The innervation of the caudal neurosecretory system of Poecilia sphenops (black molly) was studied by use of the retrograde horseradish peroxidase (HRP) method. The structure of the caudal neurosecretory system in this species was well suited for application of HRP procedures. Acrylamide/HRP gel implants were placed in the nucleus of the caudal neurosecretory system. Two neuronal groups which contained HRP filled cells were found in the brains tem. Bilateral projections originate from the dorsal tegmentum of the midbrain and the reticular nucleus of the medulla.
Kyphotic deformity arising from failure of the formation of a vertebral body is described in nine patients. The study of the natural history demonstrates the progressive nature of this disorder resulting in severe deformity and neurological embarrassment. Early radiological recognition of this congenital deformity is essential and tomography is important in the early assessment. Tomography combined with myelography is essential to delineate the extent of bony abnormality and cord compression in severe kyphosis in order that adequate decompression and reconstructive surgery can be performed.
Surgically removed discs from patients with either spondylolisthesis or tears of the posterior annulus fibrosus were analysed for water, proteoglycan, and collagen content and compared with post-mortem control material. Discs from patients with spondylolisthesis had a reduced proteoglycan content in all section sampled and less collagen in the outer annular layers. In contrast discs containing tears in the posterior annulus were unaltered biochemically, although extended studies on 2 patients indicated that there may be localised biochemical changes in the region of the tear itself. Collagen types I, II, and III and proteoglycan distributions were studies qualitatively by immunofluorescence. Collagen types I and III appeared to be reduced in discs from patients with spondylolisthesis, but again little change was found in patients with tears in the posterior annulus fibrosus.
The structural and conformational features of the "anomeric" DL-trans- and DL-cis-5-(3-hydroxytetrahydrofuran-2-yl)uracils (3a, 4a) and five similar analogues were studied in order to determine their applicability as models of beta- and alpha-pseudouridine. The 270-MHz proton NMR spectra were measured for all analogues to define their ring geometries in solution and to estimate the solution population of model N, S conformers in a two-state dynamic equilibrium treatment. Two sets of calculations were employed to evaluate the relative contributions of these states to the observed vicinal coupling constants related to the C(3')-C(4') fragment. In the first, similar geometries were assumed for each pair of conformers, while in the second, limited to 3, the geometries were those derived from X-ray crystallographic data; both gave comparable results. The cis analogues 4a and 4b are excellent conformational models for alpha-pseudouridine. In the trans series (3a-c), the equilibrium is weighted toward the N conformer (approximately 80%), differing from that found in beta-pseudouridine for which each model conformer is equally populated. Possible implications of the conformational effects upon the pairing properties of pseudouridine in tRNA are discussed.
