High-performance liquid chromatography of bile pigments: application to the determination of unconjugated bilirubin in serum.

A rapid, sensitive and specific method for the determination of unconjugated bilirubin in serum is described. Reversed-phase chromatography on short-chain alkylsilica (C2) or octadecylsilica (C18) with acetonitrile--dimethylsulphoxide--water as eluent is used for the separation of bilirubin. The method is applied to the analysis of bilirubin in patients with disorders associated with jaundice.

Determination of 5-aminolaevulinic acid and porphobilinogen by high-performance liquid chromatography.

A strong anion-exchange column coupled to a strong cation-exchange column with acetate buffer as eluent or reversed-phase ion-pair chromatography on octadecylsilica with methanol-water containing 1-heptanesulphonic acid as the mobile phase is described for the simultaneous separation of 5-aminolaevulinic acid and porphobilinogen. The separation is applied to the development of a fast and simple method for determining the activity of the enzyme aminolaevulinic acid dehydrase in human erythrocytes. 5-Aminolaevulinic acid is used as the enzyme substrate and the enzyme activity is expressed as micromoles of porphobilinogen formed per ml of erythrocytes in 1 h at 38 degrees. 5-Aminolaevulinic acid and porphobilinogen can also be separated from the urine of prophyric patients but the UV detector has insufficient sensitivity for the determination of 5-aminolaevulinic acid.

Radioaffinity assay, a new approach to binding assays applied to the measurement of serum albumin.

The affinity adsorbent Cibacron Blue F3GA-Sepharose 4-B has been used to develop a binding assay for human serum albumin. The method is simple, accurate and precise. It agrees well with the 'rocket' immunoelectrophoretic method and is proposed as an alternative technique for the re-estimation of albumin levels in the low range (less than 30 g/l) where the bromocresol green dye-binding method overestimates. Bilirubin, several drugs, gammaglobulin, haemoglobin and heparin do not interfere with the estimations.

A sensitive and specific high performance liquid chromatographic assay for imidazole dipeptides and 3-methylhistidine in human muscle biopsies, serum and urine.

A sensitive and specific high performance liquid chromatographic method is described for measuring imidazole dipeptides and 3-methylhistidine in human muscle biopsies, serum and urine. Muscle extract, serum or urine was reacted with o-phthaldialdehyde and the derivatives were separated by reversed phase chromatography with column switching and fluorescence detection.
