Alpha-hydroxy acid oxidase: localization in renal microbodies.

Differential and density equilibrium centrifugation have established the presence of alpha-hydroxy acid oxidase in microbodies of the kidney of the rat. The enzyme has been demonstrated in cells of the distal convoluted tubule by a microscopic cytochemical method. This enzyme, like certain others in microbodies, produces hydrogen peroxide.

Calcitonin gene-related peptide (CGRP) in the female rat urogenital tract.

CGRP-immunoreactivity was found throughout the female rat urogenital tract by specific radioimmunoassay, and shown to be present in nerve fibres by immunocytochemistry. The highest concentrations of CGRP-like immunoreactivity were found in the urinary tract, with lower levels in regions of the genitalia. Chromatographic analysis of bladder and vaginal extracts on Sephadex G-50 columns and HPLC revealed at least three CGRP-immunoreactive peaks. The major peak emerged in the same position as synthetic rat CGRP. CGRP nerve fibres were associated mainly with blood vessels, non-vascular smooth muscle, squamous epithelium and uterine and cervical glands, and were particularly abundant in the ureter and bladder. CGRP-immunoreactivity was depleted by neonatal treatment with capsaicin and after surgical section of pelvic and/or hypogastric nerves. Immunocytochemistry demonstrated that depletion occurred predominantly in the mucosal layer of the urogenital tract. These findings indicate a sensory function for most of the CGRP-immunoreactive nerves in the rat urogenital tract.

Effect of 6-hydroxydopamine on neuropeptides in the rat female genitourinary tract.

The occurrence and distribution of neuropeptide Y has been determined in the rat female genitourinary tract by radioimmunoassay and chromatographic analysis. Within the bladder, higher concentrations of neuropeptide Y were found in the trigone (48.8 +/- 5.2 pmol/g) than in the dome (36.0 +/- 2.1 pmol/g). In the genital tract, highest concentrations were identified in the vagina (41.4 +/- 2.1 pmol/g). Treatment of rats with 6-hydroxydopamine resulted in significant depletion of neuropeptide Y concentrations in both parts of the bladder, together with vagina, uterine horn and fallopian tube. No change was observed in the cervix, uterine body and ovary. Concentrations of vasoactive intestinal polypeptide were unaffected by treatment with 6-hydroxydopamine except in the area of the cervix where concentrations rose from 64.1 +/- 5.7 pmol/g to 133.6 +/- 15.1 pmol/g (p less than 0.05). There was a generalised, but statistically insignificant rise in substance P concentrations.

Substance P and gastrin releasing peptide in bovine mesenteric lymphatic vessels: chemical characterization and action.

Alcoholic extracts of bovine mesenteric lymphatic vessels were assayed for the presence of SP, GRP, VIP, PHI, GIP and NT using specific radioimmunoassays. SP and GRP immunoreactivities were detected at concentrations of 190 +/- 20 and 1,000 +/- 130 pg.g-1, respectively. No significant levels of immunoreactivity were detected for any of the other peptides. SP and GRP immunoreactivities coeluted with their synthetic counterparts from both Sephadex G-50 and reversed phase HPLC columns. Synthetic SP (10(-9)-10(-7) M) and the naturally occurring analogue of GRP, bombesin (10(-9)-10(-7) M), increased spontaneous contraction rate in isolated vessel segments. This excitatory effect was not blocked by the alpha-adrenoceptor antagonist phentolamine (3 x 10(-6) M).

Effects of some neurokinin A analogues on tachykinin-induced contraction of guinea pig trachea.

A series of analogues of neurokinin A (NKA) has been synthesized and characterized by testing for their abilities, in vitro, to contract guinea pig tracheal smooth muscle or to antagonize NKA-, NKB- and SP-induced contraction of this tissue. Substitution of NKA residues Gly8 or Leu9 by conformationally restricting amino acids produced peptides that were antagonists of NKA action, but the type and specificity of the antagonism depended on the size of the peptide. Thus, while [Ala5, Aib8, Leu10]NKA(2-10) showed no agonism and was a specific, competitive antagonist of NKA, [Ala5, Aib8, Leu10]NKA(4-10) was a noncompetitive antagonist of NKA and substance P (SP) and was itself a weak agonist at concentrations above 10(-7) M.

Photochemical formation of singlet molecular oxygen in illuminated aqueous solutions of several commercially available sunscreen active ingredients.

Evidence is presented for the photochemical formation of singlet molecular oxygen (1O2) in air-saturated aqueous solutions of several sunscreen active ingredients using sunlight-range illumination. This is of significance because (1) 1O2 is known to be cytotoxic, and (2) there have been several reports of toxic effects associated with the use of some sunscreens; most notably, with p-aminobenzoic acid (PABA). Illuminated aqueous solutions of PABA, 2-ethylhexyl p-(dimethylamino)benzate (ODPABA), 2-hydroxy-4-methoxybenzophenone (BZ3), 2,2'-dihydroxy-4-methoxybenzophenone (BZ8), 2-ethylhexyl 2-cyano-3,3-diphenylacrylate (OCR), 2-ethylhexyl p-methoxycinnamate (OMC), and 2-ethylhexyl salicylate (OCS) were evaluated individually for 1O2 formation. Furfuryl alcohol (FFA), a well-known chemical trap for 1O2, was added to each of the aqueous sunscreen solutions. The FFA was consumed when solutions of PABA, ODPABA, OMC, and OCR were illuminated, but no loss of FFA other than by direct photolysis occurred in solutions of BZ3, BZ8, or OCS. There was also no significant loss of FFA in any of these solutions kept in the dark. Further evidence for the formation of 1O2 in illuminated aqueous sunscreen solutions is provided by the results of experiments in which individual solutions containing sunscreen active ingredients and FFA that were diluted with D2O exhibited an increased rate of FFA consumption while the addition of azide ion (N3-) reduced the rate of FFA consumption. Continuous sunlight-range illumination of aqueous PABA solutions produced significantly higher steady-state concentrations of 1O2 than in solutions containing any of the other sunscreen active ingredients evaluated. The substituted benzophenone compounds (BZ3 and BZ8) and the salicylate-based compound (OCS) not only appear to produce no 1O2, but they also appear to produce no other reactive oxidant species that are capable of consuming FFA. This indicates that BZ3, BZ8, and OCS may be peferable, from the standpoint of toxic oxidant formation, for use as sunscreen active ingredients when compared to the other compounds evaluated in this study.

Neuropeptide Y distribution in human brain.

Tatemoto and Mutt recently used the presence of a C-terminal NH2 group to identify and isolate a new peptide, neuropeptide Y (NPY), from porcine brain. This 36 amino acid peptide was subsequently shown to be active on isolated vas deferens, vascular smooth muscle and pancreatic acinar cells in very low molar concentrations. In view of these potent effects we have now investigated its distribution in the human brain by radioimmunoassay and immunocytochemistry. High concentrations of NPY have been found, exceeding those of cholecystokinin and somatostatin, hitherto considered to be the most abundant neuropeptides. The distribution of NPY was different from that of any other peptide system described, being particularly concentrated in the basal ganglia, amygdala and nucleus accumbens. Immunocytochemistry demonstrated a large number of NPY neuronal cell bodies especially in the caudate and putamen. Immunoreactive neuronal cell bodies were also clearly localized in cortical areas, particularly layers V and VI. NPY, a newly discovered peptide with potent biological activity, thus seems to be among the most abundant of human neuropeptides. The massive numbers of NPY neurones in the basal ganglia suggest NPY to be of fundamental importance in the control of human motor function.

Stress concentrations at the apex of pinned, implanted teeth.

Stress in the alveolus at the apex is reduced when the implant is held by a pin through the alveolar wall. This stress reduction is effected by transfer of part of the coronal force to the alveolar wall by the pin. Pin positions centrally located between the apex and the alveolar crest provide the greatest reduction of stress concentration in the alveolus at the apex. Since the bone is especially fragile near the alveolar crest, care must be taken when the pin is positioned in this region. Positioning the pin near the apex has the least effect on reducing the stress in the alveolus at the apex. Although no data were obtained relating to stress in the alveolar wall at points of pin penetration, results given here do indicate that from 7 to 30% of the intrusion component of the coronal force is transmitted to the alveolar wall by the pin. Additional research is needed to determine the effects of stress at the pin through the alveolar wall because of load transfer by the pin from the implant to the alveolar wall. The situation in the alveolar wall would be one of creation of a completely unnatural stress rather than reduction of an existing stress, as was the case at the apex. Whether the reduction of stress at the apex and the creation of stress at the pin through the alveolar wall are of physiological benefit is a question that must yet be answered.

The interaction between human Fc gamma RI and the gamma-chain is mediated solely via the 21 amino acid transmembrane domain of Fc gamma RI.

We have established a biological assay to investigate the nature of the non-covalent interaction between two integral type I membrane proteins, Fc gamma RI and gamma-chain. Fc gamma RI, the human high affinity receptor for immunoglobulin G (IgG), is expressed on the surface of macrophages and monocytes and mediates a broad range of important immunological functions. Fc gamma RI relies on a functional interaction with a second integral type I membrane protein, gamma-chain, to mediate many of these functions. For example, Fc gamma RI can only mediate phagocytosis of IgG-coated particles in COS cells when co-expressed with gamma-chain. We have previously shown that the cytoplasmic domain of Fc gamma RI is not necessary for this functional interaction. In this study we have used the phagocytosis assay to investigate the role of the transmembrane region of Fc gamma RI in mediating this functional interaction with gamma-chain by using mutant and chimeric forms of the receptor. Three mutants, which introduce or remove charged residues from a conserved 10 amino acid stretch of amino acids in the proximal transmembrane region of Fc gamma RI, were able to mediate phagocytosis of IgG-coated particles. In contrast, two chimeric receptors, In which 21 of the amino acids in the distal transmembrane region of Fc gamma RI were replaced with the transmembrane region of the related receptors CD2 or LFA3, were expressed but failed to interact functionally with gamma-chain to mediate phagocytosis. Thus, these mutants demonstrate that the interaction between human Fc gamma RI and gamma-chain is mediated solely via these 21 amino acids in the transmembrane domain of Fc gamma RI.

Work sampling: a means of determining physiotherapy activities and reviewing caseloads.

Work sampling is a tool used to examine the activities involved in performing a job. A work sampling study was designed to examine the components of physiotherapy care in a 350-bed teaching hospital. The components were related to a physiotherapist's activities and included an examination of the time related to patient treatment, attendance at meetings, record-keeping and preparation, travel, teaching, continuing education (in-service), talking to colleagues (doctors, occupational therapists), and other activities. The purpose of the study was to quantify a therapist's nontreatment activities, and to compare observations concerning treatment with the work sampling data obtained in compliance with the Canadian Schedule of Unit Values and, in particular, its weighted units. The study showed that there was no difference between the "observed" time (sampling time in percentage of total time) spent in direct patient contact, and the weighted units, when the latter was converted to a percentage of time. The Nova Scotia government's standard of 50 000 work units per year was found to be useful in justifying staffing levels; a useful indicator of productivity was also developed using weighted units expressed as a percentage of worked or paid hours. Work sampling would thus appear to be a useful tool for physiotherapy department managers.
