The Monolayer Behavior of Amphiphilic Polymer and Heterostructure of Polymer LB Film/CdS Clusters.

In this paper we report the behavior of an amphiphilic polymer monolayer on pure water and Cd2+ subphase. This polymer was composed of hydrophilic ethylene diamine epichlorohydrin slightly crosslinked microgel and hydrophobic stearic chains, noted as ES-1. The introduction of Cd2+ ions in subphase had a marked effect on the process of the organization of the amphiphilic polymer at the air/water interface due to the association of Cd2+ ions with the hydrophilic network, which could be indicated by the pressure-area isotherms and Brewster Angle Microscopy. Ordered ES-1/Cd2+ LB multilayers were fabricated. After the multilayers reacted with H2S gas, CdS clusters were synthesized within the film, which was characterized by X-ray diffraction and UV-visible spectroscopy. Copyright 1999 Academic Press.

Prenatal diagnosis of glycogen storage disease type IV using PCR-based DNA mutation analysis.

Deficiency of glycogen branching enzyme activity causes glycogen storage disease type IV (GSD-IV). Clinically, GSD-IV has variable clinical presentations ranging from a fatal neonatal neuromuscular disease, to a progressive liver cirrhosis form, and to a milder liver disease without progression. Current methods for prenatal and postnatal diagnosis are based on an indirect method of measuring the enzyme activity, which has a limited sensitivity and cannot be used to distinguish patients with these variable clinical phenotypes. In this study, a GSD-IV family with a non-progressive hepatic form of the disease requested prenatal diagnosis. Determination of the branching enzyme activity in cultivated amniocytes showed 20 per cent residual activity overlapping with the level detected in the heterozygotes. Mutation analysis revealed that the fetus carried two mutant alleles, L224P and Y329S, the same as the proband of this family. The fetus was predicted to be affected and postnatally his clinical presentation is consistent with the diagnosis. We conclude that DNA mutation analysis should be used in the prenatal diagnosis of GSD-IV, especially in the situation of high residual enzyme activity.

Post-column metal complexation of quinolone antibiotics in a quadrupole ion trap.

Post-column addition of a metal salt and an auxiliary chelating ligand via a sheath flow offers an alternative strategy for promoting electrospray ionization of analytes via metal cationization. In the present study, a high-performance liquid chromatography (HPLC) mass spectrometer interface that incorporates a modified ionspray source that allows either online metal cationization or protonation is reported. The analytical utility of metal complexation is evaluated relative to protonation as a means for efficient ionization and generation of structurally diagnostic ions during HPLC separation. To improve metal cationization, an auxiliary chelating ligand is added to the sheath flow to coordinate the metal and stabilize the resulting complexes. The methodology was verified using a series of quinolone antibiotics as model analytes. Relative reaction efficiencies were compared for the protonation vs. metal complexation modes, and it was found that metal complexation with an auxiliary chelating ligand gave a three to five times better detection limit than protonation for the quinolones. Detector linearity and optimal reaction conditions are also reported.

In vivo chemical shift imaging of gamma-aminobutyric acid in the human brain.

A gradient-based multiple quantum filtering method is presented for in vivo chemical shift imaging of gamma-aminobutyric acid (GABA) in the human brain, which provides effective suppression of the overlapping creatine singlet with close to optimal detection efficiency. It is shown by product operator calculations and coherence pathway analysis that under conditions of no B1 and B0 inhomogeneity gradient filtering retains 75% of the two outer resonance lines of the GABA-4 triplet with no creatine contamination. A variation of the method with 100% retention of the GABA-4 outer resonance lines but higher sensitivity to B1 inhomogeneity is also discussed. By using a localized version of the sequence with an 8-cm surface coil for transmission and detection, it was found in phantom experiments at 2.1 T that a 69% signal retention of the two outer resonance lines of the GABA-4 triplet was achieved relative to a spin echo sequence with inhibition of GABA J modulation. A creatine suppression ratio of 2000:1 was measured. The use of the method for chemical shift imaging of GABA is demonstrated by coronal images obtained from phantoms and from the occipital lobe of a healthy volunteer.

Linear projection method for automatic slice shimming.

A fast, reliable automatic slice shimming method is described. In-slice shim adjustments are based on one-dimensional phase mapping of four in-slice linear projections through the slice center. For axial, coronal, and sagittal slices it is shown that all in-slice first-, second-, and third-order spherical harmonic terms of B(0) inhomogeneity can be unequivocally determined and corrected. Through-slice shim adjustment is achieved using a one-dimensional projection of the entire slice or ROI along the slice-selection direction. Applications of this method to single-slice in vivo spectroscopic imaging of human brain have resulted in reproducible, high-quality spectroscopic data. Magn Reson Med 42:1082-1088, 1999.

Does retrograde warm blood cardioplegic perfusion provide better protection of ischemic areas than antegrade warm blood cardioplegic perfusion? A magnetic resonance study in pig hearts.

OBJECTIVE: The purpose of this study was to determine whether retrograde continuous normothermic blood cardioplegic perfusion provides better protection to ischemic areas of the left and right ventricles than does antegrade continuous normothermic blood cardioplegic perfusion. Localized phosphorus 31 magnetic resonance spectroscopy was used to monitor the changes in energy metabolism and intracellular pH in the ventricles of pig hearts. METHODS: Ten isolated pig hearts received 20 minutes of antegrade continuous normothermic blood cardioplegic perfusion for collection of control (baseline) data, followed by 60 minutes of either antegrade continuous normothermic blood cardioplegic perfusion (n = 5) or retrograde continuous normothermic blood cardioplegic perfusion (n = 5) with occlusion of the left anterior descending and the right coronary arteries. The hearts were then subjected to antegrade continuous normothermic blood cardioplegic perfusion for 20 minutes. The perfusion pressures were maintained between 80 and 100 mm Hg and between 38 and 43 mm Hg during antegrade and retrograde continuous normothermic blood cardioplegic perfusions, respectively. Intracellular pH and creatine phosphate, inorganic phosphate, and adenosine triphosphate levels were measured continuously in each ventricle by means of localized phosphorus 31 magnetic resonance spectroscopy with 2 surface coils. RESULTS: Both antegrade and retrograde continuous normothermic blood cardioplegic perfusion resulted in a significant increase in inorganic phosphate level and decreases in creatine phosphate level, adenosine triphosphate level, and intracellular pH. No significant differences in these changes were observed between the two groups. The creatine phosphate and adenosine triphosphate levels were significantly lower in the right ventricle than in the left ventricle during retrograde continuous normothermic blood cardioplegic perfusion. On reperfusion, the inorganic phosphate level, creatine phosphate level, and intracellular pH recovered completely; however, no recovery in the adenosine triphosphate level was seen in the ventricles of either group. CONCLUSIONS: Retrograde continuous normothermic blood cardioplegic perfusion does not provide better protection to ischemic areas of the ventricles than does antegrade continuous normothermic blood cardioplegic perfusion under our experimental conditions.

Opioid receptor upregulation in mu-opioid receptor deficient CXBK and outbred Swiss Webster mice.

Chronic in vivo treatment with opioid antagonists increases opioid receptor density and the potency of opioid agonists without altering receptor mRNA levels. To determine if basal receptor density affects opioid receptor upregulation, we examined the effect of chronic naltrexone treatment on mu-opioid receptor density and mRNA in two mice strains that differ in mu-opioid receptor density. CXBK mice (mu-opioid receptor deficient) and outbred Swiss Webster mice were implanted s.c. with a placebo or 15 mg naltrexone pellet for 8 days, the pellets removed and 24 hr later opioid receptor density (mu, delta) and receptor mRNA level (mu) determined in whole brain; or morphine dose-response studies conducted. In placebo-treated CXBK mice, mu-opioid receptor density was approximately 40% less than in Swiss Webster mice, although mu-opioid receptor mRNA abundance was similar in both strains. In placebo-treated CXBK mice, morphine potency was approximately 6-fold less than Swiss Webster mice. Naltrexone treatment increased morphine potency (1.7-fold) and mu- (approximately 90%) and delta- (approximately 20-40%) opioid receptor density in CXBK and Swiss Webster mouse brain similarly. Mu-opioid receptor mRNA was unchanged by naltrexone treatment in either strain. There was no difference in the basal or naltrexone-treated whole brain G(i alpha2) protein levels in CXBK or Swiss Webster mouse. These data indicate that a deficiency in mu-opioid receptors does not alter the regulation of opioid receptors by opioid antagonists in vivo, and suggest that adaptive responses to chronic opioid antagonist treatment are independent of opioid receptor density.

A vitamin E-deficient diet affects nerve regeneration in rats.

Degenerative changes in the neuromuscular system have been found in animals and humans with vitamin E (E) deficiency. This morphologic study examined the effect of dietary E on the regeneration of peripheral nerves in male Sprague-Dawley rats. After feeding an E-sufficient diet (dl-alpha-tocopheryl acetate 50 mg/kg diet) for 6 d, 24 rats were randomly and equally assigned to one of three groups: control (CTRL) fed an E-sufficient diet for 43 d without surgery, normal (NE) fed an E-sufficient diet, or low (LE) fed an E-deficient diet (dl-alpha-tocopheryl acetate 0 mg/kg diet). After 22 d of feeding, NE and LE had surgical compression of the right sciatic nerve and continued eating for 15 d. On day 43, the right triceps surae muscles and a segment of the right sciatic nerve were removed, then all rats were euthanized. The nerve and muscles were processed for morphologic analyses. Presurgery and postsurgery LE ate less food (P < 0.048 and P < 0.001, respectively), which resulted in a lower body weight gain (P < 0.0002). LE had irregularly shaped and less myelinated axons than NE (P < 0.0001) and CTRL (P < 0.0001). The LE plantaris muscle had less type II fibers when compared with NE (P < 0.007) and CTRL (P < 0.03). The results suggest that an E-deficient diet affects food intake, impairs nerve regeneration, and decreases type II fibers, whereas an E-sufficient diet contributes to normal axon regeneration.

Organic cation transport in rabbit alveolar epithelial cell monolayers.

PURPOSE: To characterize organic cation (OC) transport in primary cultured rabbit alveolar epithelial cell monolayers, using [14C]-guanidine as a model substrate. METHODS: Type II alveolar epithelial cells from the rabbit lung were isolated by elastase digestion and cultured on permeable filters precoated with fibronectin and collagen. Uptake and transport studies of [14C]-guanidine were conducted in cell monolayers of 5 to 6 days in culture. RESULTS: The cultured alveolar epithelial cell monolayers exhibited the characteristics of a tight barrier. [14C]-Guanidine uptake was temperature dependent, saturable, and inhibited by OC compounds such as amiloride, cimetidine, clonidine, procainamide, propranolol, tetraethylammonium, and verapamil. Apical guanidine uptake (Km = 129 +/- 41 microM, Vmax = 718 +/- 72 pmol/mg protein/5 min) was kinetically different from basolateral uptake (Km = 580 +/- 125 microM, Vmax = 1,600 +/- 160 pmol/mg protein/5 min). [14C]-Guanidine transport across the alveolar epithelial cell monolayer in the apical to basolateral direction revealed a permeability coefficient (Papp) of (7.3 +/- 0.4) x 10(-7) cm/sec, about seven times higher than that for the paracellular marker [14C]-mannitol. CONCLUSIONS: Our findings are consistent with the existence of carrier-mediated OC transport in cultured rabbit alveolar epithelial cells.

Modulation of IL-10, IL-12, and IFN-gamma in the epidermis of hairless mice by UVA (320-400 nm) and UVB (280-320 nm) radiation.

We have observed recently that the suppression of contact hypersensitivity (CHS) induced in mice by UVB irradiation may be prevented by suberythemal exposure to UVA radiation. Because the UVB-immunosuppressed state is associated with an upregulation of the Th2-associated cytokines IL-10 and IL-4, and a deficiency in Th1-associated IL-2, IL-12, and IFN-gamma, and because UVA photoimmunoprotection appeared to be IFN-gamma- dependent, we tested the hypothesis that UVA immunoprotection results from an ability to prevent the UVB-induced cytokine disarray. This study describes changes in epidermal IL-10, IL-12 and IFN-gamma for 5 d following irradiation of hairless mice with the CHS-modulating doses of UVB, UVA, or UVA + UVB, using immuno-histochemical detection in paraffin embedded skin sections, followed by image analysis quantitation. We found that UVB, but not UVA exposure, caused an increase in epidermal IL-10 expression, peaking at 3 d. UVA irradiation, but not UVB, resulted in increased epidermal IL-12 expression, peaking at 3 d, and increased epidermal IFN-gamma expression peaking earlier at 1 d. Irradiation with UVA + UVB abrogated the UVB-enhanced expression of IL-10, and caused small but significant increases in IL-12 and IFN-gamma at 3 d and 1 d, respectively. These findings suggest that UVA photoimmunoprotection is mediated via prevention of IL-10 release, and thus the maintenance of the Th1/Th2 balance, probably by upregulation of IL-12 and IFN-gamma, which are known to antagonize IL-10 in numerous models. The time course suggests that IFN-gamma responds initially to UVA radiation, and may stimulate the increased expression of IL-12.

Moral values of only and sibling children in mainland China.

Seventh graders (N = 346) in Beijing and Shanghai were administered the Chinese Values Survey (M. H. Bond & the Chinese Culture Connection, 1987) and the Rokeach Values Survey (modified version; R. A. Cole, 1972) in 1992. Results showed no statistically significant difference in scores between only and sibling children who rated Chinese values and Rokeach terminal values. The 2 groups appeared to be significantly different in rating Rokeach instrumental values, but the effect sizes accounted for less than 2% of the variance. The stereotype of only children as being "spoiled" was not supported by the data.

Gender differences in reasons for entering and leaving education administration: discriminant function analyses.

Using discriminant function analyses on survey data collected from 457 students in 29 education administration programs across the United States, the authors inquired into whether men and women decide to enter and possibly leave education administration for different reasons. There were statistically significant functions separating men and women in their reasons for entering and leaving education administration. Implications of the findings are discussed.

Coarsening kinetics from a variable-mobility Cahn-Hilliard equation: application of a semi-implicit Fourier spectral method.

An efficient semi-implicit Fourier spectral method is implemented to solve the Cahn-Hilliard equation with a variable mobility. The method is orders of magnitude more efficient than the conventional forward Euler finite-difference method, thus allowing us to simulate large systems for longer times. We studied the coarsening kinetics of interconnected two-phase mixtures using a Cahn-Hilliard equation with its mobility depending on local compositions. In particular, we compared the kinetics of bulk-diffusion-dominated and interface-diffusion-dominated coarsening in two-phase systems. Results are compared with existing theories and previous computer simulations.

Inhibition of cytomegalovirus in vitro and in vivo by the experimental immunosuppressive agent leflunomide.

Despite progress in antiviral chemotherapy, cytomegalovirus (CMV) remains a major cause of morbidity and mortality among pharmacologically immunosuppressed transplant recipients, frequently engaging the clinician in a struggle to balance graft preservation with control of CMV disease. Leflunomide, an inhibitor of protein kinase activity and pyrimidine synthesis, is an experimental immunosuppressive agent effective against acute and chronic rejection in animal models. Herein we summarize our recent studies demonstrating that leflunomide inhibits the production of multiple clinical CMV isolates (including multi-drug-resistant virus) in both human fibroblasts and endothelial cells. In contrast to all other anti-CMV drugs currently in use, leflunomide does not inhibit viral DNA synthesis, but rather appears to interfere with virion assembly. Finally, preliminary studies in a rat model suggest that this agent reduces viral load in vivo. These findings imply that leflunomide, an effective immunosuppressive agent, shows potential to concurrently attenuate a major complication of immunosuppression, CMV disease, by a novel mechanism of antiviral activity.

Combined spinal-epidural analgesia in the management of labouring parturients with mitral stenosis.

We report the use of combined spinal-epidural analgesia during labour in three parturients with moderately severe mitral stenosis. In each case, rapid analgesia was achieved using intrathecal fentanyl 25 micrograms without major haemodynamic changes. Maintenance analgesia was then established gradually using a dilute epidural infusion of bupivacaine 0.1% and fentanyl 0.0002%, with the avoidance of large or rapid boluses of local anaesthetic. Supplementary analgesia in the latter stages of labour was provided using slow epidural boluses of fentanyl, with or without a low concentration of bupivacaine, which was sufficient to allow controlled instrumental deliveries. We conclude that combined spinal-epidural analgesia is a useful technique for providing analgesia and maintaining haemodynamic stability in parturients with mitral stenosis.
