Effect of structural modification on photodynamic activity of hypocrellins.

Aluminum ion complexed 5,8-di-Br-hypocrellin B is a new water-soluble perylenequinonoid derivative with enhanced absorption over hypocrellin B (HB) in the phototherapeutic window (600-900 nm). Electron paramagnetic resonance and 9,10-diphenyl-anthracene bleaching methods were used to investigate the photosensitizing activity of [AL2(5,8-di-Br-HB)Cl4]n in the presence of oxygen. Singlet oxygen, superoxide anion radical and hydroxyl radical can be generated by [AL2(5,8-di-Br-HB)CL4]n photosensitization. Singlet oxygen (1O2) is formed via energy transfer from triplet-state [AL2(5,8-di-Br-HB)CL4]n to ground-state molecular oxygen. 1O2 participates in the generation of a portion of superoxide anion radical (O2.-). Besides superoxide anion radical (O2.-) may originate from the electron transfer between the triplet-state [AL2(5,8-di-Br-HB)CL4]n and the ground-state molecular oxygen. OH is formed through the Fenton-Haber-Weiss reaction and the decomposition of DMPO-1O2 adduct. Compared with HB [AL2(5,8-di-Br-HB)CL4]n primarily remains and enhances the generation efficiency of superoxide anion radical and hydroxyl radical but that of singlet oxygen decreases.

Fast three-point dixon MR imaging using low-resolution images for phase correction: a comparison with chemical shift selective fat suppression for pediatric musculoskeletal imaging.

OBJECTIVE: The purpose of this study is to describe and to implement a new fast three-point Dixon MR imaging sequence with online image reconstruction, and to compare this sequence with conventional chemical shift selective (CHESS) suppression of fat in pediatric musculoskeletal imaging. SUBJECTS AND METHODS: A three-point Dixon technique using a fast spin-echo sequence with a new phase-correction algorithm providing online image reconstruction was implemented on a 1.5-T scanner. Twelve pediatric patients and young adults were imaged with both the new three-point Dixon and conventional CHESS sequences. Three radiologists un-aware of imaging parameters and clinical information independently scored the homogeneity of fat suppression and conspicuity of abnormality using a four-point system. An additional comparison between the two techniques was made using a phantom. RESULTS: The three-point Dixon method showed superior fat suppression and lesion conspicuity (p < 0.001), particularly in the hands and feet, where CHESS is prone to inconsistent fat suppression. The phantom study showed no significant difference in the ratio of suppressed fat signal to background noise and more homogeneous fat suppression using the three-point Dixon method. CONCLUSION: Compared with CHESS, the new fast three-point Dixon sequence with online image reconstruction provides superior fat suppression and lesion conspicuity and can be routinely used in pediatric musculoskeletal imaging.

[Mitochondrial DNA sequence variations of Zhuang ethnic group in Guangxi].

The mtDNA hypervariable segment I sequences (515bp) were sequenced in 83 Zhuangs from Gunngxi Province, with the aim to learn more about the origin and genetic structure of the current Zhuangs. 66 haplotypes were identified in the samples, with 71 sites showing polymorphism. Phylogenetic analysis of the 66 haplotypes suggests that there are geographic differentiation in current Zhuangs, and those from the 4 geographic regions (Nanning, Hechi, Baise and Liuzhou) have different distribution frequencies in the cluster I, II and III in the tree. More than 50% individuals from Liuzhou and Hechi converge into cluster II, while those from Nanning and Baise have high frequency in cluster I. Combined with the analysis of the reported data, the Zhuang ethnic group shows remote affinity to those from North China, whereas it is close to those in South China. The frequencies of the radiation groups in Zhuangs, together with the phylogenetic relationship of the Zhuang ethnic group in the tree suggest that the Zhuang is a typical south population.

Expression, purification, and MALDI analysis of RPE65.

PURPOSE: RPE65 is preferentially expressed in the retinal pigment epithelium (RPE) and is essential for retinal function. The purpose of the study was to develop methods for the expression of the protein, determine the accurate molecular weight of this expressed protein, and quantitate the amount of RPE65 in the bovine RPE. METHODS: Human RPE65 was expressed in Sf9 cells using the baculovirus system. The subcellular localization was determined by Western blot analysis and immunocytochemistry. An ELISA was developed for RPE65 and used to measure levels in bovine RPE. Recombinant and native RPE65 were purified by affinity chromatography. Molecular mass was determined by matrix-assisted laser desorption/ionization (MALDI) mass spectrometry. RESULTS: Recombinant human (rH)RPE65 was expressed as a major protein associated with cell membrane in Sf9 cells. The recombinant protein was purified to apparent homogeneity from both the membrane and nonmembrane fractions. The identity of the purified protein was confirmed by Western blot analysis and by partial peptide sequencing. rHRPE65 from the nonmembrane fraction has a mass of 64,867 +/- 80 which is close to the calculated molecular weight from the amino acid sequence including the His-tag (64,663), whereas the membrane-associated rHRPE65 has a molecular mass of 65,380 +/- 150, which is significantly higher than that of the non-membrane-associated form and the calculated molecular weight, suggesting posttranslational modifications. Similarly, native RPE65 was detected in the cytosolic and microsomal fractions of the bovine RPE, with an average level of 3.8 +/- 1.3 and 7.2 +/- 0.4 microg RPE65 per eye, respectively. The cytosolic form had a molecular mass of 61,161 +/- 60, which is close to the calculated value (60,944), whereas that of the microsomal form was 61,961 +/- 170. CONCLUSIONS: RPE65 is expressed in two forms, one of which is membrane associated and contains significant posttranslational modifications, similar to the native membrane-associated form.

Peripheral effector mechanism hypothesis of postflight cardiovascular dysfunction.

Studies on the mechanisms of cardiovascular dysfunction after space-flight are important to illustrate the cardiovascular effect of microgravity and develop appropriate multi-system countermeasures for future long-duration spaceflights. Over the past 10 yr, we have systematically studied the adaptational changes in structure and function of both the heart and vessels, using the tail-suspension rat model to simulate microgravity effects. Our results indicate that simulated microgravity induced atrophic changes and reduced contractility of the heart muscle, and upward- and downward-regulation in structure, function, and innervation state of vessels in the brain and hind body of the rat. In addition, more recent advances in relevant ground-based and space-flight studies from different laboratories have also been reviewed. Based on these studies, it has been speculated that, in addition to hypovolemia, the microgravity-induced adaptational changes in the structure and function of the two main effectors of the cardiovascular system, i.e., the arterial smooth muscle and the cardiac muscle, might be among the most important mechanisms responsible for postflight cardiovascular dysfunction and orthostatic intolerance. In this paper we will review the available evidence with comments.

[Experimental and mechanism analyses on HCl emission control during PVC combustion in fixed beds].

Incineration of chloridated plastic (PVC) in solid wastes contributes much to HCl pollutant in air environment. The characteristics of HCl emissions and reduction during PVC combustion were conducted in a nearly isothermal, externally heated, horizontal quartz-tube reactor. With increasing temperature from 700 to 900 degrees C, the conversion of HCl increased leniently from 82.5% to 88.3%. High excess air factor enhanced a small amount of HCl existing in form of Cl2. The calcium-based sorbents including CaCO3, Ca(OH)2, Ca(CH3COO)2 exhibited high HCl capture efficiencies from 68% to 79%, while magnesium-based sorbent less than that of 3%. Influences of operational variables, i.e., size of sorbent, Ca/Cl molar ratio and combustion fume compositions, on the HCl reduction were discussed. From a point of view of equilibrium constant of dechloridization reaction the experimental results were explained in detail.

The relationship between a polymorphism in CYP17 with plasma hormone levels and prostate cancer.

The A2 allele of the CYP17 gene has been thought to be associated with increased functional activity of this steroidogenic enzyme. Consequently, the A2 allele has been examined as a biomarker of individual susceptibility to hormone-related diseases among men and women. We prospectively assessed the association between the A2 allele of CYP17 and prostate cancer risk among 590 cases and 782 controls in a case-control study nested within the Physicians' Health Study cohort. We also evaluated associations between CYP17 genotype and plasma steroid hormones among controls and the potential interaction between CYP17 and SRD5A2 V89L polymorphisms in relationship with prostate cancer risk and circulating steroid hormone levels. We observed a borderline significant association between the A2 allele and prostate cancer risk (odds ratio, 1.23; 95% confidence interval, 0.99-1.54), however, we did not observe evidence of a gene-dosage effect (versus A1/A1 genotype: A1/A2 genotype; odds ratio, 1.26; 95% confidence interval, 0.99-1.59; A2/A2 genotype: odds ratio, 1.17; 95% confidence interval, 0.85-1.61). The A2 allele was not overrepresented among cases with advanced prostate cancer. Among controls, carriers of the A2 allele had steroid hormone levels similar to noncarriers. We also found no evidence of a gene-gene interaction between CYP17 and SRD5A2 V89L polymorphisms on prostate cancer risk or endogenous steroid hormone levels. These results suggest that CYP17 genotype may possibly confer a small increased susceptibility to prostate cancer but is not a strong predictor of endogenous steroid hormone levels in men.

[The development of multi-lead EEG telemetry and tele-monitoring system].

This paper introduces a kind of EEG Telemetry and Tele-monitoring system. With the use of the public telephone network to transport signals and the portable EEG recording system as the sender of system, the EEG data of 20 standard leads can be transmitted to the receiver of system (monitoring center) synchronously. And at the monitoring center, the waves can be displayed real-time, freezing, stored, reviewed, analyzed and printed, thus providing aid in remote diagnosis and treatment consultations.

Time course of pulmonary response of rats to inhalation of crystalline silica: histological results and biochemical indices of damage, lipidosis, and fibrosis.

Previous studies have determined that alpha-quartz (crystalline silica) can cause pulmonary inflammation, damage, and fibrosis. However, the temporal relationship between silica inhalation and pulmonary inflammation, damage, and fibrosis has not been fully examined. To address this gap in our knowledge of silica-induced pulmonary fibrosis, a chronic inhalation study using rats was designed. Specifically, rats were exposed to a silica aerosol (15 mg/m3 silica, 6 h/d, 5 d/wk, 116 d), and measurements of pulmonary inflammation, damage, and fibrosis were monitored throughout the study. We report (1) data demonstrating that the silica aerosol generation and exposure system produced a consistent silica aerosol of respirable size particles; (2) the time course of silica deposition in the lung; (3) calculations that demonstrate that the rats were not in pulmonary overload; (4) histopathological data demonstrating time-dependent enhancement of silica-induced alveolitis, epithelial hypertrophy and hyperplasia, alveolar lipoproteinosis, and pulmonary fibrosis in the absence of overload; and (5) biochemical data documenting the development of lipidosis, lung damage, and fibrosis.

[Analysis of several genes using primer extension preamplification based on individual cell].

OBJECTIVE: To analyze several genes at the same time in one individual cell using primer extension preamplification (PEP). METHODS: The whole genome of individual cell was preamplified by PEP using 10 base random oligonucleotides. The aliquots(5 mu liter) from PEP were used to amplify SRY, ZP(3), RhCE, RhD and F VIII genes by nested PCR. RESULTS: The five genes could be amplified following PEP-PCR. Lymphocyte of four genotypes were diagnosed by PEP-PCR; correct diagnoses were obtained in 95.6%. CONCLUSION: At least five genes can be analyzed by PEP-PCR based on one individual cell in this laboratory. This method could be used in preimplantation genetic diagnosis and noninvasive prenatal diagnosis.

Fast three-point Dixon MR imaging of the retrobulbar space with low-resolution images for phase correction: comparison with fast spin-echo inversion recovery imaging.

A new phase-correction algorithm for three-point Dixon (3PD) MR imaging allows on-line image reconstruction of three images per section: pure water, pure fat, and water plus fat. When combined with fast spin-echo acquisition, the sequence is suitable for routine MR imaging of the retrobulbar space. The 3PD pure water images have double the image signal-to-noise ratio of fast spin-echo inversion recovery images. The dramatic contrast-to-noise ratio of the 3PD pure fat images may offer improved lesion detection.

Telomerase activity and homozygous deletions of the p16 gene in liver metastases of colorectal carcinoma.

OBJECTIVE: To study the interaction between telomerase activity and abnormalities of the p16 gene in liver metastases of colorectal carcinoma. METHODS: Telomerase activity was detected by a non-isotopic PCR-based telomeric repeat amplification protocol (TRAP) assay, and homozygous deletions of the p16 gene were detected by a semiquantitative multiplex polymerase chain reaction in tissue samples from 24 liver metastases of colorectal carcinoma and 5 primary colorectal carcinomas. RESULTS: Telomerase activity was observed in 19 (79.2%) of 24 liver metastases of colorectal carcinoma. Telomerase activity was also observed in all 5 primary colorectal carcinomas and in 3 of their liver metastatic samples. The incidence of telomerase activity in liver metastases of colorectal carcinoma was not significantly correlated to tumor diameter, number of tumors, cirrhosis, and HBsAg. Homozygous deletions of the p16 gene were found in 9 of 24 (37.5%) liver metastases of colorectal carcinoma. Homozygous deletions of the p16 gene were observed in 2 of the 5 primary colorectal carcinomas and in 1 of the matching liver metastatic cancers. There was a correlation between telomerase activity and homozygous deletions of the p16 gene. CONCLUSIONS: There is a correlation between telomerase activity and homozygous deletions of the p16 gene in liver metastases of colorectal carcinoma, suggesting its crucial role in liver metastases. However, telomerase activation and homozygous deletions of the p16 gene might not be the initiating event in liver metastases of colorectal carcinoma.

[Counteraction effect of intermittent -Gx on changes of responsiveness of basilar artery in tail-suspended rats].

OBJECTIVE: To investigate counteracting effects of intermittent -Gx exposure on changes of responsiveness of basilar artery in tall-suspended rats. METHOD: Rats tail-suspended for 3 wk were daily subjected to 1 h 1.5 G or 2.6 G centrifugation, simulating -Gx exposure of various G values. Responses of perfused isolated basilar arterial rings to KCl was examined. RESULT: Maximal isometric contractile response to KCl were significantly enhanced in arterial rings isolated from 3 wk tail-suspended rats as compared with that from simultaneous control rats (P < 0. 05). However, maximal isometric contractile responses to KCl of arterial rings isolated from 3 wk tail-suspended plus daily -Gx exposure rats were no difference as compared with that from simultaneous control rats, and were significantly decreased (P < 0.05) as compared with that from 3 wk tail-suspended rats. CONCLUSION: Daily intermittent -Gx exposure as a countermeasure to prevent the effects of simulated weightless on changes in responsiveness of basilar artery from tail-suspended rats was valid.

[Pathological observation of 16 cases with non-epithelial tumor and tumor-like lesions of the prostate].

OBJECTIVE: To investigate the diagnosis, differential diagnosis and prognosis of stromal tumors and tumor-like lesions of the prostate. METHODS: By means of HE, immunohistochemistry and electron microscopy, the clinical and histological data of 16 cases with stromal tumors and tumor-like lesions of the prostate were observed. Some of them were followed-up. RESULTS: Of the 16 cases, 6 presented stromal nodular hyperplasia, 3 were stromal sarcoma, 1 malignant phyllodes-type tumor, 3 leiomyosarcoma, 1 fibrosarcoma, and one non-Hodgkin's lymphoma. CONCLUSIONS: Nearly all the leiomyomas of the prostate diagnosed previously are, in fact, stromal nodular hyperplasia. The phyllodes-type tumors are often confused with stromal sarcomas in needle biopsy specimens and cause difficulty in differential diagnosis. Diagnostic criteria of leiomyosarcoma, fibrosarcoma, non-Hodgkin's lymphoma of the prostate are the same as their counterparts in the soft tissue. Non-epithelial malignant tumors in prostate are of poor prognosis.

[Dynamic surveillance of risk behaviors facilitating sexually transmitted disease/acquired immunodeficiency syndrome transmission among permanent residents in Kunming city in 1996-1999].

OBJECTIVE: To monitor the trends of risk behaviors promoting sexually transmitted disease/Acquired immunodeficiency syndrome (STD/AIDS) transmission among general public in order to develop appropriate intervention strategies and to evaluate impact of intervention programs. METHODS: Two hundred subjects were randomly selected and interviewed every other month in Kunming city from 1996 to 1999. STD-AIDS-related information on knowledge, attitudes and behavior were collected during the interview. RESULTS: The knowledge of STD and AIDS transmission increased from 49.2% to 62.8% (chi 2 = 96.5, P < 0.01) and from 16.6% to 35.2% (chi 2 = 146.5, P < 0.01), respectively. Knowledge on condom use and attitude to AIDS did not change much. Self-reported prevalence of risk behaviors remained low. Residents aged between 15 and 39 had unprotected sex behavior (0.8%-2.5%) and drug use (0.3%-2.4%). CONCLUSION: The awareness on STD and AIDS transmission increased gradually. Risk behavior of STD/HIV infection existed among residents aged between 15 and 39 years, which called for intervention programs in the future.

[A case-control study on the risk factors of stomach cancer in Tianjin city].

OBJECTIVE: To explore the risk factors of stomach cancer in Tianjin. METHODS: A matched case-controls study was carried out. A total number of 189 patients with stomach cancer who were individually matched and interviewed. RESULTS: Multivariate conditional logistic analysis showed that stomach cancer was closely relate to the four factors: smoked food (OR = 2.34, 95% CI: 1.60-4.98), cigarettes smoking (OR = 6.07, 95% CI: 1.26-7.16), heavy salt intake (OR = 1.95, 95% CI: 1.27-3.23) and excessive intake of meats (OR = 1.46, 95% CI: 1.05-2.02) were risk factors for stomach cancer. CONCLUSION: Frequent eating smoked food, cigarettes smoking, heavy salt intake and over intake of meats were risk factors for stomach cancer.
