Short estrous cycles and associated progesterone in serum of beef heifers aborted at various stages of gestation.

Estrous cycles were observed and concentrations of progesterone were measured after abortion in crossbred Angus heifers that were injected with a prostaglandin F(2) alpha analogue about day 50 (15 heifers), 75 (14 heifers), 100 (13 heifers) or 125 (49 heifers) of gestation. The average interval from injection to first estrus was 3.6, 4.2, 3.7 and 6.4 d for heifers injected at an average of 50, 75, 100 or 125 d of gestation, respectively. Cycles of fewer than 13 d were classified as short, those of 13 to 16 d as intermediate, those of 16 to 24 d as normal, and those of more than 24 d as long. Heifers aborted at 50 d of gestation had seven short and eight normal first cycles; heifers aborted at 75 d had 11 short cycles, two normal cycles, and one long cycle; heifers aborted at 100 d had 12 short cycles and one normal cycle; whereas heifers aborted at 125 d had 25 short cycles, 13 normal cycles, two long cycles, six intermediate cycles. Three heifers failed to exhibit a second estrus by 50 d after abortion. Of those heifers having short first cycles, short second cycles occurred in one of seven heifers aborted at Day 50, three of 11 at Day 75, seven of 12 at Day 100 and 18 of 25 at Day 125. Serum progesterone was measured at 0, 1 and 2 d after injection and at 5 and 10 d after the first estrus. Concentration of progesterone in serum declined before estrus, then increased by 5 d after estrus in all cases. Progesterone concentration increased between 5 and 10 d after estrus in heifers exhibiting normal estrous cycles but docreased in heifers having short cycles. Data indicate that the percentage of heifers having single or repeated short cycles after abortion increases as the duration of gestation increases to 100 d.

Influence of environmental temperature on reproductive performance of bovine embryo donors and recipients in the southwest region of the United States.

Superovulation and embryo transfer records on performance of embryo donor (n = 3,908; beef 99%, dairy 1%) and recipient (n = 19,936; beef 92%, dairy 8%) cattle from a commercial transfer unit were analyzed for environmental effects. Embryos (n = 42,428) were recovered on Days 5 to 8 postestrus from superovulated donors. Numbers of ova, fertilized ova and embryos of transferable quality were recorded. Transferable embryos were classified according to stage of development and morphological quality. Embryos (n = 19,936) were transferred nonsurgically. Responses were modeled with maximum-likelihood procedures using log-linear functions of independent variables and ANOVA. Fluctuations in daily maximum temperature (1 to 43 degrees C), for Days 0 to 7 of embryo development, had no effect on distribution of embryos classified as good (48%), fair (40%) and poor (12%). Temperature did not affect the percentage of donors flushed with recoverable ova (89%), mean number of ova (12.2 +/- 0.3), fertilization rate (76%) or percentage of transferable embryos (57%). Recipient pregnancy rate (56%) was not affected by mean maximum temperature for Days 0 to 10 posttransfer. Interactions between temperature and breed type (dairy vs beef), parity (cow vs heifer), or lactational status (lactating vs dry) on pregnancy rate were recorded. Elevated environmental temperature does not appear to adversely affect reproductive responses of donor and recipient cattle intensely managed in a commercial transfer unit.

Reproductive characteristics of cyclic beef heifers treated with the prostaglandin analog luprostiol.

One hundred and twenty crossbred Angus heifers, after exhibiting a 17- to 23-d estrous cycle, were placed into six groups of 20 heifers each and administered 2 ml i.m. propylene glycol containing either 0 (controls), 3.75, 7.5, 15.0 or 30.0 mg of luprostiol, or saline containing 0.5 mg cloprostenol (Groups 1 through 6, respectively). Heifers were observed for estrus every 6 h and all treatments were given 6.5 to 8.0 d after heifers were observed in standing estrus. Blood samples were collected after treatments from 10 heifers in each groups. Blood serum was assayed for progesterone. The synchronization period was considered to be 120 h after administration of luprostiol or cloprostenol. There were 0, 16, 17, 18, 20 and 18 heifers observed in estrus during the synchronization period in Groups 1 through 6, respectively. Progesterone concentrations in blood serum dropped below 1 ng/ml in 0, 8, 9, 10, 10 and 10 of the heifers from which blood samples had been taken in the six groups. All heifers observed in estrus were artificially inseminated. During the synchronization period, 0, 12, 14, 15, 16 and 10 heifers conceived in Groups 1 through 6, respectively. The interval from injection to estrus for the 89 heifers that exhibited estrus in the synchronization period averaged 49.0 h and was not different among the luprostiol and cloprostenol treated groups. Control heifers returned to estrus an average of 13.2 d after the treatment. The number of heifers that conceived at first insemination, regardless of when estrus occurred, was 16, 15, 16, 16, 16 and 12, and the total number that conceived at the first and second inseminations was 18, 18, 17, 19, 19 and 16 for Groups 1 through 6, respectively. Based on serum progesterone concentration and/or interval from treatment to estrus, 15 and 30 mg of luprostiol effectively regressed corpora lutea (100%) when administered between 6.5 and 8.0 d after estrus, and the estrous response and conception rate for these two groups equalled or exceeded that of the control and cloprostenol groups.

Pseudodementia in schizophrenia and mania.

The importance of differentiating depressive pseudodementia from true dementia seems to be well recognized by psychiatrists, yet there seems less recognition that other functional psychoses in the elderly can present with symptoms of cognitive impairment. In this report, we describe two patients--one suffering from late onset schizophrenia and one from a manic illness--who were diagnosed as suffering from dementia in the early stages of their illnesses. We discuss some of the difficulties in making diagnoses in these cases which include an historical tendency for clinicians, including psychiatrists, to assume an organic aetiology for first presentation psychoses in old age. Although the concept of pseudodementia has been criticised in recent years, we conclude that it retains its clinical utility in that it orientates the clinician to the importance of recognizing treatable functional psychoses in the elderly.

Intraoral melanoacanthoma: a reactive melanocytic hyperplasia. Case report.

Melanoacanthoma was originally described as a benign skin tumor of keratinocytes and dendritic melanocytes. There is now evidence that the intraoral lesions are unlike those occurring on skin. This article documents the reactive and reversible nature of the oral condition in a patient with these lesions.

Solitary eosinophilic granuloma (histiocytosis X) of the gingiva. A report of two cases.

Two cases are reported in which the patients presented with solitary ulcers on the gingiva. The lesions were excised and examined histologically. A connective tissue infiltrate of large mononuclear cells was identified with immunohistochemical staining as Langerhans cells and a diagnosis of histiocytosis X was made. Further medical evaluation revealed no other lesions in one of the patients but did locate an infiltrate in the right mastoid of the other patient. Solitary lesions of histiocytosis in oral mucosa are extremely rare. The manifestations of the disease, pathogenesis, and treatment are discussed.

Factors associated with shortened estrous cycles after abortion in beef heifers.

Forty-six crossbred Hereford heifers were allotted into five experimental groups. Fenprotalene, a prostaglandin analogue, was administered to induce abortion and (or) to regress the corpus luteum (CL) in four groups of heifers about 75 d after conception. The four groups were 1) pregnant heifers, 2) pregnant heifers with the ovary contralateral to the gravid uterine horn removed 24 h after fenprostalene injection, 3) same as Group 2 but with the ipsilateral, rather than the contralateral, ovary removed and 4) heifers with uterus removed (ovaries intact) about 35 d after conception. A fifth group of nonpregnant heifers received implants containing norgestomet from 10 to 76 d after estrus and were given fenprostalene 24 h before removal of the implant. Average length of the first estrous cycle after the fenprostalene injection was 11.2, 8.3, 9.7 and 19.1 for Groups 1, 2, 3 and 5, respectively. Estrous cycles were longer (P less than .05) in norgestomet-treated heifers. Hysterectomized heifers (Group 4) did not exhibit a second estrus by 50 d after fenprostalene treatment; otherwise, all first estrous cycles after the fenprostalene injection were either "short" (7 to 13 d) or "normal" (17 to 23 d). Twenty-two of 24 heifers that aborted had short estrous cycles and two had normal estrous cycles. Short estrous cycles occurred after abortion regardless of whether the ovary ipsilateral or contralateral to the previously gravid uterine horn ovulated. Early regression of the CL (short luteal phase) did not occur in nonpregnant heifers after long-term progestogen stimulation or after hysterectomy, but it did occur in heifers with a previously gravid uterus.(ABSTRACT TRUNCATED AT 250 WORDS)

Treatment of adult lower extremity lymphedema with the Wright linear pump: statistical analysis of a clinical trial.

Seventy-three adult lymphedema patients with 78 affected lower extremities were treated with the Wright Linear Pump. All patients were admitted for a 48-hour clinical trial. Comparison of circumferential limb measurements before and after a 48-hour treatment period using a repeated measures analysis of variance showed statistically significant improvement (p less than 0.00005). Although the improvement appeared to be somewhat more pronounced for men than for women, both sexes experienced an important reduction in leg circumference after therapy. Other factors such as age, etiology, previous use of the Jobst pump, or duration of lymphedema did not appear to influence the extent of improvement. These data clearly indicate that the Wright Linear Pump is an effective treatment for lower extremity lymphedema in adults.

Use of protein blotting to study the DNA-binding properties of histone H1 and H1 variants.

Sub-types of histone H1 have been observed in a variety of tissues from several organisms. One of the best characterized H1 variants is H5 from avian erythrocytes. Several lines of evidence suggest that H5 has a greater affinity for DNA than H1 and is thus thought to account, in part, for the highly condensed and transcriptionally repressed state of avian erythrocyte chromatin. In trout there is an analogous erythrocyte-specific H1 variant, previously termed 'H5' [B.L.A. Miki and J.M. Neelin (1975) Can. J. Biochem. 53, 1158-1169). Using a sensitive and rapid protein-blotting procedure which is specific amongst the histones for histone H1 and its variants, we compared DNA-binding properties of the trout erythrocyte histone 'H5' and chicken H5. By increasing the NaCl concentration of the binding buffer, a gradual decrease in the amount of DNA that bound to chicken H1, trout H1 and trout erythrocyte 'H5' variant was observed, such that at concentrations above 0.37 M, negligible amounts of DNA were bound. By contrast, chicken H5 bound a significantly greater amount of DNA even at a concentration of 0.4 M NaCl. Based on the DNA-binding, properties, we conclude that the trout erythrocyte variant 'H5' is more closely related to H1 than to H5. By assaying the DNA-binding affinity of calf thymus H1 DNA-binding affinity of calf thymus H1 peptide fragments, generated by protease and chemical cleavage, and the sperm-specific H1 variants of the annelid, Platynereis dumerilii, which possess greatly shortened C-terminal tails, we conclude that a domain that includes a very small portion of the C-terminal tail and part of the globular domain is sufficient for the binding of H1 to DNA.

A controlled trial of isoniazid therapy for action tremor in multiple sclerosis.

Ten patients with clinically definite multiple sclerosis (MS) and action tremor were treated with isoniazid (INH) in a double-blind single crossover trial. The daily dose of INH administered during the 4-week treatment phase of the trial was determined by acetylator phenotype with slow acetylators receiving 12 mg/kg per day and rapid acetylators 20 mg/kg per day. Six of eight patients who completed the trial showed clinical improvement in the postural (alternating) tremor while on INH but the degree was minimal in all cases. Results of tremograms indicated that improvement also occurred in the intentional (synchronous) component of three patients while on INH, but this did not achieve statistical significance. Cerebrospinal fluid (CSF) levels of gamma-aminobutyric acid (GABA) homocarnosine and ornithine were markedly elevated with INH therapy (providing evidence for substantial inhibition of GABA aminotransferase activity and increase in GABA in the CNS), but no correlation was found between the degree of GABA elevation in the CSF and the clinical response. Side effects were minimal and well tolerated. Although INH appears to have a limited therapeutic role, a trial is warranted in MS patients with postural tremor.

Alpha-tocopherol and beta-carotene do not protect marmosets against the dopaminergic neurotoxicity of N-methyl-4-phenyl-1,2,3,6-tetrahydropyridine.

Idiopathic Parkinson's disease (PD) may possibly be caused by one or more unidentified neurotoxins present in the environment, or formed endogenously, which progressively damage dopaminergic nigrostriatal neurons. N-Methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) is an experimental neurotoxin which produces biochemical and neuropathological changes in humans, lower primates and mice that closely resemble those found in PD. Because the mechanisms of neuronal damage in both idiopathic PD and in the MPTP model of PD may involve free radical formation in the substantia nigra, antioxidants might protect dopaminergic neurons. Previously, we found that both alpha-tocopherol and beta-carotene partially protected mice against MPTP. However, in the experiments described in this paper, neither alpha-tocopherol nor beta-carotene, each administered in massive doses, had any demonstrable protective effect for dopaminergic nigrostriatal neurons in marmosets injected with low doses of MPTP. Without more knowledge about the identity of the neurotoxin(s) causing idiopathic PD, and their mechanism of action, it is not possible at this time to predict whether these 2 antioxidants might be clinically useful in preventing or ameliorating PD.

Human monoclonal antibodies to thyroid antigens derived by hybridization of lymphocytes from a diabetic patient.

Human monoclonal antibodies to human endocrine cells have been obtained following the generation of immunoglobulin-secreting interspecies lymphocyte hybridomas. Peripheral blood lymphocytes from an adult patient presenting with acute onset, Type I, diabetes mellitus were fused in vitro with mouse myeloma cells of the NS1 cell line. Initial selection of resulting hybridomas was made by their ability to proliferate in HAT medium. Those hybridomas secreting human immunoglobulins were identified by radioimmunoassay and, thereafter, cloned at frequent intervals to ensure continued antibody production. Human monoclonal antibodies selected in this manner are being employed to identify those epitopes which are common antigenic targets during initial stages of autoimmune-mediated diabetes mellitus and associated multiple endocrinopathies. Of these antibodies, one (HML 3.22) recognizes an epitope present on the human TSH receptor and a second (HML 3.21) identifies a component of thyroglobulin. The potential value of human monoclonal antibodies as probes for analyzing autoimmune-mediated endocrine diseases is discussed.

Thermoregulatory responses of guinea pigs with anteroventral third ventricle lesions.

Guinea pigs with anteroventral third ventricle region (AV3V) lesions fail to develop fever and the associated rise in acute-phase plasma protein levels following systemic injections of lipopolysaccharide (LPS). Since endogenous pyrogen (EP) injected directly into the preoptic area of animals with AV3V lesions causes appropriate elevations in core temperature (Tco) and acute-phase plasma proteins levels, the blocked responses to LPS probably are not due to damage to the adjacent preoptic area. We proposed, therefore, that EP may pass from blood into brain in the AV3V, presumably through the organum vasculosum laminae terminalis. However, the possibility that a more generalized impairment due to damaged pathways within the AV3V could account for the observed effects was not examined. To investigate this possibility, guinea pigs were given AV3V lesions. Pending histological verification of the ablated sites, AV3V lesions were presumed to be placed correctly if the animals did not develop fever following LPS (Salmonella enteritidis, 2 micrograms/kg i.p., at ambient temperature (Ta) 22 degrees C); those failing to meet this criterion were designated as sham-operated. Two experiments were conducted. In the first, metabolic rates, Tco, and two skin temperatures (Tsk) were measured at Ta 12 degrees, 22 degrees, and 32 degrees C over an 8-month postlesion period during which failure to fever persisted; the data were collected during a 30-min period after thermal balance had been achieved at any given Ta. There were no differences in the variables measured between sham-operated and AV3V-lesioned animals at Ta 22 degrees C.(ABSTRACT TRUNCATED AT 250 WORDS)

Influence of avian aflatoxicosis on the synthesis of polyamines.

Effects were examined of inanition, dietary aflatoxin (2.5 mg/kg), and dietary supplements of threonine, lysine, and arginine on the activities of renal arginase and hepatic ornithine decarboxylase and on the accumulation of polyamines in liver and brain of 24 or 26-day-old broiler cockerels. Aflatoxicosis and inanition lowered the activity of renal arginase by 58 and 37%, respectively. Supplemental dietary threonine (.4%) did not suppress the activity of renal arginase, while fortification of diets of controls with lysine (.53%), but not diets containing aflatoxin, elevated the activity of renal arginase. Supplements of dietary lysine and/or arginine did not influence the hepatic content of putrescine but lowered the concentrations of spermidine and spermine. Aflatoxicosis, but not inanition, increased the activity of hepatic ornithine decarboxylase (ODC; 22-fold), increased hepatic concentrations of putrescine and spermidine, but decreased spermine concentrations. The elevation of hepatic ODC, putrescine, and the ratio of spermidine to spermine parallels the enlargement of the liver caused by aflatoxicosis. Cadaverine and putrescine were not detected in avian brain, while cerebral concentrations of spermidine and spermine were not altered by aflatoxin, inanition, or by supplements of dietary lysine, arginine, or both lysine and arginine.

Manipulation of glutathione contents fails to alter dopaminergic nigrostriatal neurotoxicity of N-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) in the mouse.

Administration of glutathione monoethyl ester to mice increased hepatic glutathione (GSH) levels modestly, while administration of butylated hydroxyanisole increased hepatic glutathione content markedly. Yet neither substance protected mice from the toxic effects of N-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) on dopaminergic nigrostriatal neurons, as shown by marked depletion of striatal dopamine content when animals were sacrificed. Conversely, marked lowering of GSH levels in the livers of mice by administration of buthionine sulfoximine, or in both liver and brainstem following the injection of diethyl maleate, failed to accentuate the neurotoxicity of a low dose of MPTP. Thus, although MPTP produces a drop in brainstem GSH content, this GSH deficiency may not be casually related to the neurotoxic effects of MPTP.

Structure of pseudobactin A214, a siderophore from a bean-deleterious Pseudomonas.

Bean-deleterious Pseudomonas A214 produced the extracellular yellow-green, fluorescent siderophore [microbial iron(III) transport agent] pseudobactin A214 under iron-limiting conditions. Pseudobactin A214 has a molecular formula of C46H64N13O22 and a molecular mass of 1151 g/mol. Pseudobactin A214 contained an N-blocked linear octapeptide with the amino acid sequence Ser-Ala-Gly-Ser-Ala-threo-beta-OH-Asp-L-allo-Thr-N delta-OH-Orn with a yellow-green, fluorescent quinoline derivative attached via an amide bond to the amino terminus. A succinamide group was linked to carbon 3 of the quinoline derivative. Sequencing was accomplished by two-dimensional NMR spectroscopy and by Edman degradation of smaller peptides obtained from partial acid hydrolysis. Since pseudobactin A214 was not affected by nonspecific proteolytic enzymes, it might contain D-amino acids. The three bidentate iron-(III)-chelating groups consisted of a 1,2-dihydroxy aromatic group in the quinoline chromophore, an alpha-hydroxy acid group present as beta-hydroxyaspartic acid, and a hydroxamate group derived from N delta-acetyl-N delta-hydroxyornithine. The chemical structure of pseudobactin A214 is remarkably similar to those of pseudobactin and pseudobactin 7SR1, the siderophores of plant growth promoting and plant-deleterious Pseudomonas B10 and Pseudomonas 7SR1, respectively.
