Characterization of mouse thymocyte and peripheral lymphocyte xenoantigens by two-dimensional electrophoresis.

Two-dimensional electrophoresis was used to analyze the surface glycoproteins of murine thymocytes and lymph node cells. Two-dimensional maps of unselected, radioiodinated lymphocyte surface proteins were complex, showing at least 20 different components, but simpler patterns were obtained by using rabbit antibodies directed against the surface proteins of a T lymphoma cell line, to precipitate xenoantigens from lysates of radioiodinated or biosynthetically labeled thymocytes and lymph node cells. These xenoantibodies precipitated 12-13 distinct components from each cell type, of which all but 3 were sialoglycoproteins. Two types of difference between the surface glycoproteins of thymocytes and peripheral lymphocytes could be detected. First, higher mol. wt. glycoproteins and Thy-1 are more acidic in peripheral lymphocytes than in thymocytes, and this difference disappears after neuraminidase treatment. One additional high mol. wt. glycoprotein is also detectable in peripheral lymphocytes, probably reflecting the greater carbohydrate complexity of these molecules, when expressed on such cells. Second, 3 glycoproteins are strongly labeled only on thymocytes, and 3 others only on peripheral lymphocytes. These 6 glycoproteins might represent genuine differentiation antigens.

Analysis of the alloimmune properties of a recombinant genotype in the major histocompatibility complex of the chicken.

In a population of (CB X CC)F1 X WB hybrids, a chicken was found with a presumably recombinant haplotype, BR1, whose antigenic products detectable by hemagglutination contained determinants derived from both parental haplotypes, i.e. B1 (from CB) and B2 (from CC). This recombinant bird and its progeny from different crosses were tested by skin grafting, graft-vs.-host (GVH) and mixed lymphocyte reactions (MLR). The results define two regions of the B system, the B-F and the B-G. The B-F region determined serologically defined antigens, histocompatibility antigens, and controlled the GVH and MLR reactions, while the B-G region was responsible only for synthesis of serologically detectable antigens.

Complementation between genetic variants affecting the response of chicken leukocytes to concanavalin A.

Birds of the partially inbred G-B1 chicken line can be classified as either high or low responders to Con A, on the basis of the amount of 3H-thymidine incorporated by Con A-containing cultures of their peripheral blood leukocytes. The pattern of inheritance of the high and low responder traits suggests that the variation in response is due to genetic polymorphism at a single autosomal locus. However, the allele responsible for the low responder trait of the G-B1 line is not identical to the allele of the previously described Mr1 locus carried by the inbred low responder CC line, since (CC x G-B1 low responder)F1 birds are uniformly high responders to Con A. The responses to Con A of mixtures of CC and G-B1 low responder cells are not significantly higher than the responses of either component of the mixture alone. Thus, the gene products responsible for the complementation observed in F1 hybrids cannot complement extracellularly, and are not readily transferable from one cell to another.

Close linkage between genes coding for allotypic markers on chicken IgG and IgM.

The allotypic markers Gla and Gld (present on chicken IgG molecules) and Mla and Mlc (present on IgM) have previously been defined. The inheritance of these markers was studied in the F2 progeny of a cross between birds of the inbred lines CB (Gla+,d-,Mla+,c-) and WA (Gla-,d+,Mla-,c+). The results suggest that antigens Gla and Gld, and Mla and Mlc, are genetically controlled by alleles at two loci (Gl and Ml, respectively) and also show that these loci are closely linked.

Paraproteinaemia in inbred chickens.

Sera from seventy chickens of several inbred lines were screened by cellulose acetate electrophoresis for abnormalities of immunoglobulin production. IgG levels in two sera (both from apparently healthy birds) were unusually high. The IgG proteins purified from these two sera were shown, by isoelectric focusing, to be of restricted heterogeneity.
