Fatty acid composition was analysed in the producer of avermectins, Streptomyces avermitilis C-18 grown in chemically defined medium with different nitrogen sources. Significant differences in nitrogen regulation of fatty acid biosynthesis were found in this strain in comparison with other streptomycetes studied so far. This finding could be explained at the level of regulation of branched-chain amino acid metabolism.
Development of resistance to ivermectin in vitro is reported for the first time. Two strains of Caenorhabditis elegans D5 and D6 able to grow on agar plates after treating with up to 3 mg ivermectin per L were selected. The parent strain N2 was extremely sensitive to ivermectin, its growth being inhibited by treatment with 0.1-0.2 mg IVM per L.
The authors report the results of treatment and a one year follow-up study of 1000 burned patients (1982-1986). Of 482 patients with superficial and deep dermal burns 246 patients had been operated (135 of 168 patients with deep burns). Impairment of function has been observed in 52 and complete disability in 7 cases. The authors suggest planned collecting of data on the remaining complications following thermal injury.
After implantation of intraocular lenses (IOL) Purkinje images 3 and 4 (Pi3 and Pi4) are clearer on the anterior and posterior surface of its optical part (subsequently only lens). Along with the corneal Pil this phenomenon was already used in clinical work for more accurate calculations of the degree of decentration and tilt of the lens in the eye. The authors describe a simplified method for assessment of the position of the lens by means of Pil, 3 and 4, using apparatuses with an approximately coaxial light. The method uses overlapping of Pi1, 3 and 4 in perfect centering of the lens in the axis of the eye (it is assessed by drawing a perpendicular line on the centre of the cornea) and marked dislocation of Pi3 in the direction of decentration of the planoconvex lens with the convexity facing the cornea. The inclination of the lens must be eliminated according to positions Pi1 and Pi4. The method is suitable for peroperative centering and for assessment of the direction and extent of decentration of these lenses. The authors discuss the possible use of this method also in biconvex and planoconvex lenses with the convexity facing the retina. In a group of 10 implanted planoconvex lenses the authors demonstrate the possibility of perfect peroperative centring of the lens and other applications of the method. Incorrect position of the IOL at the end of the operation, which can be one of the factors of later complications, did not change during the early postoperative period.
Scleroplastic surgery is known to encounter the pathogenetical agent when the progressive myopia develops. In the first part, the implemented surgical approaches as well as up-to-date surgical techniques are presented. The experimental part is reserved for the selection of material which is necessary to perform scleroplasties. Both the allogenic and xenogenic grafts have been studied for histocompatibility on 20 rabbits. Following materials were used: 2 types of collagenic sponge, native and deproteinated human sclera, modified dura mater as well as both the native and devitalized sclera from rabbits. The best results were obtained with allogenic sclera, deproteinated xenogenic sclera and collagenic sponge. The application of other materials is discussed.
The entire amino acid sequence of human cytomegalo-virus (HCMV) 150K matrix phosphoprotein (pp150), consisting of 1048 amino acid residues, was divided into 95 overlapping 20 amino acid peptides which were synthesized on polyethylene rods. The rods were subjected to ELISA with pooled anti-HCMV-positive and anti-HCMV-negative sera. Four peptides recognized by the anti-HCMV-positive pool only were synthesized by the solid-phase method and their reactivity in a conventional ELISA, using a panel of 14 individual anti-HCMV-negative and 20 anti-HCMV-positive antisera, was evaluated; three peptides were found to be specifically reactive. Results obtained with one of these peptides (residues 595 to 614) in ELISA showed a good correlation with those obtained using a routinely performed complement fixation test.
The treatment of burned patients is frequently carried out under compulsion in more phases. The prognosis may be favourably influenced by the expert intervention in due time, on the other side deteriorated by transport in critical condition without adequate preparation. The optimal time of the secondary transport corresponds approximately with the end of the critical condition. Authors describe the criteria on the basis of which the condition of the burned patient may be considered as stable during the period of the transport to be expected. The necessary measures during the primary hospital treatment are enumerated.
Multiple continuous-flow solid-phase peptide synthesis has been adapted for synthesis of peptides on a cellulose carrier (Whatman 3MM paper). Paper-bound synthetic peptides that represent antigenic determinants of particular proteins detected antibodies against the respective proteins in an enzyme-linked immunosorbent assay. The method is applied to the synthesis, and use in site-directed serology, of four peptides derived from the gp41 glycoprotein of HIV, the Epstein-Barr virus-determined nuclear antigen-1 and VCA proteins of the Epstein-Barr virus, and the early region of human papillomavirus type 11.
