Chemotherapy of human non-Hodgkin lymphoma (NHL) xenografts.

Three human NHL (B-cell type) were established successfully as serially transplantable xenografts in artificially immunesuppressed CBA mice. All of them preserved the phenotypic characteristics of the original tumour even after several passages. The transplanted tumours were highly sensitive to cyclophosphamide and methotrexate, reflecting the results obtained in clinical practice, and relatively sensitive to dianhydrogalactitol.

Increased SCE inducibility by low doses of methylcholanthrene in lymphocytes obtained from patients with Down's disease.

The inducibility of sister-chromatid exchanges (SCEs) following 3-methylcholanthrene (MC) treatment of normal human peripheral blood lymphocytes and of in vitro cultured normal human embryo fibroblasts, as well as peripheral blood lymphocytes of patients with Down's disease and of fibroblasts of an embryo with trisomy 21 has been investigated. 10(-6) M MC treatment increased the frequency of SCEs by 2.5 in the case of Down lymphocytes when compared to the healthy control. The fibroblasts with trisomy 21, however, did not show an increased sensitivity to MC treatment when compared with normal fibroblasts, expressed in the number of SCEs per nucleus found in the cells.

Bombesin-like immunoreactivity in the pituitary gland.

Bombesin has been shown to stimulate release of anterior pituitary hormones both in vivo and in vitro. The aim of this study was to determine whether bombesin-like immunoreactivity could be detected in the human pituitary. Significant concentrations were found in the human anterior gland (4.6 +/- 1.5 pmoles/g), posterior gland (1.5 +/- 0.4 pmoles/g) and stalk (8.1 +/- 0.8 pmoles/g). Significant amounts were also observed in the guinea-pig pituitary. Gel permeation chromatography revealed the presence of 2 major molecular forms of bombesin-like immunoreactivity, one co-eluting with porcine gastrin-releasing peptide and the other with amphibian bombesin.

Measurement of IgG antibody and airborne antigen to control an industrial outbreak of hypersensitivity pneumonitis.

An outbreak of hypersensitivity pneumonitis in a textile plant was attributed on epidemiological grounds to a chilled water air-conditioning system where a slime was growing in the chilled water sump and on demister vanes. All of the patients and about 80% of their asymptomatic co-workers had strongly positive precipitin tests to extracts of the slime. An adaptation of a radioiodinated staphylococcal protein A solid-phase radioimmunoassay was applied to monitor antigen and specific antibody. To control the outbreak, a variety of cleaning and water treatment measures were taken between 1977 and 1979 to reduce the amount of antigen in the water and in the air. The amount of slime in the water was greatly reduced though the antigen content per gram of slime did not change. Airborne antigen in the affected work areas decreased progressively.

[New type of combined surgical treatment of frontobasal injuries (author's transl)].

The surgical treatment of frontobasal injuries by a new, combined method has been reported. The frontobasal injury was exposed by bifrontal craniotomy. After the intradural examination and the dressing of the brain injuries the dural defect was also extradurally searched and dressed. Thereafter--taking the advantage of the bifrontal exploration--the paranasal sinus (frontal sinus, ethmoideal system) was cleaned, then the base of the anterior scala was covered with pedicled galea flap or periosteum plate. In case of the treatment of the open injuries, following the removal of the bone fragments, the dural brain injuries and the paranasal sinus injuries were similarly treated, according to the above mentioned principles. This method makes possible to prevent the liquorrhoeic complications as well as the delayed septic complications of the paranasal sinus. The method is recommended to the operative treating of the frontobasal injuries after many years of favourable experiences and supported by good follow-up results.

Characterization of an adenylate cyclase system sensitive to histamine H2-receptor excitation in cells from dog gastric mucosa.

A method of preparing viable cells from dog gastric mucosa is described. Cyclic AMP in these cells is elevated by histamine and 4-methyl histamine but 2-methyl histamine is only a weak agonist. The effects on cyclic AMP levels are inhibited competitively by metiamide and burimamide which give apparent KBvalues of 3.5x10-7 M and 2.3x10-6 M, respectively. These values are similar to those reported for other histamine H2-receptor systems. The H1-receptor antagonists, mepyramine and chlorpheniramine, have no inhibitory effect on the histamine induced elevation of cyclic AMP: promethazine inhibits the system but not by a competitive mechanism. It is concluded that the histamine stimulated adenylate cyclase system is probably located in the parietal cell component.

A quantitative immunocytochemical approach to the analysis of type I cells in the cat carotid body.

Digital image analysis of immunostained semithin plastic sections indicates that experimentally induced changes in levels of transmitter-related reaction product in single cells fails to support the concept of clearly defined subsets of type I cells in the carotid body. This objective approach to the quantitation of staining product on a cell-by-cell basis appears to indicate that the observed changes are related to global shifts in the expression of a given neuronal marker throughout a single population of highly labile chemoreceptor elements.

Genotoxic effects of occupational exposure in the peripheral blood lymphocytes of pesticide preparing workers in Hungary.

Venous blood samples of 240 donors including 33 industrial and 60 historical controls were investigated in order to assess the genotoxic risk of pesticide preparing workers manufacturing monochlorinated benzene in Hungary. Mutation frequencies were determined in the hypoxanthine-(guanine)-phosphoribosyl transferase genes located on the X chromosome. DNA repair capacity was estimated following hydroxyurea treatment with subsequent UV irradiation of separated lymphocytes. Smoking as confounding factor of genotoxicity was also taken into consideration. Mutation frequencies were increased among the workers exposed to monochlorinated benzene in correlation with the duration of working time, compared to the controls. Mutation frequencies were lower than expected among non-smoker, long-exposed workers. Smoking itself proved to be an effective confounding factor in the enhancement of point mutations in the case of long-exposed workers. Smoking, however, caused no significant increase in the mutation frequency among the controls, and did not influence the DNA repair capacity of any of the groups.

Transplantable human non-Hodgkin lymphoma line in artificially immunesuppressed mice.

Diffuse non-Hodgkin lymphoma of B-cell origin has been established as a serially transplantable xenograft line in artificially immunesuppressed mice. The take rate and growth rate increased with repeated passages compared to the first transplant generation. However the xenografted tumor preserved many of its characteristics, including morphology, cell surface markers and DNA index. Chromosome analysis proved the human origin of tumors grown in mice and revealed translocation 8,14. Cyclophosphamide, Methotrexate and Vincristine produced substantial inhibition of tumor growth, while Dianhydrogalactitol and Adriamycin were less effective. Human alpha interferon also produced a delay in tumor growth.
