Eikenella corrodens: a clinical problem.

E corrodens occurs as a significant clinical infection more frequently than suspected or cultured at the present time. It is essential that the dentist or physician work closely with the laboratory clinician in order to use proper techniques for its isolation. Inconsistencies between in vitro disk sensitivity and clinical drug response are noted. Because of clinical response, the clinician must carefully follow the patient's daily progress to recognize antibiotic treatment failure so that combination therapy or appropriate incision and drainage procedures may be initiated.

Lysosomal enzyme cytochemistry in acute leukaemia.

A cytochemical study of the lysosomal enzyme beta-glucuronidase in 60 cases of acute leukaemia has shown a qualitative difference in the cytoplasmic distribution of the enzyme between blast cells of the lymphoid and myeloid cell series. This difference provides a useful additional method for cytochemical classification of cell type and is superior in this respect to the other lysosomal enzymes studied (aryl sulphatase and acid phosphatase). The beta-glucuronidase reaction is recommended in those cases of acute leukaemia in which the periodic acid-Schiff reaction is negative or equivocal.

Dehydrogenase enzyme cytochemistry of unfixed leucocytes.

An inert, synthetic, sucrose polymer (Ficoll) has been found to have a protective effect on leucocytes during incubation for cytochemical reactions at 37 degrees C. This permits enzyme cytochemistry to be performed on unfixed bone marrow and peripheral blood smears, and on leucocytes cultured in vitro, thus avoiding enzyme inhibition by the use of a fixative. The technique is suitable for the demonstration of both cytoplasmic and intramitochondrial dehydrogenase enzyme activity.
