[Proteases and protease antagonists in neoplasms].

The maintenance of equilibrium between proteases and their inhibitors has substantial significance for the pathogenesis of the neoplastic disease. It concerns especially the invasiveness of the neoplasms, which increases with rising of the proteases activity or with the decrease of the inhibitory activity of antiproteases. The neoplastic cell has the faculty of producing numerous proteases which by damaging connective tissue facilitates the invasion by the neoplasm. Invasiveness of the neoplasms may be augmented by the production of proteases by the host cells stimulated by the neoplasm. The condition of the proteases--antiproteases balance is also influenced by the production of antiproteases by neoplastic cells. Among numerous antiproteases alpha-2-macroglobulin has most intensive cytotoxic action in relation with the neoplasm. According to data collected from the literature, antiproteases can be regarded as anticarcinogenic agents. On account of this, the estimation of the antiproteases level can be regarded as a measure of the ability of the body system to resist the invasiveness of neoplasms.

Low molecular organic inactivators in differentiating organ origin of alpha-amylase in humans. Part I.

Alpha-amylases derived from pancreatic juice, saliva and granulocytes show different sensitivity to the low molecular organic inactivators. Pancreatic alpha-amylase is more susceptible to inhibitory activity of d-glyceraldehyde and d-mannitol and glycerol in comparison to alpha-amylase from saliva and granulocytes. Iodoacetate and urea exhibited inhibitory activity against granulocytic alpha-amylase and only a minute inactivation of salivary and pancreatic alpha-amylases was found. The most effective inhibition against all tested alpha-amylases was noted with dihydroxyacetone. We should stress, no evident differences were noted in the degree of sensitivity to this inactivator of the tested enzymes. The presumable mechanism of the inactivation of alpha-amylases with low molecular organic inactivators is briefly discussed. The procedure for the differentiation of organ origin of alpha-amylases is suggested.

[Phagocytic activity of platelets in patients with pneumonia].

In 22 patients with pneumonia the per cent of phagocytizing platelets and the phagocytosis index of the platelets were determined before treatment and in 14 patients this was done after treatment. Before treatment the per cent of phagocytizing platelets was 1.37%, on average, and after treatment it was 1.20, and it was thus much lower than in the control group X 2.33. The phagocytosis index before treatment was 1.39 on average, and after treatment it was 1.28, while in the control group it was 1.54. The obtained results seem to indicate that in pneumonia platelets participate in the antibacterial processes and the lungs as a thrombopoietic organ contribute to this phenomenon.

[Human serum colony-stimulating factor in mouse bone marrow culture].

Activity of CSF was measured in human serum after chromatography on Sephadex G-200. Agar culture of bone marrow cells was done in Petri dishes with agar in Eagle'a medium containing fetal calf serum and antibiotics. This agar-medium containing mouse bone marrow cells, was allowed to gel and the dishes were incubated at 37 degrees C in a fully humidified atmosphere of 10% CO2 in air. Our data indicate that chromatography procedure allows to get CSF free from its inhibitors. All activity was localized in one peak, in the region of low molecular weight proteins.

[Colony-stimulating factor (CSF) and granulopoietic activity (GA) in various mouse organs].

The colony stimulating factor and granulopoietic activity were determined in the homogenates of mouse kidney, liver, spleen and bone marrow. The activity of CSF and GA were highest in the kidney. The homogenate of this organ was fractionated then by ultracentrifugation. The distribution of CSF and GA in subcellular fractions and homogenates may suggest that kidney cells are the source of a substance causing, both, colony stimulation and granulopoietic activity.
