Enhancement of viral transformation for evaluation of the carcinogenic or mutagenic potential of inorganic metal salts.

Thirty-eight metal salts were tested for their capacity to enhance transformation of Syrian hamster embryo cells by a simian adenovirus, SA7. All of the metal salts with known carcinogenic potential in animals or mutagenic activity in microbial or mammalian cells increased the SA7 transformation frequency. Metals were classified into three groups according to the concentration necessary to produce significant enhancement. Those showing highest activity (positive at less than 0.05 mM) were the salts of antimony, arsenic, cadmium, chromium, and platinum. The second group (positive from 0.05 to 0.6 mM) included beryllium, cobalt, copper, lead, manganese, mercury, nickel, silver, thallium, and zinc. Iron salts were placed in a third group (only positive at concentrations greater than 0.9 mM). With the exception of ZnCl2 and ZnSO4, enhancement was demonstrated by both a relative increase in the viral transformation frequency and an absolute increase in the number of transformed foci among treated cells. The latter observation and the demonstration of enhancement in the absence of overt cell killing negate the possibility that enhancement resulted from the selection of transformation-sensitive cells.

Comparison of I-bleomycin prepared by two methods stability and pharmacokinetics in tumor-bearing mice.

Bleomycin, a chemotherapeutic antibiotic agent, was radio-iodinated by the ICI and chloramine-T methods; the radiochemical stabilities and pharmacokinetics of the two I-bleomycins in tumor-bearing mice were compared. The ICI preparation was more stable with respect to deiodination in vitro. Both products were sufficiently stable in vivo that high body background due to free isotope, a disadvantage of 111In- and 99mTc-bleomycin, was not encountered. Tumor uptake of the ICI preparation was constant from 2 to 24 hrs., and the tumor/blood ratio increased with time; with chloramine-T, this ratio decreased, and was less than that for ICI. The two preparations are chemically and biologically different; the ICI product is the superior tumor radiodiagnostic agent.

Xanthogranulomatous pyelonephritis in childhood.

We report the tenth case of xanthogranulomatous pyelonephritis, which is a rare cause of supperative proliferative pseudo tumor in children. The disease is more segmental in the child than in the adult and should be considered in the differential diagnosis of renal masses. Early recognition and conservative operation can result in a permanent cure.

Consultee-centered consultation with a teacher as a technique in behavior management.

Client-centered consultation was effective in helping a teacher to decrease one child's disruptive classroom behavior, and consultee-centered consultation helped the teacher improve her control of the class as a whole. Initially, a client-centered reinforcement program effectively decreased the disruptive behavior of one target child. Later, attention to the teacher's feelings about being the authority figure in the classroom was used in an attempt to modify the behavior of the entire class. The data describing the behavior of a second child in the classroom suggested that this consultee-centered consultation was followed by less classroom disruption. In addition, anecdotal observations indicated that consultee-centered consultation was effective in modifying the behavior of both the teacher and the rest of the class.

Auditory discrimination, intelligence, and reading achievement at grade 1.

To determine the relations of auditory discrimination and intelligence to reading achievement in first grade the California Test of Mental Maturity, the California Achievement Test (reading), and the Buktenica Modification of the Wepman Auditory Discrimination Test were administered to 78 first grade students. Correlations suggested a stronger relation between auditory discrimination and reading than between IQ and reading as measured here; however, range of reading scores were restricted. Results support Wepman's developmental theory. The methodological approach includes simultaneous consideration of effects of auditory discrimination and intelligence.

Preparation and chemical characterization of radioiodinated bleomycin.

None of the radionuclides with which bleomycin has been labeled have chemical and nuclear properties that are entirely satisfactory for in vivo tumor localization. Bleomycin has been radioiodinated by the iodine monochloride, chloramine-T, and lactoperoxidase methods. Iodine monochloride proved to be the preferred method and conditions were developed whereby 80% of radioiodide was covalently bound to bleomycin. Bleomycin (140 mug) was added to 200 mul of saline/citrate buffer (pH 7.0) followed by radioiodide and iodine monochloride. This reaction mixture was incubated for 1 hr and purified by Sephadex G-10 chromatography. The iodine monochloride reaction product underwent hydrolytic deiodination in vitro at a rate of about 1.2%/day (0.15 M NaCl, 37 degrees C). Bleomycin A and B components were radioiodinated with equal efficiency on a mole fraction basis.

The effects of infection prevention regimens on early infectious complications in marrow transplant patients: a four arm randomized study.

Three hundred and forty-two patients with hematological malignancies underwent allogeneic marrow transplantation from family donors and were allocated to receive 1) no specific infection prophylaxis in a conventional hospital room (control, 100 patients), 2) prophylactic systemic antibiotics (PSA) in a conventional hospital room (PSA group, 101 patients), 3) decontamination and isolation in a laminar air flow (LAF) room (LAF group, 65 patients) and 4) PSA in an LAF room (LAF+PSA group, 76 patients). Patients were studied for bacterial and fungal complications from the day of admission and until engraftment. LAF isolation was discontinued before engraftment in 27% (LAF+PSA group) to 32% (LAF group) of isolated in 26% (LAF+PSA group) to 27% (PSA group) of patients on prophylactic antibiotics. Septicemia occurred in 41%, 22%, 25% and 10% of patients in the control, PSA, LAF and LAF+PSA group, respectively. The incidence of septicemia was significantly less in the LAF+PSA group than in the control and LAF group with the incidence of septicemia significantly higher in the control group than in any of the other three groups. No other risk factors analyzed in proportional hazards regression tests were associated with septicemia acquisition. It is concluded that effective infection prevention modalities significantly reduce infection morbidity in transplant patients. Since most granulocytopenic transplant patients not receiving PSA will receive empiric or therapeutic broad spectrum antibiotics. The use of PSA in or out of LAF isolation is recommended as an effective modality to reduce septicemia acquisition.

Rural attitudes, opinions, and drug use.

There are two primary purposes of this paper. The first is to summarize the results of a survey conducted in a rural area of Kentucky on attitudes and behaviors regarding substance use. The second purpose is to examine differences in drug attitudes and behaviors for higher and lower density rural areas. Participants for this study were contacted by telephone in March 1996. The sample was generated by random digit dialing for eight county telephone exchanges. The sample included 334 respondents from the eight counties who were grouped into respondents from higher density areas (n = 132) and lower density areas (n = 202). Results indicated that respondents from both higher density and lower density areas were similar on drug use and attitudes. However, lower density areas reported their county as a worse place to live and less safe than respondents from higher density areas. Respondents from lower density areas were also more likely to report there was more alcohol/drug use and more alcohol bootlegging in the past year, and that bootlegging alcohol is a serious problem in their county. Future research could include the examination of heterogeneity of rural areas using different indices.
