Activity of pirlimycin (U57930E) against strains of the Bacteroides fragilis group.

The activities of pirlimycin (U57930E), lincomycin, clindamycin, erythromycin, josamycin, oleandomycin, and spiramycin were compared against strains of the Bacteroides fragilis group. Pirlimycin was the most active; the 90% minimal inhibitory concentration was 1 microgram/ml, and the activity range was 0.125 to 4 micrograms/ml. This drug was fourfold more active than any of the other drugs, including clindamycin. The minimal bactericidal concentration (range, 0.5 to 16 micrograms/ml) shows that pirlimycin behaves as a bacteriostatic antibiotic.

Dopaminergic activity in a series of N-substituted 2-aminopyrimidines.

A series of N-substituted 2-aminopyrimidines were prepared and tested for pharmacological activity associated with stimulation of central and peripheral dopamine receptors using piribedil as the reference standard. Most of the new compounds showed some degree of dopaminergic activity although in many cases central activity was not accompanied by peripheral activity and vice versa. Clear relationships between structure and activity were not apparent and none of the new compounds possessed dopamine receptor blocking properties.

Sensitivity of Bacteroidaceae to fosfomycin.

The sensitivity of 116 strains of Bacteroides fragilis and 60 strains of Sphaerophorus to fosfomycin was studied. Of the 178 strains that were tested, 156 were isolated from pathological products in our department and the remainder came from the following collections: NCTC, ATCC and the Institut fur Mikrobiologie of the University of Bonn. Identification was made morphologically and biochemically in accordance with the scheme which we usually employ and which has previously been published. The sensitivities were determined by the progressive dilution method in a solid medium with a surface inoculation of the strain. The results obtained show that no matter what kind of B. fragilis was tested, none of them were sensitive to concentrations of 200 mug/ml fosfomycin. On the other hand, all strains of Sphaerophorus were sensitive to lesser concentrations than the aforementioned. The majority of these concentrations subsequently lie within therapeutic margins. The conclusion drawn from our results is that the sensitivity to fosfomycin constitutes a test for differentiating Bacteroides from Sphaerophorus and that fosfomycin is ineffective in injections caused by Bacteroides, although it is an active antibiotic against infections caused by Sphaerophorus.

[Potential anti-inflammatory agents II. 7-Methyl-s-triazo(4,3-a)pyridines].

A synthesis of eight derivatives of 7-methyl-s-triazo-[4,3-a]pyridine substituted in position 3, has been made with a view to studying their possible antiinflammatory and analgesic action. Among the obtained compounds only the 3-(alpha-hydroxyethyl)-7-methyl-s-triazo[4,3-a]pyridine presents an activity similar to aspirin.

Study of the effect of ACTH and cortisol on the plasms lipids in man using thin-layer chromatography.

This is a study of the action of ACTH and cortisol on plasma lipids using thin-layer chromatography and evaluation by densitometry. We have studied the action of 50 I.U. of ACTH in eight healthy individuals. We found an increase of free fatty acids after four hours and of triglycerides and esterified cholesterol after six hours. Free cholesterol levels declined after another six hours, the decrease becoming more intense after twenty-four hours. After twenty-four hours lecithin underwent a decline which is accompanied by an increase of lisolecithin. There was a decrease in the total amount of phospholipids. Seven patients received 100 mg. of cortisol intravenously over a period of ten minutes. We obtained an increase in free fatty acids after four hours and an increase of cholesterol after half and hour. We concluded that ACTH activates peripheral lipolysis thereby facilitating the hepatic synthesis of triglycerides and that it exerts a stimulating action on the lecithin-cholesterol-acyl-transferase system. Cortisol acts by facilitating the synthesis of triglycerides in the liver and increasing the hydrolysis of triglycerides in the plasma.

Serum ferritin assay and iron status in chronic renal failure and haemodialysis.

Forty-four patients with chronic renal failure on haemodialysis for four months to eight years were studied. All recieved intravenous iron dextran 100 mg on alternate weeks. Serum ferritin concentrations correlated well with body iron stores estimated by grading the bone marrow stainable iron. Altogether 34 patients showed increased bone marrow iron stores and serum ferritin concentrations greater than controls; four patients showed absence of iron in the marrow, and three of these had subnormal serum ferritin concentrations. Serum ferritin assay represents the best method of repeatedly monitoring the exact amount of iron therapy needed by patients with chronic renal failure, particularly those on regular haemodialysis.

Serum ferritin in patients with iron overload and with acute and chronic liver diseases.

An immunoradiometric assay was used to determine serum ferritin in patients with iron overload disorders and in patients with liver disease. In patients with iron overload, serum ferritin was closely correlated in an exponential manner with quantitative measurements of storage iron; however, a different relationship applied when storage iron levels were within normal range, suggesting that circulating ferritin is in equilibrium with two functionally distinct tissue ferritin pools. High ferritin levels were common in patients both with acute and chronic liver disease, normal values being virtually confined to women and to subjects with a history of recent hemorrhage. In liver disease generally, serum ferritin varied both with the serum transaminase level and with liver iron concentration, but correlated well with neither factor separately. There was no correlation with the serum iron or total iron-binding capacity. An extremely close correlation was found between serum ferritin and an empirical index derived from the product of the serum transaminase times liver iron concentration, implying that the circulating level depended on both the degree of hepatocellular injury and liver iron store. There was a close linear correlation between the serum ferritin-transaminase ratio and liver iron concentration in all disorders studied, and this index may prove to be the most useful value for diagnostic purposes.
