IgE synthesis in man. I. Development of specific IgE antibodies after immunization with tetanus-diphtheria (Td) toxoids.

Specific IgE and IgG antibodies were measured in 39 adolescent and adult humans preselected to have less than 0.125 antitoxin units (AU) ml to tetanus antitoxin (IgG) irrespective of previous immunization history. Mean antitoxin values (IgG) measured by sheep red cell hemagglutination (SRBC) increased by 2 logs to 1.49 AU/ml sera for tetanus and by 1 log to 0.025 AU/ml sera for diphtheria. Specific tetanus IgE was noted to increase after immunization from a mean 2.0 +/- 0.2% counts bound to 3.1 +/- 0.6% counts bound, but was not statistically significant. Diphtheria-specific IgE rose significantly (p less than 0.05) from a mean 1.5 +/- 0.2% counts bound to a mean 4.0 +/- 1.2% counts bound in atopic individuals and from a mean 1.2 +/- 0.0% counts bound to a mean 2.2 +/- 0.5% counts bound in nonatopics. These data document the development of tetanus and diphtheria IgE antibodies after booster immunization.

[Life expectancy and frequency of infarct after aorto-coronary bypass].

Survival rate and incidence of myocardial infarction after aortocoronary bypass operation in 274 patients are presented. Mortality was 5.1% within the first postoperative month and the 5-year-survival rate was 86%. There were 24 perioperative myocardial infarctions and another 21 infarctions over the next 5 years. Comparison of these data with the natural history of coronary heart disease suggests a possible prolongation of survival after bypass surgery. The incidence of myocardial infarction appears to be unchanged.

Serum testosterone, body composition, and strength of young adults.

The relationship between serum testosterone (ST), body composition, and static strength was studied in 26 college females and 16 male college football players. In addition, the transient effect of weight training on ST was studied in 10 college males (5 skilled and 5 unskilled weight trainers), 12 male high school students, and 5 female college students. ST, measured by radioimmunoassay, ranged from 333.7 to 848.1 ng/100 ml in males, and from 32.8 to 121.5 n/100 ml in females. The high school subjects had significantly lower levels of ST. Correlations between serum testosterone, body composition (measured by densitometry), and static strength (grip and backlift) were nonsignificant in males and females. Comparisons of subjects with the highest and lowest levels of testosterone within each sex showed no significant differences in strength or body composition. Serum testosterone increased 111.4 +/- 96.l ng/100 ml (X +/- SX) following a weight training session in the male college group, but failed to increase in the college females or high school males. There were no significant differences in testosterone increases between the skilled and unskilled male weight trainers. Maximal exertion may be necessary for an increase in serum testosterone to occur. Lack of an increase in testosterone by high school males or college females may have been due to a submaximal effort during the weight training exercise.

Two injections of diphtheria-tetanus-pertussis-polio vaccine as the backbone of a simplified immunization schedule in developing countries.

From studies of antibody levels induced against poliovirus types 1, 2, and 3 and against diphtheria and tetanus toxins as well as from epidemiologic studies comparing the protective effect in children of two and three doses of diphtheria-tetanus-pertussis (DTP) vaccine, respectively, it can be concluded that two injections of DTP-polio vaccine administered at least four months apart will induce immunity against the four diseases. An immunization schedule can be built up in which bacille Calmette-Guérin (BCG) vaccine is given simultaneously with the first DTP-polio injection at the age of three to eight months and vaccination against measles and--if needed--yellow fever is performed simultaneously with the second DTP-polio immunization at the age of nine to 14 months.

Evaluation of proteinuria by two-dimensional polyacrylamide gel electrophoresis after kidney transplantation.

Urinary proteins were studied in patients after kidney transplantation during various functional states (normal function, acute rejection, chronic rejection) by two-dimensional polyacrylamide gel electrophoresis. In the first dimension, the proteins were separated according to their electric charge by isoelectric focusing, and in the second dimension according to their molecular weight by sodium dodecyl sulfate polyacrylamide gel electrophoresis. After electrophoresis the proteins were visualized using a highly sensitive silver stain technique. The combination of these methods allowed the discrimination of up to 250 protein spots in human urine, most of them unidentified up to now. Functional changes of the kidney transplants were accompanied by complex changes of the protein pattern in urine. These changes cannot be simply compared to the tubular and glomerular pattern of proteinuria which can be identified by one-dimensional sodium dodecyl sulfate electrophoresis. The 2D electrophoresis of urinary proteins may develop into a useful tool in localizing of kidney damage and in the evaluation of renal disease.

Glucocorticoids and evolution of haemopoietic tissue in the liver of the rat foetus.

The number of haematopoietic cells per unit of volume decreases after day 18 and falls rapidly until day 21. On day 21 it is reduced by three quarters as compared to the day 17 value. Maternal laparotomy causes, in the 24 h period following the operation, an anticipated and accelerated decrease. This effect is prevented by previous adrenalectomy. After maternal adrenalectomy, rat foetuses submitted to decapitation in utero, and therefore completely deprived of corticosteroids, do not present in their liver the involution of the haematopoietic tissue normally observed in their intact littermates. Cortisol administration restores the normal involution. These results show the importance of the role played by corticosteroids in the regulation of foetal liver hematopoietic function. The evolutions of proportions and frequencies of the different cellular types composing the haematopoietic tissue studied during normal pregnancy and after either overload or deprivation of corticosteroids, suggest that corticosteroids exert their action essentially at two levels of the maturation process. They decrease the ability of self-maintenance of the precursor cell pool and accelerate the maturation of the polychromatophilic cells, resulting in a progressive exhaustion of the haematopoietic tissue.
