Aliphatic chain length specificity of the polyamine transport system in ascites L1210 leukemia cells.

A series of diamine homologues of putrescine and triamine homologues of spermidine was used to determine the structural specificity of the polyamine transport system in ascites L1210 leukemia cells by measuring their ability to compete with [3H]-putrescine, [3H]spermidine, or [3H]spermine for uptake. Transport specificity among the diamines (as indicated by K1 constants) was greatest for those having chain lengths similar to that of spermidine and least for those similar to putrescine. Among the triamines, transport specificity was greatest for those having an overall chain length similar to those of spermidine and spermine. The homologue competition profiles were relatively the same for [3H]putrescine, [3H]spermidine, or [3H]spermine, suggesting that all three polyamines utilize the same transport system. This was further substantiated by uptake kinetic plots which showed that the three polyamines were competitive inhibitors of one another. In terms of receptor specificity, the ranking order among the polyamines was as follows: spermine (apparent Km, 1.6 microM) greater than spermidine (apparent Km, 2.2 microM) greater than putrescine (apparent Km, 8.5 microM). This information should prove useful in designing anticancer agents which are intended to utilize this transport system.

Wisconsin Behavior Rating Scale: measure of adaptive behavior for the developmental levels of 0 to 3 years.

The development of the Wisconsin Behavior Rating Scale was examined. We hypothesized that the extensive content of the scale as well as its arrangement of items with progressive difficulty would be effective in discriminating a rather homogeneous group of individuals functioning between 0 and 4.0 years. The scale was standardized on a sample of 325 institutionalized mentally retarded persons and a sample of 184 nonretarded infants and young children. Highly acceptable levels of reliability and validity were established for both samples. With respect to factorial validity, cognitive and psychomotor abilities accounted for approximately 83% of the explained variance. The hypothesis was supported in terms of adequate results of high concurrent and construct validity of the scale.

Canine intestinal transit and digestion of radiolabeled liver particles.

Using conscious dogs with chronic gastrointestinal fistulas, we studied the intestinal transit of radiolabeled liver particles relative to the transit of a liquid phase marker, polyethylene glycol. All studies were performed during a "fed" intestinal motility pattern induced by feeding or by intestinal perfusion with a fat-containing solution. Median transit times were determined from duodenum to fistulas at the midintestine, or at the terminal ileum, by recovery of polyethylene glycol and radiolabel in effluent from the fistulas. We also sized recovered particles of liver to determine the extent of digestion or fragmentation, or both, in transit. After a steak meal, when liquid flows were very low, the liquid marker and small (0.5-0.7 mm) and large (2-3 mm) particles moved through the proximal intestine together at a slow rate. When intestinal liquid flows were progressively increased from 2 to 24 ml/min by intestinal perfusion, transit was progressively accelerated; but 2-mm particles moved at 25%-20% of the rate of polyethylene glycol or 0.5-mm particles. Increasing transit with increasing flows was associated with decreasing digestion or fragmentation of liver particles. Under most conditions, 2-mm particles were well digested or fragmented than 0.5-mm particles. We conclude that fluid flows have a major effect on transit and digestion of food particles in the small intestine.

Three constructions of transference in Freud, 1895-1915.

Three distinct constructions of transference which have related though differentiable histories can be identified in Freud's writings. The postulation of discrete metapsychological, clinical, and universal constructions was completed by 1915, although many significant revisions within those constructions were made thereafter. Most significant is the distinction drawn between the clinical and universal constructions which (while both are founded on the metapsychological construction) facilitates recognition of cultural critical uses of transference as distinct from the clinical transference phenomena and as applicable far beyond the confines of psychopathology.

Function and fidelity of aging tRNA: in vivo acylation, analog discrimination, synthetase binding, and in vitro translation.

The liver transfer RNAs for valine and lysine were completely acylated in vivo, as judged by periodate oxidation, at 4 and 24 months of age in male Sprague-Dawley rats. In vitro acylation capacity for whole tRNA populations from rat livers is decreased, but this is evidently not deleterious in vivo. Several halogenated phenylalanines were synthesized and their effects upon acylation capacity for phenylalanine were examined. Synthetases bound to young (3 month) and old (24 month) tRNAs discriminated differently between p-chlorophenylalanine and authentic phenylalanine; synthetase with young tRNA was less able to discriminate than with old tRNA. Purified tRNAphe from old rats did not form ultraviolet-induced crosslinks to purified phenylalanyl tRNA synthetase as well as young tRNAphe. In vitro translation of encephalomyocarditis virus, hemoglobin, and ovalbumin mRNAs was effective, using tRNAs of young or old Sprague-Dawley or Fischer 344 rat livers, although, when the old tRNA was supplied, the product synthesized per unit tRNA was reduced. All of the protein products were synthesized with all tRNAs, as shown by sodium dodecyl sulfate polyacrylamide gel electrophoresis. We conclude that tRNA is capable of normal functions in livers of aging rats, is probably modification deficient, and is unlikely to produce protein errors.

Atraumatic panmural rupture of the esophagus: Boerhaave syndrome.

Three cases of spontaneous rupture of the distal esophagus are presented. All three patients presented in acute distress, exhibiting epigastric pain and signs of cardiovascular collapse. They all underwent surgery for repair of the lesion. One died postoperatively, and the other two patients recovered after a complicated postoperative period. Because Boerhaave syndrome has a high mortality rate and its diagnosis can be elusive, a high index of suspicion should be maintained by the attending physician.

Can response preparation begin before stimulus recognition finishes?

The present experiments were designed to determine whether information obtained early in the process of recognizing a stimulus can be used to begin preparing keypress responses before recognition of the stimulus has completely finished. This question is relevant to the recent debate between discrete (e.g., Sternberg, 1969a) and continuous (e.g., McClelland, 1979) models of human information processing. Stimulus sets were chosen so that recognition processes could extract incomplete preliminary information about a stimulus much faster than they could extract secondary information needed for unique stimulus identification. Discriminability of the secondary information was manipulated to vary the opportunity for response preparation based on preliminary information, with difficult secondary discriminations providing more time for response preparation than easy ones. Precues were given on some trials to allow response preparation to occur before the stimulus was presented, thereby reducing any difference in response preparation as a function of discriminability. Continuous models predict that precues should facilitate response preparation less when the secondary discrimination is difficult than when it is easy, and discrete models predict equal facilitation regardless of secondary discrimination difficulty. Evidence of response preparation was obtained with some but not all stimulus sets. The results were interpreted as support for the asynchronous discrete coding model (Miller, 1982a), in which response preparation can begin only after recognition processes have completely activated a code used in categorizing the stimulus.

Bioavailability of egg yolk iron measured by hemoglobin regeneration in anemic rats.

Biological availability of egg yolk iron and effect of egg yolk on absorption of iron from the reference salt, ferrous sulfate, were evaluated in hemoglobin repletion assays with anemic rats. Three measures of response, hemoglobin concentration, gain in hemoglobin iron content, and gain in carcass iron content were related to total dietary iron intake by regression analyses. Relative biological value (RBV) of yolk iron, in diets without ascorbic acid, compared to that of the reference salt was 85%. RBV of egg iron was equivalent to ferrous sulfate iron in diets containing 1 g ascorbic acid per kilogram of diet. Increasing increments of egg yolk in diets containing ascorbic acid had no significant effect on utilization of iron from the reference salt as measured by gain in hemoglobin or carcass iron content during the regeneration period. Stimulation of iron utilization from egg yolk by ascorbic acid was mild in these studies and was less in diets containing 32 mg/kg of egg yolk iron than in those containing 15 mg/kg of iron from egg.

Bioavailability of iron in cooked egg yolk for maintenance of hemoglobin levels in growing rats.

The relative biological value (RBV) of iron in egg yolk was compared with that of iron in ferrous sulfate in prophylactic assays with weanling rats. Egg yolk cooked by one of three different methods was used in each experiment: (1) yolk of eggs boiled in the shell, lyophilized, and mixed with other dry diet ingredients; (2) pasteurized yolk and other diet components steamed with 8 liters of water per kilogram of diet; and (3) pasteurized yolk and other diet ingredients baked with 0.5 liters of water per kilogram of diet. Diets containing three levels of egg yolk and four levels of ferrous sulfate were fed to different groups of rats for 1, 2, and 3 weeks. Response of dietary iron was calculated as the regression of hemoglobin iron gain on iron intake and the nutritional quality of egg iron relative to that of ferrous sulfate iron was evaluated by slope-ratio analysis. RBV was 61, 64, and 90%, respectively, for iron of egg yolk cooked by the three methods. The presence of ascorbic acid during heat treatment of the diets significantly increased RBV of yolk iron from 64 to 92% in experiment 2, and increased it to a value equivalent to that of ferrous sulfate in experiment 3.
