Design and synthesis of histidine analogues of folic acid and methotrexate as potential folylpolyglutamate synthetase inhibitors.

Folypolyglutamate synthetase (FPGS) is responsible for the conversion of naturally occurring folates and antifolates to their poly-gama-glutamyl derivatives, which are the forms required for intracellular retention of folates and are also the preferred substrates (cofactors) for most folate-dependent enzymes. Folate and methotrexate analogues 6 and 4, with L-histidine in place of L-glutamate, were designed and synthesized as potential FPGS inhibitors. Target compound 5, the N tau-(carboxymethyl)-L-histidine derivative of 4, was also prepared. Compounds 4 and 5 inhibited the growth of L1210 cells (IC50 values: 0.091 and 0.15 microM, respectively) and were potent inhibitors of L1210 dihydrofolate reductase. No significant inhibition of FPGS by 4, 5, or 6 was observed at the high pH of the standard enzyme assay. This could be the consequence of a lack of protonation of the basic side chains, which is likely to be required for FPGS inhibitory activity. The observed cytotoxicity indicates that partial protonation of the imidazole ring permits cellular uptake of the analogues.

Hepatocellular and hepatic peroxisomal alterations in mice with a disrupted peroxisomal fatty acyl-coenzyme A oxidase gene.

Peroxisomal genetic disorders, such as Zellweger syndrome, are characterized by defects in one or more enzymes involved in the peroxisomal beta-oxidation of very long chain fatty acids and are associated with defective peroxisomal biogenesis. The biologic role of peroxisomal beta-oxidation system, which consists of three enzymes: fatty acyl-CoA oxidase (ACOX), enoyl-CoA hydratase/3-hydroxyacyl-CoA dehydrogenase (HD), and thiolase, has been examined in mice by disrupting ACOX gene, which encodes the first and rate-limiting enzyme of this system. Homozygous (ACOX -/-) mice lacked the expression of ACOX protein and accumulate very long chain fatty acids in blood. However, these homozygous mice are viable, but growth-retarded and infertile. During the first 3-4 months of age, the livers of ACOX -/- mice reveal severe microvesicular fatty metamorphosis of hepatocytes. In such steatotic cells, peroxisome assembly is markedly defective; as a result, they contain few or no peroxisomes. Few hepatocytes in 1-3-month-old ACOX -/- mice contain numerous peroxisomes, and these peroxisome-rich hepatocytes show no fatty change. At this stage, the basal mRNA levels of HD, thiolase, and other peroxisome proliferator-induced target genes were elevated in ACOX -/- mouse liver, but these mice, when treated with a peroxisome proliferator, showed no increases in the number of hepatic peroxisomes and in the mRNAs levels of these target genes. Between 4 and 5 months of age, severe steatosis resulted in scattered cell death, steatohepatitis, formation of lipogranulomas, and focal hepatocellular regeneration. In 6-7-month-old animals, the newly emerging hepatocytes, which progressively replaced steatotic cells, revealed spontaneous peroxisome proliferation. These livers showed marked increases in the mRNA levels of the remaining two genes of the beta-oxidation system, suggesting that ACOX gene disruption leads to increased endogenous ligand-mediated transcription levels. These observations demonstrate links among peroxisomal beta-oxidation, development of severe microvesicular fatty liver, peroxisome assembly, cell death, and cell proliferation in liver.

A transcription map of the major histocompatibility complex (MHC) class I region.

We have applied cDNA hybridization selection to nine YACs spanning 3 Mb of genomic DNA from a region centromeric to HLA-A to the histone cluster that lies telomeric to the human major histocompatibility complex (MHC). In addition to Class I genes and pseudogenes, we describe over 63 genes and 23 additional expressed sequence tags distributed throughout the region. Many of the full-length genes belong to gene families. Prominent among these are a group of genes encoding proteins showing homology to the carboxyl-terminal sequences of butyrophilin and an additional group of zinc finger genes. We also detected several previously undefined genes that are specifically expressed in cells of the immune system, indicating a more complex role of the MHC in the immune response than has been appreciated.

Interleukin 4 or oncostatin M induces a prolonged increase in P-selectin mRNA and protein in human endothelial cells.

During acute inflammation, P-selectin is transiently mobilized from Weibel-Palade bodies to the surface of histamine-activated endothelial cells, where it mediates rolling adhesion of neutrophils under hydrodynamic flow. During chronic or allergic inflammation, sustained expression of P-selectin on the endothelial cell surface has been observed. We found that the cytokines interleukin 4 (IL-4) or oncostatin M (OSM) induced a five- to ninefold increase in P-selectin messenger RNA (mRNA) in human umbilical vein endothelial cells (HUVEC) that persisted as long as 72 h. IL-4 elevated P-selectin mRNA by increasing its transcription rate rather than by prolonging its already long half-life. Stimulation of P-selectin transcription by IL-4 or OSM required new protein synthesis and tyrosine phosphorylation of cellular proteins. Tumor necrosis factor alpha, IL-1 beta, lipopolysaccharide, or IL-3 did not increase P-selectin mRNA in HUVEC, and did not augment the IL-4-induced increase in P-selectin transcripts. IL-4 or OSM increased P-selectin protein on the cell surface as well as in Weibel-Palade bodies. Under flow conditions, neutrophils rolled on P-selectin expressed by IL-4-treated HUVEC, and even more neutrophils rolled on P-selectin after IL-4-treated HUVEC were stimulated with histamine. These data demonstrate that IL-4 or OSM stimulates endothelial cells to synthesize more P-selectin over prolonged periods. The increased expression of P-selectin may facilitate the emigration of leukocytes into sites of chronic or allergic inflammation.

Anxiety and depression in general practice patients: prevalence and management.

OBJECTIVE: To determine the prevalence of anxiety and depression in general practice patients and assess management of these conditions by general practitioners (GPs). METHOD: A random sample of 212 GPs were approached to be interviewed and to conduct a patient survey and audit on 50 consecutive patient consultations during 1993. PARTICIPANTS: 117 GPs (55% response rate) and 4867 patients (85%) who completed questionnaires suitable for analysis. SETTING: General practices in two areas (divisions of general practice) in Sydney, New South Wales. RESULTS: Thirty-six per cent of patients had abnormal scores on a General Health Questionnaire (GHQ-12); they were more likely to be women or to be unemployed. Twenty per cent of these patients had been treated for depression or anxiety in the previous 12 months; 52% were prescribed drug therapy, and were more likely to be older, male or unemployed. Seventy per cent of patients reported having been offered therapy by their GP that did not involve drugs. Twenty-four per cent had been referred to another health professional; they were more likely to be younger, or men, or patients attending their usual doctor. CONCLUSIONS: A brief screening instrument may improve GPs' detection rate of patients with anxiety or depression. The high prevalence of these conditions in unemployed people deserves particular attention by GPs. Both drug and non-drug therapies are being more appropriately applied in general practice than previously.

Isolation of a novel gene down-regulated by v-src.

We have isolated a novel gene which was expressed in normal rat cells, but completely suppressed in cells transformed by v-src. The molecularly cloned cDNA was about 1.8 kb in size, containing an open reading frame composed of 464 amino acid residues. DNA sequence analysis showed that there was no corresponding gene in the data bases. Besides the suppression of gene expression in the v-src transformed cells, its expression was also strongly suppressed in cells transformed by other oncogenes such as v-abl, v-fps, v-mos, v-sis, v-K-ras, and polyomavirus middle T, but not affected in cells transformed by human papillomavirus type 16 E6E7 and polyomavirus large T. We named the gene drs for a gene down-regulated by v-src.

Tyrosine phosphorylation of inducible nitric oxide synthase: implications for potential post-translational regulation.

The activation of cultured Raw 264.7 murine macrophages with interferon gamma and lipopolysaccharide results in the expression of inducible nitric oxide synthase (i_NOS) and the subsequent production of nitric oxide. In the present study, the i-NOS expressed in these activated cells was characterized for possible post-translational protein modification by endogenous tyrosine protein kinases. Western-blot analysis using phosphotyrosine antibodies revealed that i-NOS was phosphorylated on tyrosine residues and that this was an early event coinciding with the appearance of newly synthesized i-NOS. A brief exposure of activated cells to vanadate, a tyrosine phosphatase inhibitor, significantly increased the level of i-NOS tyrosine phosphorylation, suggesting that tyrosine phosphatases are dynamically involved in the regulation of this process. Vanadate treatment of activated cells also resulted in a rapid increase in enzyme activity, occurring within 5 min of exposure. Taken together, these results demonstrate that tyrosine kinases and phosphatases are involved in the post-translational modification of i-NOS and may potentially play a role in modulating the functional activity of the enzyme in macrophages.

Hydrogen peroxide toward enhanced oxide growth on titanium in PBS solution: blue coloration and clinical relevance.

Oxide films formed on titanium exposed to a phosphate-buffered saline solution with and without hydrogen peroxide (H2O2) addition were investigated by means of electrochemical impedance spectroscopy (EIS) and X-ray photoelectron spectroscopy (XPS). The oxide growth at the titanium/electrolyte interface was monitored in situ by daily EIS measurements during periods of several weeks. The results suggest that the oxide film can be described by a two-layer model with a barrier inner layer and a porous outer layer. H2O2 addition results in an increased dissolution/oxidation rate that leads to an enhanced oxide growth of the porous outer layer. As a result, the total oxide film can reach a thickness corresponding to an interference blue color. Based on XPS results, H2O2 addition furthermore seems to facilitate the incorporation of phosphate ions into the thicker porous layer. This observation may be related to the so-called osseointegration properties of titanium.

Abnormal gastric myoelectrical activity and delayed gastric emptying in patients with symptoms suggestive of gastroparesis.

Gastric myoelectrical activity modulates gastric motor activity. Abnormalities in gastric myoelectrical activity may be associated with gastric motility disorders. The aim of this study was to investigate the correlation of gastric myoelectrical activity with gastric emptying in symptomatic patients with and without gastroparesis. Ninety-seven patients with symptoms suggestive of gastroparesis participated in the study. Gastric myoelectrical activity was recorded using surface electrogastrography. The electrogastrogram (EGG) was recorded for 30 min in the fasting state and for 120 min after a solid test meal. Gastric emptying of the solid meal was simultaneously monitored for 120 min. Patients with delayed gastric emptying showed a significantly lower percentage of normal gastric slow waves (P < 0.03) and a significantly reduced increase of the dominant power in the postprandial EGG (P < 0.02). Postprandial EGG parameters were found to be able to predict delayed emptying of the stomach. Postprandial gastric dysrhythmia predicts delayed gastric emptying with an accuracy of 78%, while the abnormality in postprandial EGG power predicts delayed gastric emptying with an accuracy of 75%. All patients with abnormalities in both the rhythmicity and the power had delayed gastric emptying. Patients with delayed gastric emptying have a lower percentage of normal gastric slow waves in the EGG and a lower postprandial increase in the dominant power. Abnormalities in the postprandial EGG seem to be able to predict delayed emptying of the stomach. However, a normal EGG does not seem to guarantee normal emptying of the stomach.

Role of sham feeding in postprandial changes of gastric myoelectrical activity.

The aim of this study was to evaluate the role of sham feeding in postprandial changes of gastric myoelectrical activity. Eighteen asymptomatic healthy volunteers (10 men, 8 women; mean age: 31), with no history of gastrointestinal disease were studied. Gastric myoelectrical activity was recorded for 30 min at baseline, 30 min after sham feeding, and 1 hr after eating, using surface electrogastrography. The electrogastrogram (EGG) was analyzed by spectral analysis. It was found that the changes of postprandial EGG parameters were significantly correlated with those after sham feeding (EGG dominant power: r = 0.6, P < 0.01; dominant frequency: r = 0.8, P < 0.001; percentage of regular slow waves: r = 0.7, P < 0.003). We concluded that intrinsic gastric electrical activity can be altered by sham feeding and the cephalic phase of digestion plays an important role in the postprandial response of gastric myoelectrical activity.

Flow cytometric characterization of isolated porcine hepatocyte suspensions for liver support.

The high yield hepatocyte isolation necessary for hybrid liver assist devices (LAD) unavoidably increases contamination by nonparenchymal cells and depresses hepatocyte viability and functions. We have developed a flow cytometric procedure that improves quality control of the isolations. Cells present in these preparations were labeled by immunofluorescent antibody staining against cytokeratin 8, 18 as well as vimentin to identify hepatocytes, fibroblasts, and endothelial cells. Antibody staining against albumin and carbamoylphosphate synthetase allowed assessment of levels of albumin and carbamoylphosphate synthetase based on the hepatocyte relative fluorescence intensity. Hepatocyte P450 enzyme activity was measured by its ability to convert 5,6-methoxycarbonylfluorescein, a nonfluorescent substrate, to an intracellular fluorescent product. Flow cytometric methods of cell type identification and cell function assessment are fast and accurate and can be applied to commercial cell production. They may also provide an avenue for the enrichment of otherwise heterogeneous hepatocyte suspensions with cells presenting the specific functions desired for an hybrid liver assist devices.

[The role of TNF alpha, IL-1 beta and MIP-1 alpha in LPS-induced organ injury].

OBJECTIVE: To investigate the role of inflammatory cytokines in endotoxemia models and to explore the possible anti-cytokine therapy of endotoxemia. METHODS: TNF alpha in plasma was measured by ELISA, and the mRNA of cytokines was assessed by slot blot analysis. RESULTS: LPS-induced TNF alpha release was in a dose:dependent manner in human whole blood. Dexamethasone (> or = 10(-8)mol/L) exhibited inhibitory effect on TNFa release. Ibuprofen 10(-7)-10(-9) mol/L had inhibitory effect on TNFa production, whereas 10(-3)-10(-4)mol/L stimulated TNFa release. The rat model of acute lung injury was made by LPS intraperitoneal injection. Both dexamethasone and ibuprofen that injected at 1 hour before LPS injection could decrease the contents of Evans blue in the lung (t 2.80 and 7.31 respectively, P < 0.05 vs LPS control). After LPS administered, there was a progressive up regulation in transcripts of TNF alpha, IL-1 beta and MIP-1 alpha in whole lung homogenates of rats. The mRNA peaked at 2, 6, 12 hours respectively. The TNF alpha, IL-1 beta and MIP-1 alpha mRNA levels decreased markedly when dexamethasone or ibuprofen given at 1 hour before LPS injection. CONCLUSION: TNF alpha, IL-1 beta and MIP-1 alpha play an essential role in the inflammatory response. LPS-induced acute lung injury may be prevented by dexamethasone and ibuprofen which have inhibitory effect on the gene expression of cytokines in the lung.

[A comparison of the modified gamma nail and the ender nail for treatment of intertrochanteric fractures].

We made a comparison between the modified Gamma nailing and the Ender nailing in the treatment of 102 patients with intertrochanteric fracture. Sixty patients were treated with the modified Gamma nailing and forty-two patients with the Ender method. The preoperative conditions of the patients in the two groups were similar as showed by statistical analysis. More intraoperative bleeding was recorded in the modified Gamma nail group. However, there were a earlier full weight -bearing, a better hip function and a lower rate of operative complication in the patients treated with the modified Gamma nailing. We conclude that with the modified technique and the modified femoral component the Gamma nail is advanced in the treatment of intertrochanteric fracture. However, it is danger to apply in elderly patients accompanied with certain vital problems. For these patients the Ender nail may be considered.

The second space experiment of protein crystallization with domestic facilities.

The second experiment of protein crystallization was performed on domestic re-entry satellite FSW-2 in 1994-07. The results are superior to the ones of the first mission in 1992: 9 of 10 different proteins were crystallized in space, and 70% of the total 48 samples yielded single crystals. Besides hen egg-white lysozyme which grew high-quality crystals on the first mission, an acidic phospholipase A2(aPLA2) from snake venom and hemoglobin from Anser Indicus produced good-quality crystals suitable for X-ray diffraction analyses. The positive effect of microgravity on protein crystal growth is verified again at this time.
