL-immunopositive cells were observed in close association with insulin-positive cells in the islets of Langerhans and in the exocrine pancreas. Stimulation of isolated pancreatic segments with either ANP or Leu-ENK resulted in increased protein secretion and amylase output. The Leu-ENK-evoked amylase secretion was antagonized by naloxone. Pancreastatin was effective at all concentrations, but low concentration had more marked secretory effects whereas GAL failed to evoke any significant increases in either protein or amylase secretion. The results of the study have demonstrated a close association of peptidergic fibers with the secretory cells of the pancreas. The nerve fibers can release peptides that in turn can stimulate protein and amylase secretion.

A radioimmunoassay combined with solid-phase extraction for the determination of a novel anti-obesity agent, ARL 15849XX, in dog plasma.

A radioimmunoassay has been developed for the determination of ARL 15849XX, a cholecystokinin-8 (CCK-8) analogue, in dog plasma. The method incorporates solid-phase sample extraction and is suitable for the determination of the analyte at picogram per millilitre concentrations. The antiserum was raised in Suffolk-cross sheep following primary and booster immunisations with an immunogen prepared by conjugating ARL 16935XX, an analogue of ARL 15849KF, to bovine serum albumin. The radioligand was prepared by the no-carrier-added 125I iodination of a non-sulphated derivative, ARL 15745XX. The solid-phase extraction procedure, carried out using ion-exchange aminopropyl and octadecyl sorbents sequentially, was introduced to remove matrix interferences in the plasma and to enhance the method sensitivity. The calibration range is 20-1000 pg ml-1, using a 1 ml sample of undiluted dog plasma.

Nephrotic-range proteinuria in renal atheroembolic disease: report of four cases.

The protean clinical manifestations of atheroembolic disease (AED) mimic systemic disorders with kidney involvement. Acute or chronic renal failure develops spontaneously or more frequently after an inciting event in patients with AED. Significant proteinuria and nephrotic syndrome, however, constitute uncommon findings. We present four patients with AED documented histopathologically who developed nephrotic-range proteinuria. The mechanisms of proteinuria are discussed, and it is suggested that AED be considered in the differential diagnosis of nephrotic syndrome in elderly patients with serious vascular disease.

Comparison of intracranial self-stimulation evoked from lateral hypothalamus and ventral tegmentum: analysis based on stimulation parameters and behavioural response characteristics.

The comparison of intracranial self-stimulation (ICSS) derived across the anteroposterior axis of medial forebrain bundle (MFB) from the anterior border of lateral hypothalamus (LH) to the ventral mesencephalon including ventral tegmental area-substantia nigra (VTA-SN) in Wistar rats was assessed through stimulation parameters and behavioural response characteristics. The interpretation of response rate/charge consumption (muC/min) with respect to rectangular wave and sine wave electrical stimulation parameters suggests that the rectangular wave parameters are better in order to get the maximum responding rates. The most vigorous and robust responding was observed in the VTA or VTA-SN boundary placements, followed by placements in medial sector of LH. The acquisition of ICSS was fastest in the case of VTA-stimulation. The next site with respect to rapidity of ICSS was posterio-ventral LH. The extinction curves indicated that it is faster and exponential in case of VTA-SN, but it is slower with longer duration in case of LH-MFB. ICSS of SN were accompanied by exploratory locomotion and head bobbing. Thirty-one percent subjects with SN/VTA stimulation showed rotational behaviour. Seventy-eight percent of subjects with LH stimulations showed stimulus-bound ejaculations. Thirty-two percent of subjects with posterior LH stimulations showed biting of pedal edges. LH stimulations were accompanied by induced seizures and increased grooming in 18% and 13% of subjects, respectively. There was lateralisation of cerebral hemispheric function as right paw preference was noted in majority of rats, whether sites of stimulation were in the left or right cerebral hemisphere. The various other modes of pedal pressing operants like use of paw and mouth, alternate paw dribbling, use of head electrode assembly to manipulate the pedal were also recorded and analysed.

Cracked cartridges during foldable intraocular lens implantation.

We describe five routine phacoemulsification procedures in which a longitudinal split in the injector cartridge occurred during foldable intraocular lens (IOL) implantation. In some cases, this was associated with a broken lens. All lenses (STAAR AA-4203VF) were folded, and implantation was attempted through a 3.5 mm corneal or scleral incision. Visual outcome was not adversely affected in any case. The systems for introducing foldable IOLs have not been perfected, and further refinements in materials and techniques are required.

Secretory activity and trophic effects of epidermal growth factor in the rat pancreas.

This study was to investigate whether epidermal growth factor (EGF) may induce any long-term effect on pancreatic exocrine function in vivo as well to evaluate the chronic effects of EGF on pancreatic growth in rats. Rats were treated with EGF (10 micrograms/kg) for 5 or 7 days. EGF infused intravenously (2 micrograms/kg/h) in anaesthetized and pretreated rats for 5 or 7 days with EGF caused a slight decline flow rate after 1 h of EGF infusion compared to control values. In contrast, EGF evoked a increase in amylase secretion. This stimulatory effect was much larger in EGF-pretreated rats for 7 days, whereas the total protein output was unchanged. The trophic parameters which include pancreatic weight, total protein and total contents of DNA and RNA relative to body weight were not significantly different in any treated group. Only the pancreatic amylase content was increased significantly after 7 days of treatment with EGF. The present study fails to observe a stimulatory role of EGF on pancreatic growth in rats, but may participate in the regulation of pancreatic exocrine function in vivo.

Effect of the separate and combined administration of mestranol and phenobarbital on the development of altered hepatic foci expressing placental form of glutathione S-transferase in the rat.

The stages in the carcinogenesis process include initiation, promotion and progression. Although many characteristics of tumor promotion and promoting agents have been reported, relatively few studies on the effects of combinations of promoting agents have been detailed. For study of the combined effects of phenobarbital (PB) and mestranol (MS) in multistage rat hepatocarcinogenesis, an initiation-promotion protocol was developed. Female rats were injected i.p. at 5 days of age with either diethylnitrosamine (DEN) (10 mg/kg) or the solvent tricaprilyn. At weaning, approximately 10 rats from both the DEN-initiated and the solvent control groups were provided basal diet alone, PB (10, 100 or 500 mg/kg diet), MS (0.02 or 0.2 mg/kg diet) or various combinations of both PB and MS in the basal diet. At 8 months of age, the rats were killed and the livers removed, sectioned and fixed in ice-cold acetone. Sections of 5 microgram thickness were stained for placental glutathione S-transferase (PGST) expression, and the volume fraction of liver occupied by altered hepatic foci was determined by stereology. In addition, incorporation of bromodeoxyuridine into nuclei of PGST-expressing (focal) and non-PGST-expressing (non-focal) hepatocytes was determined. Administration of the highest dose of PB resulted in a significant decrease in non-focal hepatocyte labeling index, with a 4-fold differential between the focal and non-focal hepatocyte labeling index. Administration of 0.2 mg/kg diet MS resulted in effective promotion. The non-focal labeling index was increased and the focal labeling index was further enhanced (3-fold) relative to the non-focal index by this dose of MS. Combination of the lower MS dose with PB resulted in at least an additive promoting effect; however, a lower volume fraction was noted for the combination of low MS dose plus the highest PB dose. Combination of the higher MS dose with PB resulted in an elevation of volume fraction only for the middle PB dose. These findings indicate that the potency of promotion by mixtures is modulated by the dose of each component as well as by pharmacodynamic and pharmacokinetic properties of each component of the mixture.
