Diagnosis of "Maladie des feuilles cassantes" or Brittle leaf disease of date palms by detection of associated chloroplast encoded double stranded RNAs.

The "Maladie des feuilles cassantes" (MFC) or "Brittle leaf disease" of date palms is associated with the accumulation of two populations of small, chloroplast-encoded RNAs. A plasmid vector containing a cDNA with partial sequences of both of these RNA populations was used to synthesize a DIG-labeled bifunctional probe by PCR. The probe has been tested to detect, by molecular hybridization, MFC-associated RNAs from dsRNA-enriched palm leaflet preparations. Leaflet samples from MFC-affected date palm trees consistently gave a positive hybridization signal regardless of the date palm cultivar, severity of symptoms, or geographical location, whereas samples from date palm trees affected by other biotic and abiotic stresses tested negative. The assay is specific for MFC and can be used for early diagnostic purposes.

[Hypothyroidism and myocardial damage in cardiac donors].

BACKGROUND AND OBJECTIVES: Brain death may induce myocardial contractile dysfunction and hypothyroidism. This myocardial dysfunction is a limiting factor when considering donor heart for transplantation. Myocardial damage has shown to be the mechanism in which contractile dysfunction is established in donors. On the other hand, hypothyroidism has been evoked as a major cause of cardiovascular disorders in brain dead subjects. It is not known whether any relationship exists between hypothyroidism and myocardial damage that appear in brain death. In this study myocardial damage and thyroid status are quantified in a donor population, and whether a correlation between hypothyroidism and myocardial damage exists is analyzed. PATIENTS AND METHOD: Serum samples from 45 cardiac donors were examined for cardiac troponins T and I as myocardial damage markers, as well as thyroid hormones. Correlations between cardiac troponins and thyroid hormones were assessed. RESULTS: One or both cardiac troponins were elevated in 19 donors (42.2%). Hypothyroidism was a very frequent finding, since it was present in 39 donors (86.7%). No correlation was observed between any of the myocardial damage markers and thriiodothyronine levels. CONCLUSIONS: Hypothyroidism and myocardial damage are common in brain dead donors. No association exists between both disorders.

Modulation of the cellular immune response by a carbohydrate rich fraction from Echinococcus granulosus protoscoleces in infected or immunized Balb/c mice.

Infection of Balb/c mice with Echinococcus granulosus protoscoleces constitutes the model for secondary hydatid infection. The immune response of Balb/c mice infected with E. granulosus is characterized by secretion of antibodies specific for carbohydrate epitopes and production of type-2 cytokines. A role for glycoconjugates in the induction of type-2 responses has been suggested in other host--parasite systems. Although glycoconjugates are immunogenic in E. granulosus infection, the role of these molecules in the establishment of the type-2 response has never been analysed. In this study, a carbohydrate rich fraction (E4+) from E. granulosus protoscoleces was obtained using the monoclonal antibody E492/G1 specific for the moiety Galalpha(1,4)Gal which is widely represented in protoscoleces and other E. granulosus antigenic preparations. The results showed that E4+ was immunogenic in Balb/c mice evoking an antibody response mainly directed against carbohydrate epitopes. In addition, splenocytes from E4+-immunized mice showed suppressed proliferative responses to Con A and E4+ induced IL-10 secretion by E4+-primed and naive splenocytes. The fraction E4+ also was immunogenic in infected mice during early infection. In this case also, splenocytes from infected mice as well as peritoneal cells from infected or naive mice, when stimulated in vitro with E4+, secreted IL-10. Collectively, these results suggest that E4+ may be involved in immunosuppression phenomena and, by stimulating IL-10 secretion, may contribute to the induction and sustaining of the type-2 cytokine response established in early experimental infection.
