Characteristics and Diversity of beta-d-Glucosidase (EC 3.2.1.21) Activity in Marine Snow.

Large amorphous aggregates commonly described as marine snow were sampled in the water column of the northern Adriatic Sea in August 1991. beta-d-Glucosidase activity associated with these aggregates was characterized. Enzymatic activity was measured with the fluorogenic substrate analog 4-methylumbelliferyl-beta-d-glucoside and found to be mainly associated with particles; on average, only 24% of the whole beta-d-glucosidase activity remained in the supernatant after centrifugation. Although the temperature optimum for beta-d-glucosidase activity was approximately 40 degrees C, incubation of the previously liberated particle-attached enzyme at 50 degrees C for 20 min caused a >90% reduction of enzymatic activity relative to the activity at 40 degrees C. The level of inactivation of beta-d-glucosidase was much lower, however, when whole marine snow was incubated, indicating qualitative modifications of beta-d-glucosidase in marine snow. Separation of beta-d-glucosidase by different approaches indicated that the diversity of isoenzymes is restricted. In samples taken from the pycnocline, only one major isoenzyme was present in noticeable amounts. This isoenzyme contributed up to 70% of the whole beta-d-glucosidase activity detectable by two different chromatographic separations (anion-exchange chromatography and size exclusion chromatography). Although the same isoenzyme was dominant in marine snow taken from surface waters (0.5-m depth), we found a second peak of activity which eluted at lower NaCl concentrations from the anion-exchange column. Generally, the diversity of isoenzymes exhibiting beta-d-glucosidase activity seems to be surprisingly small in amorphous pelagic aggregates.

Thermal artifacts and inter-gynecologist variation of laser cone biopsies of the cervix.

OBJECTIVE: To evaluate the extent of thermal artifacts and the inter-gynecologist variation in CO2 laser cone biopsies for cervical intraepithelial neoplasia (CIN) using high power density. METHODS: All 1180 histologic sections from 93 CO2 laser cone biopsies performed by nine gynecologists were evaluated by morphometry. Power density of 19,700 W/cm2 and spot size of 0.43 mm were used. The lengths of different epithelia, the distance from CIN to the excision margins, the thermal artifacts, and the extension of denudation were analyzed. Mean values and standard deviations were analyzed for all indices. The inter-gynecologist variation was analyzed using one-way analysis of variance, Tukey-Kramer Honestly Significant Difference test, and chi 2 contingency table analysis regarding cones classified as "good" or "not good". RESULTS: The mean length of analyzed surface per cone was 134 mm. In 3% of the sections, the sum of artifacts at the external excision margin exceeded the average "minimum free distance" from CIN to the margin (2.88 mm). At the endocervical excision margin, the free distance was exceeded in 17% of the sections. Eighteen percent of the cones had CIN at the excision margins, with no significant differences between gynecologists (P = .83). Differences existed between gynecologists regarding free minimum distance and minor thermal artifacts at the external margin, but when clinically relevant groups were applied, no significant differences were found (P > .05). CONCLUSION: We found that a high power density at CO2 laser cone biopsy resulted in specimens with minimal thermal artifacts compared to other authors who used low power density. No clinically relevant differences were found between the gynecologists regarding free margins or artifacts.

Steroid-induced dermal thinning: discontinuous application of clobetasol-17-propionate ointment.

The skin thinning effect of discontinuous topical clobetasol-17-propionate applications was tested in human volunteers. Application frequencies were daily (1/0), every third day (1/2), every fifth day (1/4), every seventh day (1/6) and every ninth day (1/8). Clobetasol-17-propionate was administered topically under occlusion for 1 h. The treatment period was 41 days. There were no differences of the skin thinning effect of daily and 1/2 administration. The skin thinning effect of 1/4, 1/6 and 1/8 was smaller but also significant compared to controls. The curves demonstrating skin thinning effects initially were dropping off and reached a plateau within about 2 weeks. After finishing application, skin thickness normalized within 2 weeks. Because of these findings, a treatment interval of 3 days is discussed as therapeutically efficient.

Collaborative Behavioral Teratology Study: programmed data entry and automated test systems.

The automated laboratory systems described in this paper were developed for use at the National Center for Toxicological Research (NCTR) and each of five collaborating laboratories. These laboratories participated in a study designed to evaluate the intra- and interlaboratory reliability and sensitivity of several behavioral test methods used in developmental toxicity studies. This paper describes two microcomputer systems. System I was designed to control stimulus delivery to and record behavioral responses from rats during a visual discrimination learning task and to accept data from physical landmark, negative geotaxis, and olfactory discrimination evaluations. The microprocessor system was an S-100 bus, Z-80 processor with 8K PROM, 16K RAM, nine 8-bit parallel I/O ports, and one serial I/O port. For discrimination learning, the animal testing apparatus consisted of six operant chambers, equipped with two stimulus lights, two nose-poke apertures each containing a phototransmitter and receiver, an electromechanical liquid dispenser unit, and a reward bowl also containing a photodiode. Each chamber was interfaced with the microprocessor at a parallel I/O port which permitted the delivery of response- or time-contingent control signals for the lights and liquid dispenser units. System II controlled stimulus delivery and recorded behavioral responses from rats during an auditory startle habituation task, and also recorded rodent activity levels in figure 8 mazes for periods of 1, 4, or 23 hours.
