Recombinant human tumor necrosis factor-alpha: evidence of an indirect mode of antitumor activity.

The antitumor activity of recombinant human tumor necrosis factor (rTNF-alpha) was examined on murine tumors in mice and in cultured cells in vitro. Mice were implanted intradermally with Meth A fibrosarcoma (Meth A) on day 0. rTNF-alpha caused tumor necrosis and inhibited the tumor growth when given i.v. on day 7 or 10, but not when given on day 3. When rTNF-alpha was given i.v. in doses of 0.1-3.2 micrograms/mouse twice a week for 3 weeks beginning on day 7 or 11, the growth of solid Meth A, Colon 26 adenocarcinoma, Colon 38 carcinoma, Sarcoma-180, and M5076 reticulum cell sarcoma tumors implanted s.c. or intradermally was markedly inhibited, and the life of the mice bearing these tumors, except M5076 reticulum cell sarcoma, was prolonged. The growth of Meth A implanted i.m. was also markedly inhibited by rTNF-alpha given i.v. However, the life of mice bearing i.p. Colon 26 adenocarcinoma, MH134 hepatoma, Sarcoma-180, and Ehrlich carcinoma was not prolonged by rTNF-alpha given i.p. nine times (days 1-9) in doses up to 1.0 or 3.2 micrograms/mouse. Only in the case of mice bearing i.p. Meth A, the life was slightly prolonged by i.p. treatment with rTNF-alpha but not by i.v. treatment. In experiments against in vitro cultured cells, rTNF-alpha did not show any direct cytotoxicity against mouse tumor cells: Meth A, Colon 26 adenocarcinoma, Colon 38 carcinoma, and Sarcoma-180, but had a cytotoxic effect against L929 mouse fibroblast. The results suggest that rTNF-alpha is a unique antitumor drug with potent necrotizing activity against solid tumors in mice, and that this activity may derive from indirect mechanisms related to the growth of tumors and not to the direct cytotoxicity of the drug.

Correlation of hepatitis B virus DNA and antigens in the liver. A study in chronic liver disease.

Hepatitis B virus deoxyribonucleic acid (DNA) and antigens (HBsAg and HBcAg) were studied in liver biopsy specimens from 105 HBsAg-positive patients with chronic liver diseases. Free or integrated viral DNA, or both, was detected in 83 of 105 (79%) patients, whereas HBsAg and HBcAg were demonstrated immunohistologically in 96 (91%) and 39 (37%), respectively. Of 60 patients with detectable free viral DNA, 38 (63%) were positive for HBcAg, whereas only 1 of 45 (2%) with either integrated viral DNA alone (n = 23) or no detectable viral DNA (n = 22) was positive for HBcAg (p less than 0.001). Furthermore, the amount of HBcAg was positively correlated with the amount of free viral DNA in the liver tissue. In contrast, HBsAg was well expressed not only in the liver with free viral DNA, but also in the liver with integrated DNA. These data suggest that the synthesis of HBcAg is primarily directed by free viral DNA, whereas that of HBsAg may be directed by free as well as integrated viral DNAs.

Structure and mapping of the DNA of human parvovirus B19.

DNA from human parvovirus B19 was prepared from infected serum and examined by electron microscopy. Double-stranded molecules were seen, often with characteristic 'fold-back' ends that were assumed to be due to the inverted terminal repeats of the genome DNA. This double-stranded DNA was mapped with 13 restriction enzymes. More than 40 isolates, including the virus from the original B19 serum, were compared. Although isolates could be grouped by this method, there was no correlation between a particular restriction endonuclease map and any of the several disease presentations of the virus.

Antitumour activity of purified arabinogalactan-peptidoglycan complex of the cell wall skeleton of Rhodococcus lentifragmentus.

Antitumour activity of arabinogalactan peptidoglycan (AP) complex (peptidoglycan and arabinogalactan liberated by an acid or alkaline treatment from Rhodococcus lentifragmentus AN-115 cell wall skeleton) was examined in mice and compared with that of the cell wall skeleton. The growth of syngeneic fibrosarcoma Meth A cells after implantation in BALB/c mice was significantly suppressed by AP complex, and also regressed after intratumoral injection of AP complex on days 1, 4 and 7 after tumour implantation. Although the activity of peptidoglycan was less than that of AP complex, peptidoglycan also showed both tumour-suppressive and regressive activities. Arabinogalactan did not show antitumour activity. It is interesting that peptidoglycan has an important role in the effect against tumours.

[Host mediated anti-tumor effect of Nocardia rubra cell wall skeleton on syngeneic tumor in mice].

The mode of action of Nocardia rubra cell wall skeleton (N-CWS) on Meth A fibrosarcoma (Meth A) was studied in BALB/c mice. N-CWS suppressed or regressed the intradermal growth of syngeneic Meth A cells in normal BALB/c and athymic BALB/c mice. The intradermally and subcutaneously infiltrated cells harvested from injection sites of N-CWS in normal mice showed in vitro cytotoxic activity against Meth A cells. Pretreatment of normal BALB/c mice with immunosuppressing agents such as hydrocortisone, carrageenan, or silica particles significantly reduced the anti-tumor effect of N-CWS. The growth of Meth A cells, rechallenged into BALB/c mice in which Meth A cells had once been suppressed or regressed by N-CWS treatment, was also inhibited, but not in similarly treated athymic nude mice. This resistant mechanism was shown to be dependent out cellular components but not on humoral components by the Winn Assay. The present results suggest that N-CWS exerts its anti-tumor activity by mediation of the immune system of the host and that the main effector cells in the early stage of tumor rejection are macrophages; T cells may also be involved in the later stage.

[Studies on cardiac ingredients of plants (IV). Effect of reduced proscillaridins on guinea-pig hearts].

Five hydrogenated proscillaridins (bufa-4,20-dienolide (PH21), bufa-4,20 (22)-dienolide (PH22), 20R-bufa-4-enolide (PH4-R), 20S-bufa-4-enolide (PH4-S), chola-4-enoate (PH4-E) ) were prepared semi-synthetically, and their pharmacological effect was studied using guinea pigs. The positive inotropic effect (PIE) was examined in papillary muscles isolated from guinea-pig hearts. These compounds produced PIE, dose-dependently; the Dose-PIE curve was characterized by a faster PIE development than that by the parent compound proscillaridin (PS). The concentrations at which half maximum PIE just developed (pD2) were as follows: PS, 7.4; PH22, 6.2; PH21, 5.8; PH4-R, 5.3; PH4-E, 5.0; and PH4-S, 4.9. The time required to the half maximum effect (T50) at a given concentration of pD2 was measured in min: PS, 33; PH4-E, 22; PH22, 15; PH21, 7; PH4-S, 7; and PH4-R, 2. The speeds of inotropy of these compounds depended on the position of lactone ring reduction, though they have the same lipophilicity (Rm). Concentration-PIE curves of reduced PS-compounds were characterized by a decrease in potency and an increase in efficacy. PH21 and PH4-R produced PIE over wider range of concentrations than those of the other compounds. When PH21 at the dose of 11.9 mg/kg, an equivalent dose of 4.4 times the pD2 values, was administrated intravenously by bolus injection into guinea pigs, no arrhythmias were observed on the ECG.(ABSTRACT TRUNCATED AT 250 WORDS)

A new antitumor antibiotic, FR-900482. III. Antitumor activity in transplantable experimental tumors.

FR-900482 (4-formyl-6,9-dihydroxy-14-oxa-1,11-diazatetracyclo[7.4.1.02,7, O10,12]tetradeca-2,4,6-triene-8-ylmethyl carbamate), a new antibiotic with antitumor activity was isolated from fermentation broth of Streptomyces sandaensis. Its antitumor activities were studied and compared with that of mitomycin C (MMC) in animals. FR-900482 in doses of 0.32 approximately 10 mg/kg (ip) prolonged the life of mice bearing ascitic P388, L1210, B16, MM46, Ehrlich or EL4 tumors and rats bearing ascitic AH130 or AMC60 tumors. FR-900482 in doses of 5.6 approximately 18 mg/kg (iv) inhibited human LX-1, MX-1, SC-6 and LC-6 tumors xenografted sc in nude mice. FR-900482 was more effective than or equally effective to MMC in all the tumors used. FR-900482 was ineffective against cyclophosphamide-resistant P388, but was effective against MMC- or vincristine-resistant P388. The results suggest that FR-900482 may have clinical potential.

A new antitumor antibiotic, FR-900482. IV. Hematological toxicity in mice.

The new antibiotic FR-900482 (4-formyl-6,9-dihydroxy-14-oxa-1,11-diazatetracyclo-[7.4.1.02,7. O10,12]tetradeca-2,4,6-triene-8-ylmethyl carbamate) possesses an antitumor activity equal to or greater than that of mitomycin C (MMC). The hematotoxicity of equivalent effective doses of the two compounds was compared in mice. A single iv injection of either compound similarly decreased the number of white blood cells (WBC) in the peripheral blood, whereas FR-900482 had no effect on the number of platelets (PTL). Both drugs slightly reduced the number of red blood cells (RBC). The effect of FR-900482 on the bone marrow cells (BMC) measured by cfu in spleen and cfu in culture was weaker than that of MMC. The results suggest that FR-900482 is a promising antitumor agent both in efficacy and safety.

Effect of auranofin on autoimmune disease in a mouse model.

The effect of an oral gold preparation, auranofin, on the autoimmune disease mouse MRL/l was examined. Oral administration of auranofin on consecutive days from 6 weeks of age reduced anti-DNA antibody production, IgM rheumatoid factor production, hypergammaglobulinemia, polyclonal B cell activation and renal disease, but did not prevent massive lymphadenopathy or restore the low level of either IL-2 production or mitogen response associated with 1pr gene. In contrast to the effect on autoantibody production, little suppressive activity on the immune response to exogenous antigen SRBC was observed. These results indicate that autoimmune disease in MRL/l mice can be prevented without abrogation of T cell abnormalities and that autoimmune-selective suppression can be induced by chemical compound(s) like auranofin.

[Augmentation by serrapeptase of tissue permeation by cefotiam].

Cefotiam (CTM) is a new cephalosporin with a broad spectrum of activity against both Gram-positive and Gram-negative microorganisms. Cephalosporins are widely used for prophylaxis of infections in patients undergoing thoracotomy. Augmentation by serrapeptase on tissue permeation of CTM was examined in 35 thoracotomy patients with lung cancer. The subjects were divided into two groups according to the method of the administration of CTM. Group I consisted of 17 subjects, each of whom received a single dose of 2 g of CTM alone by an instillation for 30 minutes. Group II consisted of 18 subjects, each of whom received a combination of CTM and serrapeptase; serrapeptase was given 2 tablets (10 mg) each time for three times/day until the day before surgery, and then CTM was administered by the same procedure. The following results were obtained: Individual difference was observed for the permeation of CTM into tissues. Pathologic differences also affected the permeation. Nevertheless, the CTM levels in pulmonary tissues reached about a half of those in the blood in both the single dose group and the combination group, hence sufficient concentrations exceeding MIC80 for main microorganisms that caused infections in the lung were obtained. The concentrations of CTM in inflammatory tissues have showed lower levels than those of normal tissues in both CTM single dose and the combination groups. Decrease of blood flow volume may have contributed to the reduction in levels of CTM in the inflammatory tissues. The ratio of the concentration of the drug in pulmonary tissues to that in the blood was 29.1 +/- 2.5% in the single dose group, and 44.2 +/- 6.0% in the combination group, the latter showing quite a significant increase (P less than 0.05). Combined administrations of CTM and serrapeptase deserves more trials in the case when surgical treatments of the lung are performed. An antiinflammatory effect of serrapeptase in the respiratory system is expected, and in addition, the combined use of CTM and serrapeptase should stimulate permeation of the antibiotic into tissues.
