2'-nor-cGMP, a new cyclic derivative of 2'NDG, inhibits HSV-1 replication in vitro and in the mouse keratitis model.

The present study examined the antiherpetic effect of 2'-nor-cGMP, a new cyclic phosphate derivative of 2'NDG, in vitro and in the mouse keratitis model. The 50% inhibitory dose (ID50) was determined with HSV-1 RE strain in Vero cell monolayers for 2'-nor-cGMP (6.9 mcg./ml), 2'NDG (.06 mcg/ml), and trifluridine (F3T) (.72 mcg/ml). Balb C mice underwent bilateral ocular inoculation with HSV-1 RE strain, and then were treated with different therapeutic regimens. The antiviral efficacy of each drug was evaluated by ocular virus titers, clinical grading of epithelial keratitis, and histological evaluation of stromal keratitis. 2'-nor-cGMP was the most effective drug (P = .0001) in reducing ocular viral titers. Both 2'-nor-cGMP and 2'NDG were significantly more effective (P = .0001) than F3T in reducing epithelial keratitis, and as effective as F3T in reducing stromal keratitis.

Circadian temperature rhythm and circadian-circaseptan (about 7-day) aspects of murine death from malaria.

About-7-day (circaseptan) and circadian rhythms were sought and found in host-parasite relations of mice infected with Plasmodium berghei. Five inbred male DBA mice, about 18 weeks of age, were implanted with transsensors for temperature telemetry. Core temperature, monitored every 10 min for 3 days before the intravenous or intraperitoneal inoculation of 10(5) infected erythrocytes and thereafter until death, was analyzed by cosinor. A statistically highly significant circadian rhythm exhibited similarly synchronized acrophases. Core temperatures on the days immediately after malarial infection were mostly within the range of temperatures observed before injection. A mesor-hypothermic stage preceded death by several days. In a second study, 24 male BALB/c and 42 male DBA mice, 12 weeks of age, housed in three rooms on different regimens of light and darkness (alternating at 12-hr intervals), staggered by 8 hr, were inoculated ip with 10(4) infected erythrocytes, one-half at noon, the other half at midnight, within 0.5 hr of blood withdrawal. Thus, one endeavored to cover six circadian host stages (02, 06, 10, 14, 18, and 22 hr after light-on). At 54 and 51% overall mortality (irrespective of inoculation time), a circadian rhythm in susceptibility to malaria was demonstrated in these mice by the single cosinor fit of a 24-hr period (P less than 0.003 and less than 0.020, respectively). The single cosinor fit of a 7-day period further demonstrated a circaseptan rhythm (P = 0.014) in the mortality of both strains following the inoculation of P. berghei. The acrophase (360 degrees = 168 hr) was at -325 degrees from the inoculation time with 95% confidence limits extending from -276 to -378 degrees. Such predictable time relations of P. berghei to its murine host await the exploration of mechanisms underlying the circadian and infradian (7-day) rhythmicities here demonstrated and quantified with their uncertainties. Irrespective of mechanisms, information on such periodicities may also guide attempts to optimize treatment by timing according to the interactions of plasmodial virulence and host resistance that remain to be quantified separately.

Preliminary results of AS 791/792 artificial sphincter for urinary incontinence.

Twenty-seven patients with incontinence have been treated by implantation of the AS 791 or 792 artificial sphincter over the past 24 months. All but four were incontinent as a result of myelodysplasia or prostatic ablation: 22 are currently completely continent, 2 await activation and 3 have failed. There has been only one mechanical failure, though 3 patients have required the insertion of a higher pressure reservoir to resolve stress incontinence, reflecting our conservative approach to the initial choice of reservoir.

Influence of palmitic acid on mouse lymphocyte function in vivo and in vitro.

The effect of palmitic acid on the immune response of mice to sheep erythrocytes was studied. It was found to increase the number of background plaques in normal mice but it had no significant effect on primed cells. When given with antigen it functioned as a weak adjuvant but it had no effect on response of mouse lymphocytes to the mitogens phytohemagglutinin or bacterial lipopolysaccharide. While it is clear that palmitic acid does not have a profound effect on lymphocyte reactivity, it may have a subtle modulating influence on the immune system.

Studies on the generation of biologically active substances by Trypanosoma theileri in vitro.

Trypanosoma theileri was grown in bovine primary spleen cell cultures. These organisms were tested for phospholipase activity which was found to be very low, for haemolytic activity which was very weak and for lymphocyte mitogenicity which was found to be absent. High concentrations of T theileri were found to activate bovine complement probably by both the alternate and classical pathways. There was no evidence that these organisms exerted any significant toxic effect in vitro or in vivo. This is compatible with the known low pathogenicity of this species.

Complement activating factor(s) of Trypanosoma lewisi: some physiochemical characteristics of the active components.

Of the complement activating factors present in Trypanosoma lewisi, the major component, a carbohydrate containing substance was further investigated. This component was found to have a lag time of complete activation of 2 CH50 units of bovine complement of approximately 15 minutes while 1% trypsin (a known activator of complement, used as a control system) was capable of instant consumption of a similar quantity of complement. In addition, the complement activating factor of trypanosomes was observed to be stable at 100 degrees C for 15 minutes and over a pH range of 3.0 to 11.0. Thin layer chromatography studies suggested that at least part of the active component contained lipid, perhaps indicating that it may be glycolipid in nature.

Experimental bovine trypanosomiasis. Changes in the catabolism of serum immunoglobulins and complement components in infected cattle.

The turnover of serum proteins of calves experimentally infected with Trypanosoma congolense was compared to that of normal uninfected cattle. All proteins examined had much increased catabolic rates in infected animals. In normal animals the average half-lives in days for each protein were: IgG1 17.4, IgG2 22.4, IgM 4.8, IgA 3.4, IgE 1.9, C1 5.6 and C3 2.9. In trypanosome infected cattle the average half-lives were IgG1 1.9, IgG2 1.7, IgM 0.9, IgA 1.2, IgE 0.9, C1 1.2 and C3 1.1 days.
