Characteristics of DSM-III borderline MMPI profiles.

Examined the distinction between the MMPI mean profile (8-4-2) obtained for a sample of 27 psychiatric inpatients diagnosed as borderline on the basis of DSM-III criteria, and the actual frequency distribution of scale elevations and code-types within the sample. Results of this study support the findings of previous investigations of a peak on scale 8, but suggest that the heterogeneity of this population will probably preclude the specification of any two- or three-point code type which would be obtained by even a sizable minority of a given sample.

Bromperidol radioimmunoassay: human plasma levels.

A sensitive radioimmunoassay (RIA) procedure was developed to assay for bromperidol levels in human plasma after therapeutic drug administration. The antisera used in the RIA procedure was generated in rabbits against a haloperidol-bovine serum albumin conjugate. Tritiated haloperidol was used as the radioligand in the assay. A single ether extraction of alkalinized plasma was used to separate bromperidol from its more polar metabolites and to reduce assay variability encountered with a direct plasma assay. The lower limit of detection was approximately 0.5 ng/mL of parent drug in plasma. The assay exhibited within- and between-assay variabilities of approximately 9 and 14%, respectively. A 103-106% recovery of bromperidol from quality control plasma samples was observed over the concentration range of 1-150 ng/mL. A correlation coefficient of 0.9999 with respect to measured versus expected bromperidol content in the quality control plasma samples was exhibited. Cross-reactivity characteristics of the antisera indicated that dehydrobromperidol could significantly interfere (approximately 25% cross-reactivity) with the RIA procedure. However, biotransformation studies have not suggested this compound as a metabolite of bromperidol. Predose ( Cmin ) plasma levels of bromperidol in schizophrenic patients maintained on drug therapy are also reported.

Display of prolonged EEG recordings.

An interactive minicomputer system for displaying, storing and providing hard copy output of prolonged EEG recordings has been described. Perhaps the most significant feature of the system is the versatility gained by using a minicomputer. The routines developed initially solely for the display and storage of the EEG have recently been readily modified to control a different playback unit. They also form an integral part of a software package being written to set up a data base for prolonged EEG recordings and also for the study of artefacts in ambulatory recordings.

Localization of copper-zinc superoxide dismutase in the endocrine pancreas.

Immunoelectron microscopy and cellular fractionation on sucrose density gradients have been used to examine the intracellular distribution of copper-zinc superoxide dismutase (SOD) in the canine and rat endocrine pancreas. Using rabbit anti-canine copper-zinc SOD as the primary antiserum, immunostaining in canine beta and non-beta islet cells was significantly greater than that in serial sections of the same islet incubated with preimmune serum from the same rabbit. Within the cells, immunostaining was associated with spherical and crystalloid granules as well as with the cytoplasm. Radioimmune assays of cellular fractions or rat islet-cell preparations showed that the granule-rich fraction containing large amounts of insulin was also rich in SOD. This interesting observation of association of SOD with hormone-containing granules suggests that SOD may play a role in protecting these oxidation-sensitive proteins.

Short-term clinical, nutritional, and functional effects of continuous elemental enteral alimentation in children with cystic fibrosis.

Ten children with cystic fibrosis, aged 3.5 to 12 years, whose weights were lower than 90% of the expected weight for height, received high-calorie elemental enteral alimentation for four weeks. Clinical, anthropometric, and biochemical evaluations as well as blood gas analyses and chest radiograph scoring were performed in all. Pulmonary function tests were performed in the five older children, and progressive exercise tests in three. These evaluations were done before, immediately after, and two months after termination of therapy. Nutritional therapy resulted in an increase of caloric intake and in dramatic weight gain, which persisted only for a short time and was mainly related to adipose tissue accretion. No functional improvement accompanied the amelioration in nutritional status. This short-term nutritional therapy in malnourished children with cystic fibrosis was effective in increasing relative weight and energy stores, but there was no evidence of any long-term functional benefit.

The effect of age on corneal and lens superoxide dismutase.

The enzymatic activity of copper-zinc superoxide dismutase within the cornea and lens of the juvenile (1 week) and adult (26 week) rat has been measured relative to both total soluble protein and immunoreactive protein. Enzymatic activity of corneal superoxide dismutase, localized in the epithelium and endothelium, remains constant with age relative to total soluble protein and the ratio of catalytically active enzyme to immunoreactive enzyme is near unity indicating that the superoxide dismutase present is enzymatically active. Likewise, in the juvenile rat, the majority of lens superoxide dismutase, which is located in the capsule epithelium, is enzymatically active. However, in the adult rat a large portion of the enzyme present is catalytically inactive. The enzymatic activity also decreases relative to total soluble protein with age in both the rat and the rabbit lens. This decrease in protection against toxic radicals and/or the accumulation of enzymatically defective enzyme may be contributing factors to age related lens disorders.

Leukocyte zinc in the assessment of zinc status.

The problems of assessment of human zinc status are reviewed, with particular emphasis on the limitations to each of the current available measurements. The advantages and limitations of leukocytes are then described. Methods of preparation and potential problems in the assay for zinc are described in detail. The data so-far produced by this method are reviewed.

Effect of maternal ethanol ingestion on fetal breathing movements, gross body movements, and heart rate at 37 to 40 weeks' gestational age.

The effect of maternal ingestion of ethanol (0.25 gm/kg) on fetal breathing movements, gross fetal body movements, and fetal heart rate was studied in 11 healthy pregnant women at 37 to 40 weeks' gestation. Fetal breathing movements were almost abolished within 30 minutes of the alcoholic drink and remained significantly decreased for 3 hours. The incidence of gross fetal body movements before or after ethanol was not different from that on the control day, and the fetal heart rate was not changed after maternal ingestion of ethanol.

Rorschach presentation of borderline personality disorder: primary process manifestations.

Obtained WAIS and Rorschach protocols for a sample of 21 psychiatric inpatients diagnosed as borderline personality disorder. Responses were analyzed according to Holt's (1977) and Rapaport, Gill, and Schafer's (1945-46) test correlates of primary process thinking and thought disturbance. Results were discussed in the context of Kernberg's (1975) structural and genetic-dynamic analyses of borderline personality organization and seemed to correspond with his characterization of this population as possessing contradictory personality traits, lacking in normal repressive defenses, and regressing toward primary process thinking. Interrater agreement on the assignment of Rorschach responses within the above scoring categories was assessed, with a high degree of concordance found for primary process content variables. It was concluded that the frequency and type of primary process content identified in the present study probably represent highly reliable Rorschach indicators of borderline personality disorder for purposes of differential diagnosis.

The effect of maternal hyperglycemia on gross body movements in human fetuses at 32-34 weeks' gestation.

This study demonstrated that fetal gross body movements were not significantly altered by fluctuations in maternal plasma glucose concentration. Fetuses moved episodically a relatively constant amount of time whether mother received water orally, 50 g of glucose orally or a 25 g bolus of glucose intravenously. In addition, depending on the length of the observational epoch, the incidence of fetal gross body movements differed. As the length of the observational period increased, the chance of finding no fetal gross body movements decreased and at intervals of 24 min duration, fetal gross body movements were absent in only 2% of 24 min periods examined, irrespective of whether they had received water orally, 50 g of glucose orally or 25 g of glucose intravenously. It is concluded that maternal carbohydrate intake was not an important determinant of fetal body movements in healthy fetuses at 32-34 weeks' gestation.

Nucleotide and deduced amino acid sequences of Torpedo californica acetylcholine receptor gamma subunit.

The nucleotide sequence has been determined of a cDNA clone that codes for the 60,000-dalton gamma subunit of Torpedo californica acetylcholine receptor. The length of the cDNA clone is 2,010 base pairs. The 5' and 3' untranslated regions have respective lengths of 31 and 461 base pairs. Data suggest that the putative polyadenylylation consensus sequence A-A-T-A-A-A may not be required for polyadenylylation of the mRNA corresponding to the cDNA clone described in this study. From the DNA sequence data, the amino acid sequence of the gamma subunit was deduced. The subunit is composed of 489 amino acids giving a molecular mass of 56,600 daltons. The deduced amino acid sequence data also indicate the presence of a 17-amino acid extension or signal peptide on this subunit. From these data, structural predictions for the gamma subunit are made such as potential membrane-spanning regions, possible asparagine-linked glycosylation sites, and the assignment of regions of the protein to the extracellular, internal, and cytoplasmic domains of the lipid bilayer.

Metabolism of acetylcholine receptors on embryonic amphibian muscle.

The functional properties of acetylcholine receptors are altered during the development of Xenopus myotomal muscle. A similar change also occurs in dissociated cell cultures of embryonic myotomal muscle. Metabolism of acetylcholine receptors was examined in order to ascertain whether the turnover rate changed at a time when the functional changes were detected. Incorporation of newly synthesized receptors occurred at a constant rate over a 24-hr period. The degradation of receptors followed first order kinetics with a half-life of 52 hr. Incorporation and degradation occurred at approximately equal rates such that 1 to 1.5% of the total receptor number was replaced every hour at equilibrium. Addition of neural tube cells to muscle cell culture, which altered the functional properties of channels, had no effect on either the incorporation or the degradation rate. We conclude that the changes in the functional properties of receptors occur independent of changes in the turnover rate of receptors.

Influence of maternal heart rate and gross fetal body movements on the daily pattern of fetal heart rate near term.

Continuous measurements of fetal heart rate (FHR), gross fetal body movements, fetal breathing movements, and maternal heart rate (MHR) were made for 24-hour observation intervals in 11 pregnant women at 38 to 40 weeks. There was a significant positive correlation between each mother's daily mean MHR and her fetus' daily mean FHR. There was a trough in mean hourly FHR between 0200 and 0600 hours and a trough in mean hourly MHR between 2400 and 0700 hours. Mean hourly FHR both during and between times of gross fetal body movements was significantly correlated to mean hourly MHR. At term, the mean FHR is strongly influenced by the mean MHR and the presence or absence of gross fetal body movements.
