An inhibition ELISA method was used to test culture filtrate antigens of Aspergillus fumigatus for the avidity of antibodies to them. Four different culture media were used to check the ability of this assay to distinguish between similar antigenic preparations. Differences between antigens exhibiting similar crossed immunoelectrophoresis patterns were observed by inhibition ELISA.

Three children had pustular patches develop over the occipital areas of the scalp. Colonies of Trichophyton mentagrophytes grew on cultures with Sabouraud's dextrose agar with antibiotics. After a week of topical treatment with 1:10,000 potassium permanganate solution and oral treatment with griseofulvin, typical nodules of erythema nodosum appeared over the shins, ranging in diameter from 1 to 4 cm. Nodules on the legs resolved spontaneously within 12 days in the three children. Erythema nodosum occurring in patients with kerion may be interpreted as individual allergic hypersensitivity reactions as a result of the immune host response to allergenic products from the primary site of infection. This possibly may cause immune complexes that precipitate erythema nodosum.
A case of cutaneous and renal geotrichosis in a giant tortoise, Geochelone elephantopus, at the Zoological Park of Barcelona is reported. Fungal hyphae and spores were seen in skin and kidney. Culture of these tissues yielded Geotrichum candidum. This fungus was isolated from the faeces of 5 other giant tortoises that were housed with the dead animal and from specimens of corn hydroponic culture which is part of their diet. Arthrospore suspensions of the 2 strains isolated from the dead animal's skin and kidney were experimentally inoculated into mice and turtles (Testudo horsfiedi) in order to determine the pathogenicity of G. candidum for animal tissues. Our results confirm its low pathogenicity.
The susceptibility of 21 strains of Aspergillus (11 of A. fumigatus, 8 of A, niger, and 2 of A. flavus) isolated from human pathologic specimens to Amphotericin B and Miconazole has been comparatively studied. Determination of the minimal inhibitory concentration of both drugs in a liquid medium showed a noticeably variability for the different strains. The values obtained for Amphotericin B varied between 0.25 microgram/ml (2 strains) and 1.25 microgram/ml (5 strains) after 48 hours, and between 1,25 microgram/ml (1 strain) and 50 microgram/ml (1 strain) after 10 days. For Miconazole the results varied between 0.1 microgram/ml (1 strain) and 25 microgram/ml (1 strain) after 48 hours of incubation, and between 0.5 microgram/ml (5 strains) and greater than 100 microgram/ml after 10 days. The variability of these results indicates the usefulness of carrying our in vitro sensitivity studies whenever it is possible.

Limited numbers of antigens were detected in the structural components of Echinococcus granulosus cysts by double diffusion and immunoelectrophoresis tests using homologous and heterologous antisera without antibody activity to host serum contaminants. Although antigens common to the germinal membranes, laminated layers and protoscoleces were demonstrated, no evidence was found of antigen sharing between the laminated layer and hydatid fluid from fertile and sterile cysts. These fluids, however, had antigenic determinants in common with protoscoleces. Antigens responsible for the formation of a characteristic cathodic are were detected but only in the three structural cyst components. In contrast, E. granulosus arc 5 antigens were found solely in hydatid fluid. These observations suggest that these two antigen groups are of somatic and metabolic origin, respectively. The results are discussed in terms of current knowledge of the immunobiology of hydatid cysts.
Hydatid cyst fluid from Echinococcus granulosus (HCF) and cyst fluid from Taenia hydatigena (TCF) cysts were compared in reciprocal immunoelectrophoresis (IEP) tests using homologous and heterologous antisera which were free of antibodies to host serum contaminants. The antigens for the E granulosus arc 5 were demonstrated in TCF. Antibody activity to these and other antigens common to HCF and TCF was removed from homologous antisera by absorptions with the heterologous antigenic preparation. Antigens not shared by the two metacestodes fluids were then demonstrated by IEP tests. These findings are discussed in terms of their significance to phylogenetic and immunodiagnostic studies of these parasites in their immediate hosts.
The extraordinarily high rate of amyotrophic lateral sclerosis (ALS) and parkinsonism-dementia (PD) in the Chamorro linguistic group on Guam suggested a genetic etiology for these diseases. Results of three studies are described. Two involved comparisons of the rates of disease in relatives and spouses of index patients and controls. In the third we examined offspring of doubly affected parents, 10 spouse pairs with ALS, 3 spouse pairs with PD and 13 spouse pairs in which 1 spouse had ALS and the other PD. Within the limitations of numbers and age of offspring, we found a tendency among siblings and offspring of ALS cases to have a higher risk than control relatives. Numbers were small and in no instance were significant differences encountered. Spouses of patients had at least as high risk of developing ALS or PD as any of the groups of blood relatives. A slightly higher age-specific rate of ALS and PD occurred in the offspring of doubly affected parents. The only group large enough for present analysis are 60 offspring of doubly affected ALS parents over the age of 20. Thus far only 3 have developed ALS. Within our entire patient population of 350 ALS cases and 219 PD cases there were 12 instances in which one parent of the patient was not Chamorro. In addition there is a consistent two- to threefold excess of affected males which could not be accounted for by competitive risks of death from other causes among females. While a familial tendency does exist these findings and observations are not compatible with any simple Mendelian form of inheritance. It is possible that genetic influences affect the occurrence of these diseases but our studies suggest that environmental factors play at least as important a role.
