Interface dynamics of lipid membrane spreading on solid surfaces.

As a model system for two-dimensional interface dynamics, we study the wetting front of a lipid membrane moving over a solid substrate that is structured with regularly spaced pinning centers. By analyzing the contour of the front, we derive the normal growth rate and the relaxation coefficient. Both exhibit a 1/t(1/2) time dependence. Moreover, the friction coefficient and the line tension can be determined. Randomly distributed pinning centers cause a fractal contour line, whereas on surfaces that are artificially roughened, self-affine contour lines are observed. The latter exhibit an anomalous roughness exponent of zeta = 0.81+/-0.05.

1H NMR of compounds with low water solubility in the presence of erythrocytes: effects of emulsion phase separation.

When lipophilic compounds like diethyl phthalate (DEP) were added to water, two sets of resonances appeared in the 1H NMR spectrum, whereas when added in concentrations above approximately 3.5 mM to erythrocytes in a high haematocrit suspension, only one set of resonances was observed at the low-frequency position. The appearance of one set of resonances at lower frequency was found to be common to a series of lipophilic compounds in erythrocytes. The appearance of the NMR spectra is ascribed to the existence of an emulsion, meaning two different phases of a compound: a "droplet" (resonances to lower frequency) and aqueous dissolved phase (resonances to higher frequency). The absence of the resonances from the dissolved phase in erythrocyte solution is ascribed to exchange broadening. The absolute chemical shift of the compound in its "droplet" phase was also measured using a cylindrical/spherical microcell. This arrangement mimicked the geometry of the dissolved versus the phase-separated species and thus obviated the effect of a difference in magnetic susceptibility between the "droplet" solute and its aqueous solution. Factors influencing the formation of emulsion phases such as erythrocytes, haemoglobin and smaller proteins were investigated; they are found to be effective in the order given.

Purification and characterization of an astrocyte GABA-carrier inducing protein (GABA-CIP) released from cerebellar granule cells in culture.

A glycoprotein that induces gamma-aminobutyric acid (GABA) carriers in cultured cerebellar astrocytes was isolated and purified from conditioned media from cultured cerebellar granule cells by anion exchange chromatography, affinity chromatography, and gel filtration. Following gel filtration three fractions corresponding to M(r) 30,000, 60,000, and 240,000 exhibited GABA carrier inducing activity. SDS-PAGE of the M(r) 30,000 fraction revealed under non-reducing conditions three bands corresponding to M(r) 30,000, 60,000, and 120,000. Under reducing conditions only the band corresponding to an M(r) of 30,000 was visible. An identical N-terminal amino acid sequence and amino acid composition was found in the M(r) 30,000 and the M(r) 60,000 fraction from the gel filtration. These results suggest that the protein polymerizes into di- and tetramers. Computer base analysis of the N-terminal amino acid sequence revealed no obvious homology with previously reported N-terminal amino acid sequences. Application of the glycoprotein to cerebellar astrocytes led time and dose dependently to an increased GABA uptake. The effect became maximal after 24 h exposure of the cells. Kinetic analysis of the GABA uptake showed that exposure of the astrocytes to the glycoprotein led to an increase in Vmax for GABA uptake without affecting Km, suggesting an increase in the number of GABA carrier molecules. Addition of actinomycin D together with the glycoprotein abolished this effect suggesting that the glycoprotein acts by stimulating de novo synthesis of GABA carriers. Hence, the newly purified protein secreted from neurons is named GABA-carrier inducing protein (GABA-CIP).

A clinical comparison of optical and ultrasonic pachometry.

Central corneal thickness (CCT) of normal and oedematous corneae was measured in a study comparing a modified Haag-Streit optical pachometer and an ultrasonic pachometer (Teknar Ophthasonic, preset velocity 1630 m/s). Sixty-eight patients were examined before and after cataract extraction with implantation of an anterior chamber lens. Mean values of CCT before operation were 531 +/- 4.9 (SEM) micron by optical pachometry and 524 +/- 4.7 microns when measured by ultrasound (not significantly different). On the first post-operative day the values were 618 +/- 8.4 and 602 +/- 7.6 microns for optical and ultrasonic measurements, respectively (significantly different, 2P less than 0.001). Correlation analysis showed a high dependence between the methods with coefficients of correlation being 0.955 before and 0.958 after the operation. Linear regression analysis revealed small, but significant differences between the techniques. The difference between the two methods increased with increasing corneal hydration, whereas it could not be ascribed to sex, age, or intraocular pressure. It is concluded that for clinical purposes optical and ultrasonic pachometry techniques are comparable.

Life events and social support in prediction of stroke outcome.

A prospective study of 252 patients (average age 73, range 26-95) admitted to a regional general hospital over a 12-month period was carried out. 241 patients had stroke verified by the initial neurological examination and CT scan, and of these baseline data were not available on 27%. 34% died before or were not willing or able to provide data at follow-up. 39% survived and completed the study. Prestroke life events and social support could not predict the outcome of stroke rehabilitation measured as survival, length of stay, functional recovery (Barthel's Index) or placement at the follow-up 12 months after the onset of stroke. Age and arteriosclerotic heart disease predicted poor survival at follow-up. Premorbid hypertension, stroke, diabetes, obesity, tobacco smoking, and alcohol consumption did not significantly influence the outcome. Problems in stroke rehabilitation research are discussed.

Circadian variation of serum thyrotropin in endogenous depression.

The circadian variation of serum thyrotropin (thyroid-stimulating hormone; TSH) was studied in nine patients with endogenous depression before and after recovery. Depressed state did not appear to influence the pattern of TSH. When 2 mg of dexamethasone was administered, serum TSH was significantly reduced for 18 hours, whereafter the effect leveled off. The TSH response to thyrotropin-releasing hormone (TRH) was evaluated 25 hours after the administration of dexamethasone and the response was found to be unchanged.
