Antitumor and antimetastatic activities of human recombinant interferon alpha A/D.

Human recombinant interferon alpha A/D (alpha A/D) was examined for its antitumor activity in several mouse tumor models using metastatic tumors, such as B16 melanoma F1, BL6 and F10, UV2237m fibrosarcoma, and K1735m melanoma. Therapeutic treatment with alpha A/D reduced the incidence of pulmonary metastasis and inhibited the tumor growth resulting in an increase of the mean survival time. Since alpha A/D also showed a prophylactic activity against the metastasis, its antitumor activity was suggested to be due to augmentation of the host defense systems. This was confirmed by the fact that alpha A/D inhibited the in vivo growth and incidence of pulmonary metastasis of B16 F1 sublines regardless of their sensitivity to the direct antiproliferative activity of the IFN in vitro.

High constitutive expression of heat shock protein 90 alpha in human acute leukemia cells.

The constitutive expression of the genes for four heat shock proteins (hsps) was studied in leukemia cell lines, cells obtained from patients with acute leukemia, and normal blood cells by means of Northern-blot analysis. Western-blot analysis with hsp90 antibody showed that the leukemia cells contained larger amounts of hsp90 than the normal peripheral mononuclear cells. The expression of the hsp90 alpha gene was enhanced in the leukemia cell lines and the acute leukemia cells from patients as compared with the normal blood cells. In contrast, the expression of the hsp90 beta gene could hardly be recognized in either the acute leukemia cells or the normal blood cells. An increased expression of hsp70 gene was observed in only one patient. The expression of the hsp27 gene was enhanced in one-half the patients with common acute lymphoblastic leukemia. Thus, exclusively the hsp90 alpha gene was expressed highly in the leukemia cells, indicating its association with cellular proliferation.

Design and properties of photocurable electroconductive polymers for use in biosensors.

The authors report preparation of a new hydrophilic photocurable electroconductive polymer to be used as a matrix for an enzyme based biosensor. Polymers, prepared by radical terpolymerization, consisted of N,N-dimethylacrylamide (DMAA) as the water soluble part, m-azidostyrene (AzSt) as the photochemically active center to entrap an enzyme and to fix it on an electrode, and vinylferrocene (VFe) as the charge transfer mediator between an enzyme and an electrode. Ultraviolet light (UV) irradiation to the terpolymer coated on a carbon electrode produced a water swellable cross-linked polymer that was chemically fixed on a carbon electrode. Cyclic voltammetry on a hydrogelated electrode in a phosphate buffered solution exhibited single oxidation/reduction reproducibly, indicating that the formed conducting hydrogel was stable. An increase in VFe content and a decrease in AzSt content of a terpolymer resulted in larger electrochemical responses on normal pulse voltammetry. When glucose oxidase was immobilized in a hydrogel, a dose dependent electrochemical response to glucose was observed.

A case of infectious pseudoaneurysm of the abdominal aorta associated with infectious spondylitis due to Klebsiella pneumoniae.

A 67-year-old woman with pneumonia and diabetes mellitus was admitted with the complaints of abdominal and back pain. Sputum culture was positive for Klebsiella pneumoniae. Computed tomographic scanning (CT) of the abdomen and spinal radiograph of the lumber column revealed a paraventebral space-occupying lesion, abdominal aortic aneurysm and destructive change of L3 and L4. Pseudoaneurysm of the abdominal aorta associated with infectious spondylitis with paravertebral abscess was suspected and confirmed by aortography. Klebsiella pneumoniae was cultured from the abscess. The patient's condition improved rapidly after drainage of the abscess and administration of LMOX and gentamicin. Infectious pseudoaneurysm of the abdominal aorta associated with infectious spondylitis has rarely been reported. These two in combination due to Klebsiella pneumoniae has not been reported to our knowledge. The pathologic changes were found easily by CT scan. When infectious aneurysm or infectious spondylitis is diagnosed alone, possible combination of these diseases should be kept in mind.

Case of a child with paroxysmal nocturnal hemoglobinuria diagnosed by flow cytometry.

A case of a child with paroxysmal nocturnal hemoglobinuria (PNH) is characterized by an increased sensitivity of the erythrocyte to hemolytic action of complement. The widely used Ham test may not always be reliable. Recently, a panel of monoclonal antibodies has become available to detect various glycosylphosphatidylinositol (GPI)-linked proteins by flow cytometry (FCM) and the deficiency of GPI-anchored proteins on the various kinds of cell membranes is implicated as the pathogenesis of PNH. We diagnosed a case of a child with PNH by FCM and complement lysis sensitivity (CLS) test, which showed the increased sensitivity of PNH erythrocytes to complement. His diagnosis was delayed because of Ham test negativity and rarity of PNH cases in children.
